Does the magnitude of mitral regurgitation determine hemodynamic response to vasodilation in chronic congestive heart failure?

Although it is usually assumed that direct-acting vasodilators improve cardiac function in patients with congestive heart failure (CHF) by altering left ventricular preload and afterload, several studies have suggested that most of the benefit occurs as a result of a reduction in associated mitral regurgitation (MR), which is present in the majority of patients with severe CHF. To test his hypothesis, the hemodynamic response to oral hydralazine was examined in CHF patients with competent mitral prostheses (group 1) and patients with CHF due to severe MR and left ventricular dysfunction (group

Renal function during consciousness and anesthesia in sheep: effects of renal vasodilation by papaverine and acetylcholine.

Adult ewes were studied to determine the effects of barbiturate anesthesia on renal function and the natriuretic response to renal vasodilation by papaverine and acetylcholine. During a period of consciousness, renal blood flow, glomerular filtration rate, urinary Na excretion (UNaV) and arterial blood pressure were monitored. Renal blood flow was maximally increased by renal arterial infusion of papaverine or acetylcholine and changes in renal function were observed. The sheep were subsequently anesthetized and the experimental protocol was repeated. Barbiturate anesthesia resulted in statist…

A redox sensor in protein kinase A regulatory subunit Iα regulates vasodilation and protects against hypertension.

The regulatory RIα subunit of type I protein kinase A (PKARIα) can be oxidized to an interprotein disulfide homodimer, linking cellular oxidant signals with downstream phosphorylation-mediated physiological regulation. Although PKA and oxidants each have well-established roles in vasodilation and blood pressure lowering, whether the oxidation of RIα to the disulfide state regulates these physiological processes is unknown. Here, we demonstrate that PKARIα contains a thiol redox sensor, the oxidation state of which is dynamic and modulates systemic arterial blood pressure by regulating oxidant-…

Postmenopausal vasomotor symptoms are associated with heightened cutaneous vasodilation despite preserved nitric oxide-mediated endothelial function.

OBJECTIVE: To evaluate whether postmenopausal females with vasomotor symptoms (VMS) exhibit altered nitric oxide synthase (NOS)-dependent cutaneous vasodilation during local heating, and whether these responses vary by body region. METHODS: Cutaneous vascular conductance normalized to maximum vasodilation (percentage of the maximal conductance [%CVCmax]) was assessed at the chest, abdomen, forearm, and calf using laser-Doppler flowmetry in 18 postmenopausal females (9 with current VMS, 9 without VMS). Local skin temperature was increased from 33 °C to 42 °C (~30 min) to elicit a vasodilatory …

Variability in flow-induced vasodilation mechanisms in cerebral arteries: the impact of different hyperbaric oxygen protocols.

The present study aimed to assess the mechanisms of flow-induced dilation (FID) altered by acute/intermittent hyperbaric oxygenation (HBO 2 ) in isolated middle cerebral arteries of healthy male Sprague‒Dawley rats ( n = 96) and randomized to the Ac-HBO 2 group (exposed to a single HBO 2 session, 120 minutes of 100% O 2 at 2.0 bars), the 4Dys-HBO 2 group (4 consecutive days of single HBO 2 sessions, analyzed on the fifth day), and the CTRL (untreated) group. Results demonstrated increased vascular oxidative stress and decreased vascular nitric oxide bioavailability, as measured by direct fluor

Maternal Gestational Diabetes Impairs Fetoplacental Insulin-Induced Vasodilation via AKT/eNOS Pathway and Reduces Placental Efficiency.

Gestational Diabetes Mellitus (GDM) increases the long-term risk for metabolic and cardiovascular diseases in the offspring. However, the un … ctional analysis using a Mulvany myograph demonstrated a significant impairment of insulin-mediated vasodilation in fetoplacental vessels of GDM patients compared to NG controls. This vascular dysfunction was associated with a reduction in total insulin receptor protein expression. Further investigation revealed an impaired PI3K/AKT/eNOS signaling pathway, as …

Blunted baroreflex-mediated sympathoinhibition and vasodilation to countdown before exercise in young women compared to men.

PURPOSE: In our previous study (Manabe et al., J Appl Physiol 128:1196-1206, 1985), we demonstrated that the cerebral and cardiovascular responses induced by a pre-exercise countdown elicited peripheral vasodilation via baroreflex-mediated sympathetic withdrawal, which is likely advantageous for rapid oxygen delivery to contracting skeletal muscles in young men. Whether this is also true in young women, who generally show different neuro-cardiovascular responses to stress compared to men, remains unknown. Thus, we examined whether biological sex would affect the neuro-cardiovascular responses …

An Activatable Chemiluminescent Self-Reporting Sulfur Dioxide Donor for Inflammatory Response and Regulation of Gaseous Vasodilation.

Sulfur dioxide (SO2), being a novel gaseous signaling molecule, exhibits significant potential for application in the field of cardiovascular diseases. SO2 donors serve as crucial tools for the transportation and regulation of SO2 in vivo, facilitating the investigation of physiological roles associated with this molecule. However, the current therapeutic SO2 donors lack the capability to monitor the real-time release of SO2, thereby hindering accurate assessment of their therapeutic efficacy and target localization. Herein, we present an activatable chemiluminescent self-reporting SO2 donor (

Dieckol induces vasodilation via endothelial-smooth muscle crosstalk in co-culture and in vivo zebrafish models.

Dieckol (DK), a phlorotannin isolated from Ecklonia cava, has been suggested to exert vasodilatory effects through endothelial signaling. In this study, the endothelial-smooth muscle signaling pathway involved in DK-mediated vasodilation was recapitulated using a multistep experimental system that integrates a co-culture of human coronary artery endothelial cells (HCAEC) and human coronary artery Smooth Muscle Cells (HCASMC) with an in vivo zebrafish model. DK activated calcium-dependent signaling pathways in HCAEC, notably via muscarinic acetylcholine receptor M3 (AChM3R) and voltage-dependen…

Dapagliflozin induces vasodilation in resistance-size mesenteric arteries by stimulating smooth muscle cell KV7 ion channels.

Dapagliflozin is a sodium-glucose cotransporter 2 (SGLT2) inhibitor that, in addition to glucose reduction, lowers systemic blood pressure. … large-conductance Ca2+-activated K+ (BKCa) channels, ATP-sensitive (KATP) channels did not abolish vasodilation. Dapagliflozin-evoked vasodilation remained unaltered by pharmacological inhibition of endothelium-derived nitric oxide (NO) signaling, prostacyclin (PGI2), as well as by endothelium denudation. Our Western blotting data revealed that SGLT2 protein …

Sex Differences in the Vasodilation Mediated by G Protein-Coupled Estrogen Receptor (GPER) in Hypertensive Rats.

BACKGROUND: The protective effect of estrogen on the vasculature cannot be explained only by its action through the receptors ERα and ERβ. G protein-coupled estrogen receptors (GPER)-which are widely distributed throughout the cardiovascular system-may also be involved in this response. However, little is known about GPER actions in hypertension. Therefore, in this study we evaluated the vascular response mediated by GPER using a specific agonist, G-1, in spontaneously hypertensive rats (SHR). We hypothesized that G-1 would induce a relaxing response in resistance mesenteric arteries from SHR

Relationship between impaired parasympathetic vasodilation and hyposalivation in parotid glands associated with type 2 diabetes mellitus.

We examined the relationship between hemodynamics in the three major salivary glands and salivary secretion in urethane-anesthetized and sym … mpared with the nondiabetic rats. Our results indicate that type 2 diabetes impairs parasympathetic vasodilation and salivary secretion in the parotid gland and suggest that disturbances in the cholinergic vasodilator pathway may contribute to the underlying mechanisms involved in the disruption of parasympathetic nerve-mediated glandular vasodilation.

Pharmacological Characterizations of anti-Dementia Memantine Nitrate via Neuroprotection and Vasodilation in Vitro and in Vivo.

We have previously designed and synthesized a series of novel memantine nitrates, and some of them have shown neuroprotective effects; however, the detailed mechanisms remain unknown. In this study, we demonstrated that MN-12, one of the memantine nitrates, concentration-dependently protected against glutamate-induced neurotoxicity in rat primary cultured cerebellar granule neurons (CGNs). Western blotting assays revealed that MN-12 might possess neuroprotective effects through the inhibition of ERK pathway and activation of PI3K/Akt pathway concurrently. Moreover, MN-12 concentration-dependen

Metoprolol Impairs β1-Adrenergic Receptor-Mediated Vasodilation in Rat Cerebral Arteries: Implications for β-Blocker Therapy.

The practice of prescribing β-blockers to lower blood pressure and mitigate perioperative cardiovascular events has been questioned because of reports of an increased risk of stroke. The benefit of β-blocker therapy primarily relies on preventing activation of cardiac β1-adrenergic receptors (ARs). However, we reported that β1ARs also mediate vasodilator responses of rat cerebral arteries (CAs), implying that β-blockers may impair cerebral blood flow under some conditions. Here, we defined the impact of metoprolol (MET), a widely prescribed β1AR-selective antagonist, on adrenergic-elicited dia
