A nonimmune library, containing single chain variable fragments (scFv) of immunoglobulin human genes displayed on the surface of M13 filamentous phages, was used to recognize molecules exposed on Histoplasma capsulatum yeasts' surface, during their growth in synthetic medium. The scFv clones were checked in their consistency by Dot-ELISA using HRP/anti-M13 conjugate, and they were tested to recognize molecules on H. Capsulatum yeasts' surface by ELISA in plates. Three out of 80 scFv cones (C2, C6, and C52) reacted consistently with H. capsulatum molecules, and they recognized molecules from bo…

The prevalence of dermatophytes and yeasts (Candida spp. and Pityrosporum spp.) was studied in 40 former drug-addicts, all of whom were HIV seropositive but otherwise had no other symptoms (2nd Stage CDC Atlanta, 1987). We considered 7 skin areas for dermatophytes and Pityrosporum spp. (scalp, forehead, nose, back, chest, groin, toe webs) and the mouth for yeasts. Dermatophytes were found in 8 (20%) and tinea pedis was the most common dermatophytosis: Tricophyton rubrum was the fungus most frequently isolated (6 cases or 15%). The HIV+ group showed almost the same rate of dermatophytes colonis…

In microbial cytoplasmic reaction compartments, competitive pathways and metabolic interactions often hinder the efficient synthesis of target compounds. In contrast, eukaryotic cells can control metabolic complexity by utilizing organelles to segregate biochemical pathways. Mitochondria, known as the "powerhouses of the cell," are rich in a unique set of enzymes, intermediate metabolites, and cofactors that can be exploited for metabolic engineering. However, wider application of mitochondria in metabolic engineering is limited by an insufficient understanding of their unique metabolic traits

The addition of yeast cultures to diets has been demonstrated to maintain the homeostasis of the rumen endo-environment and to enhance the body's immunity. However, the effect of yeast cultures on alleviating transportation stress remains unclear. A total of 16 goats were randomly distributed into 2 dietary treatments (8 replicates/treatment) as follows: Basic diet group (CON); basic diet + 0.3% YC group (YC). The experiment lasted for a total of 46 days, consisting of a 30-day growth trial, followed by a one-day transportation stress treatment, and subsequently a 14-day feeding trial. Samples

Xanthophyllomyces dendrorhous is a basidiomycetous yeast capable of synthesizing astaxanthin, a powerful antioxidant carotenoid pigment. Despite appreciable research efforts, molecular tools for the genetic manipulation of this yeast remain limited. Inducible promoters are essential tools to control gene expression in response to external stimuli, such as molecules or stress signals. This regulation enables precise control of gene expression in synthetic biological systems, minimizing unwanted effects caused by constant gene expression. In the present work inducible promoters from the L-arabin

Backbone-cyclized conotoxin Vc1.1 is a potent GABAB receptor agonist and analgesic drug lead for treating neuropathic pain. Here, we describe two yeast-based strategies for the direct recombinant production of this cyclic, disulfide-rich peptide. In the first approach, using Pichia pastoris, a fusion protein comprising the conotoxin precursor and a full-length asparaginyl ligase connected by a poly-His linker was secreted into the culture medium. Cyclization of the secreted α-conotoxin was initiated by a two-step pH adjustment protocol for generating the correctly folded conotoxin. In the seco

In Saccharomyces cerevisiae, cytosolic NADH generated during biomass production must be efficiently reoxidized to sustain metabolic flux. Under aerobic conditions, this process is primarily mediated by the oxidative phosphorylation and specifically via external NADH dehydrogenases (Nde1p/Nde2p) and the glycerol-3-phosphate (G3P) shuttle. Although these pathways perform equivalent redox functions, their relative contributions vary with physiological conditions, indicating the existence of regulatory mechanisms governing electron partitioning. Here, we combined experimental measurements and kine

This study investigated the effect of Penicillium (Penicillium aethiopicum & Penicillium chrysogenum, PP), yeast (Candida parapsilosis & Rhodotorula mucilaginosa, CM) and mixture (MI) inoculation on the taste development of Jinhua ham. The results showed that Penicillium and yeast significantly increased acidic protease activity and reduced TVB-N values compared to the control. Three kinds of inoculation promoted the degradation of both myofibrillar and sarcoplasmic proteins, leading to a accumulation of peptides and free amino acids. It was strongly detected by the electronic tongue sensors P

Carbohydrates are involved in energy and material metabolism, playing an essential role in plant growth, development, and stress response. This study used Sorbus aucuparia suspension cells(SASCs) as a model to investigate the effects of yeast extract(YE) induction on the carbohydrate components of SASCs. Polysaccharides were extracted using an orbital shaker, and total and soluble sugar content was determined. The types and quantities of oligosaccharides were analyzed by ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS) and ultra-performance liq

The increasing prevalence of antifungal resistance represents a major clinical challenge. To explore potential new therapeutic avenues, we investigated fitness trade-offs associated with azole and echinocandin resistance in Nakaseomyces glabratus (syn. Candida glabrata), a priority yeast pathogen showing growing incidence of drug and multidrug resistance. For this, we comprehensively phenotyped a large collection (n = 77) of azole- and echinocandin-resistant strains to uncover resistance-associated stress sensitivity trade-offs. Our results show that increased stress sensitivity is a common tr

Tartrazine is a synthetic dye commonly used in the food industry to enhance the visual appeal of food products. However, its instability under specific conditions, such as changes in pH, exposure to UV or sunlight, or increased temperature, may lead to adverse effects, raising concerns about its toxicity. Thus, ensuring the safety, controlled release, and stability of these colorants in food matrices remains a significant challenge. This study aimed to evaluate inactivated brewer's yeast (Saccharomyces cerevisiae) as a promising biosorbent matrix for the adsorption and stabilization of tartraz

Despite growing trend in sustainable protein sources, yeast proteins have mainly been explored as a source of bioactive peptides using a monospecies and general protein approach. The contribution of highly abundant protein fractions in the yeast proteome to peptide formation remains insufficiently investigated, limiting a comprehensive understanding of yeast proteins as optimized peptide sources. The current review presents a systematic analysis of yeast proteins as emerging protein sources and evaluates the suitability of high-abundance proteins as bioactive peptide precursors by in silico te

Kauniolide, a parthenolide derivative, serves as a key biosynthetic intermediate for various guaianolide-type sesquiterpenoids, such as agrabin, lactucin, and lactupicrin. However, its chemical synthesis is often hampered by demanding reaction conditions and high reagent consumption, which severely limit further development and application. To address this, this study employed systematic metabolic engineering strategies to construct an efficient yeast cell factory for kauniolide production. First, the biosynthetic pathway genes for costunolide-the direct precursor of kauniolide(HaGAS, TpGAO, T

Interactions between biological oscillators and environmental periodic signals are crucial for the correct regulation of cellular behaviors. Most synthetic biological oscillators only respond to one signal that initiates the oscillation, limiting the study of the interaction between the biological oscillator and multiple environmental cues. Here, we present a detailed protocol for constructing a synthetic oscillatory circuit that can respond to both α-factor and ethanol inputs. Based on the mammalian NF-κB negative feedback loop, this circuit produces sustained and tunable oscillation in the b

BACKGROUND: Sucrose Non Fermenting1 (SNF1) constitutes a family of protein kinases conserved in eukaryotes, plants, and fungi. SNF1 has been known to play a crucial role in stress adaptation and metabolism, enabling organisms to respond to changing environmental conditions. Initially identified in yeast, SNF1 is essential for shifting from the primary carbon source, glucose, to secondary carbon sources like sucrose. Homologs of this protein family were identified in Leishmania major, a protozoan parasite and we aimed to determine their role in this parasite.
METHODS: In the present study, we

Inhibition of the target of rapamycin (TOR/mTOR) protein kinase by the drug rapamycin extends lifespan and health span across diverse species. However, rapamycin has potential off-target and side effects that warrant the discovery of additional TOR inhibitors. TOR was initially discovered in Saccharomyces cerevisiae (yeast) which contains two TOR paralogs, TOR1 and TOR2. Yeast lacking functional Tor1 are viable but are hypersensitive to growth inhibition by TORC1 inhibitors, which is a property of yeast that can be exploited to identify TOR inhibitors. Additionally, yeast lacking FK506-sensiti
