Prevalence and molecular characterizations of Toxoplasma gondii and Babesia microti from small mammals captured in Gyeonggi and Gangwon Provinces, Republic of Korea.

A survey was conducted to determine the distribution and prevalence of Toxoplasma gondii and Babesia microti infections in small mammals captured in Gyeonggi and Gangwon Provinces, Republic of Korea (ROK). The serological prevalence of T. gondii (ELISA) and B. microti (IFAT) was 2.3% (15/667) and 2.1% (14/667), respectively. DNA extracts from small mammal heart tissues were screened by PCR for T. gondii and B. microti targeting regions of the GRA5 gene and the 18S rRNA and β-tubulin genes, respectively. Only 0.17% (1/578) of Apodemus agrarius was positive of T. gondii by PCR, while 0.52% (3/57

Emerging babesiosis in the mid-Atlantic: autochthonous human babesiosis cases and Babesia microti (Piroplasmida: Babesiidae) in Ixodes scapularis (Acari: Ixodidae) and Ixodes keiransi (Acari: Ixodidae) ticks from Delaware, Maryland, Virginia, West Virginia, and the District of Columbia, 2009 to 2024.

The range of Babesia microti (Franca, 1910)-infected ticks is expanding, resulting in locally acquired human babesiosis cases occurring in new areas: Maryland (2009), the District of Columbia (2013), Virginia (2016), and West Virginia (2017). We collected host-seeking ticks from old fields, ecotones, forested habitats and animal hosts in Delaware, Maryland, and Virginia, 2010 to 2024. Ixodes scapularis Say, the tick vector of babesiosis, was captured in all 3 states. PCR revealed B. microti in 2.7% (36/1310) of I. scapularis, with site prevalence ranging from <1% to 12.5% infected. The first B

Conserved regional 3' grouping of rare codons in the coding sequence of ocular prosecretory mitogen lacritin.

PURPOSE: Lacritin is a prosecretory mitogen in tears and, although a tear protein, it promotes basal tearing and lacrimal gland secretion. Since scale up is relevant to its potential use in the treatment of dry eye, we explored various mutagenic strategies to alter the stability, solubility, and translational efficiency of nascent lacritin, and discovered 3' clustering of rare human codons. METHODS: Site-directed mutagenesis of lacritin coding cDNA "pLAC" generated 24 different nonsynonymous and 13 synonymous mutations. Nonsynonymous mutations altered amino acids with nonpolar, basic or acidi

DNA of Ehrlichia muris, hyperendemicity of Babesia microti in Ixodes ricinus and age-related detection of nuclear mitochondrial DNA (NUMTs) in Dermacentor reticulatus from an urban, marshy biotope of South-central Europe.

The present study was initiated to analyze ticks collected periodically from the vegetation in an urban marshy biotope of central Europe. During the one-year-long study period, 1960 ticks were found, including Ixodes ricinus (n = 1037), Dermacentor reticulatus (n = 610) and Haemaphysalis concinna (n = 313). DNA was extracted from 199 Dermacentor reticulatus and 47 Ixodes ricinus, selected from the beginning of their questing period. Molecular analysis of the cytochrome c oxidase subunit I (cox1) barcoding gene with the classical Folmer primers revealed that among 37 D. reticulatus all except t

Description of Rhipicephalus hibericus phenology and main hosts in its type locality.

We conducted a study on different aspects of the biology and ecology of the recently described Rhipicephalus hibericus Millán, Rodríguez-Pastor and Estrada-Peña (Ixodida: Ixodidae) in agricultural and riparian habitats of its type locality (Zaragoza, north-east Spain), with a focus on seasonal dynamics of questing individuals and the identification of the main hosts (both through live-trapping and molecular blood meal analysis in questing adults). We further evaluated the effect of potential wildlife hosts (micro and meso-mammals, and birds), habitat (agrarian and natural) and climate variable

Chimeric vaccine design against the epidemic Langya Henipavirus using immunoinformatics and validation via immune simulation approaches.

In July 2022, a new virus called Langya virus (LayV) was discovered in China in patients who had a fever. This virus is a type of Henipavirus (HNV) and is considered a potential threat as it could spread from animals to humans. It causes respiratory disease with symptoms including fever, coughing, and fatigue and is closely linked to two other henipaviruses that are known to infect humans, namely Hendra and Nipah viruses. These viruses may cause fatal respiratory illnesses. Investigators believe that the LayV is spread by shrews, and may have infected humans directly or via an intermediary spe

Human-animal interactions and bat coronavirus spillover potential among rural residents in Southern China.

Human interaction with animals has been implicated as a primary risk factor for several high impact zoonoses, including many bat-origin viral diseases. However the animal-to-human spillover events that lead to emerging diseases are rarely observed or clinically examined, and the link between specific interactions and spillover risk is poorly understood. To investigate this phenomenon, we conducted biological-behavioral surveillance among rural residents in Yunnan, Guangxi, and Guangdong districts of Southern China, where we have identified a number of SARS-related coronaviruses in bats. Serum

Identification of pheasant ghrelin and motilin and their actions on contractility of the isolated gastrointestinal tract.

Motilin and ghrelin were identified in the pheasant by molecular cloning, and the actions of both peptides on the contractility of gastrointestinal (GI) strips were examined in vitro. Molecular cloning indicated that the deduced amino acid sequences of the pheasant motilin and ghrelin were a 22-amino acid peptide, FVPFFTQSDIQKMQEKERIKGQ, and a 26-amino acid peptide, GSSFLSPAYKNIQQQKDTRKPTGRLH, respectively. In in vitro studies using pheasant GI strips, chicken motilin caused contraction of the proventriculus and small intestine, whereas the crop and colon were insensitive. Human motilin, but n

Molecular cloning and baculovirus expression of the rabbit corneal aldehyde dehydrogenase (ALDH1A1) cDNA.

Most mammalian species express high concentrations of ALDH3A1 in corneal epithelium with the exception of the rabbit, which expresses high amounts of ALDH1A1 rather than ALDH3A1. Several hypotheses that involve catalytic and/or structural functions have been postulated regarding the role of these corneal ALDHs. The aim of the present study was to characterize the biochemical properties of the rabbit ALDH1A1. We have cloned and sequenced the rabbit ALDH1A1 cDNA, which is 2,073 bp in length (excluding the poly(A+) tail), and has 5' and 3' nontranslated regions of 46 and 536 bp, respectively. Thi

Identification and mapping of the UL56 gene transcript of herpes simplex virus type 1.

The herpes simplex virus type 1 (HSV-1) strain HFEM is apathogenic for tree shrews and mice by the intraperitoneal application route. This is due to a 4.1 kbp deletion [0.762 to 0.789 map units (mu)] within the BamHI DNA fragment B of the viral genome. With exception of 71 bp the DNA sequences of the deleted region are located within the repetitive DNA sequences of the inverted repeat of the L segment of the HSV-1 genome (IRL). A 1.5 kb RNA hybridizing to the DNA sequences of the HSV-1 genome at map position 0.760-0.762 (BssHII DNA fragment F, part of the BamHI DNA fragment B) was found to be

Immunohistochemical distribution and colocalization of regulatory peptides in the carotid body.

Current investigations on the immunohistochemical occurrence and co-occurrence of biogenic polypeptides in the mammalian carotid body were reviewed and extended by our own recent findings. The family of chromogranins and related peptides in glomus cells appears to have a widespread interspecies distribution, whereas other peptides investigated occur in a species-specific pattern. Immunoreactivity to antisera against opioids, which derive from the proenkephalin sequence, appears to be present in glomus cells of the rabbit, cat, dog, and a shrew. Conversely, glomus cells of pig and guinea pig pr

Male/female differences in drug-induced emesis and motion sickness in Suncus murinus.

In order to elucidate possible male/female differences in emesis, the effects of various emetogenic drugs (cisplatin, copper sulfate, veratrine, nicotine, serotonin) and motion stimulus were compared between male and female Suncus murinus. Cisplatin (IP), nicotine (SC), veratrine (SC) and copper sulfate (PO) induced dose-dependent emesis in either sex, and there was no apparent difference in estimated ED50 values. However, male animals tended to be more susceptible to serotonin-induced emesis. The ID50 values for tropisetron, a 5-HT3 receptor antagonist, to block serotonin-induced emesis were

Mapping Scrub Typhus Across Multiple Hosts in India.

Scrub typhus is a significant public health concern in India. Despite numerous studies on its prevalence across hosts and vectors, country-specific data remain unavailable. This systematic review and meta-analysis provide comprehensive insights into the prevalence of scrub typhus across humans, animal hosts, and vectors to aid disease control programs. The PECOS framework was used to identify eligible studies. A systematic search across six databases (PubMed, Scopus, Web of Science, Google Scholar, Krishikosh, and Shodhganga) was conducted in March 2024. Data were classified by diagnostics and

Synanthropic and Wild Animals as Sentinels of Zoonotic Agents: A Study of Leptospira Genotypes Circulating in Northeastern Italy.

Leptospirosis is an infectious disease widely reported in veterinary practice and a worldwide zoonosis. In Northeastern Italy, different serogroups and genotypes of Leptospira have been described in ill dogs, the most commonly detected being Icterohaemorragiae (ICT) ST 17, Australis (AUS) ST 24 and ST 198, Pomona (POM) ST 117 and ST 289, and Sejroe (SEJ) ST 155. However, there is little information available on the environmental exposure to Leptospira of wild and synanthropic animals. The aim of this study was to identify the circulating genotypes in potential reservoirs to fill this gap of kn

Screening red foxes (Vulpes vulpes) for possible viral causes of encephalitis.

BACKGROUND: Next to various known infectious and non-infectious causes, the aetiology of non-suppurative encephalitis in red foxes (Vulpes vulpes) often remains unclear. Known causes in foxes imply rabies, canine distemper, toxoplasmosis, Aujeszky's disease, as well as parvovirus, adenovirus, circovirus and flavivirus infections. In this study, particular attention was paid on bornaviruses, since red foxes are predators of bicoloured white-toothed shrews, a reservoir of Borna disease virus 1 (BoDV-1). In addition, foxes are known to be highly susceptible for viruses of the order Mononegavirale

Genomic epidemiology and evolutionary analysis of Lassa virus from small mammals suggest bidirectional viral movement across humans and animals.

Lassa fever is a viral haemorrhagic fever that poses a persistent public health threat in several West African countries, particularly Nigeria. The scarcity of Lassa virus (LASV) sequences isolated from small mammal reservoirs limits our knowledge and understanding of LASV genomic diversity and transmission dynamics. To address this knowledge gap, we sampled 1189 small mammals, including mice, rats, and shrews, from two LASV-endemic states in southern Nigeria (Ondo and Ebonyi States) and tested them for the presence of LASV RNA using reverse transcription-quantitative polymerase chain reaction

[Hantavirus Disease: An Update].

In addition to the well-known clinical early symptoms of hantavirus disease (fever, flank and abdominal pain as well as arthralgia), unusual neurological changes in the context of infection come into focus. The spectrum of neurological symptoms ranges from transient myopia to severe pareses in the context of Guillain-Barré syndrome. In endemic areas, rapid IgM tests for initial assessment are of certain value for differential diagnosis. For therapeutic approaches, only supportive measures up to transient dialysis are available.Molecular genetic analysis and comparison of hantavirus strains of
