Frailty phenotype in adults with sickle cell disease.

BACKGROUND: People with sickle cell disease (SCD) are at risk for accelerated biological aging and functional decline due to both age and SCD-related stressors. Frailty is characterized by decreased physiologic reserve and increased vulnerability to stressors that can lead to disability and death. The purpose of this study was to evaluate frailty phenotype in adults with SCD and identify factors associated with frailty. METHODS: Frailty was defined by meeting ≥3 of the following criteria: slowness, weakness, weight loss, low physical activity, and exhaustion. Participants meeting 1-2 criteria…

Inflammation modifies breathing phenotype in mice with epilepsy.

Impaired CO₂ responsiveness in epilepsy can result in hypoventilation and hypercapnia and these respiratory disturbances are key contributors to Sudden Unexpected Death in Epilepsy (SUDEP). While mild to moderate inflammation is known to modulate respiratory function, its specific role in regulating respiratory responses in the context of epilepsy remains unclear. We studied the effects of lipopolysaccharide (LPS)-induced inflammation and microglial inhibition via minocycline during the acute and chronic phases of epilepsy on hypercapnic ventilatory responses (HCVR) in the intrahippocampal kai

Genotype-phenotype interaction in Alzheimer's disease immune activation.

INTRODUCTION: Alzheimer's disease (AD) is a neurodegenerative disorder influenced by genetic factors, particularly related to immune activation. This study examines genotype-phenotype interactions affecting immune responses in AD as a reaction to neurodegeneration. METHODS: We computed AD polygenic risk scores (PRSs) informed by shared AD-autoimmunity genetics (AD PRSINFL), AD-independent immune activation score from 10 autoimmune diseases (sum PRSIMMUNE), and standard AD PRSs for 294 individuals. Cerebrospinal fluid (CSF) immune markers (sTREM2, clusterin, fractalkine, and chitinase 3 like p…

Insights Into Aggregation-Delayed Phenotype of Dictyostelium discoideum.

Dictyostelium discoideum is a cellular model that has been widely used in cell signal research focusing on different cellular and developmen … e thought to be included in this paradigm, as blocking these signals leads to the aggregation-minus phenotype. Recently, Dictyostelium was used to detect the effect of natural products, some of which caused a noticeable delay of the aggregation stage. However, no clear evidence has been obtained that explains why aggregation is delayed or the science behind …

Phenotype Variability in Interstitial Cystitis Clinical Trial Recruitment.

IMPORTANCE: There is a need to understand phenotype-specific therapies for interstitial cystitis/bladder pain syndrome (IC/BPS) at the clinical trial level to move beyond trial-and-error treatment approaches. OBJECTIVE: The objective of this study was to characterize IC/BPS clinical trials that incorporate patient phenotype into the eligibility criteria in accordance with the American Urological Association (AUA) guidelines. STUDY DESIGN: Registered IC/BPS clinical trials were identified from ClinicalTrials.gov. Trials were included if they enrolled patients with IC/BPS and addressed pain-re…

Rare subgroup of A phenotype-a case series.

Testing for ABO incompatibility between a potential blood donor and recipient is the cornerstone of pretransfusion testing. Occasionally, weak agglutination reactions occur due to low expression of A and B antigens on the surface of red blood cells. This can lead to inconsistencies in blood group typing, as seen in a case where a patient's forward and reverse typing showed discrepancies. Following a comprehensive analytical assessment, the patient was identified as having a distinctive variant of blood group A, belonging to the less common subgroups-A3, Aend, Ax, Am, Ay, and Ael. Herein, we pr

Bone Phenotype in Autosomal Dominant Polycystic Kidney Disease.

Autosomal dominant polycystic kidney disease (ADPKD) is a systemic ciliary disorder, yet the impact on bone health remains insufficiently defined. Given the expression of polycystins in osteocytes, osteoclasts and extracellular matrix, a disease-specific bone phenotype has been hypothesized. This study aimed to comprehensively characterize the bone phenotype in ADPKD across stages of chronic kidney disease (CKD) and Mayo Imaging Classification (MIC). In this cross-sectional study, 81 patients with ADPKD, 15 with CKD of other etiologies, and 21 healthy individuals were included. Bone phenotype …

Bone phenotype of patients with telomere biology disorders.

Telomere biology disorders (TBD) are a group of diseases that interfere with normal maintenance of telomeres, the ends of chromosomes that protect them from DNA damage. The most common manifestations are bone marrow failure, pulmonary and liver fibrosis. Systematic data on bone health in patients with TBD are scarce. Our aim was to elucidate the bone phenotype of these patients. The cohort comprises 36 adult patients (20 women and 16 men), with genetically confirmed TBD. Demographic, clinical and genetic characteristics, dual-energy X-ray absorptiometry (DXA) scans, spine X-rays and treatment …

Structural Phenotype of High-Frequency Recurrent Facial Palsy.

OBJECTIVES: High-frequency recurrence of peripheral facial palsy suggests an underlying structural predisposition. We aimed to identify the anatomical phenotype associated with ≥ 3 recurrences using high-resolution CT (HRCT). METHODS: We retrospectively analyzed 63 patients with high-frequency recurrence (mean 3.5 episodes) and 62 normal controls. Four HRCT parameters were quantified: geniculate ganglion (GG) enlargement, labyrinthine segment (LS) constriction, intracanalicular bony exostosis, and greater superficial petrosal nerve (GSPN) thickening. Inter-parameter correlations were also eva…

The electroclinical phenotype of epilepsy following autoimmune encephalitis.

OBJECTIVE: To describe the temporal and spatial dynamics of electroencephalographic (EEG) recordings in epilepsy following autoimmune encephalitis (AE) compared to temporal lobe epilepsy (TLE) without signs of inflammatory etiology. METHODS: We assessed electroencephalographic properties using continuous long-term (72-h) EEG recordings in combination with neuroimaging markers and cerebrospinal fluid (CSF) parameters of 25 patients with epilepsy following AE and 30 patients with TLE. We specifically investigated the presence of focal slowing (FS) and epileptiform discharges (EDs). Additionally

Antiparasitics discovery: from genotype to phenotype to compounds.

For decades, the discovery of antiparasitics was dominated by whole-organism screening of intact parasite organisms or surrogate parasite models, such as Caenorhabitis elegans, using in vivo animal models or in vitro parasite assays, the latter also known as phenotypic screening. Molecular target-based screening played only a minor role, if at all. While publications using phenotypic screening are abundant in the literature, publications of successful, marketed, antiparasitic drugs discovered using the molecular target-based approach are scarce. This approach, therefore, is often perceived as
