Interfacial Electrostatic Properties of Hydrated Mesoporous and Nanostructured Alumina Powders by Spin Labeling EPR.

Acid-base equilibria and interfacial electrostatic properties of hydrated mesoporous and nanostructured alumina powders are determining factors for the use of these materials in heterogeneous catalysis and as a sorption media for filtration and chromatographic applications including life sciences. Here spin probe electron paramagnetic resonance spectroscopy of pH-sensitive nitroxides was employed to evaluate the surface charge and interfacial acid-base equilibria at the pore surface of mesoporous powders of α-Al2O3, γ-Al2O3, Al2O3 × nH2O, and basic γ-Al2O3 and nanostructured Al2O3 in the form

Structural and functional analysis of the ASM p.Ala359Asp mutant that causes acid sphingomyelinase deficiency.

Niemann-Pick disease (NPD) type A and B are recessive hereditary disorders caused by deficiency in acid sphingomyelinase (ASM). The p.Ala359Asp mutation has been described in several patients but its functional and structural effects in the protein are unknown. In order to characterize this mutation, we modeled the three-dimensional ASM structure using the recent available crystal of the mammalian ASM as a template. We found that the p.Ala359Asp mutation is localized in the hydrophobic core and far from the sphingomyelin binding site. However, energy function calculations using statistical pot

Achieving successful delivery of oligonucleotides--From physico-chemical characterization to in vivo evaluation.

RNA interference is one of the most promising fields in modern medicine to treat several diseases, ranging from cancer to cardiac diseases, passing through viral infections and metabolic pathologies. Since the discovery of the potential therapeutic properties of non-self oligonucleotides, it was clear that it is important to develop delivery systems that are able to increase plasma stability and bestow membrane-crossing abilities to the oligonucleotides in order to reach their cytoplasmic targets. Polymer therapeutics, among other systems, are widely investigated as delivery systems for therap

Slow Internal Dynamics and Charge Expansion in the Disordered Protein CGRP: A Comparison with Amylin.

We provide the first direct experimental comparison, to our knowledge, between the internal dynamics of calcitonin-gene-related peptide (CGRP) and amylin (islet amyloid polypeptide, IAPP), two intrinsically disordered proteins of the calcitonin peptide family. Our end-to-end contact formation measurements reveal that in aqueous solution (i.e., in the absence of structure-inducing organic solvents) CGRP preferentially populates conformations with short end-to-end distances. However, the end-to-end distance of CGRP is larger than that of IAPP. We find that electrostatic interactions can account

The Kaposi Sarcoma Herpesvirus Latency-associated Nuclear Antigen DNA Binding Domain Dorsal Positive Electrostatic Patch Facilitates DNA Replication and Episome Persistence.

Kaposi sarcoma-associated herpesvirus (KSHV) has a causative role in several human malignancies. KSHV latency-associated nuclear antigen (LANA) mediates persistence of viral episomes in latently infected cells. LANA mediates KSHV DNA replication and segregates episomes to progeny nuclei. The structure of the LANA DNA binding domain was recently solved, revealing a positive electrostatic patch opposite the DNA binding surface, which is the site of BET protein binding. Here we investigate the functional role of the positive patch in LANA-mediated episome persistence. As expected, LANA mutants wi

Role of N-terminal residues in Aβ interactions with integrin receptor and cell surface.

beta-Amyloid (Aβ) is the primary protein component of senile plaques in Alzheimer's disease (AD) and is believed to play a role in its pathology. To date, the mechanism of action of Aβ in AD is unclear. We and others have observed that Aβ interacts either with or in the vicinity of the α6 sub-unit of integrin, and believe this may be important in its interaction with neuronal cells. In this study, we used confocal microscopy and flow cytometry to explore the residue specific interactions of Aβ40 with the cell surface and the α6 integrin receptor sub-unit. We probed the importance of the RHD se

Study on conformational stability, molecular structure, vibrational spectra, NBO, TD-DFT, HOMO and LUMO analysis of 3,5-dinitrosalicylic acid by DFT techniques.

In this work we analyzed the vibrational spectra of 3,5-dinitrosalicylic acid (3,5DNSA) molecule. The total energy of eight possible conformers can be calculated by Density Functional Theory with 6-31G(d,p) as basis set to find the most stable conformer. Computational result identify the most stable conformer of 3,5DNSA is C6. The assignments of the vibrational spectra have been carried out by computing Total Energy Distribution (TED). The molecular geometry, second order perturbation energies and Electron Density (ED) transfer from filled lone pairs of Lewis base to unfilled Lewis acid sites

Variations within class-A β-lactamase physiochemical properties reflect evolutionary and environmental patterns, but not antibiotic specificity.

The bacterial enzyme β-lactamase hydrolyzes the β-lactam ring of penicillin and chemically related antibiotics, rendering them ineffective. Due to rampant antibiotic overuse, the enzyme is evolving new resistance activities at an alarming rate. Related, the enzyme's global physiochemical properties exhibit various amounts of conservation and variability across the family. To that end, we characterize the extent of property conservation within twelve different class-A β-lactamases, and conclusively establish that the systematic variations therein parallel their evolutionary history. Large and s

CdTe/CdSe quantum dots improve the binding affinities between α-amylase and polyphenols.

People exposed to engineered nanomaterials have potential health risks associated. Human α-amylase is one of the key enzymes in the digestive system. There are few reports about the influence of quantum dots (QDs) on the digestive enzymes and their inhibition system. This work focused on the toxic effect of CdTe/CdSe QDs on the interactions between α-amylase and its natural inhibitors. Thirty-six dietary polyphenols, natural α-amylase inhibitors from food, were studied for their affinities for α-amylase in the absence and presence of CdTe/CdSe QDs by a fluorescence quenching method. The magnit

Development of a Coulombimetric immunosensor based on specific antibodies labeled with CdS nanoparticles for sulfonamide antibiotic residues analysis and its application to honey samples.

A new electrochemical immunosensor has been developed to detect sulfonamide antibiotic residues in food samples. The immunosensor presented uses immunoreagents specifically developed for the broad recognition of the sulfonamide antibiotic family, a graphite composite electrode (GEC), biofunctionalized magnetic μ-particles and electrochemical nanoprobes prepared by labeling the specific antibodies with CdS nanoparticles (CdSNP). After the immunochemical reaction, the CdSNP are dissolved and the metal ions released are reduced at the electrode and read as in the form of current or charge signal,

Sorption of Cu(II) complexes with ligands tartrate, glycine and quadrol by chitosan.

The sorption by chitosan in Cu(II) solutions containing tartrate, glycine (amino acetic acid) and quadrol (N,N,N',N'-tetrakis(2-hydroxypropyl)ethylenediamine) as ligands has been investigated. The degree of sorbate removal strongly depends on pH. In solutions containing tartrate almost complete sorption of both Cu(II) and tartrate proceeds in mildly acidic and neutral solutions. The sorption of Cu(II) is also complete in alkaline solutions containing glycine; meanwhile a substantial sorption of glycine proceeds at pH approximately 6. The Cu(II) sorption in solutions containing quadrol is insig

Development of potent purine-derived nitrile inhibitors of the trypanosomal protease TbcatB.

Human African trypanosomiasis (HAT), a major health concern in sub-Saharan Africa, is caused by the protozoan parasite Trypanosoma brucei. Recent studies have shown that a cathepsin B like protease, TbcatB, is essential to the survival of T. brucei in vitro (Mackey, Z. B.; O'Brien, T. C.; Greenbaum, D. C.; Blank, R. B.; McKerrow, J. H. J. Biol. Chem. 2004, 279, 48426-48433). Herein, we describe the first inhibitors of TbcatB, a series of purine nitriles. The compounds are potent trypanocides, killing the parasite with a high degree of selectivity over a panel of three human cell lines. In addi

SGTx1, a Kv channel gating-modifier toxin, binds to the interfacial region of lipid bilayers.

SGTx1 is a gating-modifier toxin that has been shown to inhibit the voltage-gated potassium channel Kv2.1. SGTx1 is thought to bind to the S3b-S4a region of the voltage-sensor, and is believed to alter the energetics of gating. Gating-modifier toxins such as SGTx1 are of interest as they can be used to probe the structure and dynamics of their target channels. Although there are experimental data for SGTx1, its interaction with lipid bilayer membranes remains to be characterized. We performed atomistic and coarse-grained molecular dynamics simulations to study the interaction of SGTx1 with a P

Microemulsion electrokinetic chromatography of drugs varying in charge and hydrophobicity: I. Impact of parameters on separation performance evaluated by multiple linear regression models.

The separation of anionic, cationic and neutral drugs in microemulsion electrokinetic chromatography (MEEKC) was studied with a statistical experimental design. The concentration of sodium dodecyl sulfate (SDS, surfactant), 1-butanol (co-surfactant) and borate buffer and the factors Brij 35 (surfactant), 2-propanol (organic solvent) and cassette temperature were varied simultaneously, while the parameters pH (9.2), the concentration of octane (oil, 0.8% w/w), the voltage (10 kV) and the dimension of the fused-silica capillary, were kept constant. Eight different model substances were chosen wi

NMR structure of a gemcitabine-substituted model Okazaki fragment.

Gemcitabine (2'-deoxy-2',2'-difluorodeoxycytidine; dFdC) is a potent anticancer drug that exerts cytotoxic activity, in part, through incorporation of the nucleoside triphosphate dFdCTP into DNA and perturbations to DNA-mediated processes. The structure of a model Okazaki fragment containing a single dFdC substitution, [GEM], was determined using NMR spectroscopy and restrained molecular dynamics to understand structural distortions that may be induced in replicating DNA resulting from dFdC substitution. The electrostatic surface of [GEM] was also computed to determine how the geminal difluoro

Butyrylcholinesterase-catalysed hydrolysis of aspirin, a negatively charged ester, and aspirin-related neutral esters.

Although aspirin (acetylsalicylic acid) is negatively charged, it is hydrolysed by butyrylcholinesterase (BuChE). Catalytic parameters were determined in 100 mM Tris buffer, pH 7.4, in the presence and absence of metal cations. The presence of Ca2+ or Mg2+ (<100 mM) in buffer did not change the Km, but accelerated the rate of hydrolysis of aspirin by wild-type or D70G mutant BuChE by 5-fold. Turnover numbers were of the order of 5000-12000 min-1 for the wild-type enzyme and the D70G and D70K enzymes in 100 mM Tris, pH 7.4, containing 50 mM CaCl2 at 25 degreesC; Km values were 6 mM for wild-typ

Reconciling Cryo-EM and Crystallographic Views of RNA Folding through Atomistic Structure-Based Simulations: Roles of Ionic Conditions and Truncation of Flexible Elements.

Discrepancies between biomolecular structures resolved by cryo-electron microscopy (cryo-EM) and X-ray crystallography (XRD) arise from differences in ionic conditions and construct design, yet how these shape RNA folding remains unresolved. The SARS-CoV-2 frameshifting stimulatory element provides a representative case: cryo-EM captures a conformation with the slippery segment, whereas XRD reveals a higher-resolution, coaxially stacked structure lacking this segment but displaying base-triple interactions absent in cryo-EM. To reconcile these condition-dependent views, we integrate explicit-s

Size-dependent ruthenium/ceria nanozymes synchronize catalytic ROS scavenging and electrostatic mtDNA sequestration for periodontitis therapy.

Mitochondrial oxidative stress and release of mitochondrial DNA (mtDNA) are increasingly implicated in amplification of inflammatory responses in chronic diseases such as periodontitis. While cerium dioxide (CeO2)-based nanozymes have been explored as antioxidants, their therapeutic efficacy remains limited by suboptimal catalytic performance and weak interactions with intracellular inflammatory mediators. Here, we report a structure-engineered ruthenium-sensitized ceria nanozyme platform designed to modulate mitochondrial redox imbalance and mtDNA-driven innate immune activation in periodonti

Computational and experimental approaches for chitosan-based nano PECs design: Insights on a deeper comprehension of nanostructure formation.

Nanostructured polyelectrolyte complexes (nano PECs) based on biopolymers are an important technological strategy to target drugs to the action and/or absorption site in a more effective way. In this work, computational studies were performed to predict the ionization, spatial arrangement and interaction energies of chitosan (CS), hyaluronic acid (HA), and hypromellose phthalate (HP), for the design of nano PEC carriers for methotrexate (MTX). The optimal pH range (5.0-5.5) for preparing nano PECs was selected by experimental and computational methodologies, favoring the polymers interactions.
