A subhuman primate model for fetal alcohol syndrome research.

The object of this project was to develop and evaluate new methods for the establishment of a subhuman primate model (SPM) of the fetal alcohol syndrome (FAS). Adult Rhesus monkeys (5.5--16.0 kg) were used as subjects. All were proven breeders and all matings in this project were repeats of matings that had previously produced normal offspring. The animals received alcohol (ALC) via permanently implanted intragastric cannulae (IGC) at doses to establish blood alcohol concentrations (BAC) within 2 selected ranges. Initially the high dose group received ALC doses designed to maintain BAC of appr

The development and nature of immune deficit in primates in response to malnutrition.

Within 2--3 days of establishing protein-energy malnutrition (PEM) in primates there was a significant reduction (P less than 0.01) of the serum transferrin and C3 concentrations. No such changes occurred in the serum albumin until 21 days later. There was an early marked development of immune deficiency, at least 3 weeks before any signs of infection. Lymphocyte E and EAC rosetting, DNA stimulation, in vivo delayed hypersensitivity, passive cutaneous anaphylaxis and skin-graft rejection were all markedly decreased as early as Day 7 of the restricted diet. There was a disproportionately high p…

Endotoxic shock in the primate: effects of aspirin and dipyridamole administration.

A primate model was utilized to study the cardiovascular and coagulation effects of endotoxic shock. The therapeutic effectiveness of drugs such as aspirin and dipyridamole, which diminish platelet aggregation and adherence, were evaluated. From the data, it appears that the kidney is a target organ in endotoxic shock, at least when a bolus injection of endotoxin is administered. The precipitate falls in the renal artery flow (p less than 0.01) and platelet count (p less than 0.01), which occur 3 minutes after the intravenous injection of endotoxin, can be prevented in part by pretreatment wit

Responses to experimental subarachnoid hemorrhage in the spontaneously breathing primate.

The pathophysiological responses to experimental subarachnoid hemorrhage (SAH) were investigated in 20 spontaneously breathing cynomolgus monkeys. Four different volumes of fresh autogenous blood were used: 1.0, 1.33, 1.67, and 2.0 cc/kg. Five other animals had injection of 1.67 cc/kg of mock cerebrospinal fluid. Cerebral blood flow (CBF) was measured using the xenon-133 clearance technique. Respiratory rate and tidal volume were monitored by way of a Vertek pneumotach. The reduction of CBF after the SAH became more pronounced with increasing volumes of subarachnoid blood. The CBF remained red

Monoamines in the C cells of the thyroid gland of callithricid primates.

The thyroid gland of three species of marmosets were studied with the Falck and Hillarp fluroescence technique for monoamines. Two types of fluorescent C cells were observed. The predominant type displayed the greenish (or blue) fluorescence characteristic of catecholamines. The other type displayed a yellowish fluorescence most probably due to 5-HT. The presence of monoamines in C cells now reported for the first time in Primates offers an interesting possibility for studying their presumed role in calcitonin secretion.

Neuronal processing of facial expressions in the primate superior colliculus.

Fast processing of emotionally salient facial expressions is widely believed to depend on a subcortical visual circuit from the superior colliculus through the thalamus to the amygdala, but the specific processing steps remain unclear. Here we identified two largely nonoverlapping groups of neurons in the superior colliculus significantly modulated by facial expressions. One group exhibited a short-latency preference for threat and the other exhibited a delayed preference for fear.

BRIDGING COTYLEDON PATHOLOGY AND PERFUSION IN HEALTHY PRIMATE PREGNANCY.

INTRODUCTION: Healthy and diseased placentae alike often display some degree of pathology. However, quantitative techniques to characterize common pathologies and their relationship to local maternal hemodynamics in healthy primate placentae are currently limited. METHODS: Placentae from seven rhesus macaques were imaged by MRI at three time points across mid- to late-gestation, to quantify placental blood volume, flow, and perfusion from maternal spiral arteries across pregnancy. Near term, we collected placental cotyledons, digitized hematoxylin/eosin-stained slides, then segmented and anno

GABAergic interneurons in local circuits of primate prefrontal cortex.

Cortical GABAergic interneurons exhibit remarkable diversity of morphological, electrophysiological and molecular features, which underlies their specialized roles in circuit functions. Molecular characteristics have proven to be a reasonable starting point in the past for classification, grouping them into parvalbumin (PV), somatostatin (SST), vasoactive intestinal polypeptide (VIP) and "others" (e.g. LAMP5). While Patch-seq studies provide valuable insight into human cortical interneurons, similar results from non-human primates are lagging behind. This is problematic because the studying of…

Defining the cell and molecular origins of the primate ovarian reserve.

The primate ovarian reserve is established during late fetal development and consists of quiescent primordial follicles in the ovarian cortex each composed of granulosa cells surrounding an oocyte in dictate. As late stages of fetal development are not routinely accessible using human tissues, the current study exploits the evolutionary proximity of the rhesus macaque to investigate follicle formation in primates. Like in humans, the rhesus prenatal ovary develops multiple types of pre-granulosa cells in time and space, with primordial follicles deriving from later emerging pre-granulosa subty…

Nonhuman primate antigenic cartography of SARS-CoV-2.

Virus neutralization profiles against primary infection sera and corresponding antigenic cartography are integral part of the COVID-19 and i … or influenza virus. We evaluated neutralization profiles against 23 SARS-CoV-2 variants in nonhuman primates (NHPs) after single variant exposure and generated an NHP-derived antigenic map. We identified a distant antigenic region occupied by BA.2.86, JN.1, and the descendants KP.2, KP.3, and KZ.1.1.1. We also found that the monovalent XBB.1.5 mRNA vaccine …

The Use of Nonhuman Primate Models for Advancing HIV PrEP.

The global fight against HIV/AIDS has been significantly bolstered by the development and implementation of pre-exposure prophylaxis (PrEP), yet innovation in PrEP interventions, improved adherence and greater access are still needed to maximize its benefit. Nonhuman primate (NHP) infection with simian immunodeficiency virus (SIV) has served as an instrumental animal model in advancing HIV PrEP research. This review comprehensively examines the utility of NHP models in evaluating the efficacy, pharmacokinetics, and safety of diverse PrEP strategies, including oral, injectable, implantable, and

Transcriptional Profiling of Filovirus-Infected Nonhuman Primate Samples.

Transcriptional analysis is a powerful tool to help decipher the host response to filovirus infections. In this chapter, we describe how transcriptional analysis can be performed on samples obtained from nonhuman primates infected with filoviruses, specifically Marburg virus, for assessment of the RNA changes over the course of the infection and disease.

A non-human primate model for human norovirus infection.

Norovirus infection can cause gastrointestinal disease in humans. Development of therapies and vaccines against norovirus have been limited by the lack of a suitable and reliable animal model. Here we established rhesus macaques as an animal model for human norovirus infection. We show that rhesus macaques are susceptible to oral infection with human noroviruses from two different genogroups. Variation in duration of virus shedding (days to weeks) between animals, evolution of the virus over the time of infection, induction of virus-specific adaptive immune responses, susceptibility to reinfec

General mechanisms of task engagement in the primate frontal cortex.

Staying engaged is necessary to maintain goal-directed behaviors. Despite this, engagement exhibits continuous, intrinsic fluctuations. Even in experimental settings, animals, unlike most humans, repeatedly and spontaneously move between periods of complete task engagement and disengagement. We, therefore, looked at behavior in male macaques (macaca mulatta) in four tasks while recording fMRI signals. We identified consistent autocorrelation in task disengagement. This made it possible to build models capturing task-independent engagement. We identified task general patterns of neural activity

Aging hallmarks of the primate ovary revealed by spatiotemporal transcriptomics.

The ovary is indispensable for female reproduction, and its age-dependent functional decline is the primary cause of infertility. However, the molecular basis of ovarian aging in higher vertebrates remains poorly understood. Herein, we apply spatiotemporal transcriptomics to benchmark architecture organization as well as cellular and molecular determinants in young primate ovaries and compare these to aged primate ovaries. From a global view, somatic cells within the non-follicle region undergo more pronounced transcriptional fluctuation relative to those in the follicle region, likely constit

A revised perspective on the evolution of the lateral frontal cortex in primates.

Detailed neuroscientific data from macaque monkeys have been essential in advancing understanding of human frontal cortex function, particularly for regions of frontal cortex without homologs in other model species. However, precise transfer of this knowledge for direct use in human applications requires an understanding of monkey to hominid homologies, particularly whether and how sulci and cytoarchitectonic regions in the frontal cortex of macaques relate to those in hominids. We combine sulcal pattern analysis with resting-state functional magnetic resonance imaging and cytoarchitectonic an

An ON-type direction-selective ganglion cell in primate retina.

To maintain a stable and clear image of the world, our eyes reflexively follow the direction in which a visual scene is moving. Such gaze-stabilization mechanisms reduce image blur as we move in the environment. In non-primate mammals, this behaviour is initiated by retinal output neurons called ON-type direction-selective ganglion cells (ON-DSGCs), which detect the direction of image motion and transmit signals to brainstem nuclei that drive compensatory eye movements1. However, ON-DSGCs have not yet been identified in the retina of primates, raising the possibility that this reflex is mediat…
