Aqueous synthesis and characterization of TGA-capped CdSe quantum dots at freezing temperature.

CdSe quantum dots (QDs) have traditionally been synthesized in organic phase and then transferred to aqueous solution by functionalizing their surface with silica, polymers, short-chain thiol ligands, or phospholipid micelles. However, a drastic increase in the hydrodynamic size and biotoxicity of QDs may hinder their biomedical applications. In this paper, the TGA-capped CdSe QDs are directly synthesized in aqueous phase at freezing temperature, and they prove to possess high QY (up to 14%).

Toxicity of quantum dots and cadmium salt to Caenorhabditis elegans after multigenerational exposure.

To fully understand the biological and environmental impacts of nanomaterials requires studies that address both sublethal end points and multigenerational effects. Here, we use a nematode to examine these issues as they relate to exposure to two different types of quantum dots, core (CdSe) and core-shell (CdSe/ZnS), and to compare the effect to those observed after cadmium salt exposures. The strong fluorescence of the core-shell QDs allowed for the direct visualization of the materials in the digestive track within a few hours of exposure. Multiple end points, including both developmental an…

Synthesis and application of intercellular Ca2+-sensitive fluorescent probe based on quantum dots.

A novel Ca(2+)-sensitive fluorescent probe was synthesized and characterized with a coupled method that coupled di[2-(N,N-dicarboxylmethyl)amino]ethyl ether (EGTA) to the surface of mercaptoethylamine-modified CdTe quantum dots (CdTe/MA-EGTA QDs). The application of this probe to detect intercellular Ca(2+) change in the leaf cells of Arabidopsis thaliana was studied. Results from transmission electron micrographs showed that the particle size of CdTe/MA-EGTA was about 3-4 nm; the fluorescent spectrum indicated that the excitation spectral ranged from 350 to 490 nm with a narrow and symmetric …

Toxicity evaluation of hydrophilic CdTe quantum dots and CdTe@SiO2 nanoparticles in mice.

Quantum dots have drawn tremendous attention in the field of in vitro and small animal in vivo fluorescence imaging in the last decade. However, concerns over the cytotoxicity of their heavy metal constituents have limited their use in clinical applications. Here, we report our comparative studies on the toxicities of quantum dots (QDs) and silica coated CdTe nanoparticles (NPs) to mice after intravenous injection. The blood cells analysis showed significant increased level of white blood cells (WBCs) in groups treated with CdTe QDs as compared to the control while red blood cells (RBCs) and p…

Interaction of β-cyclodextrin-capped CdSe quantum dots with inorganic anions and cations.

A facile method was developed for the preparation of water soluble β-Cyclodextrin (β-CD)-modified CdSe quantum dots (QDs) (β-CD-QDs) by directly replacing the oleic acid ligands on the QDs surface with β-CD in an alkaline aqueous solution. The as-prepared QDs show good stability in aqueous solution for several months. Oxoanions, including phosphoric acid ion, sulphite acid ion and carbonic acid ion, affect the fluorescence of β-CD-QDs. Among them, H(2)PO(4)(-) exhibited the largest quenching effect. For the polyprotic acids (HO)(3)AO, the effect of acidic anions on the fluorescence of β-CD-QDs…

Liposome encapsulation of thiol-capped CdTe quantum dots for enhancing the intracellular delivery.

Although water soluble thiol-capped quantum dots (QDs) have been widely used as photoluminescence (PL) probes in various applications, the negative charges on thiol terminals limit the cell uptake hindering their applications in cell imaging. The commercial liposome complex (Sofast®) was used to encapsulate these QDs forming the liposome vesicles with the loading efficiency as high as about 95%. The cell uptakes of unencapsulated QDs and QD loaded liposome vesicles were comparatively studied by a laser scanning confocal microscope. We found that QD loaded liposome vesicles can effectively enha…

Preparation of stable, water-soluble, highly luminescence quantum dots with small hydrodynamic sizes.

This work presents a simple method to prepare water-soluble alloyed CdSe-ZnS quantum dots, which photoluminescence are tunable from green to red continuously, through replacing the hydrophobic oleic acid stabilizers with hydrophilic thiol molecules. 3-Mercaptopropionic acid and 2-mercaptoethylamine have been used respectively as the surface substitutes to obtain water-soluble quantum dots with negative and positive surface charges. The methods achieved highly efficient phase transfer (approximately 100%) of quantum dots from non-polar media to water. Different techniques including photolumines…

Trophic transfer of amphiphilic polymer coated CdSe/ZnS quantum dots to Danio rerio.

To investigate the trophic transfer of nanomaterials along the food chain, we examined the potential trophic transfer and biomagnification of CdSe/ZnS quantum dots (QDs) in a simple freshwater food chain. Our results indicate that QDs can transfer from zooplankton to Danio rerio (zebrafish) by dietary exposure. No significant biomagnification of QDs was observed and the biomagnification factors for both adult and juvenile zebrafish were both less than one (0.04 and 0.004 respectively). The assimilation efficiency was 8% and 4% for adult and juvenile zebrafish respectively. This study is the fi…

Rapid detection of lily symptomless virus with CdTe quantum dots by flow cytometry.

Lily symptomless virus (LSV) is the most common lily virus, being detected in many species and hybrids. We established a microsphere-based fluorescent immunoassay for the determination of LSV, using a polyclonal antibody against LSV covalently bound to carboxy-modified microspheres able to capture LSV antigen. A monoclonal antibody against LSV conjugated to quantum dots (QDs) was used as a fluorescent probe, enabling LSV to be fluorescently detected by a combination of encoded beads and QDs. This method was 16 times more sensitive than ELISA in the detection of LSV, and could potentially be ap…

The inhibition of fluorescence resonance energy transfer between quantum dots for glucose assay.

Fluorescence resonance energy transfer (FRET) between two quantum dots of different sizes causes fluorescence quenching. Hereby a binding site pre-blocking approach is proposed to avoid this effect. Pre-binding of glucose on the donor occupies the binding sites and thus blocks resonance energy transfer between the two quantum dots, protecting the fluorescence from being quenched. A glucose assay is developed based on this approach. The glucose content is correlated with the fluorescence difference in the absence and in the presence of glucose. In practice, Green QDs-Con A conjugates are used a…

An upper bound to carrier multiplication efficiency in type II colloidal quantum dots.

We experimentally investigate carrier multiplication (CM) in type II CdTe/CdSe quantum dot (QD) heterostructures by the means of a simple and robust subnanosecond transient photoluminescence spectroscopy setup. Experimental conditions were set to minimize the blurring of the CM signature by extraneous effects. The extracted photon energy threshold for CM is consistent with previous studies in CdSe and CdTe QDs (around 2.65 times the type II energy band gap) and we can infer an upper bound to CM yield. This study indicates that, while CM is probably present in type II QD heterostructures below

Preparation of photocontrollable, nontoxicity complex of phenylazophenylalanine modified CdS quantum dots with cyclodextrin.

A novel complex of beta-cyclodextrin with phenylazophenylalanine (PAP)-conjugated CdS quantum dot (Fazo-CdS QD) was fabricated for a photocontrollable toxicity study of CdS. Photocontrolling release of Fazo-CdS QDs from the complex was confirmed under UV irradiation by the steady state photoluminescence (PL) spectroscopy. The cytotoxicity of the surface modified CdS QDs was evaluated in vivo. The results suggest that the Fazo-CdS QDs conjugated with beta-CD could be almost nontoxicity, which may have great potential applications in biological labeling and imaging.

One-pot large-scale synthesis of robust ultrafine silica-hybridized CdTe quantum dots.

A facile one-pot strategy for synthesis of silica-hybridized CdTe quantum dots (SiO(2)-h-CdTe QDs) in aqueous solution is presented, and subkilogram scale fluorescent SiO(2)-h-QDs can be readily produced in one batch. This approach also makes the tuning of emission wavelength and absorption bandgap of SiO(2)-h-QDs accessible for the first time. In the case of using MPA as ligand, the emission wavelength and absorption bandgap can be tuned in the range of 546-584 nm (the corresponding diameter of QDs increased from 2.0 to 3.2 nm) and 2.55-2.27 eV, respectively. The content of QDs in the resulti…

Biokinetics and in vivo distribution behaviours of silica-coated cadmium selenide quantum dots.

Recently, quantum dots derived from trace elements like cadmium and selenium have attracted widespread interest in biology and medicine. They are rapidly being used as novel tools for both diagnostic and therapeutic purposes. In this report, we evaluated the distribution of silica-coated cadmium selenide (CdSe) quantum dots (QDs) following intravenous injection into male Swiss albino mice as a model system for determining tissue localization using in vivo fluorescence and ex vivo elemental analysis by inductively coupled plasma optical emission spectroscopy (ICP-OES). "Cloud" assemblies: quantum dots form electrostatically bound dynamic nebulae around large gold nanoparticles.

Dynamic self-assembled structures of nanoparticles can be produced using predominantly electrostatic interactions. Such assemblies were made from large, positively charged Au metal nanoparticles surrounded by an electrostatically bound cloud of smaller, negatively charged CdSe/ZnS or CdTe quantum dots. At low concentrations they are topologically similar to double electric layers of ions and corona-like assemblies linked by polymer chains. They can also be compared to the topological arrangement of some planetary systems in space. The great advantages of the cloud assemblies are (1) their high…

[Characterization of biocompatible CdTe/znte quantum dots and its application in cell labeling].

Water-soluble CdTe/ZnTe core-shell quantum dots (QDs) coated with L-cysteine were synthesized in low-temperature aqueous-phase one-pot approach. The authors measured the spectral characteristics of QDs at different pH in various buffer solutions and under different excitation laser powers. The primary results show that the absorption spectra of QDs approximately overlap and the fluorescence spectra peaks have no shift in different pH solution. The fluorescence intensity increased linearly with increasing pH. With the incubation time in borate buffer solution, the fluorescence intensity decreas…

In vivo NIR imaging with CdTe/CdSe quantum dots entrapped in PLGA nanospheres.

Luminescent near-infrared (NIR) CdTe/CdSe QDs were synthesized and encapsulated in poly(lactic-co-glycolic acid) (PLGA) nanospheres to prepare stable and biocompatible QDs-loaded nanospheres for in vivo imaging. QDs were encapsulated with PLGA nanospheres by a solid dispersion method and optimized to have high fluorescence intensity for in vivo imaging detection. The resultant QDs-loaded PLGA nanospheres were characterized by various analytical techniques such as UV-Vis measurement, dynamic light scattering (DLS), fluorescence spectroscopy, and transmission electron microscopy (TEM). Finally,

Fluorescent recognition of potassium and calcium ions using functionalised CdSe / ZnS quantum dots.

Schiff base receptor 1a has been synthesised and attached to the surface of preformed CdSe/ZnS Quantum Dots (QDs) to form QD-conjugate 2a. While 1a was determined to be selective for Mg2+, 2a demonstrated selectivity for both K+ and Ca2+ when tested against a range of physiologically and environmentally relevant cations by changes in the fluorescence spectra. Thus, the nanoparticle surface functions as a scaffold for the organisation of receptors enabling semi-selective binding. The fluorescence response was shown to be linear between 15-50 microM for K+ and 2-35 microM for Ca2+. It was also d…
