Fetal-Maternal Hemorrhage Detected by Sudden Disappearance of Rh Immune Globulin-Related Anti-D.

BACKGROUND: Fetal-maternal hemorrhage is usually spontaneous and goes undetected but can be associated with adverse perinatal outcomes. CASE: We describe the detection of a fetal-maternal hemorrhage by abrupt disappearance of prophylactic anti-D on antibody screen in an Rh-negative mother with dichorionic twins admitted for atrial flutter of one twin. Both rosette and Kleihauer-Betke tests were positive. The diagnosis was confirmed by anemia in one twin at birth. CONCLUSION: Fetal-maternal hemorrhage requires a high index of suspicion for diagnosis. An unexpected sudden decline in Rh immune

Independent brain cortical signatures of risk for adolescent cannabis use and consequences of such use are moderated by sex.

There is an accumulation of evidence linking adolescent cannabis use with variations in brain structure and function, however it remains poorly understood whether cannabis-associated variations in brain structure represent pre-existing risk factors or consequences of cannabis use. We investigated whether cannabis use propensity and within-person variations of cannabis use were associated with cortical thickness during adolescence. Adolescents (n = 136, 74 female) completed three neuroimaging sessions and annual assessments from 12 until 17 years of age (with 90% follow-up). Cannabis use was di

Cardiac fibrosis and heart failure: Metabolic stress, circadian dysregulation, and extracellular matrix remodeling.

Diabetes is a major risk factor for heart failure and drives a distinct cardiomyopathic phenotype characterized by oxidative stress, metabolic inflexibility, mitochondrial dysfunction, and progressive myocardial remodeling. Central to this process is cardiac fibrosis, marked by excessive extracellular matrix deposition, increased myocardial stiffness, and impaired mechanotransduction. Emerging evidence identifies circadian rhythm disruption as an under recognized modifier of diabetic cardiac fibrosis. The circadian clock regulates daily oscillations in myocardial metabolism, redox balance, inf

The Application of Rho Kinase Inhibitors in the Management of Glaucoma.

Glaucoma is a chronic neurodegenerative disease that poses a significant threat of irreversible blindness worldwide. Current treatments for glaucoma focus on reducing intraocular pressure (IOP), which is the only modifiable risk factor. Traditional anti-glaucomatous agents, including carbonic anhydrase inhibitors, beta-blockers, alpha-2 agonists, and prostaglandin analogs, work by either improving uveoscleral outflow or reducing aqueous humor production. Rho kinase (ROCK) inhibitors represent a novel class of anti-glaucomatous drugs that have emerged from bench to bedside in the past decade, o

Inflammation modulates intercellular adhesion and mechanotransduction in human epidermis via ROCK2.

Aberrant mechanotransduction and compromised epithelial barrier function are associated with numerous human pathologies including inflammatory skin disorders. However, the cytoskeletal mechanisms regulating inflammatory responses in the epidermis are not well understood. Here we addressed this question by inducing a psoriatic phenotype in human keratinocytes and reconstructed human epidermis using a cytokine stimulation model. We show that the inflammation upregulates the Rho-myosin II pathway and destabilizes adherens junctions (AJs) promoting YAP nuclear entry. The integrity of cell-cell adh

The serotonin hypothesis of pulmonary hypertension revisited.

The serotonin hypothesis of pulmonary arterial hypertension (PAH) arose after an outbreak of PAH in patients taking the anorexigenic drugs aminorex and dexfenfluramine. Both of these drugs are serotonin transporter (SERT) substrates and indirect serotinergic agonists. There is now a wealth of evidence to support a role for serotonin in the pathobiology of PAH. Synthesis of serotonin can occur in pulmonary artery endothelial cells by the enzyme tryptophan hydroxylase 1 (TPH1). Serotonin then acts at the 5-HT(1B) receptor and the SERT to mediate constriction and proliferation of pulmonary artery

Genetically encoded photoswitching of actin assembly through the Cdc42-WASP-Arp2/3 complex pathway.

General methods to engineer genetically encoded, reversible, light-mediated control over protein function would be useful in many areas of biomedical research and technology. We describe a system that yields such photo-control over actin assembly. We fused the Rho family GTPase Cdc42 in its GDP-bound form to the photosensory domain of phytochrome B (PhyB) and fused the Cdc42 effector, the Wiskott-Aldrich Syndrome Protein (WASP), to the light-dependent PhyB-binding domain of phytochrome interacting factor 3 (Pif3). Upon red light illumination, the fusion proteins bind each other, activating WAS

N-WASP activation by a beta1-integrin-dependent mechanism supports PI3K-independent chemotaxis stimulated by urokinase-type plasminogen activator.

Urokinase-type plasminogen activator (uPA)-uPA receptor (uPAR) and epidermal growth factor (EGF)-EGF receptor (EGFR) expression is highly correlated with breast cancer metastasis. Phosphoinositide 3-kinase (PI3K), small Rho GTPases, such as Cdc42 and Rac1, and neuronal Wiskott Aldrich syndrome protein (N-WASP) are key effectors that regulate dynamic changes in the actin cytoskeleton and cell migration. uPA- and EGF-stimulated chemotaxis, cytoskeletal rearrangements and activation of Cdc42, Rac1 and N-WASP were studied in the highly metastatic human breast cancer cell line MDA MB 231. Quantification of Lewy body pathology by cerebrospinal fluid endpoint dilution RT-QuIC in a neuropathological autopsy cohort of clinically heterogeneous participants.

The identification of biomarkers predicting the burden of brain alpha-synuclein (α-syn) pathology in vivo represents a research priority in Lewy body disease (LBD). Recently, some kinetic parameters of seed amplification assays (SAAs) for α-syn showed associations with measures of clinical progression. However, preanalytical and analytical factors significantly affect these parameters, reducing reproducibility. The Endpoint Dilution (ED) SAA Real-time Quaking-induced Conversion (RT-QuIC) is emerging as an alternative, more accurate tool for seed quantification. Still, the approach needs valida

Reproducibility of quantitative cervical strain elastography in nonpregnant patients and the effect of vaginal misoprostol on measured parameters.

Quantitative strain elastography (SE), referred to as E-Cervix (Samsung Medison), is a novel method for assessing parameters that indirectly describe the elasticity of the cervix. This evaluation yields quantitative parameters based on the analysis of the elastographic map image, which represents a new approach in the context of SE. In our study, we aimed to examine the reproducibility of this method in evaluating the cervix of non-pregnant women, the impact of basic demographic parameters of the patient on the results, and the responsiveness of the method, i.e., the assessment of changes occu

Sex differences in the coherence of PCL5 and CAPS-5 assessments for post-traumatic stress disorder.

The clinician-administered Post Traumatic Stress Disorder (PTSD) scale (CAPS-5) is a clinical tool for the diagnosis and assessment of the severity of PTSD symptoms based on DSM-5, while the PTSD Checklist for DSM-5 (PCL-5) is a self-assessment tool. Both assessments are widely used, but CAPS-5 is time and resource intensive, while PCL-5 is quicker. Although PCL-5 has been validated for PTSD symptom severity assessment, PCL-5 scores tend to show higher symptom severity and sometimes leading to higher prevalence of PTSD diagnosis. The contributing factors for such discrepancies in reporting are

Unprecedented Surge of Melioidosis after Typhoon Events in Taiwan 2024.

Melioidosis, caused by Burkholderia pseudomallei, is a potentially fatal bacterial infection endemic to Southeast Asia and northern Australia. The disease often resurges after extreme weather events, such as typhoons. In this study, we investigated the epidemiological trends of melioidosis in Taiwan from 2015 to 2024, focusing on the impact of typhoon-associated precipitation. A total of 396 confirmed cases were reported over the decade, with 123 cases (31.1%) occurring in 2024 alone. Statistical analysis revealed a weak but significant correlation between precipitation and melioidosis inciden

Tyrosine-modifying glycosylation by Yersinia effectors.

Mono-O-glycosylation of target proteins by bacterial toxins or effector proteins is a well-known mechanism by which bacteria interfere with essential functions of host cells. The respective glycosyltransferases are important virulence factors such as the Clostridioides difficile toxins A and B. Here, we describe two glycosyltransferases of Yersinia species that have a high sequence identity: YeGT from the zoonotic pathogen Yersinia enterocolitica and YkGT from the murine pathogen Yersinia kristensenii. We show that both modify Rho family proteins by attachment of GlcNAc at tyrosine residues (T

Hands train the brain-what is the role of hand tremor and anxiety in undergraduate microsurgical skills?

INTRODUCTION: Physiological hand tremor occurs naturally, due to oscillations of the upper extremities. Tremor can be exacerbated by stress and anxiety, interfering with fine motor tasks and potentially impact on surgical performance, particularly in microsurgery. We investigated the link between tremor, anxiety and performance in a neurosurgical module as part of an international surgical course. METHODS: Essential Skills in the Management of Surgical Cases (ESMSC) course recruits medical students from European Union (EU) medical schools. Students are asked to suture the dura mater in an ex

Gastric and pyloric sphincter muscle function and the developmental-dependent regulation of gastric content emptying in the rat.

Feeding intolerance is a common issue in the care of preterm neonates. The condition manifests as delayed emptying of gastric contents and represents a therapeutic challenge, since the factors accounting for its manifestations are unknown. The main goal of this study was to comparatively investigate the age-related function of rat gastric and pyloric smooth muscle and their putative regulators. We hypothesized that a reduced gastric muscle contraction potential early in life contributes to the delayed gastric emptying of the newborn. Newborn and adult rat gastric (fundus) and pyloric sphincter

D-dimer, FVIII and thrombotic burden in the acute phase of deep vein thrombosis in relation to the risk of post-thrombotic syndrome.

BACKGROUND: Post-thrombotic syndrome (PTS) is the most common complication of deep vein thrombosis (DVT), but few data are available on the risk factors for PTS. AIMS: To assess whether the time-course of D-dimer, FVIII, and thrombotic burden are related to PTS development. METHODS: Patients (n=59) with proximal DVT of the lower limbs (age 64; range:20-88years; male 56%) were enrolled on the day of diagnosis (D0) and all received heparin for 5-7days, overlapped and followed by vitamin K antagonists (VKA) for 3months. Whole-leg compression ultrasound examination was conducted on D0 and 7 (D7)

ROCK-regulated cytoskeletal dynamics participate in the inhibitory effect of melatonin on cancer cell migration.

Cell movement is generated by a driving force provided by dynamic cytoskeletal organization. Two main cytoskeletal-dependent features, essential for migration, are the highly cell polarized structure and focal adhesion complexes. Cell migration and substrate anchorage are finely regulated by external signaling exerted by growth factors and hormones. In particular, the serine threonine kinase activated by the small GTPase Rho, the Rho-associated protein kinase (ROCK), participate in both processes through regulation of actin rearrangements in lamellipodia, filopodia, ruffles, and stress fibers.

DOCK10-mediated Cdc42 activation is necessary for amoeboid invasion of melanoma cells.

BACKGROUND: Tumor cells can move in a three-dimensional (3D) environment in either mesenchymal-type or amoeboid modes. In mesenchymal-type movement, cells have an elongated morphology with Rac-induced protrusions at the leading edge. Amoeboid cells have high levels of actomyosin contractility, and movement is associated with deformation of the cell body through the matrix without proteolysis. Because signaling pathways that control the activation of GTPases for amoeboid movement are poorly understood, we sought to identify regulators of amoeboid movement by screening an siRNA library targeting
