Acquisition of 3-D arterial geometries and integration with computational fluid dynamics.

A system for acquisition of 3-D arterial ultrasound geometries and integration with computational fluid dynamics (CFD) is described. The 3-D ultrasound is based on freehand B-mode imaging with positional information obtained using an optical tracking system. A processing chain was established, allowing acquisition of cardiac-gated 3-D data and segmentation of arterial geometries using a manual method and a semi-automated method, 3D meshing and CFD. The use of CFD allowed visualization of flow streamlines, 2-D velocity contours and 3-D wall shear stress. Three-dimensional positional accuracy wa

Cancer-specific transgene expression mediated by systemic injection of nanoparticles.

The lack of safe and efficient systemic gene delivery vectors has largely reduced the potential of gene therapy in the clinic. Previously, we have reported that polypropylenimine dendrimer PPIG3/DNA nanoparticles are capable of tumor transfection upon systemic administration in tumor-bearing mice. To be safely applicable in the clinic, it is crucial to investigate the colloidal stability of nanoparticles and to monitor the exact biodistribution of gene transfer in the whole body of the live subject. Our biophysical characterization shows that dendrimers, when complexed with DNA, are capable of

3D vs 2D qualitative and semiquantitative evaluation of the glaucomatous optic disc atrophy using computer-assisted stereophotography.

BACKGROUND AND PURPOSE: Real colour documentation of the optic nerve head (ONH) is one of the most important methods to identify the early progression of glaucomatous optic nerve damage. This study compares the qualitative and semiquantitative evaluation of ONH photographs, using a 3D time-multiplexing system and conventional 2D photography, visualised on a computer monitor. PATIENTS AND METHODS: Stereophotographs of the ONH were converted by special software in a 3D file. The same images were shown in 2D and 3D to 22 subjects with different training grades (residents and ophthalmologists). T

Quantification of facial motion for objective evaluation using a high-speed three-dimensional face measurement system--a pilot study.

OBJECTIVE: To estimate asymmetry in normal and pathological facial functions using an established computer generated objective evaluation technique. METHODS: Analysis was performed on three-dimensional (3-D) data captured using a specially designed 3-D face shape measurement system. Six healthy volunteers and six patients with Bell's palsy were analyzed for forced eye closure and grinning actions. The asymmetry was computed at locations that had the greatest effect in each action on both the left and right sides of the face, in reference to a relaxed condition. The patients' data were capture

Heart rate variability and arterial blood pressure variability show different characteristic changes during hemorrhage in isoflurane-anesthetized, mechanically ventilated dogs.

UNLABELLED: We assessed the changes in heart rate variability (HRV) and blood pressure variability (BPV) as indices of autonomic nervous system and volume status during hemorrhage in isoflurane-anesthetized, mechanically ventilated dogs. Nine dogs were used. They were sequentially subjected to withdrawal of 30% estimated blood volume and graded isoflurane inhalation of 1% and 2% followed by discontinuation of isoflurane and retransfusion. The power spectra of HRV and BPV were computed using the fast Fourier transformation, and were quantified by determining the areas of the spectrum in two com

The ability of a computer program based on the Marquette Matrix-12 short measurement matrix to replicate coding by the Minnesota ECG coding laboratory.

The study was undertaken to determine whether a computer program that uses "short measurement matrix" data from the Marquette Matrix-12 system can replicate Minnesota electrocardiogram (ECG) coding laboratory interpretations. An agreement was found between coding of median complex ECGs at the Minnesota ECG coding laboratory and coding based on Marquette Matrix-12 short measurement matrix. The comparison was based on 763 ECGs plus chest pain history and serum enzyme values for a stratified random sample of 141 patients hospitalized in 1990 or 1991 for an event coded as HICDA 410.x (acute myocar

A Functional Investigation of Antibody Fc-FcRn Variant Binding Guided by In Silico Free Energy Perturbation Methods.

Accurate calculation of energy changes upon mutation is a key requirement for the effective use of computational methods in protein design. In this study, we applied free energy perturbation (FEP) calculations to predict the effects of mutations on the binding free energy between the immunoglobulin G (IgG) antibody fragment-crystallizable (Fc) region and the neonatal Fc receptor (FcRn), an interaction that is primarily responsible for antibody half-life. We assembled an extensive experimental dataset of Fc-FcRn binding affinities for wild-type (wt) and mutant complexes, including values from l

Screw perforation features in 129 consecutive patients performed computer-guided cervical pedicle screw insertion.

STUDY DESIGN: A cross-sectional study of the data retrospectively collected by chart review. OBJECTIVES: This study aimed to clarify screw perforation features in 129 consecutive patients treated with computer-assisted cervical pedicle screw (CPS) insertion and to determine important considerations for computer-assisted CPS insertion. CPS fixation has been criticized for the potential risk of serious injury to neurovascular structures. To avoid such serious risks, computed tomography (CT)-based navigation has been used during CPS insertion, but screw perforation can occur even with the use of

A recursive enzymatic competition network capable of multitask molecular information processing.

Living cells understand their environment by combining, integrating and interpreting chemical and physical stimuli. Despite considerable advances in the design of enzymatic reaction networks that mimic hallmarks of living systems, these approaches lack the complexity to fully capture biological information processing. Here we introduce a scalable approach to design complex enzymatic reaction networks capable of reservoir computation based on recursive competition of substrates. This protease-based network can perform a broad range of classification tasks based on peptide and physicochemical in

Extracellular-Matrix-Anchored Click Motifs for Specific Tissue Targeting.

Local presentation of cancer drugs by injectable drug-eluting depots reduces systemic side effects and improves efficacy. However, local depots deplete their drug stores and are difficult to introduce into stiff tissues, or organs, such as the brain, that cannot accommodate increased pressure. We present a method for introducing targetable depots through injection of activated ester molecules into target tissues that react with and anchor themselves to the local extracellular matrix (ECM) and subsequently capture systemically administered small molecules through bioorthogonal click chemistry.

Towards X-ray free endovascular interventions - using HoloLens for on-line holographic visualisation.

A major challenge during endovascular interventions is visualising the position and orientation of the catheter being inserted. This is typically achieved by intermittent X-ray imaging. Since the radiation exposure to the surgeon is considerable, it is desirable to reduce X-ray exposure to the bare minimum needed. Additionally, transferring two-dimensional (2D) X-ray images to 3D locations is challenging. The authors present the development of a real-time navigation framework, which allows a 3D holographic view of the vascular system without any need of radiation. They extract the patient's su
