The neurotoxicity of pesticides: Implications for Parkinson's disease.

Parkinson's disease (PD) is the fastest-growing neurodegenerative disorder worldwide, and no effective cure is currently available. Neuropat … nt contributor. While the effects of many are still uncharacterized, it has already been shown that rotenone, paraquat, maneb, and dieldrin affect critical cellular pathways, including mitochondrial and proteasomal dysfunction, aSyn aggregation, autophagy dysregulation, and disruption of dopamine metabolism. With the constant rise in pesticide usage to mee …

Computational classification of mitochondrial shapes reflects stress and redox state.

Dynamic variations in mitochondrial shape have been related to function. However, tools to automatically classify and enumerate mitochondrial shapes are lacking, as are systematic studies exploring the relationship of such shapes to mitochondrial stress. Here we show that during increased generation of mitochondrial reactive oxygen species (mtROS), mitochondria change their shape from tubular to donut or blob forms, which can be computationally quantified. Imaging of cells treated with rotenone or antimycin, showed time and dose-dependent conversion of tubular forms to donut-shaped mitochondri…

A mitochondrial encephalomyopathy with cardiomyopathy. A case revealing a defect of complex I in the respiratory chain.

We describe a 16-year-old Japanese girl with a mitochondrial encephalomyopathy who presented with progressive dementia, limb weakness and at … re impairment of oxidation of NADH-linked substrates in contrast to normal succinate oxidation. The rotenone-sensitive NADH-coenzyme Q reductase activity was markedly decreased in heart, skeletal muscle and liver mitochondria. The biochemical investigations have led to the identification of a defect of complex I in the respiratory chain. Reported cases of …

Respiratory effects and ascorbate reactions with misonidazole and other recently developed drugs.

A number of nitro compounds behave as electron acceptors with ascorbate in a non-enzyme-catalyzed reaction and with NADPH as electron donor … nced by protein and drug concentrations. Various inhibitors of electron transfer reactions, such as rotenone, antimycin A, and sodium azide, had no effect on misonidazole-stimulated oxygen utilization. However, cyanide was found to be stimulating in this system and this may be due to the inhibition of enzymes such as catalase and superoxide dismutase known …

Development and evaluation of S-carboxymethyl-L-cystine-loaded solid lipid nanoparticles for Parkinson's disease in murine and zebrafish models.

Parkinson's disease (PD) is an advanced neurodegenerative condition distinguished by the rapid decline of dopamine neurons in the midbrain, leading to an imbalance in dopamine and acetylcholine levels, precipitating associated symptoms. The main objective of this work was to fabricate solid lipid nanoparticles (SLNs) loaded with S-carboxymethyl-L-cystine (SC) for enhanced delivery to the brain. This study examines the impact of these SLNs on rotenone (RT) caused Parkinson's disease (PD) in both rat and zebrafish models. The process of loading SC into SLNs was achieved through the solvent evapo…

Chemical Characterization and Beneficial Effects of Walnut Oil on a Drosophila melanogaster Model of Parkinson's Disease.

A nutritional approach could be a promising strategy to prevent or decrease the progression of neurodegenerative disorders such as Parkinson's disease (PD). The neuroprotective role of walnut oil (WO) was investigated in Drosophila melanogaster treated with rotenone (Rot), as a PD model, WO, or their combination, and compared to controls. WO reduced mortality and improved locomotor activity impairment after 3 and 7 days, induced by Rot. LC-MS analyses of fatty acid levels in Drosophila heads showed a significant increase in linolenic (ALA) and linoleic acid (LA) both in flies fed with the WO-e…

Effect of thyroid state on H2O2 production by rat liver mitochondria.

It has been suggested that activation of mitochondrial respiration by thyroid hormone results in oxidative tissue injury secondary to increased reactive oxygen species production. In order to throw light on this subject, the effects of thyroid state on O2 consumption and H2O2 release by rat liver mitochondria were investigated. Hypothyroidism decreased the rates of O2 consumption and H2O2 release by succinate or pyruvate/malate-supplemented mitochondria during both State 4 and State 3 respiration, whereas hyperthyroidism increased such rates. Conversely, with both substrates and during either

A requirement for reversible binding between aggregating embryonic cells before stable adhesion.

Chick embryonic liver and neural retina cells aggregate in a two-step process. Initially, cells formed a loose association in a step that apparently did not require metabolic energy. Cells bound in this manner were dissociable by mild shear forces or by simple dilution. The results of the dilution experiments suggest a readily reversible binding of single cells to form these types of aggregates. In a second step, which required metabolic energy, the cells became firmly, or stably attached. The formation of both types of bond was temperature-dependent. Kinetic studies indicated that the formati

Midazolam prevents motor neuronal death from oxidative stress attack mediated by JNK-ERK pathway.

Midazolam is a sedative used by patients with mechanical ventilation. However, the potential clinical value is not fully explored. In this report, we made use of a neuroblastoma-spinal cord hybrid motor neuron-like cell line NSC34, and elucidated the potential role of Midazolam on these cells under the insult of oxidative stress. We found the protective effect of Midazolam on motor neurons against cytotoxicity induced by the combination of oligomycin A and rotenone (O/R) or phenylarsine oxide. The characteristics of apoptosis, such as the ratio of TUNEL+ cells or the expression level of cleave…

Mitochondrial genetic control of assembly and function of complex I in mammalian cells.

Sixteen years ago, we demonstrated, by immunological and biochemical approaches, that seven subunits of complex I are encoded in mitochondri … he mtDNA-encoded subunits of complex I by exposing established cell lines to high concentrations of rotenone. Our analyses of these mtDNA mutations affecting subunits of complex I have shown that at least two of these subunits, ND4 and ND6, are essential for the assembly of the enzyme. ND5 appears to be located at the periphery of the enzyme and, while it …

Chloride-hydroxyl exchange in the guinea-pig ventricular myocyte: no role for bicarbonate.

Reduction of extracellular pH (pHo) leads to a fall of intracellular pH (pHi) in the guinea-pig ventricular myocyte. In nominally CO2/HCO3-- … 1 (AM-loaded). Possible sources of CO2 were eliminated by adding the aerobic inhibitors, cyanide or rotenone, and by equilibrating the superfusates flowing over the myocyte with a CO2-free, 100% N2 atmosphere. The fall of pHi upon reducing pHo (to 6.4) persisted after complete CO2-removal. This indicates that, in nominally CO2-free conditions, residual HCO …

Peroxisomal fatty acid beta-oxidation in HepG2 cells.

HepG2 cells, originally derived from a human hepatoblastoma, contain peroxisomes which could be separated from mitochondria and other subcel … epG2 cells. Peroxisomal beta-oxidation was independent of carnitine, insensitive to antimycin A and rotenone, and not blocked by an inhibitor of carnitine palmitoyl transferase I. HepG2 peroxisomes contained immunoreactive acyl-CoA oxidase, the first enzyme unique to the peroxisomal beta-oxidation pathway. In addition, HepG2 peroxisomes contained VLCFA-CoA …

Integrin α5β1 Activation by PHSRN Peptide Elicits Neuroprotection and Functional Recovery in Parkinson's Disease Mice.

Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration driven by oxidative stress, mitochondrial dysfunction, synaptic loss, and impaired neurotrophic signaling; however, the role of integrin α5β1 in neuronal vulnerability remains unclear. Here, the data show that rotenone-induced stress reduces integrin α5 expression in a dose- and time-dependent manner, leading to increased ROS accumulation, glutathione imbalance, synaptic degeneration, senescence-like β-gal activity, and apoptosis, whereas integrin α5 knockdown further exacerbates these deficits, supporting …

Multi-omics analysis reveals the protective role of transcriptional enhancer factor and the pathogenic mechanism of monocytes in Parkinson's disease.

Parkinson's disease (PD) is a multifactorial neurodegenerative disorder whose pathogenic mechanisms remain incompletely elucidated. This stu … emonstrated that TEF overexpression significantly enhanced the resistance of neuroblastoma cells to rotenone-induced damage, inhibited apoptosis, and preserved tyrosine hydroxylase (TH) expression. In vivo, TEF notably improved motor coordination and exploratory behavior in PD mouse models. Collectively, these findings suggest that TEF may exert neuroprote …
