Robot-assisted stereotactic brainstem biopsy in children: prospective cohort study.

Tumours located within the brainstem comprise approximately a tenth of all paediatric brain tumours. Surgical biopsy of these tumours is technically challenging and has historically been associated with considerable risk. To this end, robot-assisted surgery theoretically allows for increased accuracy and precision. In this study we report our experience using the Neuromate robot (Renishaw, Gloucestershire, UK) to perform robot-assisted stereotactic biopsy in children with tumours located within the brainstem. An uncontrolled prospective cohort study was performed (phase II) according to the ID

Recording the human brainstem frequency-following-response in the free-field.

BACKGROUND: The human auditory brainstem frequency-following response (FFR) is an objective measure used to investigate the brainstem's encoding ability of sounds. Traditionally, FFRs are recorded under close-field conditions (earphones), but free-field stimulations (loudspeaker) have yet to be attempted, which would increase the applications of FFRs by making this technique accessible to those who cannot wear inserted transducers. Here we test the feasibility and reliability of measuring speech ABRs across free and close-field. NEW METHOD: The FFR was evoked by a 40-ms consonant-vowel (cv) /

Intraparenchymal treatment with bone marrow mesenchymal stem cell-conditioned medium exerts neuroprotection following intracerebral hemorrhage.

Intracerebral hemorrhage (ICH) is a life-threatening type of stroke. Previous studies have reported that bone marrow mesenchymal stem cells (BMSCs) may exert beneficial effects on the treatment of ICH. However, it remains unknown whether the neuroprotection exerted by BMSCs on ICH is due to the differentiation of BMSCs, or the trophic factors secreted into their conditioned medium (CM). In addition, growth‑associated protein‑43 (GAP‑43) is a protein associated with neurite extension, which may be considered a prospective therapeutic target in the treatment of ICH. The present study investigate

The development of auditory perception in children after auditory brainstem implantation.

Auditory brainstem implants (ABIs) can provide useful auditory perception and language development in deaf children who are not able to use a cochlear implant (CI). We prospectively followed up a consecutive group of 64 deaf children up to 12 years following ABI surgery. The etiology of deafness in these children was: cochlear nerve aplasia in 49, auditory neuropathy in 1, cochlear malformations in 8, bilateral cochlear postmeningitic ossification in 3, neurofibromatosis type 2 in 2, and bilateral cochlear fractures due to a head injury in 1. Thirty-five children had other congenital nonaudito

Functional recovery after transplantation of induced pluripotent stem cells in a rat hemorrhagic stroke model.

Transplantation of induced pluripotent stem cells (iPSCs) has shown promising therapeutic effects for ischemic stroke. However, it is not clear if this treatment would promote recovery after intracerebral hemorrhage (ICH). In this study, we investigated the functional outcome of iPSCs transplantation in experimental ICH in rats. IPSCs were derived from an ICH patient's fibroblasts and were injected into the ipsilateral side of ICH in rats. IPSCs transplantation significantly improved the neurological functions after ICH as compared to vehicle and fibroblast injection. The grafted iPSCs migrate

Differential expression of apoptosis-related genes in the cochlea of noise-exposed rats.

Exposure to intense noise induces apoptosis in hair cells in the cochlea. To identify the molecular changes associated with noise-induced apoptosis, we used quantitative real-time PCR to evaluate the changes in 84 apoptosis-related genes in cochlear samples from the sensory epithelium and lateral wall. Sprague-Dawley rats exposed to a continuous noise at 115 dB SPL for 2 h. The exposure caused a 40-60 dB threshold shift 4 h post-exposure that decreased to 20-30 dB 7 days post-exposure. These functional changes were associated with apoptotic markers including nuclear condensation and fragmentat

Mesenchymal stem cells transplantation could be beneficial for treatment of experimental ischemic stroke in rats.

Cell therapy is prospective, modern attempt to ischemic stroke treatment. It has been being widely worked out recently. We suggest mesenchymal stem cells (MSC) as a cell therapy agent in the therapy of this disease. Experiments were carried out in inbred male Wistar-Kyoto rats. Animals were subjected to middle cerebral artery occlusion (MCAO). MSCs were isolated from rat bone marrow, expanded in culture and labelled with vital fluorescent dye PKH-26. Then 5 x 10(6) cells were injected into the tail vein on the day of MCAO and three days later. Control group animals received PBS injection (nega

Cripto as a target for improving embryonic stem cell-based therapy in Parkinson's disease.

Embryonic stem (ES) cells have been suggested as candidate therapeutic tools for cell replacement therapy in neurodegenerative disorders. However, limitations for the use of these cells lie in our restricted knowledge of the molecular mechanisms involved in their specialized differentiation and in the risk of tumor formation. Recent findings suggest that the EGF-CFC protein Cripto is a key player in the signaling pathways controlling neural induction in ES cells. Here we show that in vitro differentiation of Cripto(-/-) ES cells results in increased dopaminergic differentiation and that, upon

Differential responses to acoustic damage and furosemide in auditory brainstem and otoacoustic emission measures.

Characteristics of distortion product otoacoustic emissions (DPOAEs) and auditory brainstem responses (ABRs) were measured in Mongolian gerbil before and after the introduction of two different auditory dysfunctions: (1) acoustic damage with a high-intensity tone, or (2) furosemide intoxication. The goal was to find emission parameters and measures that best differentiated between the two dysfunctions, e.g., at a given ABR threshold elevation. Emission input-output or "growth" functions were used (frequencies f1 and f2, f2/f1 = 1.21) with equal levels, L1 = L2, and unequal levels, with L1 = L2

Composite probability modelling of increasing resolution where diagnostic covariates are unmeasurable for some subjects.

When predictive covariates for a dichotomous outcome dependent variable are undetectable or unmeasurable for some subjects, a fact in itself that may be of considerable prognostic importance, traditionally such subjects' data are dropped from multiple logistic regression analyses. An alternative analytical algorithm is offered here for such situations. First, a series of multiple logistic regressions are applied to the complete data where covariates are initially coded as dichotomous, e.g., detectable or nondetectable. Then, a second series of logistic regressions are fitted for those subjects

Compound action potentials recorded from the intracranial portion of the auditory nerve in man: effects of stimulus intensity and polarity.

Compound action potentials (CAP) were recorded from the intracranial portion of the eighth nerve in patients with normal hearing who were undergoing neurosurgical operations for cranial nerve disorders (trigeminal neuralgia and hemifacial spasm). Brain-stem auditory-evoked potentials were recorded intraoperatively to ensure that no noticeable changes occurred in conduction in the auditory nerve as a result of surgical dissections. The CAP recorded from the middle portion of the exposed intracranial portion of the eighth nerve in response to clicks of high intensity (100-110 dB peak equivalent

[Changes in the early auditory evoked potentials during premature infancy, infancy and early childhood].

We report on the derivation of the early acoustic evoked potentials (EAEP) in 498 healthy normotrophic infants between the 32nd post-conceptional week and the end of the 3rd year of life. The changes of the wave latencies and interpeak intervals in different age periods until they reached the constant values found in adults are followed by 1528 potential derivations. The varying rate of maturation of the auditory pathway in the brain stem can be followed up well by the potential patterns. The time around the 34th post-conceptional week is characterized by a maturation episode of central and pe…

Exploring the Dual Roles of Neural Stem Cells in Glioblastoma: Therapeutic Implications and Opportunities.

Glioblastoma (GBM) is recognized as the most aggressive and lethal form of primary brain tumor, characterized by rapid proliferation and significant resistance to conventional therapies. Recent studies have illuminated the complex role of Neural Stem Cells (NSCs) in both the progression and treatment of GBM. This review examines the specific molecular pathways influenced by NSCs, focusing on critical signaling cascades such as Notch, P13K, and SHH, which are implicated in tumor development and maintenance. Furthermore, we explore the dual role of NSCs in glioblastoma, where they can act as bot

Mesenchymal Stem Cells and Their Extracellular Vesicles: Therapeutic Mechanisms for Blood-Spinal Cord Barrier Repair Following Spinal Cord Injury.

Spinal cord injury (SCI) disrupts the blood-spinal cord barrier (BSCB) exacerbating damage by allowing harmful substances and immune cells to infiltrate spinal neural tissues from the vasculature. This leads to inflammation, oxidative stress, and impaired axonal regeneration. The BSCB, essential for maintaining spinal cord homeostasis, is structurally similar to the blood-brain barrier. Its restoration is a key therapeutic target for improving outcomes in SCI. Mesenchymal stromal/stem cells (MSCs) and their secreted extracellular vesicles (MSC-EVs) have gained attention for their regenerative,
