Bletilla striata polysaccharide-mediated trained immunity drives the hematopoietic progenitors' expansion and myelopoiesis.

Trained immunity represents a functional state of the innate immune response, characterized by enduring epigenetic reprogramming of innate immune cells. This phenomenon facilitates a sustained and advantageous reaction of myeloid cells to subsequent challenges. Bletilla striata polysaccharide (BSP) is the primary active component of Bletilla striata, mainly consisting of mannose and glucose in its chemical structure. Previous studies have demonstrated BSP's immunomodulatory properties, highlighting its effectiveness in enhancing the immune response. In the present study, we demonstrated that B

Beyond amyloid and tau: rethinking Alzheimer's disease through less explored avenues.

Neurodegenerative diseases, particularly Alzheimer's disease (AD), pose a significant challenge in ageing populations. Our current understanding indicates that the onset of toxic amyloid and tau protein pathologies initiates disease progression. However, existing treatments targeting these hallmark symptoms offer symptomatic relief without halting disease advancement. This review offers an alternative perspective on AD, centring on impaired adult hippocampal neurogenesis (AHN) as a potential early aetiological factor. By delving into the intricate molecular events during the initial stages of

Expressions of Notch signalling pathway members during early pregnancy in mice.

Although pregnancy is initiated and maintained through highly complex mechanisms, it is essential to understand the events that occur before and during early pregnancy to understand a healthy implantation process. The Notch signal, thought to be involved in this process, is frequently the subject of research with its different aspects. To better understand the role of Notch signaling in the peri-implantation period of the mouse uterus, we investigated the state of expression and localization of Notch 3, Notch 4, Rbp-J, Hes1, Hes7, Hey2, HeyL, and Fbw7 in the uterus and implantation sites in ea

Azithromycin induces neurotoxicity in zebrafish by interfering with the VEGF/Notch signaling pathway.

Azithromycin (AZM) is a widely used antibiotic in both human and veterinary medicine, and its use has significantly increased during the COVID-19 pandemic. However, potential adverse effects of AZM on aquatic organisms have not been well studied. In this study, we explored the neurotoxicity of AZM in zebrafish and delved into its underlying mechanisms. Our results showed that AZM exposure resulted in a spectrum of detrimental effects in zebrafish, encompassing abnormal behaviors, damaged neuronal development, aberrant lateral line nervous system development, vascular malformations and perturbe

Gamma Secretase Inhibitors as Potential Therapeutic Targets for Notch Signaling in Uterine Leiomyosarcoma.

Uterine leiomyosarcoma (uLMS) is a rare and aggressive cancer with few effective therapeutics. The Notch signaling pathway is evolutionarily conserved with oncogenic properties, but it has not been well studied in uLMS. The purpose of our study was to determine expression of Notch family genes and proteins and to investigate the therapeutic effect of γ-secretase inhibitors (GSIs), indirect inhibitors of Notch signaling, in uLMS. We determined expression of Notch genes and proteins in benign uterine smooth muscle tissue, fibroids, and uLMS samples by immunostaining and in two uLMS cell lines, S

Imaging of Cancer γ-Secretase Activity Using an Inhibitor-Based PET Probe.

PURPOSE: Abnormal Notch signaling promotes cancer cell growth and tumor progression in various cancers. Targeting γ-secretase, a pivotal regulator in the Notch pathway, has yielded numerous γ-secretase inhibitors (GSIs) for clinical investigation in the last 2 decades. However, GSIs have demonstrated minimal success in clinical trials in part due to the lack of specific and precise tools to assess γ-secretase activity and its inhibition in vivo. EXPERIMENTAL DESIGN: We designed an imaging probe based on GSI Semagacestat structure and synthesized the radioiodine-labeled analogues [131I]- or [1

Therapeutic Potential of Wnt and Notch Signaling and Epigenetic Regulation in Mammalian Sensory Hair Cell Regeneration.

Hearing loss is one of the most prevalent sensory deficits worldwide and can result from the death of mechanosensory hair cells that transduce auditory signals in the cochlea. The mammalian cochlea lacks the capacity to regenerate these hair cells once damaged, and currently there are no biological therapies for hearing loss. Understanding the signaling pathways responsible for hair cell development can inform regenerative strategies and identify targets for treating hearing loss. The canonical Wnt and Notch pathways are critical for cochlear development; they converge on several key molecules

A systematic review and critical evaluation of reported pathogenic sequence variants in hidradenitis suppurativa.

Hidradenitis suppurativa (HS) is a severe chronic inflammatory disorder characterized by recurrent painful deep-seated nodules with a predilection to the apocrine-bearing areas of skin. A minority of cases of HS are due to mutations in the γ-secretase complex. Contention exists surrounding the pathogenicity of sequence variants and their effects upon Notch signalling. This systematic review was registered with PROSPERO (CRD42016041425) and was conducted in line with the PRISMA statement. Eligibility criteria for this review included published case reports, case series and reviews that identifi

A "latent niche" mechanism for tumor initiation.

Stem cells, their niches, and their relationship to cancer are under intense investigation. Because tumors and metastases acquire self-renewing capacity, mechanisms for their establishment may involve cell-cell interactions similar to those between stem cells and stem cell niches. On the basis of our studies in Caenorhabditis elegans, we introduce the concept of a "latent niche" as a differentiated cell type that does not normally contact stem cells nor act as a niche but that can, under certain conditions, promote the ectopic self-renewal, proliferation, or survival of competent cells that it

Notch-responsive cells initiate the secondary transition in larval zebrafish pancreas.

Zebrafish provide a highly versatile model in which to study vertebrate development. Many recent studies have elucidated early events in the organogenesis of the zebrafish pancreas; however, several aspects of early endocrine pancreas formation in the zebrafish are not homologous to the mammalian system. To better identify mechanisms of islet formation in the zebrafish, with true homology to those observed in mammals, we have temporally and spatially characterized zebrafish secondary islet formation. As is the case in the mouse, we show that Notch inhibition leads to precocious differentiation

PLC-IP3 signaling promotes sensory hair cell formation in zebrafish neuromast.

Sensorineural hearing loss primarily results from irreversible damage to sensory hair cells (HCs), and regenerative strategies remain limited by inefficient HC formation and incomplete functional maturation of newly generated cells. Here, using pharmacological perturbation, single-cell transcriptomics sequencing, and genetic analysis in zebrafish neuromasts, we identify phosphoinositide-specific phospholipase C (PLC) signaling as an intracellular regulator of HC development and maturation. Activation of PLC signaling increases the number of HCs and FM4-64 labeling, whereas inhibition reduces H

Modeling Disease-Relevant Bile Duct Morphogenesis Defects in a Tunable In Vitro System.

Biliary tubulogenesis is a highly coordinated process essential for liver function, yet its mechanisms remain difficult to study due to the inaccessibility of developing ducts in vivo and the stochastic nature of conventional three-dimensional (3D) models. Here, we present a micropattern-based in vitro platform that constrains cholangiocyte growth to defined geometries, enabling reproducible formation of tubular structures with controlled folding, lumen formation, and branching. Using this system, we demonstrate that cholangiocytes carrying a Jag1 mutation (Jag1Ndr/Ndr cells) or subjected to p

Prenatal nicotine exposure induces epigenetic alterations in the Notch signaling genes in the proximal colon in rats.

BACKGROUND: Prenatal nicotine exposure (PNE) has been shown to significantly impact fetal development, leading to various pathologies in adulthood. OBJECTIVES: In this study, we analyzed the epigenetic effects of PNE on the proximal colon. RESULTS: We show here that PNE decreases the length of the colon crypts and induces a higher Ki-67 index. The genes related to metabolic pathways, tight junctions, and neurodegenerative processes were downregulated in PNE colons, and genes related to inflammation and oxygen transporter activity were upregulated. We observed a decrease in acetylated H4Ac and

Maternal Alkaline-Ionized Mineral Complex Supplementation Regulates Intestinal Epithelial Development and Promotes Glucose and Lipid Metabolism Through the Wnt/Notch Pathway in Piglets.

Breast milk is the sole nutritional source for infants during lactation, but the immaturity of infants' intestinal function elevates the risk of infant malnutrition. As a nutritional additive, maternal Alkaline Ionized Mineral Complex (AIMC) has great application potential in livestock production. This study investigated maternal AIMC supplementation on offspring glucose and lipid absorption using a piglet model. Thirty sows at 107 days of gestation were randomly divided into the Con group (basal diet) or the AIMC group (basal diet + 30 g/head/day AIMC) and fed until piglets were weaned (total

Exploring the Dual Roles of Neural Stem Cells in Glioblastoma: Therapeutic Implications and Opportunities.

Glioblastoma (GBM) is recognized as the most aggressive and lethal form of primary brain tumor, characterized by rapid proliferation and significant resistance to conventional therapies. Recent studies have illuminated the complex role of Neural Stem Cells (NSCs) in both the progression and treatment of GBM. This review examines the specific molecular pathways influenced by NSCs, focusing on critical signaling cascades such as Notch, P13K, and SHH, which are implicated in tumor development and maintenance. Furthermore, we explore the dual role of NSCs in glioblastoma, where they can act as bot

NOTCH signaling orchestrates the inflammatory-fibrotic continuum of macrophages in renal allograft rejection.

BACKGROUND: Chronic rejection is a major cause of long-term kidney allograft failure, characterized by persistent inflammation and progressive fibrosis. Macrophages are central mediators of this process, but their phenotypic heterogeneity and regulatory mechanisms in chronic rejection remain incompletely understood. METHODS: We performed single-cell transcriptomic analysis on renal allograft biopsies from patients with different types of rejection and on a time-course rat model of chronic rejection. Macrophage subsets were identified through transcriptional profiling and Pseudotime trajectory

Airway basal stem cells are necessary for the maintenance of functional intraepithelial airway macrophages.

Stem cells are known to provide signals that contribute to the maintenance and function of neighboring cells. We demonstrate that Notch signaling arising from airway basal stem cells is necessary for the function of a unique population of intraepithelial airway macrophages (IAMs) in the murine trachea. Without this stem cell signaling, IAMs lose MHC II expression, which in turn prevents antigen-induced allergic inflammation. Distal murine airways do not harbor basal stem cells, and, in this region of the lung, allergic inflammation proceeds unperturbed. We speculate that the functional couplin

Deficiency of DDX3X results in neurogenesis defects and abnormal behaviors via dysfunction of the Notch signaling.

The molecular mechanisms underlying the neurodevelopmental disorders (NDDs) caused by DDX3X variants remain poorly understood. In this study, we validated that de novo DDX3X variants are enriched in female developmental delay (DD) patients and mainly affect the evolutionarily conserved amino acids based on a meta-analysis of 46,612 NDD trios. We generated a ddx3x deficient zebrafish allele, which exhibited reduced survival rate, DD, microcephaly, adaptation defects, anxiolytic behaviors, social interaction deficits, and impaired spatial recognitive memory. As revealed by single-nucleus RNA seq

Identification of KCNQ1 as a diagnostic biomarker related to endoplasmic reticulum stress for intervertebral disc degeneration based on machine learning and experimental evidence.

Intervertebral disc degeneration (IDD) is a primary cause of low back pain and disability. Cellular senescence and apoptosis due to endoplasmic reticulum stress (ERS) are key in IDD pathology. Identifying biomarkers linked to ERS in IDD is crucial for diagnosis and treatment. We utilized machine learning on gene expression profiles from the Gene Expression Omnibus database to discover biomarkers associated with ERS in IDD. Gene set enrichment analysis (GSEA) and single-sample GSEA were applied to evaluate the immunological features and biological functions of these biomarkers. The expression o

Inaccessible LCG Promoters Act as Safeguards to Restrict T Cell Development to Appropriate Notch Signaling Environments.

T cell development is restricted to the thymus and is dependent on high levels of Notch signaling induced within the thymic microenvironment. To understand Notch function in thymic restriction, we investigated the basis for target gene selectivity in response to quantitative differences in Notch signal strength, focusing on the chromatin architecture of genes essential for T cell differentiation. We find that high Notch signal strength is required to activate promoters of known targets essential for T cell commitment, including Il2ra, Cd3ε, and Rag1, which feature low CpG content (LCG) and DNA
