Diabetes-induced vascular dysfunction in the retina: role of endothelins.

AIMS/HYPOTHESIS: Vasoactive factors like endothelins, by virtue of the microvascular regulation as well as by other effects, possibly play important parts in the pathogenesis of diabetic retinal microangiopathy. We investigated retinal vascular dysfunction in streptozotocin-induced diabetes and its relation with endothelins in short- and long-term diabetes. METHODS: Diabetic rats with or without an endothelin receptor antagonist (bosentan) treatment were investigated after 1 month and 6 months of follow-up. Retinal blood flow was measured and compared with age- and sex-matched non-diabetic co…

Circadian expression of tryptophan hydroxylase mRNA in the chicken retina.

Many aspects of retinal physiology are controlled by a circadian clock located within the eye. This clock controls the rhythmic synthesis of melatonin, which results in elevated levels during the night and low levels during the day. The rate-limiting enzyme in melatonin biosynthesis in retina appears to be tryptophan hydroxylase (TPH)[G.M. Cahill and J.C. Besharse, Circadian regulation of melatonin in the retina of Xenopus laevis: Limitation by serotonin availability, J. Neurochem. 54 (1990) 716-719]. In this report, we found that TPH mRNA is strongly expressed in the photoreceptor layer and t…

The pineal organ as a folded retina: immunocytochemical localization of opsins.

The most simple pineal complex (the pineal and parapineal organs of lampreys), consists of saccular evaginations of the diencephalic roof, and has a retina-like structure containing photoreceptor cells and secondary neurons. In more differentiated vertebrates, the successive folding of the pineal wall multiplies the cells and reduces the lumen of the organ, but the pattern of the histological organization remains similar to that of lampreys; therefore, we consider the histological structure of the pineal organ of higher vertebrates as a 'folded retina'. The cell membrane of several pineal phot…

The retinal pigment epithelium as a developmental regulator of the neural retina.

Melanin-related agents regulate the development of the mammalian neural retina, because in albinos there are a range of retinal deficits including abnormal connections between the eye and brain, an underdeveloped central retina and a rod deficit. These deficits may arise because gradients of retinal development in the albino are delayed and the retina is abnormally proliferative, but also goes through a subsequent period of excessive cell death. This may be caused by a reduction in ocular DOPA in albinos as this is in the synthetic pathway of melanin and is a known cell cycle regulator.

Identification of the bcl-2 family of genes in the rat retina.

PURPOSE: Retinal ganglion cells die by apoptosis after axotomy, and this process may reflect altered expression of cell-death genes. Several of these genes, including bcl-2, bcl-x, and bax, share homology at the amino acid level in the BH1 and BH2 domains, through which they also interact. To understand their role in the neuronal response to axotomy, the authors studied their expression in the adult rat retina and after optic nerve crush. METHODS: An initial survey was conducted with reverse transcription-polymerase chain reaction (RT-PCR), using oligonucleotides against identified members of…

Neuropeptide Y in the forebrain and retina of the killifish, Fundulus heteroclitus.

The organization of the neuropeptide Y (NPY)-immunoreactive system in the forebrain, pineal organ and retina of a biweekly spawning fish (Fundulus heteroclitus) was investigated. Immunoreactivity was encountered in neurons of the nucleus olfactoretinalis, in the large population of neurons in the floor of the telencephalon, and in the nucleus entopeduncularis. Isolated somata were encountered in the hypophysiotropic hypothalamic nuclei, viz., the nucleus preopticus periventricularis, nucleus preopticus, and nucleus lateralis tuberis. Immunoreactive somata were also seen in the nucleus dorsomed…

Nonteratoid medulloepithelioma of the retina with electron microscopic and immunohistochemical characterization.

Medulloepitheliomas are rare intraocular tumors, predominant in children, and originate mainly from undifferentiated nonpigmented epithelium of the ciliary body. These tumors rarely involve the optic nerve or the retina. They are classified as nonteratoid and teratoid types; the latter contains heterologous tissues. The teratoid variant of medulloepitheliomas involving the optic nerve or the retina is reported in four patients only. We describe the first case of a benign nonteratoid medulloepithelioma of the retina in a 3 1/2-year-old girl with immunohistochemical and electron microscopic char…

A reliable method for Golgi staining of retina and brain slices.

Although the classical Golgi method is a powerful means for structural analysis of the brain, it is generally considered to be an unpredictable technique making anatomists wary of using it. Often, even when successful staining has occurred, deposits of silver chromate crystals on the surface of the tissue obscure examination. This paper describes a simple procedure for Golgi impregnation of retina and brain slices so that good, even staining is obtained and crystal formation is avoided. The most outstanding feature of the method is the consistency of results. This consistency is due to two fac…

The relative distribution of retinal and choroidal blood in the human retina.

In this paper a model is presented to explain the relative distribution of retinal and choroidal blood to the human retina. According to this model the retina between the inner limiting membrane and the outer limiting membrane is metabolically controlled and nourished by the retinal vasculature, through Muller cells. The retina between the outer limiting membrane and Bruch's membrane is nourished by the choroidal vasculature, through the retinal pigment epithelium. Muller cells have the ability of metabolic uptake from the interphotoreceptor space as demonstrated in pathological situations suc…

Immunocytochemical demonstration of taurine in the retina and pineal organ of the pigeon.

Taurine-like immunoreactivity (TLI) is found in the pigeon retina at high levels within cones, in select cells of the inner nuclear layer and in sublaminae of the inner plexiform layer. At the ultrastructural level a high density of immuno-gold particles in visualized in the ellipsoid, but not in the paraboloid area of the inner segment. It is present in perikarya and the presynaptic endings of cones, but not in postsynaptic cell processes. The immuno-gold particles are not associated directly with the synaptic vesicles. In the pineal organ TLI was confined to the so-called dark pinealocytes w…

[Color campimetry in the diagnosis of diseases of the retina and optic nerve].

Color campimetry was realized with the use of an IBM-compatible computer display with a program permitting the registration of brightness sensitivity thresholds and the time of sensorimotor reaction to visual stimuli offered on the display in each point of the visual field. Investigation of light and color sensitivity thresholds and of the time of sensorimotor reaction to stimuli of different spectral composition in normal subjects helped study the functional topography of the retina by the time of sensorimotor reaction and by the brightness sensitivity thresholds. The methods of equalization …

Gyrate atrophy of the choroid and retina with hyperornithinaemia, cystinuria and lysinuria.

The case of a 45-year-old woman with gyrate atrophy of the choroid and retina is documented. Additional features in this case, to the authors' knowledge not previously described in gyrate atrophy, are massive cystinuria, massive lysinuria, axial hypermetropia and diabetes. Gyrate atrophy is a rare autosomal recessive degenerative disease of the choroid and retina and is accompanied by defective ornithine metabolism. Simell and Takki demonstrated the association with hyperornithinaemia in 1973. The main metabolic features are those of hyperornithinaemia and ornithuria caused by a deficiency of …

von Hippel-Lindau disease (angiomatosis of the retina and cerebellum).

von Hippel-Lindau disease is a multi-system disorder characterized by hemangioblastic lesions of the central nervous system and visceral organs. Angiomatosis retinae is often the first observable manifestation of von Hippel-Lindau disease. Optometry may be the initial point of entry into the health care system for the von Hippel-Lindau patient. As such, the eye care practitioner should be aware of the diffuse nature of the disease and initiate the search for silent, possibly lethal, lesions associated with the disease. Management may involve consultation with a neurosurgeon, internist, and ret…

Long-term natural history of lattice degeneration of the retina.

An initial series of patients with lattice degeneration was reported to the Academy in 1964 and a follow-up report given in 1973. A continuing prospective study of 276 consecutive untreated patients (423 eyes) is now reported with follow-up from 1 to 25 years (average, 10.8 years). Clinical retinal detachments (RDs) occurred in 3 (1.08%) of 276 patients and 0.7% of eyes. Tractional retinal tears were seen in eight (2.9%) patients and 1.9% of eyes; one of these led to a clinical RD. Clinical or progressive subclinical RD occurred in 3 (2%) of eyes with atrophic holes. Subclinical RD was seen in…

Ultrastructural studies of the retina in infantile neuronal ceroid-lipofuscinosis.

A 9-year-old boy who had died of infantile neuronal ceroid-lipofuscinosis had experienced retina-derived visual failure. Ophthalmologically and morphologically, his retina was severely atrophic and scarred by a dense fibrillary gliosis while photoreceptor cells had completely disappeared, cells of the bipolar layer had decreased in number and had become atrophic beyond cytologic recognition. Retinal pigment epithelial cells had undergone either atrophy or proliferation. Disease-specific granular lipopigments had accumulated in perikarya and processes of remaining cells and were infrequently as…

Interactions between the rod and the cone pathways in the cat retina.

The receptive field centers of 21 on-center X retinal ganglion cells in cat were tested with stimuli designed to detect nonlinear interactions between the rod and the cone systems. One red and one green stimulus light were always present, at a level such that modulation of the red light essentially affected only cones, and that of the green light only rods. The two lights could be superimposed spatially (overlapped configuration) or fall on separate subareas of the receptive field center (nonoverlapped configuration). In most cases, there was less complete summation of the responses to modulat…

The inadvertent injection of gas beneath the retina in a pseudophakic eye.

Gas injected through the pupil to tamponade a detached retina in a pseudophakic eye inadvertently made its way beneath the retina and augmented the detachment. A transparent membrane posterior to an iris-supported lens prevented the gas from entering the vitreous and diverted it laterally behind the iris, to the ciliary body, where is dissected between layers of ciliary epithelium and entered the subretinal space.
