Radiation-induced brain injury (RIBI) is a serious complication of cranial radiotherapy and currently lacks effective disease-modifying treatment. Persistent neuroinflammation and neurovascular dysfunction are considered central to RIBI progression, making them attractive therapeutic targets for regenerative intervention. Multilineage-differentiating stress-enduring (Muse) cells are a non-tumorigenic pluripotent-like cell population with reported reparative and immunomodulatory properties, but their therapeutic potential in RIBI has not been defined. In this study, we evaluated the effects of

BACKGROUND: Ionizing radiation (IR) generates reactive oxygen species (ROS), resulting in oxidative imbalance and tissue injury. The nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway regulates antioxidant gene expression; however, effective radiomodulatory agents remain limited. Resveratrol (RES), a natural antioxidant with anti-inflammatory and ROS-scavenging properties, has been proposed as a potential radioprotective compound.
METHODS: Forty male albino rats were randomly assigned to four groups (n = 10/group): control, RES-only (20 mg/kg/day, or

INTRODUCTION: Radiation-induced temporal lobe injury (RI-TLI) is one of the most common late-stage complications after radiotherapy for nasopharyngeal carcinoma (NPC), which seriously affects patients' cognitive function and quality of life. However, the mechanism by which RI-TLI affects the gray matter (GM) and white matter (WM) across the whole brain, leading to cognitive impairment, is currently unclear in NPC patients.
METHODS: One year after the end of radiotherapy, 32 NPC patients with RI-TLI (RI-TLI group) and 38 NPC patients without RI-TLI (nRI-TLI group) were included in this study.

Radiation therapy for head and neck cancer often causes otologic complications due to inflammation and mucosal damage in the middle ear. To address this problem, an injectable glycol chitosan-dexamethasone prodrug thermogel (HGC-Su-DEX) was developed for localized and sustained otoprotective therapy. Dexamethasone (DEX) was covalently linked to hexanoyl glycol chitosan (HGC) via a biodegradable succinate linker, yielding an amphiphilic conjugate capable of sol-gel transition near physiological temperature. The HGC-Su-DEX formulations exhibited tunable thermogelling behavior and comparable rele

UNLABELLED: Fractional ionizing radiation (FIR) is a standard cancer therapy but often induces severe central nervous system complications, including cognitive decline and physiological dysfunction. While natural flavonoids hold therapeutic promise for radiation-induced brain injury (RIBI), optimizing their combinations and elucidating the underlying molecular pathways remain challenging.
METHODS: To develop a precise therapeutic strategy, we first integrated network pharmacology and UHPLC-Q-Orbitrap MS/MS analysis to identify three highly effective flavonoid monomers from a radioprotective b

OBJECTIVE: Complications associated with nasopharyngeal carcinoma (NPC) treatment significantly impair patients' quality of life. Radiotherapy-induced xerostomia is one of the most common complications of NPC treatment. This study aimed to evaluate the utility of quantitative parameters derived from DKI, CTRW, and FROC models for the early assessment of parotid gland damage following intensity-modulated radiotherapy (IMRT) in NPC. The findings may provide theoretical support for early identification of parotid injury and timely therapeutic intervention.
METHODS: A total of 54 patients with ne

Radiation-induced heart damage (RIHD) remains a prevalent and clinically challenging late complication of thoracic radiotherapy, for which effective minimally invasive restorative interventions remain unavailable. Injectable hydrogels are attractive for myocardial repair because they can be delivered through minimally invasive procedures, undergo in situ gelation, and provide a tunable microenvironment that supports tissue regeneration. In this work, we designed an injectable, thermosensitive, and electrically conductive hydrogel composed of chitosan (CS), dextran (DEX), and polypyrrole (PPy).

Radiation-induced lung injury (RILI) is a common complication associated with radiotherapy for thoracic malignancies, and its etiology and pathogenesis remain poorly understood. This study aims to elucidate the role and its underling mechanism of heme oxygenase-1 (HO-1) in the progression of RILI. Our findings revealed that HO-1 levels were significantly elevated in the serum of RILI patients, as well as in lung tissues and macrophages of RILI mice. Meanwhile, by deleting lung macrophages with chlodronate liposomes, we demonstrated the critical involvement of macrophages in RILI development. T

BACKGROUND: Therapeutic ionizing radiation is a standard treatment modality for thoracic cancers that may affect surrounding normal tissues, including the cardiovascular system. The increasing recognition of mitochondrial health in cellular responses to oxidative stress, particularly in ionizing radiation-induced cardiac implications, highlights mitochondria as a critical target for therapeutic interventions. Optimizing mitochondrial function to attenuate ionizing radiation-induced cardiac pathology necessitates balancing killing cancer cells with sparing nonmalignant tissues.
METHODS: Using

OBJECTIVE: Radiation-induced lung injury (RILI) is a serious complication of radiotherapy, and the role of IL-17A in this process is not well understood. While IL-17A has been shown to modulate autophagy, conflicting reports exist regarding its activation or inhibition of autophagy. This study investigates the role of IL-17A in RILI and its effects on autophagy via the PP2A-mTOR pathway, with a focus on the PP2A B56α subunit.
METHODS: C57BL/6J mice and human lung epithelial cells (BEAS-2B) were exposed to radiation with or without recombinant IL-17A. Autophagy markers were analyzed using West
