A Roadmap for the Molecular Farming of Viral Glycoprotein Vaccines: Engineering Glycosylation and Glycosylation-Directed Folding.

Immunization with recombinant glycoprotein-based vaccines is a promising approach to induce protective immunity against viruses. However, the complex biosynthetic maturation requirements of these glycoproteins typically necessitate their production in mammalian cells to support their folding and post-translational modification. Despite these clear advantages, the incumbent costs and infrastructure requirements with this approach can be prohibitive in developing countries, and the production scales and timelines may prove limiting when applying these production systems to the control of pandemi

Redox-based protein persulfidation in guard cell ABA signaling.

Hydrogen sulfide (H2S) is a versatile signaling molecule that regulates multiple physiological processes in plants, including growth and development, immunity, and stress response as well. Signaling triggered by H2S is proposed to occur via persulfidation, an oxidative post-translational modification (PTM) of cysteine residues (-SH) to persulfides (-SSH). Notwithstanding the growing body of information for the plant persulfidation proteome, the gap between the molecular mechanism of H2S and physiological functions of protein persulfidation remains large. In this mini-review, we discussed the s

Thirty years of plant farnesylation research: Few substrates, many gaps, and a clear path forward.

Protein farnesylation - the covalent attachment of a 15-carbon farnesyl isoprenoid to a C-terminal CaaX motif by protein farnesyltransferase (PFT, FTase) - is one of the most functionally important post-translational modifications in eukaryotic biology. In mammals, the biomedical significance of farnesylation is well established. The FTase inhibitor (FTI) lonafarnib treats Hutchinson-Gilford progeria syndrome. FTase remains an oncology target, and chemical proteomic strategies have mapped more than 100 farnesylated substrates at single-protein resolution. In plants, genetic evidence establishe

Cell-length heterogeneity: a population-level solution to growth/virulence trade-offs in the plant pathogen Dickeya dadantii.

Necrotrophic plant pathogens acquire nutrients from dead plant cells, which requires the disintegration of the plant cell wall and tissue structures by the pathogen. Infected plants lose tissue integrity and functional immunity as a result, exposing the nutrient rich, decayed tissues to the environment. One challenge for the necrotrophs to successfully cause secondary infection (infection spread from an initially infected plant to the nearby uninfected plants) is to effectively utilize nutrients released from hosts towards building up a large population before other saprophytes come. In this s

Enhanced adoptive transfer of immunity to Listeria monocytogenes after in vitro culture of murine spleen cells with concanavalin A.

In vitro incubation of spleen cells with T-cell mitogens has been shown to augment cytotoxic and cytolytic T-cell function. The plant lectins Concanavalin A and phytohemagglutinin were employed in a similar fashion to investigate their abilities to enhance cell-mediated immunity to the microbial pathogen Listeria monocytogenes. Spleen cells from immune mice were incubated in vitro with Concanavalin A or phytohemagglutinin before passive transfer into normal recipients. Results indicated a 10- to 100-fold enhancement in the ability of these cells stimulated in vitro to transfer antilisterial re

Induction of salicylic acid-mediated defense response in perennial ryegrass against infection by Magnaporthe oryzae.

Incorporation of plant defense activators is an innovative approach to development of an integrated strategy for the management of turfgrass diseases. The effects of salicylic acid (SA), benzothiadiazole (BTH, chemical analog of SA), jasmonic acid (JA), and ethephon (ET, an ethylene-releasing compound) on development of gray leaf spot in perennial ryegrass (Lolium perenne L.) caused by Magnaporthe oryzae were evaluated. Gray leaf spot disease incidence and severity were significantly decreased when plants were treated prior to inoculation with SA, BTH, and partially by ET but not by JA. Accumu

Roles of M cells in infection and mucosal vaccines.

The mucosal immune system plays a crucial part in the control of infection. Exposure of humans and animals to potential pathogens generally occurs through mucosal surfaces, thus, strategies that target the mucosa seem rational and efficient vaccination measures. Vaccination through the mucosal immune system can induce effective systemic immune responses simultaneously with mucosal immunity compared with parenteral vaccination. M cells are capable of transporting luminal antigens to the underlying lymphoid tissues and can be exploited by pathogens as an entry portal to invade the host. Therefor

Transcription factor PbbZIP4 is targeted for proteasome-mediated degradation by the ubiquitin ligase PbATL18 to influence pear's resistance to Colletotrichum fructicola by regulating the expression of PbNPR3.

Pear anthracnose caused by Colletotrichum fructicola is one of the main fungal diseases in all pear-producing areas. The degradation of ubiquitinated proteins by the 26S proteasome is a regulatory mechanism of eukaryotes. E3 ubiquitin ligase is substrate specific and is one of the most diversified and abundant enzymes in the regulation mechanism of plant ubiquitination. Although numerous studies in other plants have shown that the degradation of ubiquitinated proteins by the 26S proteasome is closely related to plant immunity, there are limited studies on them in pear trees. Here, we found tha…

VOZ-dependent priming of salicylic acid-dependent defense against Rhizopus stolonifer by β-aminobutyric acid requires the TCP protein TCP2 in peach fruit.

Vascular plant one-zinc finger (VOZ) transcription factors (TFs) play crucial roles in plant immunity. Nevertheless, how VOZs modulate defense signaling in response to elicitor-induced resistance is not fully understood. Here, the defense elicitor β-aminobutyric acid (BABA) resulted in the visible suppression of Rhizopus rot disease of peach fruit caused by Rhizopus stolonifer. Defense priming by BABA was notably associated with increased levels of salicylic acid (SA) and SA-dependent gene expression. Data-independent acquisition proteomic analysis revealed that two VOZ proteins (PpVOZ1 and Pp…

CaASR1 promotes salicylic acid- but represses jasmonic acid-dependent signaling to enhance the resistance of Capsicum annuum to bacterial wilt by modulating CabZIP63.

CabZIP63 acts positively in the resistance of pepper (Capsicum annuum) to bacterial wilt caused by Ralstonia solanacearum or tolerance to high-temperature/high-humidity stress, but it is unclear how CabZIP63 achieves its functional specificity against R. solanacearum. Here, CaASR1, an abscisic acid-, stress-, and ripening-inducible protein of C. annuum, was functionally characterized in modulating the functional specificity of CabZIP63 during the defense response of pepper to R. solanacearum. In pepper plants inoculated with R. solanacearum, CaASR1 was up-regulated before 24 h post-inoculation

The extent to which methyl salicylate is required for signaling systemic acquired resistance is dependent on exposure to light after infection.

Systemic acquired resistance (SAR) is a state of heightened defense to a broad spectrum of pathogens that is activated throughout a plant following local infection. Development of SAR requires the translocation of one or more mobile signals from the site of infection through the vascular system to distal (systemic) tissues. The first such signal identified was methyl salicylate (MeSA) in tobacco (Nicotiana tabacum). Subsequent studies demonstrated that MeSA also serves as a SAR signal in Arabidopsis (Arabidopsis thaliana) and potato (Solanum tuberosum). By contrast, another study suggested tha

Genetic suppressors of the growth-immunity trade-off in the Arabidopsis salicylic acid-accumulating dmr6 dlo1 mutant.

Plants actively suppress growth and development upon activation of immunity. In turn, when pathogen attack has subsided, immune responses are suppressed again. Phytohormones play an important role in regulating this balance and the growth-immunity trade-off in general. The trade-off is evident in the Arabidopsis dmr6 dlo1 mutant, which accumulates the immune-activating phytohormone salicylic acid (SA) to high levels, resulting in high disease resistance but repression of growth. Little is known about the SA-induced growth trade-off mechanism. In this study, we performed a genetic suppressor sc

Exogenous application of methyl jasmonate induces a defense response and resistance against Sclerotinia sclerotiorum in dry bean plants.

Sclerotinia sclerotiorum (Lib.) de Bary is a necrotrophic fungal pathogen that causes a disease known as white mold, which is a major problem for dry bean (Phaseolus vulgaris L.) and other crops in many growing areas in Brazil. To investigate the role of methyl jasmonate (MeJA) in defending dry bean plants against S. sclerotiorum, we used suppression subtractive hybridization (SSH) of cDNA and identified genes that are differentially expressed during plant-pathogen interactions after treatment. Exogenous MeJA application enhanced resistance to the pathogen, and SSH analyses led to the identifi

An overview of antifungal peptides derived from insect.

Fungi are not classified as plants or animals. They resemble plants in many ways but do not produce chlorophyll or make their own food photosynthetically like plants. Fungi are useful for the production of beer, bread, medicine, etc. More complex than viruses or bacteria; fungi can be destructive human pathogens responsible for various diseases in humans. Most people have a strong natural immunity against fungal infection. However, fungi can cause diseases when this immunity breaks down. In the last few years, fungal infection has increased strikingly and has been accompanied by a rise in the

Decoding MicroRNA-Guided Antiviral Defense in Cucurbitaceae: Regulatory Networks, RNA Silencing Cross-Talk, and Emerging Strategies for Crop Resilience.

MicroRNAs (miRNAs) are central regulators of gene expression and play pivotal roles in plant antiviral defense. In Cucurbitaceae, a globally important crop family including cucumber, melon, and watermelon, viral pathogens such as CGMMV, CMV, and ZYMV represent major constraints on productivity. However, the regulatory complexity of miRNA-mediated antiviral responses in these species remains incompletely understood. This review provides an integrated overview of recent advances in miRNA-guided antiviral immunity in Cucurbitaceae, highlighting the dynamic reprogramming of small RNA pathways upon

New molecules in plant defence against pathogens.

Plants host a multipart immune signalling network to ward off pathogens. Pathogen attack upon plant tissues can often lead to an amplified state of (induced) defence against subsequent infections in distal tissues; this is known as systemic acquired resistance (SAR). The interaction of plants with beneficial microbes of the rhizosphere microbiome can also lead to an induced resistance in above-ground plant tissues, known as induced systemic resistance. Second messengers such as calcium (Ca2+), reactive oxygen species (ROS), and nitric oxide (NO) are necessary for cell-to-cell signal propagatio

Apoplast-Confined Nanozyme Catalysis for Selective 1O2 Generation and Host Defense in Plants.

Reactive oxygen species (ROS)-based antimicrobial strategies in plants are often constrained by limited oxidant selectivity and collateral phytotoxicity. Here, we report an apoplast-confined nanozyme that integrates selective singlet oxygen (1O2) catalysis with host defense activation. The copper-iron Prussian blue analogue nanozyme CuFePBA exhibits oxidase-like activity under physiologically relevant conditions, converting molecular oxygen into 1O2 without detectable generation of superoxide or hydroxyl radicals. Following foliar application, CuFePBA is retained within the apoplast because of

Light wavelength-specific regulation of hypersensitive response dynamics in tobacco plants during non-host interaction with Pseudomonas syringae pv. tomato.

Light quality is a key environmental signal that integrates plant immune pathways, yet how specific wavelengths regulate the execution, timing, and efficiency of the hypersensitive response (HR) remains incompletely resolved. This study investigated how light spectral composition modulates HR dynamics and resistance outcomes during a non-host interaction. Tobacco leaves were inoculated with Pseudomonas syringae pv. tomato DC3000 (PstDC3000) and incubated under red (RL), green (GL), blue (BL), white light (WL), or darkness. Monochromatic light treatments distinctly reshaped HR progression. BL a
