Enzyme histochemical analysis on cryostat sections of human bone marrow.

A recently developed procedure, that has been shown to be suitable for detailed immunohistological analysis, has been used to prepare cryostat sections of bone marrow to investigate whether enzyme-histochemical techniques are also feasible on such material. A selected group of enzymes, some of which are inhibited or destroyed in paraffin- or plastic-embedded samples, have been demonstrated. The morphological details obtained were satisfactory in the preparations. The enzymes were dipeptidyl(amino)peptidase IV (for T lymphocytes); tartrate-resistant acid phosphatase (for hairy cell leukaemia);

Lymphocyte sorting on albuminated CIBA blue dextran-staphylococcal protein A-conjugated sepharose 6MB affinity columns.

Modification of Sepharose 6MB affinity beads by conjugation with CIBA blue dextran and albumination together with siliconization and modification of the bed supports of glass chromatography columns permitted the construction of affinity columns with low non-specific binding characteristics. When used with staphylococcal protein A as the affinity ligand to fractionate antibody-coated lymphocytes, non-specific adherence was reduced to 2--3% of the viable input cells and essentially the entire input cell number could be recovered in viable, functional form. The capacity of the columns permitted q

Failure of T and B cell cooperation during graft-versus-host disease.

Possible explanations for the immune deficiency usually associated with graft-versus-host disease include defective lymphocyte differentiation and active suppressor cell mechanisms. The present observations in a patient with chronic graft-versus-host disease delineate yet another possible mechanism: a failure of successful interaction between competent patient lymphocytes. Thus, helper and suppressor T cells as well as precursor B cells for the generation of specific in vitro plaque-forming cell (PFC) responses were present among patient lymphocytes and could interact normally with cells from

Affinity-matured B cell responses neutralizing type-I interferons underlie severe viral infections.

Autoantibodies neutralizing type-I interferons (AAN-I-IFNs) emerge as global, common, and strong determinants of a growing number of severe viral diseases. We report that AAN-I-IFNs+ patients with life-threatening COVID-19 pneumonia harbor circulating type-I IFN-specific B cells indistinguishable from patients bearing T cell tolerance defects of genetic origin. This autoimmune response mobilizes a highly diverse and stable circulating B cell response that is detected prior to severe viral infection and acquires high affinity and neutralization potential to type-I IFNs through extended somatic

Dual dynamics of persistence and recruitment characterise the ACPA-expressing B cell response in rheumatoid arthritis.

OBJECTIVES: Rheumatoid arthritis, a prototypic autoimmune disease, is characterised by the presence of anticitrullinated protein antibodies (ACPAs). Unlike responses to recall antigens, ACPA-expressing B cells display a continuous phenotype of recent activation, even in patients with sustained clinical remission, indicating that these cells are continuously activated over long periods without 'dying out' or acquiring a resting or anergic phenotype. To elucidate whether this persistent activation reflects ongoing stimulation of the same B cell clones or recruitment of new cells, we longitudinal

Spatial Transcriptional Dynamics of CD74⁺ B Cells in Tertiary Lymphoid Structures Drive Immune Evolution in Penile Squamous Cell Carcinoma.

Penile squamous cell carcinoma (PSCC) is a malignancy characterized by a poor prognosis and lack of reliable biomarkers, presenting considerable therapeutic challenges. Tertiary lymphoid structures (TLSs) are critical modulators of antitumor immunity; however, the immunological dynamics, particularly the roles of B cells and their interactions with naive T cells in PSCC tissues, remain inadequately understood. This study integrates transcriptomic approaches, including spatial transcriptomics, single-cell sequencing (scRNA-seq), and bulk RNA sequencing (bulk RNA-seq), and immunohistochemistry t

Splenic marginal zone B cells restrict Mycobacterium tuberculosis infection by shaping the cytokine pattern and cell-mediated immunity.

Understanding the role of B cells in tuberculosis (TB) is crucial for developing new TB vaccines. However, the changes in B cell immune landscapes during TB and their functional implications remain incompletely explored. Using high-dimensional flow cytometry to map the immune landscape in response to Mycobacterium tuberculosis (Mtb) infection, our results show an accumulation of marginal zone B (MZB) cells and other unconventional B cell subsets in the lungs and spleen, shaping an unconventional B cell landscape. These MZB cells exhibit activated and memory-like phenotypes, distinguishing thei

Can Galectin-3 Be a Novel Biomarker in Chronic Lymphocytic Leukemia?

Galectin-3's (Gal-3) effect on the pathogenesis of chronic lymphocytic leukemia (CLL) has not yet been extensively studied. The present study aims to analyze the potential role of Gal-3 as a prognostic biomarker in CLL patients. The Gal-3 expression was evaluated in CLL cells with RT-qPCR and flow cytometry. Due to the unclear clinical significance of soluble Gal-3 in CLL, our goal was also to assess the prognostic value of Gal-3 plasma level. Because cell survival is significantly affected by the interaction between Gal-3 and proteins such as Bcl-2, the results of Gal-3 expression analysis we

CD74 as a regulator of transcription in normal B cells.

CD74 is receptor for the cytokine macrophage migration inhibitory factor (MIF). MIF binding to CD74 induces a signaling cascade resulting in the release of its cytosolic intracellular domain (CD74-ICD) that serves as a transcriptional regulator in chronic lymphocytic leukemia (CLL) cells. In the current study, we investigated the transcriptional and regulatory function of CD74-ICD in normal B cells. We show that following activation, CD74-ICD forms a complex in the cytosol with transcription factors, like PAX5, and binds the chromatin at a significantly higher number of sites compared with its

Early Events in Lymphoid Neoplasms and Indolent Follicular Lymphomas.

Technical advances in diagnostic modalities have led to the characterization of indolent lymphoid disorders similar to the in situ lesions described in epithelial malignancies. These early and indolent lymphoid lesions share clinicopathologic characteristics with well-characterized lymphoid malignancies such as chronic lymphocytic leukemia and follicular lymphoma. The in situ lesions have an indolent clinical course with only a minor subset shown to progress to frank malignancies. In addition to the in situ lesions, new indolent lymphoproliferative disorders have been recently characterized. D

Development of the Bruton's tyrosine kinase inhibitor ibrutinib for B cell malignancies.

Ibrutinib is a first-in-class oral covalent inhibitor of Bruton's tyrosine kinase that has demonstrated clinical benefit for many patients with B cell malignancies. Positive results in initial trials led the U.S. Food and Drug Administration to grant ibrutinib three breakthrough therapy designations for mantle cell lymphoma (MCL), del17p chronic lymphocytic leukemia (CLL), and Waldenström's macroglobulinemia (WM). Ibrutinib was approved for these three cancers within 14 months of the original U.S. approval. Additionally, ibrutinib is approved for patient subsets with MCL and/or CLL in >45 othe

Interleukin 35-Producing B Cells (i35-Breg): A New Mediator of Regulatory B-Cell Functions in CNS Autoimmune Diseases.

Neuroinflammation contributes to neuronal deficits in neurodegenerative CNS (central nervous system) autoimmune diseases, such as multiple sclerosis and uveitis. The major goal of most treatment modalities for CNS autoimmune diseases is to limit inflammatory responses in the CNS; immune-suppressive drugs are the therapy of choice. However, lifelong immunosuppression increases the occurrence of infections, nephrotoxicity, malignancies, cataractogenesis, and glaucoma, which can greatly impair quality of life for the patient. Biologics that target pathogenic T cells is an alternative approach tha

Monoclonal B cell lymphocytosis and "in situ" lymphoma.

The finding of monoclonal B-cell populations not fulfilling criteria for a lymphoid malignancy has given great impulse to study mechanisms involved in lymphomagenesis and factors responsible for the transition from B-cell precursor states to overt lymphoproliferative disorders. Monoclonal B cell expansions were initially recognized in peripheral blood of otherwise healthy subjects (thus defined monoclonal B-cell lymphocytosis, MBL) and in most cases share the immunophenotypic profile of chronic lymphocytic leukemia (CLL). The clinical relevance of this phenomenon is different according to B-ce

[Effect of some vitamins on activity of immunocompetent cells].

The content of vitamins A, E, B1, B2, B6 in lymphocytes and serum of blood was studied with simultaneous determination of CD3+, CD5+, CD4+, CD8+, CD16+, CD25+, CD71+, CD95+, and HLADR+ populations. According to cellular immunity response, patients were divided into two groups--with physiological norm and deficiency. It was established that, irrespective of the group, the content of vitamins A, E, B6 in blood serum does not change, while the concentration of B1 and B2 in the group with physiological norm is greater than that in the group with deficiency. In the group with deficiency of cellular

[Lymphocytic proliferation in gastric mucosa].

There is the substantial problem as how to exactly define the minimal requirements for the diagnosis of a marginal zone B cell lymphoma of MALT type in the stomach. Consequently, there is vigorous disagreement in the literature whether or not clonal B cell proliferation exists in chronic gastritis. This situation was the starting point of a retrospective multicenter study of German/Swiss lymph node registries. The panel contributed gastric biopsies of 26 cases of consensually diagnosed gastric marginal zone lymphoma, 18 borderline cases and 53 cases of consensually diagnosed chronic gastritis.

The triterpenoid CDDO induces apoptosis in refractory CLL B cells.

Chronic lymphocytic leukemia (CLL) cells develop chemo-resistance over time. Most anticancer agents function through induction of apoptosis, and therefore resistance against these agents is likely to be caused by selection for CLL cells with defects in the particular apoptosis pathway that is triggered by these drugs. Anticancer agents that function through alternative apoptotic pathways might therefore be useful in treating chemo-resistant CLL. Triterpenoids represent a class of naturally occurring and synthetic compounds with demonstrated antitumor activity. We examined the effects of CDDO (

Development and function of the early B cell repertoire.

The early B cell repertoire is characterized by extensive interconnectivity, autoreactivity and multispecificity. Our preliminary sequence analysis of some of the idiotype specific antibodies is beginning to provide molecular clues to explain the observed multireactivity and the expression of shared idiotypic determinants on immunoglobulins of early B cells. The VH gene rearrangements analyzed are typical of the early pre-B cell and CD5 B cell repertoire. Some of these include shared or identical CDR3 regions resulting from the use of germline VH, D and JH gene segments in the absence of N reg
