Plant-based models of neurodegenerative CAG repeat expansion diseases.

Expansion of cytosine-adenine-guanine (CAG) trinucleotide repeats causes Huntington's disease and other neurodegenerative disorders, yet no disease-modifying treatments exist. Plant-based CAG models expressing mutant huntingtin protein provide novel insights into expanded CAG-induced toxicity, complementing traditional systems for studying neurodegeneration and therapeutic discovery, and may offer unique opportunities for advancing plant biology research.

25+ Years of TRPV4: From Discovery to Translational Horizons.

Transient receptor potential vanilloid member 4 (TRPV4) is a calcium-permeable, nonselective cation channel first described in 2000. Polymodally-activated, also by osmotic, mechanical, thermal, actinic, and chemical stimuli, TRPV4 has emerged as a versatile molecular integrator across a remarkable diversity of cell lineages and vertebrate organ systems. Re orthology to invertebrate TRPV channels, TRPV4 rescued osmotic and mechanical sensing in ASH nociceptor neurons of C. elegans mutant for OSM-9 TRPV channels. This chapter surveys the arc of TRPV4 research from foundational cloning to transla

Potential Associations between Vascular Biology and Hodgkin's Lymphoma: An Overview.

Hodgkin's lymphoma (HL) is a lymphatic neoplasm typically found in the cervical lymph nodes. The disease is multifactorial, and in recent years, the relationships between various vascular molecules have been explored in the field of vascular biology. The connection between vascular biology and HL is intricate and the roles of several pathways remain unclear. This review summarizes the cellular and molecular relationships between vascular biology and HL. Proteins associated with various functions in vascular biology, including cytokines (TNF-α, IL-1, IL-13, and IL-21), chemokines (CXCL10, CXCL1

Primary Cultures of Spermatogonia and Testis Cells.

Spermatogonial stem cells (SSCs) maintain adult spermatogenesis in mammals by undergoing self-renewal and differentiation into spermatozoa. In order to study the biology of SSCs as related to spermatogenesis, an in vitro, long-term expansion system of SSCs constitutes an ideal tool. In this chapter, we describe a robust culture system for mouse and rat SSCs in vitro. In the presence of GDNF, GFRα1, and bFGF, SSCs maintained on STO feeder layers with serum-free medium continuously proliferate for over 6 months. Complete spermatogenesis in infertile recipient mice can be attained following trans

Molecular Epidemiology of Giardia Infections in the Genomic Era.

Giardia duodenalis is a major gastrointestinal parasite of humans and animals across the globe. It is also of interest from an evolutionary perspective as it possesses many features that are unique among the eukaryotes, including its distinctive binucleate cell structure. While genomic analysis of a small number of isolates has provided valuable insights, efforts to understand the epidemiology of the disease and the population biology of the parasite have been limited by the molecular tools currently available. We review these tools and assess the impact of affordable and rapid genome sequenci

Engineering DNA-Targeting CRISPR and CRISPR-Like Effectors: Advances from Rational Design and High-Throughput Screening to AI-Driven Development.

Clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated (Cas) proteins constitute adaptive immune systems in prokaryotes and have transformed life sciences, precision medicine, and synthetic biology as programmable genome-editing tools. Despite their broad utility, naturally occurring DNA-targeting Cas effectors remain constrained by several intrinsic limitations, including large protein size that complicates delivery, stringent protospacer adjacent motif (PAM) requirements that restrict targetable genomic space, and mismatch tolerance that can lead to off-targ

Mce transport systems in Mycobacterium tuberculosis: architecture, regulation, lipid import, and translational perspectives.

Mammalian cell entry (Mce) systems are now recognized as central determinants of lipid uptake, cell-envelope homeostasis, and host adaptation in Mycobacterium tuberculosis (Mtb). Although the term "mammalian cell entry" originated from early studies linking the Mce loci to host-cell invasion phenotypes, subsequent genetic, biochemical, and structural work has substantially reframed their biological significance. Current evidence supports a model in which Mce1 and Mce4 function as multiprotein lipid-import systems specialized primarily for fatty-acid and cholesterol uptake, respectively, wherea

Development of a replicating plasmid based on the native oriC in Mycoplasma pneumoniae.

Bacteria of the genus Mycoplasma have recently attracted considerable interest as model organisms in synthetic and systems biology. In particular, Mycoplasma pneumoniae is one of the most intensively studied organisms in the field of systems biology. However, the genetic manipulation of these bacteria is often difficult due to the lack of efficient genetic systems and some intrinsic peculiarities such as an aberrant genetic code. One major disadvantage in working with M. pneumoniae is the lack of replicating plasmids that can be used for the complementation of mutants and the expression of pro…

Construction of new synthetic biology tools for the control of gene expression in the cyanobacterium Synechococcus sp. strain PCC 7002.

Predictive control of gene expression is an essential tool for developing synthetic biological systems. The current toolbox for controlling gene expression in cyanobacteria is a barrier to more in-depth genetic analysis and manipulation. Towards relieving this bottleneck, this work describes the use of synthetic biology to construct an anhydrotetracycline-based induction system and adapt a trans-acting small RNA (sRNA) system for use in the cyanobacterium Synechococcus sp. strain PCC 7002. An anhydrotetracycline-inducible promoter was developed to maximize intrinsic strength and dynamic range.

Systems pharmacology modeling: an approach to improving drug safety.

Advances in systems biology in conjunction with the expansion in knowledge of drug effects and diseases present an unprecedented opportunity to extend traditional pharmacokinetic and pharmacodynamic modeling/analysis to conduct systems pharmacology modeling. Many drugs that cause liver injury and myopathies have been studied extensively. Mitochondrion-centric systems pharmacology modeling is important since drug toxicity across a large number of pharmacological classes converges to mitochondrial injury and death. Approaches to systems pharmacology modeling of drug effects need to consider drug…

Using clickers in nonmajors- and majors-level biology courses: student opinion, learning, and long-term retention of course material.

Student response systems (clickers) are viewed positively by students and instructors in numerous studies. Evidence that clickers enhance student learning is more variable. After becoming comfortable with the technology during fall 2005-spring 2006, we compared student opinion and student achievement in two different courses taught with clickers in fall 2006. One course was an introductory biology class for nonmajors, and the other course was a 200 level genetics class for biology majors. Students in both courses had positive opinions of the clickers, although we observed some interesting diff

A multi-objective genetic algorithm to find active modules in multiplex biological networks.

The identification of subnetworks of interest-or active modules-by integrating biological networks with molecular profiles is a key resource to inform on the processes perturbed in different cellular conditions. We here propose MOGAMUN, a Multi-Objective Genetic Algorithm to identify active modules in MUltiplex biological Networks. MOGAMUN optimizes both the density of interactions and the scores of the nodes (e.g., their differential expression). We compare MOGAMUN with state-of-the-art methods, representative of different algorithms dedicated to the identification of active modules in single

Asia-Pacific Transplant Oncology Symposium 2024: Expert Statements on the Current State and Future Direction of Hepatobiliary Surgical and Transplant Oncology.

BACKGROUND: Hepatobiliary oncology is at an inflection point. Parallel advances in surgical technique, locoregional therapy, systemic treatment, and understanding of tumour biology have expanded the boundaries of curative-intent care yet have also exposed a critical reality that technical resectability is not synonymous with oncologic resectability. Technically successful resection or liver transplantation may be futile when tumour biology is aggressive, leading to early recurrence and limited survival benefit. Modern hepatobiliary surgical and transplant oncology must therefore move beyond an

Current approaches and advances in placental toxicology.

Despite the crucial role of the placenta in supporting pregnancy and fetal development, research into its susceptibility to environmental exposures has been limited by methodological challenges. We review diverse approaches to studying placental biology and responses to chemical exposures, and provide a comprehensive assessment of traditional and emerging methodologies. Beginning with an overview of placental biology and species differences, we evaluate in vivo and in vitro models, and discuss their strengths and limitations. We examine advances, including placental transfer models, toxicokine

Mitochondrial biology and the identification of biomarkers of Huntington's disease.

Apart from finding novel compounds for treating Huntington's disease (HD) an important challenge at present consists in finding reliable read-outs or biomarkers that reflect key biological processes involved in HD pathogenesis. The core elements of HD biology, for example, HTT RNA levels or protein species can serve as biomarker, as could measures from biological systems or pathways in which Huntingtin plays an important role. Here we review the evidence for the involvement of mitochondrial biology in HD. The most consistent findings pertain to mitochondrial quality control, for example, fissi
