Multi-armored mesenchymal stem cells potentiate antitumor immunity to combat ovarian cancer.

BACKGROUND: Ovarian cancer (OC) remains one of the most lethal gynecologic malignancies often resistant to immune checkpoint blockade (ICB) due to poor infiltration of cytotoxic CD8⁺ T cells and type 1 conventional dendritic cells (cDC1s) into the tumor microenvironment (TME). To overcome this, we engineered magnetic resonance imaging (MRI)-visible mesenchymal stem cells (MSCs) to co-express interleukin-15 (IL-15) for T-cell activation and XC motif chemokine ligand 1 (XCL1) for cDC1 recruitment, aiming to remodel the TME and enhance therapeutic outcomes. METHODS: MSCs were engineered via lent

ABCA8-positive lipid-metabolic CAFs mediate immunotherapy resistance in TNBC.

BACKGROUND: Triple-negative breast cancer (TNBC) is an aggressive subtype characterized by the absence of estrogen receptor, progesterone receptor, and HER2 expression, which limits the availability of targeted therapies and results in poor prognosis. Immune checkpoint blockade (ICB) therapies have emerged as promising treatments by enhancing anti-tumor immunity; however, a substantial proportion of patients with TNBC exhibit primary or acquired resistance. This resistance is largely influenced by the tumor microenvironment (TME). This study uses integrated single-cell and spatial transcriptom

Synergistic anticancer effects of ABT-199 and Vorinostat encapsulated in PLGA nanoparticles: Formulation, characterization, and antiproliferative effects against colorectal cancer cells.

The combination of ABT-199, a BCL-2 inhibitor, and Vorinostat, a histone deacetylase inhibitor, holds great potential in colorectal cancer therapy due to their synergistic effects. However, their poor solubility and bioavailability present challenges for effective treatment. This study aimed to co-encapsulate these drugs in poly(lactic-co-glycolic acid) nanoparticles to enhance stability, control drug release, and preserve their synergistic anti-proliferative effects in colorectal cancer cells. This study initially focused on evaluating the anti-proliferative activity of free ABT-199 and Vorin

A "plus one" strategy impacts replication of felid alphaherpesvirus 1, Mycoplasma and Chlamydia, and the metabolism of coinfected feline cells.

Coinfections are known to play an important role in disease progression and severity. Coinfections are common in cats, but no coinfection studies have investigated the in vitro dynamics between feline viral and bacterial pathogens. In this study, we performed co-culture and invasion assays to investigate the ability of common feline bacterial respiratory pathogens, Chlamydia felis and Mycoplasma felis, to replicate in and invade into Crandell-Rees feline kidney cells. We subsequently investigated how coinfection of these feline cells with each bacterium (C. felis or M. felis) and the common fe

RUNX1 mutations promote leukemogenesis of myeloid malignancies in ASXL1-mutated leukemia.

BACKGROUND: Additional sex combs-like 1 (ASXL1) mutations have been described in all forms of myeloid neoplasms including chronic myelomonocytic leukemia (CMML) and associated with inferior outcomes, yet the molecular pathogenesis of ASXL1 mutations (ASXL1-MT) remains poorly understood. Transformation of CMML to secondary AML (sAML) is one of the leading causes of death in CMML patients. Previously, we observed that transcription factor RUNX1 mutations (RUNX1-MT) coexisted with ASXL1-MT in CMML and at myeloid blast phase of chronic myeloid leukemia. The contribution of RUNX1 mutations in the p

TCR Triggering Induces the Formation of Lck-RACK1-Actinin-1 Multiprotein Network Affecting Lck Redistribution.

The initiation of T-cell signaling is critically dependent on the function of the member of Src family tyrosine kinases, Lck. Upon T-cell antigen receptor (TCR) triggering, Lck kinase activity induces the nucleation of signal-transducing hubs that regulate the formation of complex signaling network and cytoskeletal rearrangement. In addition, the delivery of Lck function requires rapid and targeted membrane redistribution, but the mechanism underpinning this process is largely unknown. To gain insight into this process, we considered previously described proteins that could assist in this proc

Investigation of the significance and role of androgen receptor expression in basal/squamous type urothelial carcinoma cell line J82 cells.

BACKGROUND: Urothelial carcinoma (UC) is a prevalent malignancy that shows higher incidence in males but reportedly poorer outcomes in females. Among molecular subtypes, the basal/squamous (Ba/Sq) subtype is associated with poor prognosis and therapeutic resistance. Androgen receptor (AR) signaling has been implicated in UC pathogenesis, but its precise role in the Ba/Sq subtype remains unclear. METHODS: J82 cells stably expressing AR (J82-FLAG-AR) were established by retroviral transduction. Gene and protein expression were assessed by quantitative reverse transcription PCR (RT-qPCR) and Wes

Prognostic value and potential upstream regulator of Schwann cells in esophageal squamous cell carcinoma.

BACKGROUND: Esophageal squamous cell carcinoma (ESCC) is a major malignancy of the upper gastrointestinal tract. Owing to limited actionable therapeutic targets, long-term outcomes remain unsatisfactory. This study aimed to investigate the prognostic value of non-myelinating Schwann cells (nmSCs) infiltration in ESCC and to identify potential upstream regulators and underlying mechanisms. METHODS: nmSCs infiltration was quantified in ESCC transcriptomic datasets from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) using single-sample gene set enrichment analysis (ssGSEA). Targeting CD8+ T cells with natural products for tumor therapy: Revealing insights into the mechanisms.

BACKGROUND: Despite significant advances in cancer immunotherapy over the past decades, such as T cell-engaging chimeric antigen receptor (CAR)-T cell therapy and immune checkpoint blockade (ICB), therapeutic failure resulting from various factors remains prevalent. Therefore, developing combinational immunotherapeutic strategies is of great significance for improving the clinical outcome of cancer immunotherapy. Natural products are substances that naturally exist in various living organisms with multiple pharmacological or biological activities, and some of them have been found to have anti-

Atypical Expression of Smooth Muscle Markers and Co-activators and Their Regulation in Rheumatic Aortic and Calcified Bicuspid Valves.

OBJECTIVE: We have previously reported that human calcified aortic cusps have abundant expression of smooth muscle (SM) markers and co-activators. We hypothesised that cells in bicuspid aortic valve (BAV) cusps and those affected by rheumatic heart valve (RHV) disease may follow a similar phenotypic transition into smooth muscle cells, a process that could be regulated by transforming growth factors (TGFs). AIMS: Cusps from eight patients with BAV and seven patients with RHV were analysed for early and late SM markers and regulators of SM gene expression by immunocytochemistry and compared to

Establishment of an in vitro culture system to study the developmental biology of Onchocerca volvulus with implications for anti-Onchocerca drug discovery and screening.

BACKGROUND: Infections with Onchocerca volvulus nematodes remain a threat in Sub-Saharan Africa after three decades of ivermectin mass drug administration. Despite this effort, there is still an urgent need for understanding the parasite biology especially the mating behaviour and nodule formation as well as the development of more potent drugs that can clear the developmental (L3, L4, L5) and adult stages of the parasite and inhibit parasite reproduction and behaviour. METHODOLOGY/PRINCIPAL FINDINGS: Prior to culture, freshly harvested O. volvulus L3 larvae from dissected Simulium damnosum f

Biogenesis of P-TEFb in CD4+ T cells to reverse HIV latency is mediated by protein kinase C (PKC)-independent signaling pathways.

The switch between HIV latency and productive transcription is regulated by an auto-feedback mechanism initiated by the viral trans-activator Tat, which functions to recruit the host transcription elongation factor P-TEFb to proviral HIV. A heterodimeric complex of CDK9 and one of three cyclin T subunits, P-TEFb is expressed at vanishingly low levels in resting memory CD4+ T cells and cellular mechanisms controlling its availability are central to regulation of the emergence of HIV from latency. Using a well-characterized primary T-cell model of HIV latency alongside healthy donor memory CD4+

Competition among co-infecting potyviral chimeras with adaptive, single-nucleotide differences in HCPro: Properties, and influence of environment factors on outcomes.

We previously showed that two natural, single-amino acid mutations in the HCPro of a potyviral chimera, separately increased virulence by enhancing either its silencing-suppression or proteolytic functions. To investigate how beneficial variants generated during infection compete with non-mutant forms, or among each other, we co-expressed them in pairs, in Nicotiana benthamiana. We assessed how variants co-exist and influence each other in the cell, and whether altered abiotic (elevated temperature) or biotic (a silencing-impaired plant) environmental factors, affected local and systemic infec

Laser-based therapies in the management of non-melanoma skin cancers: a narrative review.

The rising global incidence of basal cell carcinoma (BCC) and cutaneous squamous cell carcinoma (cSCC) necessitates the exploration of tissue-sparing alternatives to surgical excision, which remains the gold standard. Laser systems (CO₂, Nd: YAG, PDL) offer a less invasive approach using selective photothermolysis, addressing functional and aesthetic concerns in anatomically sensitive areas. This narrative review summarizes the current clinical data on laser-based strategies for non-melanoma skin cancers (NMSC), focusing on treatment parameters, patient/lesion selection, oncologic outcomes (cl

Doxorubicin Intercalated Co-Al Layered Double Hydroxide Nanocarrier With pH-Responsive Controlled Release and Selective Melanoma Suppression: Toward Next-Generation Nanotherapeutics.

Clinical oncology grapples with persistent challenges, particularly the rapid onset of tumor resistance and the systemic limitations of conventional chemotherapeutics such as doxorubicin (Dox). Despite its potency, Dox suffers from poor solubility, non-specific distribution, and severe toxicity, often compromising therapeutic efficacy. Addressing these obstacles, this study explores cobalt-aluminium layered double hydroxides (Co-Al LDHs) as smart nanocarriers for targeted melanoma therapy. Capitalizing on their tunable structure, high drug-loading capacity, and biocompatibility, Co-Al LDHs fac

Inavolisib for PIK3CA-mutant non-small cell lung cancer: A case report.

INTRODUCTION: PIK3CA mutations are oncogenic drivers in 2-4% of non-small cell lung cancers (NSCLC), often co-occurring with other drivers and conferring therapeutic resistance. This report is unique in describing the efficacy of the novel, highly selective PI3Kα inhibitor inavolisib in two heavily pre-treated patients with PIK3CA-mutant NSCLC harboring divergent co-drivers (EGFR and KRAS). CASE PRESENTATION: We present a 71-year-old male with EGFR/PIK3CA-mutant adenocarcinoma and brain metastases, and a 54-year-old female with KRAS/PIK3CA-mutant squamous cell carcinoma. Both patients reporte

3D-printed magnesium/strontium-co-doped calcium silicate scaffolds promote angiogenesis and bone regeneration through synergistic bioactive ion stimulation.

Bone defects resulting from trauma, infection, or surgical resection require biomaterials that support osteogenesis and vascularization for effective regeneration. In this study, we developed a 3D-printed magnesium- and strontium-co-doped calcium silicate (MSCS) scaffold using direct ink writing to optimize its bioactivity and structural integrity. X-ray diffraction confirmed the successful incorporation of Sr and Mg, leading to phase modifications that influenced ion release and degradation. Wettability and mechanical testing showed that Sr improved the stability, while Mg accelerated degrada

Low-Resistance LiFePO4 Thick Film Electrode Processed with Dry Electrode Technology for High-Energy-Density Lithium-Ion Batteries.

LiFePO4 emerges as a viable alternative to cobalt-containing cathodes, such as Li[Ni1-x-y Mn x Co y ]O2 and Li[Ni1-x-y Co x Al y ]O2. As Fe is abundant in nature, LiFePO4 is a low-cost material. Moreover, stable structure of LiFePO4 imparts long service life and thermal stability. However, the practical implementation of LiFePO4 cathode in energy storage devices is impeded by its low energy density and high ionic/electrical resistance. Herein, the LiFePO4 electrode with high active material loading and low ionic/electrical resistance through the dry process is reported for the first time. The

Alleviating Structure Collapse of Polycrystalline LiNixCoyMn1-x-yO2 via Surface Co Enrichment.

The structure collapse issues have long restricted the application of polycrystalline LiNixCoyMn1-x-yO2 (NCM) at high voltages beyond 4.4 V vs Li/Li+. Herein, for LiNi0.55Co0.12Mn0.33O2 (P-NCM), rapid surface degradation is observed upon the first charge, along with serious particle fragmentation upon repeated cycles. To alleviate these issues, a surface Co enrichment strategy is proposed [i.e., Co-enriched NCM (C-NCM)], which promotes the in situ formation of a robust surface rock-salt (RS) layer upon charge, serving as a highly stable interface for effective Li+ migration. Benefiting from th
