Feasibility of neuromuscular electrical stimulation for knee osteoarthritis in older women: A double-blind randomized controlled trial.

This study examined the feasibility of using neuromuscular electrical stimulation (NMES)-specifically AUSSIE and functional electrical stimulation (FES) currents-for older women with knee osteoarthritis (OA). Primary outcomes included feasibility indicators such as recruitment rate, adherence, satisfaction, and safety, while secondary outcomes assessed pain and physical function. Participants were randomly assigned to three groups: AUSSIE current, pulsed FES current, or a control group without intervention. The intervention lasted four weeks, with three 20-min sessions per week. Feasibility wa

Randomized controlled trial of an oral gastrin receptor antagonist for the treatment of postmenopausal osteoporosis.

High gastrin levels may help to explain the association between several conditions and osteoporosis, such as pernicious anemia, the use of proton pump inhibitors, and atrophic gastritis. This study aimed to determine whether administering a gastrin receptor antagonist (GRA) to older women would lower their bone turnover markers (BTM) and, therefore, be a suitable preventive measure for osteoporosis. We conducted a randomized, double-blind, placebo-controlled clinical trial to assess the efficacy, safety, and tolerability of an oral GRA (netazepide) 100 mg administered daily for 90 d in postmen

Prednisone, not vamorolone, suppresses novel serum bone and cartilage biomarkers associated with growth failure in children with Duchenne muscular dystrophy.

Corticosteroids are among the most highly prescribed drugs. Chronic treatment with corticosteroids causes osteopenia, as well as growth failure in children. Our goal was to identify serum biomarkers of prednisone-related perturbations of bone and/or cartilage metabolism associated with growth failure in children. We utilized longitudinal serum samples from a double-blind, placebo- and prednisone-controlled trial of vamorolone in 4 to < 7 years children with Duchenne muscular dystrophy assayed by proteomic methods. Vamorolone is a recently approved bone-sparing corticosteroid that has been show

Effect of a 3-week program of cane training and use on gait of individuals with Parkinson's disease: Protocol for a randomized controlled trial.

INTRODUCTION: Although studies examining the immediate effects of cane use in individuals with Parkinson's disease (PD) have reported negative outcomes, the efficacy and safety of structured training programs for assistive device use remain to be investigated. Furthermore, the limited evidence regarding patient-reported outcomes hinders the implementation of patient-centered, evidence-based practices in prescribing assistive devices for this population. OBJECTIVE: To investigate the effect of cane training and use on gait speed (primary outcome), gait confidence, cadence, step length, functio

A Digital Therapeutic Intervention for Negative Symptoms of Schizophrenia: A Randomized Clinical Trial.

IMPORTANCE: Negative symptoms of schizophrenia are a major contributor to poor outcomes, yet no US Food and Drug Administration-approved pharmacotherapies are specifically indicated for their treatment. In particular, motivation and pleasure negative symptoms are especially burdensome for patients. Psychosocial interventions can be effective, but access barriers and challenges in sustaining engagement limit their impact. Digital therapeutics are a novel modality that can deliver patient-centered interventions. CT-155/BI 3972080 (CT-155), an investigational smartphone-based digital therapeutic

Effect of Epigallocatechin Gallate Supplementation on Inflammation, Iron Status, and Metabolomic Profiles in Women With Obesity: A Pilot Randomized Controlled Trial.

Obesity in women of reproductive age is often associated with low-grade inflammation and functional iron deficiency. Epigallocatechin gallate (EGCG), a major green tea catechin, has antioxidant, anti-inflammatory, and metabolic-regulating properties but may also interfere with iron absorption. This randomized, double-blind, placebo-controlled pilot study evaluated the effects of EGCG on lipid profile, inflammation, iron status, and plasma metabolomics in women with obesity. Seventeen participants (BMI ≥30 kg/m2; age 20-44 years) received 400 mg/day EGCG (green tea extract: 98% polyphenols, 60%

Effect of L‑arginine supplementation on homocysteine and asymmetric dimethylarginine in atherosclerotic lower limb ischemia.

Chronic lower limb ischemia is a manifestation of systemic atherosclerosis frequently accompanied by lipid metabolism disorders, endothelial dysfunction, and disturbances in the L-arginine-nitric oxide (NO) pathway. Asymmetric dimethylarginine (ADMA), an endogenous inhibitor of nitric oxide synthase, and homocysteine (Hcy) are recognized markers of vascular risk. This study aimed to evaluate the effect of 2 months of oral L-arginine supplementation (6g/day) on serum ADMA and Hcy concentrations in patients with atherosclerotic lower limb ischemia, depending on lipid metabolism disorder type. Redefining the real problem in psychedelic trials: Why fighting the Lessebo matters more than blinding integrity.

Imperfect blinding is not specific to psychedelic trials. In randomized trials, treatment allocation is frequently correctly guessed, yet blinding integrity is rarely assessed outside of psychedelic research and is generally not considered a barrier in regulatory evaluation. The intense debate in psychedelics may reflect a broader double standard affecting mental health research, when uncertainties arising from imperfect blinding are confounded by those linked to patient-reported outcome measures. Indeed, people living with mental disorders are often viewed as unreliable reporters, despite wel

Effects of electroacupuncture on brain functional connectivity in patients with subjective tinnitus: A randomized controlled clinical trial☆.

BACKGROUND: Subjective tinnitus (ST) often causes significant psychological and sleep disturbances, and limited treatment options are available. Neuroimaging suggests that acupuncture modulates brain activity. This study aimed to evaluate electroacupuncture (EA) for ST treatment using resting-state functional magnetic resonance imaging (rs-fMRI) to explore its central mechanisms. METHODS: This single-center, randomized, double-blind trial enrolled 90 patients with ST (aged 18-65 years), who were randomized at a 2:1 ratio to EA (n = 60) or sham acupuncture (SA, n = 30). Participants received 2

INulin-type β-fructans supplementation to modulate gut microbiota and assessment of its effects on health status and GUT-SKIN axis parameters in patients with psoriasis (INGUTSKIN): the 8-week, randomised, double-blind, placebo-controlled clinical study protocol.

BACKGROUND: A strong, bidirectional association exists between gastrointestinal health and skin homeostasis in many chronic skin inflammations, including psoriasis (PS). It is postulated that, apart from genetic predisposition, the rise of local and systemic immune response in PS could be a consequence of intestinal dysbiosis associated with increased intestinal permeability. We hypothesised that restoring gut microbiome homeostasis and proper functioning of the intestinal barrier in PS patients would alleviate the inflammatory symptoms and severity of the skin lesions. The study aims to deter

Targeted left ventricular lead placement in biventricular pacing for heart failure: a national, multicentre, double-blind, randomised controlled trial in Denmark.

BACKGROUND: Left ventricular lead position is a key determinant of optimal biventricular pacing for heart failure. Observational data indicate better outcomes in patients with late myocardial activation at the left ventricular lead site. We aimed to test whether targeting the left ventricular lead at site of latest electrical activation improves outcomes. METHODS: In this national, randomised, controlled, double-blinded, superiority trial we included patients from all five Danish university centres performing device implantation who had heart failure and wide QRS on guideline-directed medicat
