Yeast Display of Single-Chain Fc Fragments with Modulated Binding of Effector Molecules.

Yeast display platform has proven instrumental for directed evolution of diverse proteins towards altered binding specificity, affinity, or improved biophysical properties. Particularly in antibody display, this method has delivered variants that are featured today as integral parts of approved therapeutic agents. While broadly used for discovery of binding agents, yeast display has also been applied for optimization of properties of Fc fragment. Among the bispecific antibody formats, which come in a variety of different molecular architectures, the single-chain Fc is a less explored alternati

A new record of triggerfish (Tetraodontiformes: Balistidae) from China with a complete mitogenome.

We report the first confirmed record of the largescale triggerfish, Canthidermis macrolepis (Boulenger, 1888), from the South China Sea, based on a specimen collected near Wenchang, Hainan, China. Sequencing of the complete mitochondrial genome (16,527 bp) revealed typical vertebrate mitogenome organisation, comprising 37 genes and a control region. The genome displayed A + T bias (55.1%) and strong purifying selection across all protein-coding genes. Phylogenetic analyses with both cytochrome c oxidase subunit I (COI) and whole mitogenome datasets confirmed the monophyly of Canthidermis and i

CRISPR/Cas-Mediated Genome Editing for Developing Herbicide Tolerant Rice: A Step-by-Step Protocol.

Weed management in rice cultivation has predominantly relied on acetolactate synthase (ALS) and acetyl-CoA carboxylase (ACCase) inhibiting herbicides, particularly against Echinochloa spp. (watergrass). However, these herbicides carry a high risk of resistance evolution, as evidenced by the numerous resistant biotypes reported worldwide. The emergence of herbicide resistance necessitates innovative and sustainable weed control strategies. Genome editing, particularly through the CRISPR/Cas system, provides a precise and efficient platform for introducing targeted genetic modifications to devel

Dynamic conformations of fluorophores on self-labeling protein tags.

Self-labeling protein tags (SLPs) enable protein-specific conjugation of synthetic fluorophores for versatile bioimaging, yet their rational engineering has largely relied on static structural models. Here, we reveal that crystal structures of SLP-dye complexes generally bear packing artifacts due to strong π-π interactions of dyes, frequently failing to profile the chromophore microenvironment. By integrating molecular dynamics (MD) with crystallography, we elucidate distinct dynamic behaviors of fluorophores across major SLPs: quasi-free diffusion in SNAP-tag, balanced hydrophobic-polar inte

Structural and Functional Perspectives of α-L-Rhamnosidase in Glycoside Biotransformation, Therapeutic Applications, and Enzyme Engineering.

α-L-Rhamnosidase is a glycoside hydrolase (GH) that cleaves α-L-rhamnosidic bonds in diverse natural substrates, including oligosaccharides, glycoproteins, flavonoids, and glycolipids that contain terminal L-rhamnose. Owing to its broad substrate specificity, it has attracted growing interest in biotechnology, food processing, pharmaceuticals, and natural product modification. Fungal and bacterial α-L-rhamnosidases show diversity in molecular architecture, catalytic efficiency, thermostability, and pH tolerance, while sharing a conserved inverting mechanism mediated by acidic residues within t

Acute Intestinal Obstruction Caused by Dual-Component Metastasis From Combined Small Cell Lung Cancer: A Rare Case and Literature Review.

This case report describes a 75-year-old male patient who was admitted due to acute intestinal obstruction. The patient had a history of small cell lung cancer. Emergency surgery pathology confirmed the presence of primary composite small cell lung cancer with rare independent dual component metastasis: The colonic lesion was metastatic poorly differentiated adenocarcinoma, and the mesenteric lymph nodes were metastatic small cell carcinoma. Postoperatively, the patient rapidly progressed to septic shock and multiple organ failure, ultimately resulting in death. To our knowledge, this case is

Serpin-driven green camouflage and NIR fluorescence in frogs.

Biliverdin is one of the few endogenous green pigments in animals, yet its use in camouflage and fluorescence is limited by rapid degradation or excretion. Multiple arboreal frogs have overcome this constraint by evolving biliverdin-binding serpins (BBSs)-proteins that stabilize biliverdin. Here, we show that BBSs have convergently evolved high-affinity biliverdin binding comparable to hormone-receptor interactions and tune biliverdin's spectral properties to produce leaf-like green coloration. We further show that BBSs from independent lineages exhibit distinct spectral properties, including

Molecule Modification Enhancing the CO2 Electroreduction Performance on Au/Ag Bimetallic Spiky Nanostars Investigated by SERS.

The electrochemical carbon dioxide (CO2) reduction reaction is a promising approach to mitigate atmospheric CO2 accumulation and achieve carbon neutrality, but it is severely limited by low catalytic activity, poor selectivity, and the competing hydrogen evolution reaction (HER). Herein, a 3-aminopropyltriethoxysilane (APTES) molecule modification of spiky bimetallic Au/Ag nanostars is designed to enhance the catalytic activity and selectivity and suppress the HER for the CO2 electroreduction to CO. A remarkable CO Faraday efficiency was increased by 20% from 61% to 81% at -0.9 V vs reversible

Molecular mimicry in the agroecosystem: A new paradigm for understanding how pesticide residues drive the emergence of antimicrobial resistance.

Antimicrobial resistance (AMR) is a mounting global crisis, with environmental dissemination of antibiotic resistance genes (ARGs) emerging as a critical driver. Agroecosystems, chronically exposed to complex mixtures of bioactive chemicals, including pesticides, represent an underrecognized hotspot for AMR evolution. This review synthesizes established mechanisms by which pesticides select for resistance and introduces a novel hypothesis: molecular mimicry as a hidden driver. Evidence highlights three key pathways: cross-resistance via multidrug efflux pumps; coselection on mobile genetic ele

The Extreme Diversity Of Retinal Amacrine Cells Has Deep Evolutionary Roots.

Amacrine cells (ACs) comprise a heterogeneous class of inhibitory neurons in the vertebrate retina, exhibiting morphological and functional complexity rivaling that of cortical interneurons. Here, we integrate single-cell and single-nucleus transcriptomic atlases from 24 vertebrate species to reconstruct the evolutionary origins of this extreme diversity. We identify 42 orthologous AC types (oACs), most of which exhibit a one-to-one correspondence across amniotes and, in many cases, across vertebrates. While core molecular identities are conserved, AC types vary in abundance and gene expressio

Anatomical dynamics define cancer cachexia subtypes and identify systemic inflammation as a marker of lethal wasting.

Cancer cachexia is a wasting syndrome that remodels the anatomy of the patient. How this remodeling unfolds across tissues, whether it defines distinct disease states, and how these states relate to underlying biology remain unknown. We used longitudinal computed tomography imaging from 4,516 patients to quantify evolution of muscle, adipose, and organs during cachexia. Across two independent institutional cohorts, unsupervised analysis identified three reproducible anatomical subtypes of cachexia, including an inflammatory Type A marked by progressive hepatosplenic enlargement and inferior su

A 3D amphioxus brain atlas illuminates the blueprint of the ancestral chordate brain and highlights unique vertebrate innovations.

Comparative studies of vertebrate brains have elucidated the evolution of morphological diversity; yet, the fundamental chordate brain blueprint and the emergence of vertebrate-specific innovations remain elusive. To reconstruct the cellular and molecular origins of brain architecture in chordates, we combined single-nucleus RNA sequencing with spatial transcriptomics to generate a three-dimensional cell atlas of the central nervous system (CNS) in the early-branching chordate amphioxus. This atlas reveals a tripartite anterior-posterior organization comprising a rostral retinal/hypothalamic r

Cellular Models and Functional Assays for Assessing CFTR Function: A Comprehensive Review.

Cystic fibrosis (CF) is a genetic disorder caused by dysfunction of the CFTR chloride ion channel. Progress in molecular understanding and therapy development relies on advanced cellular models and robust assays for evaluating CFTR function. This review traces the evolution of in vitro models, from primary and immortalized cell lines to patient-specific induced pluripotent stem cells (iPSCs) and complex three-dimensional systems. These advanced models, including air-liquid interface (ALI) cultures, organoids, and microfluidic organ-on-a-chip platforms, enable recapitulation of tissue architect

ZMAT3 alleviates cell death in Fanconi anemia via adaptation of sphingolipid metabolism.

Fanconi anemia (FA) is characterized by defective DNA repair and chronic p53 activation, predisposing to acute myeloid leukemia through persistent genomic instability. The molecular adaptations enabling cell survival under chronic stress remain poorly understood. This study investigates the role of ZMAT3, a p53 responsive RNA binding protein overexpressed in FA deficient cells, to elucidate its role in cellular adaptation. Using FA patient fibroblasts and Fancg KO mouse models, we launched transcriptomic profiling, lipidomic, and functional analyses. RNA-seq and lipidomic analyses revealed une

SARS-CoV-2 intra-host recombination promotes epistatic spike interactions and temperature-dependent adaptation.

Prolonged infections in immunocompromized individuals are associated with accumulation of unexpected combinations of mutations, as exemplified by the Omicron variant. Here, we show that intra-host evolution during a chronic SARS-CoV-2 infection in the pre-Omicron era was associated with mutations that modulate antibody evasion, cell entry efficiency, spike processing, and fusogenicity. Single spike N-terminal mutations S13I and W64G exerted deleterious impacts on S1/S2 cleavage, syncytia formation, and cell entry but combined through an inferred recombination event with P330S to restore functi

Genomic characterisation of Crimean-Congo haemorrhagic fever virus (CCHFV) in Tajikistan identifies a novel reassortant virus.

Crimean-Congo haemorrhagic fever virus (CCHFV) is an important human tick-borne pathogen, able to cause severe haemorrhagic fever. CCHFV is endemic in Tajikistan, which records between 5-38 cases of CCHF a year from southern regions. Molecular surveillance of CCHFV is crucial to implement effective prevention and control strategies, understand viral evolution, study transmission dynamics, and develop effective diagnostics, therapeutics, and vaccines. While the presence of Asia-1 and Asia-2 genotypes has been previously reported, only two historical samples from Tajikistan have been fully seque

Comparative analysis of rhesus macaque and human placental organoids highlights evolutionary differences in placentation.

Throughout evolution, the placenta has diversified in structure and composition while maintaining its essential role in supporting fetal development. Trophoblasts, cells responsible for nutrient exchange and immune modulation, are a conserved feature of all placentas. Although primate placentas share morphological similarities, species-specific differences in gene expression remain poorly characterized, largely due to the lack of tractable in vitro models. To address this gap, we developed rhesus macaque placental organoids representing trophoblast and maternal-derived decidual cell types and

Biodegradation of Benzo(a)pyrene in Contaminated Soil: Plant and Microorganism Contributions from Isotope Tracing.

Biological degradation effectively removes benzo(a)pyrene (BaP) from contaminated soil; however, knowledge regarding the contributions of plant absorption, microbial degradation, and volatilization to BaP removal remains limited. In this study, the BaP removal pathway in contaminated soil was investigated. The structural evolution of the microbial community in contaminated soil was revealed using a comparative experimental study. BaP, as a representative of high-molecular-weight polycyclic aromatic hydrocarbons, was removed from freshly contaminated soil by microbial degradation, plant absorpt

Molecularly Crosslinked Dense Ni-N4-O/Ni Clusters for Scalable CO2 Electroreduction in Acidic and Alkali-Metal-Free Media.

Electrochemical CO2 reduction (ECO2R) in pure acid or water offers a promising solution to carbon loss and salt precipitation. However, these harsh conditions pose significant challenges, including intensified hydrogen evolution reaction (HER). Herein, guided by density functional theory (DFT) calculations, the Ni-N4-O/Ni clusters are identified as active sites that can simultaneously promote *COOH formation and suppress HER. Based on this insight, a molecular-crosslinking strategy is developed to construct a NiX@NiN4-O catalyst with high-density Ni-N4-O/Ni clusters. The resulting catalyst ach
