A new look at the etiology and pathogenesis of early-onset periodontitis. Cementopathia revisited.

For more than 20 years, investigators have unanimously agreed that human periodontitis is caused by bacterial colonization of the surfaces of the teeth and their extension apically. Recently, several investigators have demonstrated that most, if not all, individuals manifesting the early-onset, aggressive forms of periodontitis have abnormalities in their peripheral blood phagocytes which can be detected as abnormal cell motility or adherence. Because of the major role of these cells, especially the neutrophilic granulocytes, in host defense against the microbial onslaught at the gingival sulc

Cellular immunity to xenogeneic corneal grafts in rabbits.

We wished to determine whether a small orthotopic corneal xenograft could sensitize a host systemically. Ten rabbits were grafted with interlamellar bovine cornea and pre- and postoperative blood samples tested by the H3 thymidine incorporation test (micro-method). The leucocyte migration inhibition test was applied in 3 animals. A skin test was performed on all rabbits in the 4th postoperative week. The challenging antigen used was a homogenate of pooled bovine corneas. Clinically and histologically each rabbit showed a characteristic corneal graft reaction. Where performed, leucocyte migrati

Development of a gene-editing strategy to overcome genetic intractability in Lactobacillus johnsonii.

Genetic editing in lactobacilli is often constrained by their robust defense mechanisms against foreign DNA and low transformation efficiency, resulting in pronounced species- and strain-specific variability in the performance of genetic tools. In this study, we developed a markerless gene editing system for Lactobacillus johnsonii MR1, a Jackson mouse isolate with a closed genome and well-documented host immunomodulatory properties. Initial testing revealed that existing transformation and gene-editing protocols produced low transformation efficiency, poor reproducibility, and exhibited poor

Integrated dual transcriptome sequencing and experimental validation reveal potential mechanisms of baicalin against pneumocystis pneumonia in immunosuppressed rats.

BACKGROUND: Pneumocystis pneumonia (PCP) remains a major cause of morbidity and mortality in immunocompromised individuals. Although baicalin (Ba), a natural bioactive flavonoid, has demonstrated protective and therapeutic effects against PCP, its molecular mechanisms remain undefined. We employed dual RNA sequencing (dual RNA-seq) to characterize host and pathogen transcriptional responses to Ba treatment in an immunosuppressed rat model of PCP. METHODS: Comparative transcriptomic analyses identified differentially expressed genes in both the host and Pneumocystis, followed by Gene Ontology,

Integrating machine learning and WGCNA to identify core candidate genes and decipher immune-related molecular networks in TcdA/TcdB-associated CDI.

Clostridioides difficile infection (CDI) remains a leading cause of healthcare-associated diarrhea and is characterized by high recurrence rates and inadequate predictive tools despite available therapeutic options. Current toxin- or nucleic acid-based assays do not fully capture host inflammatory dynamics, highlighting the need for mechanistically informed host-response studies that may complement existing microbiological approaches. This study aims to clarify the molecular mechanisms through which Clostridioides difficile (C. difficile) toxins toxin A (TcdA) and toxin B (TcdB) induce CDI and

Varying conjunctival immune response adaptations of house finch populations to a rapidly evolving bacterial pathogen.

Pathogen adaptations during host-pathogen co-evolution can cause the host balance between immunity and immunopathology to rapidly shift. However, little is known in natural disease systems about the immunological pathways optimised through the trade-off between immunity and self-damage. The evolutionary interaction between the conjunctival bacterial infection Mycoplasma gallisepticum (MG) and its avian host, the house finch (Haemorhous mexicanus), can provide insights into such adaptations in immune regulation. Here we use experimental infections to reveal immune variation in conjunctival tiss

Systematic investigation of the Borrelia miyamotoi spirochetes in ticks, wildlife and domestic animal hosts in Yunnan province, Southwest China.

BACKGROUND: Borrelia miyamotoi is a spirochete species transmitted via hard ticks. Following its discovery in Japan, this pathogen has been detected around the world, and is increasingly confirmed as a human pathogen causing febrile disease, namely relapsing fever. Its presence has been confirmed in the Northeast China. However, there is little information regarding the presence of B. miyamotoi and other hard-tick-borne relapsing fever spirochetes in southern China including Yunnan province, where tick and animal species are abundant and many people both inhabit and visit for recreation. METH

High-Throughput Screening Identifies Mixed-Lineage Kinase 3 as a Key Host Regulatory Factor in Zika Virus Infection.

The Zika virus (ZIKV) life cycle involves multiple steps and requires interactions with host factors. However, the inability to systematically identify host regulatory factors for ZIKV has hampered antiviral development and our understanding of pathogenicity. Here, using a bioactive compound library with 2,659 small molecules, we applied a high-throughput and imaging-based screen to identify host factors that modulate ZIKV infection. The screen yielded hundreds of hits that markedly inhibited or potentiated ZIKV infection in SNB-19 glioblastoma cells. Among the hits, URMC-099, a mixed-lineage

A Novel, Extremely Elongated, and Endocellular Bacterial Symbiont Supports Cuticle Formation of a Grain Pest Beetle.

The saw-toothed grain beetle, Oryzaephilus surinamensis (Silvanidae), is a cosmopolitan stored-product pest. Early studies on O. surinamensis in the 1930s described the presence of peculiar bacteriomes harboring endosymbiotic bacteria in the abdomen. Since then, however, the microbiological nature of the symbiont has been elusive. Here we investigated the endosymbiotic system of O. surinamensis in detail. In the abdomen of adults, pupae, and larvae, four oval bacteriomes were consistently identified, whose cytoplasm was full of extremely elongated tubular bacterial cells several micrometers wi

Differential quantitative proteomic profiling of Leishmania infantum and Leishmania mexicana density gradient separated membranous fractions.

UNLABELLED: Leishmaniasis, caused by infection with Leishmania, is a major public health concern affecting more than 20million people globally. Leishmania has a digenetic lifecycle consisting of an extracellular flagellated promastigote, adapted to live in the mid-gut of the sand fly host and an aflagellated intracellular amastigote that resides within the macrophage of the mammalian host. Leishmania mexicana and Leishmania infantum are causative agents of cutaneous and visceral leishmaniasis, respectively. Membrane proteins play a pivotal role in host-pathogen interactions and in regulatory p

Evolutionary history of mammalian sucking lice (Phthiraptera: Anoplura).

BACKGROUND: Sucking lice (Phthiraptera: Anoplura) are obligate, permanent ectoparasites of eutherian mammals, parasitizing members of 12 of the 29 recognized mammalian orders and approximately 20% of all mammalian species. These host specific, blood-sucking insects are morphologically adapted for life on mammals: they are wingless, dorso-ventrally flattened, possess tibio-tarsal claws for clinging to host hair, and have piercing mouthparts for feeding. Although there are more than 540 described species of Anoplura and despite the potential economical and medical implications of sucking louse i

Novel dual-gene target and probe-based real-time PCR for the detection and differentiation of Salmonella Dublin.

Salmonella enterica serovar Dublin (S. Dublin) is a host-adapted cattle pathogen that causes systemic disease, chronic carriage, and environmental contamination. Early detection of subclinical infections remains challenging due to low bacterial loads and the limitations of conventional diagnostics. We developed a duplex TaqMan real-time PCR assay targeting a chromosomal locus (SeD_A1104) and a plasmid-encoded gene (vagD). The assay demonstrated strong linearity (R² > 0.99) with amplification efficiencies of 91%-102% and detection limits as low as 1.9 × 10¹ genome copies per reaction. Specifici

Preparedness, prevention and control related to zoonotic avian influenza.

A risk assessment framework was developed to evaluate the zoonotic potential of avian influenza (AI), focusing on virus mutations linked to phenotypic traits related to mammalian adaptation identified in the literature. Virus sequences were screened for the presence of these mutations and their geographical, temporal and subtype-specific trends. Spillover events to mammals (including humans) and human seroprevalence studies were also reviewed. Thirty-four mutations associated with five phenotypic traits (increased receptor specificity, haemagglutinin stability, neuraminidase specificity, enhan

Gut microbiota analysis reveals microbial signature for multi-autoimmune diseases based on machine learning model.

INTRODUCTION: Human microbiota is a major factor contributing to the immune system, offering an opportunity to develop non-invasive methods for disease diagnosis. In some research on Autoimmune Diseases (AIDs), gut microbiota variation has been observed. However, there remains a paucity of research that explores the potential of gut microbiota as a microbial signature for the classification and diagnosis of multi-AIDs. METHODS: In this study, we analyzed 1,954 gut microbiota sequencing datasets from public databases collected from 1,043 patients with 10 AIDs to identify common or unique micro

Rapid visual detection of hepatitis E virus combining reverse transcription recombinase-aided amplification with lateral flow dipstick and real-time fluorescence.

UNLABELLED: Hepatitis E virus (HEV) is a globally prevalent zoonotic pathogen that is primarily spread through the fecal-oral route, such as by consuming undercooked or contaminated pork. HEV infection leads to an estimated 3.3 million symptomatic cases of viral hepatitis and 70,000 deaths in human populations each year. Therefore, a rapid and accurate method for detecting HEV in serum or stool samples is essential. In this study, we aimed to develop and evaluate two methods for the rapid and convenient detection of HEV RNA: reverse transcription recombinase-aided amplification with lateral fl

Development of a visual rapid detection method for aquatic animal-derived Trypanosoma via MIRA-LFD.

Trypanosoma is a genus of flagellated protozoan parasites that commonly infect the blood of various vertebrates, including aquatic animals. Under natural conditions, these pathogens are vectored by leeches and may induce clinical manifestations such as anemia, edema, and appetite suppression in hosts. Prolonged infection can lead to severe weight loss, ultimately contributing to economic losses in aquaculture. In this study, we developed a rapid diagnostic assay combining Multienzyme isothermal rapid amplification (MIRA) with Lateral flow dipstick (LFD) visualization for detecting aquatic anim

Navigating the fungal battlefield: cysteine-rich antifungal proteins and peptides from Eurotiales.

Fungi are ubiquitous in the environment and play a key role in the decomposition and recycling of nutrients. On the one hand, their special properties are a great asset for the agricultural and industrial sector, as they are used as source of nutrients, producers of enzymes, pigments, flavorings, and biocontrol agents, and in food processing, bio-remediation and plant growth promotion. On the other hand, they pose a serious challenge to our lives and the environment, as they are responsible for fungal infections in plants, animals and humans. Although host immunity opposes invading pathogens,

Interleukin (IL)-6 and IL-10 Are Up Regulated in Late Stage Trypanosoma brucei rhodesiense Sleeping Sickness.

BACKGROUND: Sleeping sickness due to Trypanosoma brucei rhodesiense has a wide spectrum of clinical presentations coupled with differences in disease progression and severity across East and Southern Africa. The disease progresses from an early (hemo-lymphatic) stage to the late (meningoencephalitic) stage characterized by presence of parasites in the central nervous system. We hypothesized that disease progression and severity of the neurological response is modulated by cytokines. METHODS: A total of 55 sleeping sickness cases and 41 healthy controls were recruited passively at Lwala hospit
