Uniform tissue distribution of tRNA(Lys) mutation in mitochondrial DNA in MERRF patients.

We documented the presence of a point mutation in the tRNA(Lys) gene of mitochondrial DNA (mtDNA) in various postmortem tissues from two patients with myoclonus epilepsy associated with ragged-red fibers (MERRF). The percentages of the mutant mtDNA were similar (93 to 99%) in both clinically affected and unaffected tissues, suggesting that preferential clinical involvement of certain tissues in MERRF is based not only on the variation of distribution of the mutant mtDNA, but also on other factors such as differences in the threshold in various CNS regions and organs.

Inhibition of deacylation and improvement in N-hydroxysuccinimide ester modification of phenylalanyl-tRNA by dimethyl sulfoxide.

We have found that dimethyl sulfoxide significantly inhibits deacylation of Phe-tRNA. This allows a high pH in N-hydroxysuccinimide ester reactions while maintaining a high level of aminoacylated tRNA, improving the overall yield of the Phe-tRNA modification reaction.

Extracellular RNA Biomarkers for Chronic Non-Healing Wounds.

Chronic non-healing wounds represent a major clinical challenge, often associated with diabetes, vascular insufficiencies and aging. Despite the substantial burden that such wounds place on patients and healthcare systems, few biomarkers have been approved for prediction of wound healing trajectories and outcomes, limiting opportunities to inform clinical management decisions or quantify patient responses to interventions. Recent advances have identified cell-free nucleic acids as powerful tools for gaining molecular insights because they offer a non-invasive, dynamic snapshot of physiological

In vitro gene targeting in human hepatoblastoma.

Poor treatment results in advanced hepatoblastoma (HB) made alternative treatment approaches desirable. Gene-directed tumor therapy is increasingly investigated in different malignancies. The aim of this study was to analyze possible alternatives of gene transfer into HB cells and to study therapeutic applications based on different strategies. Liposomal transfection of HB cells was assessed using liver-specific promoters, and adenovirus and Sendai virus transductions were performed in vitro. Transfer efficiencies were measured via flow cytometry determining expression of vector-encoded marker

PANDORA-seq reveals human sperm sncRNA signature endowed with sperm quality assessment.

One of the leading causes of human subfertility is the continuous decline in semen quality, contributing to a global fertility crisis. Over half of subfertile men suffer from asthenozoospermia and teratozoospermia, with mechanisms still largely unknown. Traditional small noncoding RNA sequencing (sncRNA-seq) primarily targets miRNAs, failing to capture the broader spectrum of small noncoding RNAs (sncRNAs), including abundant transfer RNA-derived small RNAs (tsRNAs) and ribosomal RNA-derived small RNAs (rsRNAs). These sncRNAs possess complex RNA modifications and non-canonical terminal structu

Correcting human mitochondrial mutations with targeted RNA import.

Mutations in the human mitochondrial genome are implicated in neuromuscular diseases, metabolic defects, and aging. An efficient and simple mechanism for neutralizing deleterious mitochondrial DNA (mtDNA) alterations has unfortunately remained elusive. Here, we report that a 20-ribonucleotide stem-loop sequence from the H1 RNA, the RNA component of the human RNase P enzyme, appended to a nonimported RNA directs the import of the resultant RNA fusion transcript into human mitochondria. The methodology is effective for both noncoding RNAs, such as tRNAs, and mRNAs. The RNA import component, poly

RNA Polymerase III Subunit Mutations in Genetic Diseases.

RNA polymerase (Pol) III transcribes small untranslated RNAs such as 5S ribosomal RNA, transfer RNAs, and U6 small nuclear RNA. Because of the functions of these RNAs, Pol III transcription is best known for its essential contribution to RNA maturation and translation. Surprisingly, it was discovered in the last decade that various inherited mutations in genes encoding nine distinct subunits of Pol III cause tissue-specific diseases rather than a general failure of all vital functions. Mutations in the POLR3A, POLR3C, POLR3E and POLR3F subunits are associated with susceptibility to varicella z…

Temporal Dynamics of tsRNA Regulation Mark an Abrupt Transition After Epileptogenesis.

Epileptogenesis involves widespread molecular remodeling, including transcriptional and post-transcriptional changes that reshape neuronal networks. While microRNAs have been extensively studied in this context, the contribution of transfer RNA-derived small RNAs (tsRNAs) remains largely unexplored. Understanding how tsRNAs engage in Argonaute 2 (Ago2)-mediated regulation during epileptogenesis could uncover new layers of post-transcriptional control relevant to seizure development and progression. Recent studies increasingly recognize transfer RNA-derived small RNAs or tsRNAs, especially thos

Transient transcriptional activation of the Incl1 plasmid anti-restriction gene (ardA) and SOS inhibition gene (psiB) early in conjugating recipient bacteria.

The ardA gene of the enterobacterial plasmid CollbP-9 acts to alleviate restriction of DNA by type I systems, while psiB inhibits induction of the bacterial SOS response. Both genes are transferred early in a round of bacterial conjugation as part of the plasmid leading region. We report here that ardA and psiB are transcribed transiently after their conjugative transport into the recipient cell. Transcript levels, monitored by competitive reverse transcription-polymerase chain reaction (RT-PCR) amplification of RNA templates, started to increase about 5 min after the initiation of conjugation

Deciphering transcriptional plasticity in pancreatic ductal adenocarcinoma reveals alterations in sensory neuron innervation.

Neuronal innervation of the pancreas has historically been characterized using marker-based classification and physiological studies, but its transcriptomic landscape remains only partially explored. A detailed molecular profile of pancreatic sensory neurons could provide insights into their role in health and disease, particularly in pancreatic ductal adenocarcinoma (PDAC), where neural remodeling influences tumor progression and pain signaling. Wild-type and PDAC mice were injected with the retrotracer Fast Blue into pancreatic or cancerous tissue. Dorsal root ganglia were dissociated, and F

Release of extracellular vesicles containing small RNAs from the eggs of Schistosoma japonicum.

BACKGROUND: Schistosoma japonicum is a parasitic flatworm that causes human schistosomiasis. Secreted extracellular vesicles (EVs) play a key role in pathogen-host interfaces. Previous studies have shown that S. japonicum adult worms can release microRNA (miRNA)-containing EVs, which can transfer their cargo to mammalian cells and regulate gene expression in recipient cells. Tissue-trapped eggs are generally considered the major contributor to the severe pathology of schistosomiasis; however, the interactions between the host and parasite in this critical stage remain largely unknown. A Novel Presentation of Refractory Immune Thrombocytopenia in Anti-synthetase Syndrome: A Case Report.

Anti-synthetase syndrome (AS) is a rare autoimmune disorder classified among the idiopathic inflammatory myopathies and is characterized by antibodies directed against aminoacyl-transfer RNA synthetases and the presence of myositis, interstitial lung disease, ±arthritis. Here, we report, for the first time, immune thrombocytopenia (ITP) in a patient with AS. This case reports a new association of AS with ITP and highlights the utility of identifying the underlying driver in secondary ITP to guide therapy.

Toxic and nutritional factors trigger Leber hereditary optic neuropathy due to a mitochondrial tRNA mutation.

Leber hereditary optic neuropathy is a mitochondrial disease mainly due to pathologic mutations in mitochondrial genes related to the respiratory complex I of the oxidative phosphorylation system. Genetic, physiological, and environmental factors modulate the penetrance of these mutations. We report two patients suffering from this disease and harboring a m.15950G > A mutation in the mitochondrial DNA-encoded gene for the threonine transfer RNA. We also provide evidences supporting the pathogenicity of this mutation.

Production of triploid transgenic zebrafish, Brachydanio rerio (Hamilton).

Eggs of zebrafish Brachydanio rerio were heat-shocked just after fertilisation to induce triploidy and subsequently subjected to microinjection with the linear recombinant plasmid containing the Rous Sarcoma Virus-Long Terminal Repeat (RSV-LTR) promoter linked to the rainbow trout (Oncorhynchus mykiss) growth hormone (GH)cDNA(pRSVrtGH). Slot and Southern analyses showed the extra chromosomal transfer of pRSVrtGH. Expression of pRSVrtGH was detected in the triploid transformants by RNA slot, Northern and Western analyses.

A Single Dimer of the SARS-CoV‑2 N Protein Can Associate with Multiple Fragments of Single-Stranded and Stem-Loop RNA: A Single-Molecule FRET and FCS Investigation.

The nucleocapsid (N) protein of SARS-CoV-2 binds to the viral genomic RNA (gRNA), forming ribonucleoprotein (RNP) granules. However, the detailed molecular structures of the RNP and how they form are largely unknown. To understand the interaction between the N protein and different structural units of RNA, we used circular dichroism (CD) spectroscopy, fluorescence correlation spectroscopy (FCS), and single-molecule Förster resonance energy transfer (sm-FRET) spectroscopy. We selected polyadenylate chains of 40, 30, and 20 bases with a single-stranded structure and three stem loops of 50, 41, a

mRNA turnover dynamics are affected by cell differentiation and loss of the cytosine methyltransferase Nsun2.

Nsun2 catalyzes 5-methylcytosine (m5C) formation in several types of RNA, including messenger RNAs (mRNAs), transfer RNAs, and other non-coding RNAs. In mRNA, m5C was reported to influence transcript stability. However, it is unclear if it has stabilizing or destabilizing effects. To address the role of Nsun2 in mRNA stability, we characterized the landscape of mRNA turnover dynamics during embryonic stem cell (ESC) differentiation in wild-type and Nsun2-mutant cells. By using an RNA labeling approach combined with thiouridine-to-cytidine-sequencing (TUC-seq), we demonstrate that mRNA synthesi
