The growth and development of breast cancer are accompanied by an increase in oxidative stress. A close relationship is known to exist between the biological activity of several antioxidant enzymes and the regulation of estrogen-mediated signaling in breast cancer. The aim of this study was to study the activity of salivary antioxidant enzymes and the level of lipid peroxidation products in breast cancer before and after surgical treatment. The study included 115 patients with breast cancer (58.7 ± 10.9 years) and 60 healthy volunteers (51.8 ± 12.1 years). Saliva samples were obtained again fr

Severe infections and a complicated healing process caused by multidrug-resistant bacteria in skin wounds have urgently driven the development of novel antibacterial agents. In this study, bimetallic copper-cobalt nanoclusters (CuCo NCs) were successfully synthesized via a one-pot aqueous co-reduction method and evaluated as a potential antimicrobial platform for wound infection treatment. The peroxidase-like (POD-like) activity of CuCo NCs catalyzes H2O2 to generate hydroxyl radicals (•OH), inducing membrane disruption of bacteria. Consequently, the CuCo NCs exhibited potent and broad-spectru

Given the prevalent use of horseradish peroxidase (HRP) as an enzyme label in immunoassays, the practical importance of point-of-care testing for the portable identification of HRP-related biomarkers is evident. Accordingly, this study devised a portable immunoassay platform using a personal glucometer (PGM) to achieve rapid detection of HRP-related biomarkers. 3,3',5,5'-tetramethylbenzidine (TMB) and dopamine (DA) were used as signal-regulating substrates, and the assay principle was based on the HRP-mediated redox reaction. In the absence of HRP, TMB and DA directly reacted with [Fe(CN)6]3-

BACKGROUND: Bipolar disorder (BD) is a heterogeneous psychiatric disorder that is increasingly recognized as having immune-inflammatory pathogenesis. Previous studies have shown thyroid autoimmunity, in particular, thyroid peroxidase antibody (TPO-Ab), is associated with rapid cycling and antidepressant-induced mania in BD. We examined the relationship between TPO-Ab seropositivity and BD clinical sub-phenotypes and lithium response. METHODS: In this cross-sectional study, adult patients with BD enrolled in the Mayo Clinic BD Biobank were stratified into TPO-Ab positive versus negative groups

It is known that a percentage of patients who undergo total thyroidectomy, approximately 16-34%, complain of symptoms of hypothyroidism or altered quality of life (QoL) despite achieving normal serum TSH values. The present study aimed to identify whether the level of selenium could be responsible for this phenomenon. This pilot cohort study included 44 thyroidectomized outpatients. All patients underwent surgery for benign disease. In this study, no patients with a history of autoimmunity, malignancy, or other conditions that could affect well-being, absorption, or selenium intake were includ

In this study, a facile method for the sensitive and selective detection of sulfisoxazole (SFX) was developed using bovine serum albumin (BSA)-stabilized gold nanoclusters (BSA-AuNCs). These nanoclusters, which possess high intrinsic peroxidase-like activity, can catalyze the hydrogen peroxide (H2O2)-based oxidation of o-phenylenediamine (OPD) and rapidly generate bright yellow product: 2,3-diaminophenazine (DAP). We found that the antibiotic SFX strongly inhibits the catalytic activity of BSA-AuNCs, while other common sulfonamide compounds do not interfere. As the concentration of SFX increas

The purpose of this study was to elucidate the role of dihydrochelerythrine (DHC) in prostate cancer and to explore its targets in DU145 and PC3 cells. DHC significantly inhibited the growth of both cell lines. RNA-sequencing analysis revealed that ferroptosis-related genes, including glutathione peroxidase 4 (GPX4) and light chain 3, were significantly downregulated in DHC-treated DU145 cells. Molecular docking studies demonstrated significant interactions between DHC and the GPX4 protein, with a binding energy of -9.01 kcal/mol. Furthermore, GPX4 mRNA expression was significantly upregulated

A new gene coding for an iron-containing enzyme was identified in the genome of Acinetobacter radioresistens. Bioinformatics analysis allowed the assignment of the protein to DyP peroxidases, due to the presence of conserved residues involved in heme binding and catalysis. Moreover, Ar-DyP is located in an operon coding also for other enzymes involved in iron uptake and regulation. The crystal structure of Ar-DyP determined at 1.85 Å resolution shows that the heme pocket Ar-DyP is "wet" forming a continuous hydrogen-bond network that enables the communication between heme and distal residues. …

Tetracycline antibiotics (TCs) are widely used in medicine, agriculture, and animal husbandry. However, their overuse has led to environmental pollution, posing a significant threat to water sources, soil, and food safety. Therefore, there is an urgent need for efficient, sensitive, simple, and low-cost detection methods for environmental pollution monitoring. In this study, the catalytic activity of copper-based nanozymes was regulated by AAs. Lysine, aspartic acid, glycine, and arginine were chosen as ligands to synthesize different copper-based nanozymes. The results showed that the type of

Designing three-dimensional (3D) catalytic sites in single-atom catalysts (SACs) that mimic thyroid peroxidase (TPO) function for achieving iodotyrosine coupling, although highly desirable for the synthesis of thyroid hormones, poses a great challenge. Herein, we design and synthesize a class of SACs with 3D catalytic centers composed of Cu-N5 as catalytic sites and BO2 as binding sites (BO2/CuN5C) for mimicking TPO in activating H2O2 to facilitate tyrosine iodination and conjugation for producing thyroid hormones. We demonstrate that the as-prepared BO2/CuN5C not only provides binding sites f

Trimethylamine N-oxide (TMAO), a gut microbiota-derived metabolite, has been linked to cardiovascular, renal, and hepatic disorders, but its direct impact on mitochondrial apoptotic machinery remains unclear. Here, we show that TMAO binds cytochrome c (Cyt c), disrupting its structural integrity and converting it into an apoptotically competent species. Spectroscopic analyses revealed that TMAO destabilizes the heme-Met80 axial ligation, shifting Cyt c from its native hexacoordinate to a pentacoordinate state. This conformational change enhances peroxidase activity, exposes hydrophobic cluster

INTRODUCTION: Anti-thyroid peroxidase antibodies (TPO-Ab) are detectable in almost all patients with autoimmune thyroid disease or Hashimoto's thyroiditis (HT) but may also be present in healthy individuals. HT affects women to a greater extent than men and can lead to overt hypothyroidism, which may increase the risk of miscarriage. There are no local data available on the prevalence of TPO-Ab among healthy women in the United Arab Emirates. The objective of this study was to determine the prevalence of TPO-Ab and assess thyroid function in healthy medical students.
METHODS: This cross-secti

PURPOSE: Thyroid peroxidase (TPO) is essential for the synthesis of thyroid hormones. However, specific mutations render TPO antigenic and prone to autoimmune attacks leading to thyroid cancer, TPO deficiency, and congenital hypothyroidism (CH). Despite technological advancement, most experimental procedures cannot quickly identify the genetic causes of CH nor detect thyroid cancer in the early stages.
METHODS: We performed saturated computational mutagenesis to calculate the folding energy changes (∆∆G) caused by missense mutations and analyzed the mutations involved in post-translational mo

The human lens is composed of a monolayer of lens epithelial cells (LECs) and elongated fibers that align tightly but are separated by the plasma membrane. The integrity of the lens plasma membrane is crucial for maintaining lens cellular structure, homeostasis, and transparency. Glutathione peroxidase 4 (GPX4), a selenoenzyme, plays a critical role in protecting against lipid peroxidation. This study aims to elucidate the role of GPX4 in lens plasma membrane stability during lens development using in vitro, ex vivo, and in vivo systems. Our findings reveal that GPX4 deficiency triggers lens e

Ferroptosis is an iron-dependent form of oxidative cell death induced by lipid peroxidation accumulation. Glutathione peroxidase 4 (GPX4) plays a key role in the regulation of ferroptosis and is considered to be a promising therapeutic target for cancer and other human diseases. Herein, we describe our design, synthesis, and biological evaluation of a series of HyT-based degraders of the GPX4. One of the most promising compounds, 7b (ZX782), effectively induces dose- and time-dependent degradation of GPX4 protein and potently suppresses the growth of human fibrosarcoma HT1080 cells, which are

The development of high-performance biosensors is a key focus in the nanozyme field, but the current limitations in biocompatibility and recyclability hinder their broader applications. Herein, we address these challenges by constructing core-shell nanohybrids with biocompatible poly(ethylene glycol) (PEG) modification using a galvanic replacement reaction between orthovanadate ions and liquid metal (LM) (VOx@EGaIn-PEG). By leveraging the excellent charge transfer properties and the low band gap of the LM surface oxide, the VOx@EGaIn-PEG heterojunction can effectively convert hydrogen peroxide

Iron phosphide/carbon (FeP/C) serving as electrocatalysts exhibit excellent activity in oxygen reduction reaction (ORR) process. H2O2 catalyzed by peroxidase (POD) is similar to the formation of new electron transfer channels and the optimization of adsorption of oxygen-containing intermediates or desorption of products in ORR process. However, it is still a challenge to discover FeP/C with enhanced POD-like catalytic activity in the electrocatalytic database for biocatalysis. The discovery of FeP/carbon dots (FeP/CDs) nanozymes driven by electrocatalytic activity for enhanced POD-like ability
