An integrated subtractive genomics and immunoinformatics approach for designing a universal multi-epitope vaccine against Brucella spp.

INTRODUCTION: Brucella spp. are Gram-negative bacteria accountable for brucellosis in immunocompromised individuals and livestock. Due to the slow-growing latent phenotype, current antibiotics are insufficient to treat the infection. The lack of an approved vaccine for human use against this pathogen represents a significant public health concern and indicates the urgent need for novel prophylactic interventions. METHODOLOGY: In this study, the reverse vaccinology method was combined with pan-genome analysis to identify potential vaccine targets. Proteins have been screened for antigenicity,

A novel chimpanzee adenovirus vector vaccine for protection against infectious bronchitis and Newcastle disease in chickens.

The development of effective poultry vaccines is crucial for maintaining flock health and productivity. In this study, we developed and evaluated a recombinant chimpanzee adenovirus vaccine (PAD-S1-HN) that simultaneously expresses the infectious bronchitis virus (IBV) spike subunit protein S1 and Newcastle disease virus (NDV) hemagglutinin-neuraminidase HN protein. The recombinant virus was successfully rescued in HEK293 cells, and transmission electron microscopy confirmed its typical adenoviral morphology. The expression of the IBV S1 and NDV HN proteins was validated by indirect immunofluo

Rational mpox vaccine design: immunogenicity and protective effect of individual and multicomponent proteins in mice.

The 2022 global mpox virus (MPXV) outbreak highlights the urgent need for safer, next-generation vaccines. We compared the immunogenicity and protective efficacy of individual and multicomponent membrane proteins of MPXV virions in mice to inform the development of a recombinant subunit vaccine against mpox. BALB/c mice were immunized with eukaryotically expressed A35R, A29L, B6R, and M1R proteins, administered individually or in multicomponent combinations with an Al(OH)3 + CpG oligodeoxynucleotide adjuvant. Three multicomponent protein vaccines (A29/B6, A29/B6/M1, and A29/B6/M1/A35) provided

[Purification and identification of pili of Vibrio cholera O139].

To provide the base of manufacturing V. cholera O139 subunit vaccine, purification and identification methods of V. cholera O139 Toxin-coregulated pili (TCP) were studied. The results showed the optimal expressing condition of TCP was in AKI or CFA medium, at 30 C for 24-36 h with stationary cultivation. The molecular weight of the TCP subunit protein measured by sodium dodecyl sulfate-poly acrylamide gel electrophoresis was about 20.5 kd. By using mouse anti-TCP monoclone antibody and sheep anti-mouse IgG-HRP antibody, the TCP was confirmed by dot blot immunoassay. Rabbit immune serum of TCP

Adjuvanted SARS-CoV-2 spike protein elicits neutralizing antibodies and CD4 T cell responses after a single immunization in mice.

BACKGROUND: SARS-CoV-2 has caused a global pandemic, infecting millions of people. A safe, effective vaccine is urgently needed and remains a global health priority. Subunit vaccines are used successfully against other viruses when administered in the presence of an effective adjuvant. METHODS: We evaluated three different clinically tested adjuvant systems in combination with the SARS-CoV-2 pre-fusion stabilized (S-2P) spike protein using a one-dose regimen in mice. FINDINGS: Whilst spike protein alone was only weakly immunogenic, the addition of either Aluminum hydroxide, a squalene based

Active vaccination with vaccinia virus A33 protects mice against lethal vaccinia and ectromelia viruses but not against cowpoxvirus; elucidation of the specific adaptive immune response.

Vaccinia virus protein A33 (A33VACV) plays an important role in protection against orthopoxviruses, and hence is included in experimental multi-subunit smallpox vaccines. In this study we show that single-dose vaccination with recombinant Sindbis virus expressing A33VACV, is sufficient to protect mice against lethal challenge with vaccinia virus WR (VACV-WR) and ectromelia virus (ECTV) but not against cowpox virus (CPXV), a closely related orthopoxvirus. Moreover, a subunit vaccine based on the cowpox virus A33 ortholog (A33CPXV) failed to protect against cowpox and only partially protected mi

Development of a tripartite vector system for live oral immunization using a gram-negative probiotic carrier.

The mucosa represents the primary target site and thus the first barrier for most microbial pathogens. Nevertheless, nearly all present-day vaccines are applied by an invasive route, target the systemic immune system, and do not confer efficient mucosal protection. Currently, mucosal immunity can only be achieved by the delivery of antigens via the mucosal route. Therefore, multiple efforts are under way to develop mucosal vaccines and particularly live oral vaccines as these would confer considerable advantages. We have engineered the AIDA autotransporter system for the surface presentation a

[Eukaryotic expression of GP5 and M protein of porcine reproductive and respiratory syndrome virus and immunogenicity evaluation].

In order to understand the prevalence and evolution of porcine reproductive and respiratory syndrome virus (PRRSV) in China and to develop subunit vaccine against the epidemic lineage, the genetic evolution analysis of PRRSV strains isolated in China from 2001 to 2021 was performed. The representative strains of the dominant epidemic lineage were selected to optimize the membrane protein GP5 and M nucleotide sequences, which were used, with the interferon and the Fc region of immunoglobulin, to construct the eukaryotic expression plasmids pCDNA3.4-IFNα-GP5-Fc and pCDNA3.4-IFNα-M-Fc. Evaluation of Mycoplasma mycoides subsp. mycoides antigens capable of stimulating host IRG-47 release identifies Mmm604, Mmm605, and Mmm606 as potential subunit vaccine antigens.

Contagious bovine pleuropneumonia (CBPP), caused by Mycoplasma mycoides subsp. mycoides (Mmm), is a devastating cattle disease with high morbidity and mortality, threatening cattle productivity in Sub-Saharan Africa and potentially in parts of Asia. Cross-border livestock trade increases the risk of CBPP introduction or reintroduction. Current vaccines were developed from attenuated Mmm strains in the last century and face limitations regarding animal welfare, immunity duration, and adverse reactions, necessitating new vaccine strategies. Subunit vaccines offer a promising alternative, but ide

Proteomics and bioinformatics strategies to design countermeasures against infectious threat agents.

The potential devastation resulting from an intentional outbreak caused by biological warfare agents such as Brucella abortus and Bacillus anthracis underscores the need for next generation vaccines. Proteomics, genomics, and systems biology approaches coupled with the bacterial ghost (BG) vaccine delivery strategy offer an ideal approach for developing safer, cost-effective, and efficacious vaccines for human use in a relatively rapid time frame. Critical to any subunit vaccine development strategy is the identification of a pathogen's proteins with the greatest potential of eliciting a prote

Secreted expression of truncated capsid protein from porcine circovirus type 2 in Pichia pastoris.

OBJECTIVE: To achieve secreted expression of the truncated capsid protein from porcine circovirus type 2 (PCV2) in Pichia pastoris. RESULTS: A truncated cap gene (tcap) with a deleted N-terminal nuclear localization signal was optimized and synthesized. Effective secreted expression was achieved in P. pastoris GS115. The high-productive recombinant strain for tCap was grown in a 5 l bioreactor and the productivity of tCap in supernatant reached 250 μg/ml. Furthermore, serum antibody test demonstrated that adjuvant-assisting tCap induced a significant increase of specific PCV2-Cap antibody ove

An invariant Trypanosoma vivax vaccine antigen induces protective immunity.

Trypanosomes are protozoan parasites that cause infectious diseases, including African trypanosomiasis (sleeping sickness) in humans and nagana in economically important livestock1,2. An effective vaccine against trypanosomes would be an important control tool, but the parasite has evolved sophisticated immunoprotective mechanisms-including antigenic variation3-that present an apparently insurmountable barrier to vaccination. Here we show, using a systematic genome-led vaccinology approach and a mouse model of Trypanosoma vivax infection4, that protective invariant subunit vaccine antigens can

A foldon-free prefusion F trimer vaccine for respiratory syncytial virus to reduce off-target immune responses.

Respiratory syncytial virus (RSV) is a major cause of severe respiratory disease in infants and older people. Current RSV subunit vaccines are based on a fusion protein that is stabilized in the prefusion conformation and linked to a heterologous foldon trimerization domain to obtain a prefusion F (preF) trimer. Here we show that current RSV vaccines induce undesirable anti-foldon antibodies in non-human primates, mice and humans. To overcome this, we designed a foldon-free RSV preF trimer by elucidating the structural basis of trimerization-induced preF destabilization through molecular dynam

Balancing the influenza neuraminidase and hemagglutinin responses by exchanging the vaccine virus backbone.

Virions are a common antigen source for many viral vaccines. One limitation to using virions is that the antigen abundance is determined by the content of each protein in the virus. This caveat especially applies to viral-based influenza vaccines where the low abundance of the neuraminidase (NA) surface antigen remains a bottleneck for improving the NA antibody response. Our systematic analysis using recent H1N1 vaccine antigens demonstrates that the NA to hemagglutinin (HA) ratio in virions can be improved by exchanging the viral backbone internal genes, especially the segment encoding the po

Protective anti-idiotype vaccine against Schistosoma mansoni infection.

Vaccine development may be the most cost-effective way to ultimately control schistosomiasis, a disease that affects 200 million people worldwide. Vaccine development in schistosomiasis has centered on identifying key surface antigen(s) on schistosomula, the migrating larval form of the parasite, and cloning these antigens through molecular biology-derived technology. This strategy will be most effective if the key surface epitopes are protein in nature. An alternative strategy is to develop anti-idiotypic antibody vaccines that take advantage of the unique antigen-mimic quality of certain ant

Reverse vaccinology-based identification of immunogenic membrane proteins from zoonotic multidrug-resistant Proteus vulgaris: a one health approach to cross-species vaccine development.

BACKGROUND: Proteus vulgaris is a Gram-negative opportunistic bacterium implicated in wound infections, urinary tract infections, foodborne illnesses, pneumonia and septicemia. It poses a growing threat to human, veterinary and aquaculture health systems. In animals, P. vulgaris has been isolated from cattle, poultry, and pets, while in fish species such as Nile tilapia and Clarias gariepinus, it contributes to ulcerative and systemic diseases. Its frequent detection in raw milk, meat, and retail fish highlights its zoonotic potential. The rise of multidrug-resistant strains across clinical, v

CD38-Targeting Peptide Vaccine Ameliorates Aging-Associated Phenotypes in Mice.

Antiaging vaccines have recently been found to elicit long-term benefits in slowing the aging process. Meanwhile, high CD38 expression in organs is an aging characteristic contributing to a decreased NAD+/NADH ratio. Thus, in the current study, we systematically investigate the effects of a CD38-targeting peptide vaccine (CD38-vaccine) on aging-associated phenotypes in mice. The CD38-vaccine induces a robust T-cell immune response, selectively depletes CD38+ myeloid cells in the spleen, and ameliorates age-related physical and cognitive function decline. Metabolically, vaccination improves glu

The year that shaped the outcome of the OspA vaccine for human Lyme disease.

The expansion of Lyme borreliosis endemic areas and the corresponding increase of disease incidence have opened the possibility for greater acceptance of a vaccine. In this perspective article, we discuss the discovery of outer surface protein A (OspA) of B. burgdorferi, and the subsequent pre-clinical testing and clinical trials of a recombinant OspA vaccine for human Lyme disease. We also discuss in detail the open public hearings of the FDA Lyme disease vaccine advisory panel held in 1998 where concerns of molecular mimicry induced autoimmunity to native OspA were raised, the limitations of

Strategic Development of a Next-Generation Multi-Epitope Vaccine To Prevent Nipah Virus Zoonotic Infection.

Nipah virus (NiV) is an emerging zoonotic pathogen, reported for the recent severe outbreaks of encephalitis and respiratory illness in humans and animals, respectively. Many antiviral drugs have been discovered to inhibit this pathogen, but none of them were that much efficient. To overcome the complications associated with this severe pathogenic virus, we have designed a multi-epitope subunit vaccine using computational immunology strategies. Identification of structural and nonstructural proteins of Nipah virus assisted in the vaccine designing. The selected proteins are known to be involve

Evaluation of novel recombinant porcine circovirus type 2d (PCV2d) vaccine in pigs naturally infected with PCV2d.

INTRODUCTION: The pathogenic porcine circovirus type 2 (PCV2) causes significant economic losses in pig production. Emergence of the PCV2d genotype has been linked with PCV2-associated disease (PCVAD) outbreaks. However, no study has been conducted efficacy of an experimental PCV2d-based subunit vaccine in pigs. Therefore, PCV2b- and PCV2d-based capsid (CP) proteins were generated using a baculovirus (Bac) expression system, and we evaluated the protective immune responses in a commercial pig farm where predominant PCV2d is circulating. METHODS: Eighteen 3-week-old pigs with maternal antibodi
