Coevolution-based protein engineering of alcohol dehydrogenase at distal sites enables enzymatic compatibility with substrate diversity and stereoselectivity.

Chiral alcohols with various substituents and functional groups are attractive synthesizers in many fields. Biocatalysts have attracted great interest for their use in " sustainable chemistry". However, substrate specificity of enzymes limits their widespread use as "generalists" in biocatalysis. In addition, engineering enzymes for simultaneously improving catalytic efficiency and stereoselectivity for structurally diverse substrates is a contemporary challenge. Inspired by naturally occurring coevolution of residues dedicated to a particular function and clustered together in space, we appli…

Synergistic defect engineering in TiO2 nanotubular electrodes for advanced electrochemical oxidation of benzene-toluene mixtures: Performance and mechanisms.

TiO2 nanotube-based (TNT) anodic oxidation methods are widely used for the removal of organic compounds due to their large surface area. However, poor conductivity and limited lifetime constrain its further application. Herein, this study provided a novel strategy to induce reduced TNT surface defects by co-doping boron (B) and manganese (Mn), which promoted its electrocatalytic activity and prolonged the lifetime. The resulting nanoelectrode exhibited projected lifetime of up to 9470.0 h at a current density of 100 mA/cm2 and achieved benzene removal efficiency of 89.4 % after 3 h, outperform

Fabrication and characterization of 3D printed agarose/poly(ethylene glycol) diacrylate/hydroxyapatite nanocomposite hydrogel for cartilage tissue engineering.

Damage to the articular cartilage can be caused by osteoarthritis (OA), exercise, cancer, and etc. A major challenge is cartilage's limited ability to self-repair after injury. In this study, the new cartilage scaffold was prepared by two biocompatible materials, agarose and poly(ethylene glycol) diacrylate (PEGDA), using a 3D printer. In this work used Taguchi-based statistical optimization to determine the optimal agarose concentration for 3D scaffolds. A key innovation was the incorporation of nanohydroxyapatite (nHA), which improved the hydrogel's viscosity and shear-thinning behavior, pri

Engineering of ω-transaminase at binding pocket and access tunnel for preparation of (R)-1-phenoxypropan-2-amine.

Transaminase-catalyzed amination of prochiral ketones is regarded as a promising route for optically active amines production. These chiral amines serve as key building blocks in the synthesis of agrochemicals and pharmaceuticals. However, the limited substrate scope and substrate/(co)product inhibition have significantly hindered the industrial-scale implementation of transaminase-catalyzed processes. Engineering of TAs at binding sites is an efficient strategy to expand the capacity of binding pocket, thereby improving the catalytic efficiency. Furthermore, modification of access tunnel is c…

Fabrication and Characterization of a Porous TiO2-Modified PEEK Scaffold with Enhanced Flexural Compliance for Bone Tissue Engineering.

Bone pathologies are becoming increasingly prevalent with an aging population, often necessitating bone grafting procedures. The current gold standard for grafting uses autologous tissue; however, this approach carries limitations such as donor site morbidity. Consequently, there is a growing interest in alternative biomaterials. Polyetheretherketone (PEEK), a thermoplastic with bone-like mechanical properties, has shown promise, although its limited bioactivity remains a critical constraint. Various functionalization strategies have been employed to enhance the biological performance of other

Engineering Perovskite Hydroxide as a Cold-Adapted Oxidase Mimic for Construction of the Robust Low-Temperature Adaptive Biosensors.

Traditional biological detection methods rely on signal amplification strategies such as enzymatic catalysis or nucleic acid amplification. However, their efficiency decreases in low-temperature environments, compromising their detection sensitivity. To break the loss of enzyme catalytic activity at low temperatures, research on cold-adaptive nanozymes has attracted much attention. Till now, only a few nanozymes have been reported to have cold-resistant catalytic properties. Here, a new type of cold-adapted nanozyme was constructed by engineering a perovskite hydroxide. The nanozyme not only b…

Approaching efficient H2O2 electrosynthesis on ZnO nanorod via a combined strategy of crystal facet-engineering and surface amorphization.

Two-electron oxygen reduction reaction (2e- ORR) offers a sustainable pathway for hydrogen peroxide production, yet its efficiency relies on cost-effective electrocatalysts with tailored active sites. Herein, a hybrid ZnO@ZnO2-x nanorod with tunable oxygen vacancies (Ov) in the outer-skin peroxidate thin film was developed as a neutral electrocatalyst for hydrogen peroxide production through cation-terminated facet-engineering and surface amorphous peroxidation. The optimized ZnO@ZnO2-x (named am-ZNR-180-24) electrocatalyst with a moderate oxygen vacancy concentration (∼17.9 at.% from XPS) on …

Dual Regulation of Molecular Rigidity and Orbital Engineering of Pt(II) Emitters for High-Performance Deep-Blue OLEDs.

Blue phosphorescent organic light-emitting diodes (PhOLEDs) face critical challenges in terms of low color purity and severe efficiency roll-off. In this study, four novel tetradentate Pt(II) emitters (PtCY, PtCY-F, PtCY-tBu, PtCY-tBuF) are designed through a synergistic strategy of molecular orbital engineering and steric hindrance effect. By introducing a bulky diisopropylbiphenyl (diPrPh) group at the N-heterocyclic carbene (NHC) moiety to increase molecular rigidity and suppress intermolecular interactions, the Pt(II) emitters achieve narrowband deep-blue emission (452-457 nm) in dichlorom…

Engineering High-Performance Polymer Dielectrics via Sea-Island Phase Separation: Synergizing Toughness, Energy Density, and Low Dielectric Loss.

In modern electronics and power systems, high-performance dielectric capacitors with metallized films are indispensable for efficient energy storage and power delivery. However, commercially available polymers like BOPP and PVDF are limited by inherent structural defects, restricting their ability to achieve high energy density and low energy loss. Glassy polymers, which offer a promising balance between these attributes, are severely restricted by their brittleness, complicating film casting and coil winding processes crucial for capacitor applications. To address these challenges, we introdu

3D Flipwell Engineering for Developing Asynchronous Systems for Toxicologic and Immunomodulatory Therapies in Bacterial, Gut, and Immune Cells.

To study the roles of bacterial secretome in immune cell polarization and gut mucosal homeostasis, we developed a 3-dimensional co-culture model using commercially available inserts in a back-to-back stacked format. We termed this system 3D Flipwells, and successfully co-cultured bacteria, colon epithelial, and immune cells in stratified layers to recapitulate the gut mucosal microenvironment. This co-culture system enables the seeding of colon epithelial cells on one side of the membrane and undifferentiated non-adherent monocytes on the other, allowing for the synchronous progression of colo

Preclinical Assessment in Juvenile Sheep of an Allogeneic Bone Tissue Engineering Product with Wharton's Jelly Mesenchymal Stromal Cells.

Secondary osteonecrosis (ON) is a common complication in paediatric cancer survivors. Combining multipotent mesenchymal stromal cells (MSCs) with core decompression surgery halts disease progression and stimulates bone regeneration. However, the success of advanced therapy medicinal products (ATMPs) requires versatile "off-the-shelf" tissue engineering products (TEPs). This study evaluated the safety and efficacy of TEPs loaded with allogeneic MSCs from Wharton's jelly (WJ-MSCs) in a large-animal model of bone regeneration to support a paediatric investigational plan for ON patients. Strain engineering of Zeeman and Rashba effects in transition metal dichalcogenide nanotubes and their Janus variants: anab initiostudy.

We study the influence of mechanical deformations on the Zeeman and Rashba effects in transition metal dichalcogenide nanotubes and their Janus variants from first principles. In particular, we perform symmetry-adapted density functional theory simulations with spin-orbit coupling to determine the variation in the electronic band structure splittings with axial and torsional deformations. We find significant effects in molybdenum and tungsten nanotubes, for which the Zeeman splitting decreases with increase in strain, going to zero for large enough tensile/shear strains, while the Rashba split
