Despite the availability of many drugs and therapies to treat malaria, many countries' national policies recommend using a single first-line therapy for most clinical malaria cases. To assess whether this is the best strategy for the population as a whole, we designed an evolutionary-epidemiological modeling framework for malaria and compared the benefits of different treatment strategies in the context of resistance evolution. Our results show that the population-wide use of multiple first-line therapies (MFT) against malaria yields a better clinical outcome than using a single therapy or a c

Nosocomial infections, those acquired during receiving healthcare services, represent a significant public health challenge, particularly when caused by multidrug-resistant (MDR) bacteria. While natural products have been widely investigated for their antibacterial properties, many show limited efficacy against nosocomial pathogens and fail to adequately address the escalating problem of antimicrobial resistance. Moreover, research on the antibacterial effects of natural product combinations remains scarce. This study investigates the antibacterial activity of black cumin (Nigella sativa) esse

BACKGROUND: Leclercia adecarboxylata is an emerging clinical pathogen within the Enterobacteriaceae family and causes infections in immunocompromised and immunocompetent patients. It frequently exhibits resistance to a broad spectrum of β-lactams as well as to colistin, which is a last-resort antibiotic against multidrug-resistant Gram-negative bacteria. The global spread of mcr genes poses a serious public health concern. This study aimed to examine the prevalence and genetic contexts of mcr genes in clinical Leclercia spp. isolates from China. RESULTS: This study identified three mobile coli

Streptococcus parasuis is currently not only an underestimated zoonotic pathogen but also a bacterial source of infection in food animals, posing a potential threat to global public health. Despite increased reports in recent years, systematic studies in Northwest China remain scarce. However, the lack of complete genomic sequence information has limited in-depth bioinformatics analysis of multidrug-resistant S. parasuis isolated from pigs. This study reports the whole-genome sequencing results of S. parasuis strain A1 isolated from pigs. The A1 genome consists of a single circular chromosome

This study reports the first isolation of a strain of Levyella massiliensis (strain HGL1) from human blood samples in China. Currently, the genomic characteristics, antibiotic resistance, and pathogenic potential of this species have not been fully studied. To systematically decipher the phylogenetic position, genomic composition, and potential pathogenicity-related factors of strain HGL1, this study employed a dual-platform sequencing strategy (PacBio Sequel II and Illumina NovaSeq PE150) for whole-genome sequencing of strain HGL1 and assessed its antibiotic susceptibility through antimicrobi

The emergence of antimicrobial resistance (AMR) in Klebsiella pneumoniae poses a significant threat to public health. This study aims to elucidate the mechanisms of colistin and fluoroquinolone resistance using whole-genome sequencing in two K. pneumoniae isolates. Two clinical isolates of K. pneumoniae that phenotypically showed resistance to colistin and ciprofloxacin but did not yield any corresponding drug resistance genes were selected from a tertiary care facility in Chennai, India. Genome sequences of the isolates were generated using the NovaSeq sequencing platform (Illumina, USA). The

This review provides an integrative analysis of the current knowledge on Klebsiella pneumoniae quorum sensing (QS) network. Although it is widely recognised as a high-priority ESKAPE pathogen, the regulatory networks coordinating its virulence and persistence remain insufficiently understood. Here, we examine QS as regulatory framework linking pathogenicity, including multidrug resistance. We first synthesise the major stages of infection, virulence factors related, and molecular basis of antibiotic resistance and immune evasion. We then focus on the four main QS systems described in K. pneumo

Residual tumor cells that persist after chemotherapy, even in minimal quantities, often exhibit drug resistance and increased invasiveness, potentially leading to tumor metastasis and recurrence. This study introduces a novel reactive oxygen species (ROS)-responsive prodrug, Dex-Dox, designed to overcome multidrug resistance (MDR) in tumor cells. The prodrug is composed of the anti-inflammatory glucocorticoid dexamethasone (Dex) conjugated with doxorubicin (Dox), a widely used antitumor agent, through an oxidative stress-responsive linker. Our in vitro findings revealed that, while Dex alone d

Avian pathogenic Escherichia coli (APEC) causes severe economic losses in duck farms through complex, frequent infections and secondary infections. In this study, 73 suspected E. coli strains were isolated from diseased ducks of three provinces in China during 2019-2020, and identified using polymerase chain reaction. The O serotypes, drug susceptibility and molecular subtyping of the strains were also analyzed. The results showed that 9 of the 73 strains belong to classical APEC serotypes, including 1 serotype O1 strain, 2 serotype O2 strains, 2 serotype O18 strains, and 4 serotype O78 strain

A Streptococcus suis strain isolated from the blood of a patient in Zhejiang Province, China, was analysed using whole-genome sequencing and tested for antimicrobial resistance. The isolated strain was identified as S. suis serotype 2, and classified to ST25 on multilocus sequence typing (MLST). The minimum core genome group of the strain was identified as Group 4, and multilocus variable-number tandem-repeat analysis (MLVA) assigned it as type 2, 4.4, 0, 9, 3, 2, 0, 0. An antimicrobial resistance analysis showed that the strain was resistant to clindamycin, tetracycline, azithromycin, and ery

While repeated Helicobacter pylori eradication attempts are increasingly common due to antimicrobial resistance, the compounding effects of multiple antibiotic courses on gastrointestinal microbiota remain unquantified. We enrolled 32 treatment-naïve patients and 84 patients with refractory H. pylori infections, defined as failure of at least two prior eradication attempts. Antibiotic susceptibility testing was conducted for refractory H. pylori isolates to assess resistance patterns. Gastric mucosal biopsies and fecal samples were obtained for 16S rRNA gene sequencing. The constant evolution of Methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant S. aureus (VRSA) constitutes a major threat to human health with limited treatment options. In our previous studies, we invented an innovative cycloberberine (CBBR) scaffold to battle MRSA and other Gram-positive strains. Herein, we report the synthesis and biological evaluations of forty-four CBBR esters derivatives, of which forty-two were new, against Gram-positive pathogens. Compound 5a demonstrated superior in vitro potency against MRSA and VISA strains, with MIC values of 0.06-0.125 μg/m

Escherichia coli recovered from dogs with clinical conditions such as urinary tract infections are often used to assess populations for resistance to critically important antimicrobials (CIAs). Despite the potential importance of such strains, the number of organisms scrutinised is very small and no information is obtained from the preponderance of normal, healthy dogs. Commensal E. coli are a valuable alternative, but little is gained if the number of isolates also remains small. In this work we demonstrate novel technology reliant on laboratory robots to examine the CIA resistance status of
