Unraveling the Role of Proteinopathies in Parasitic Infections.

Proteinopathies, characterized by the misfolding, aggregation, and deposition of proteins, are hallmarks of various neurodegenerative and systemic diseases. Increasingly, research has highlighted the role of protein misfolding in parasitic infections, unveiling intricate interactions between host and parasite that exacerbate disease pathology and contribute to chronic outcomes. The life cycles of parasitic protozoa, including Plasmodium, Toxoplasmosis, and Leishmania species, are complicated and involve frequent changes between host and vector environments. Their proteomes are severely stresse

Unraveling the interplay of neuroinflammatory signaling between parenchymal and meningeal cells in migraine headache.

BACKGROUND: The initiation of migraine headaches and the involvement of neuroinflammatory signaling between parenchymal and meningeal cells remain unclear. Experimental evidence suggests that a cascade of inflammatory signaling originating from neurons may extend to the meninges, thereby inducing neurogenic inflammation and headache. This review explores the role of parenchymal inflammatory signaling in migraine headaches, drawing upon recent advancements. BODY: Studies in rodents have demonstrated that sterile meningeal inflammation can stimulate and sensitize meningeal nociceptors, culminati

Commensal gut microbiota-derived acetate and propionate enhance heart adaptation in response to cardiac pressure overload in mice.

Background: The gut microbiota plays a vital role in maintaining tissue homeostasis and regulating disease pathophysiology; however, the underlying mechanisms remain to be elucidated. We previously showed that mice depleted of gut microbiota with antibiotics (ABX mice) were more prone to cardiac rupture after infarction, suggesting that the gut microbiota impacts cardiac structural remodeling following injury. Here, we aimed to determine whether the gut microbiota is required for adaptive cardiac remodeling in response to pressure overload stress. Methods: Transverse aortic constriction (TAC)

Oxidation of Sperm DNA and Male Infertility.

One important reason for male infertility is oxidative stress and its destructive effects on sperm structures and functions. The particular composition of the sperm membrane, rich in polyunsaturated fatty acids, and the easy access of sperm DNA to oxidative damage due to sperm cell specific cytologic and metabolic features (no cytoplasm left and cells unable to mount stress responses) make it the cell type in metazoans most susceptible to oxidative damage. In particular, oxidative damage to the spermatozoa genome is an important issue and a cause of male infertility, usually associated with si

l-Glutamine Represses the Unfolded Protein Response in the Small Intestine of Weanling Piglets.

BACKGROUND: Dysfunction of the endoplasmic reticulum (ER) results in apoptosis, inflammation, and enhanced proteolysis in the small intestine of humans and animals. l-Glutamine (Gln) is required for intestinal mucosal homeostasis in piglets. However, a functional role of the ER in the enterocytes of weanling piglets and its contribution to intestinal mucosal integrity remain largely unknown. OBJECTIVE: This study was conducted to test the hypothesis that preweaning administration of Gln alleviates the activation of unfolded protein response (UPR) in the small intestine of weanling piglets. The effect of intravenous corticotropin-releasing hormone administration on esophageal sensitivity and motility in health.

Esophageal hypersensitivity is important in gastroesophageal reflux disease (GERD) patients who are refractory to acid-suppressive therapy. Stress affects visceral sensitivity and exacerbates heartburn in GERD. Peripheral CRH is a key mediator of the gut stress response. We hypothesize that CRH increases esophageal sensitivity and alters esophageal motility in health. Esophageal sensitivity to thermal, mechanical, electrical, and chemical stimuli was assessed in 14 healthy subjects after administration of placebo or CRH (100 μg iv). Perception scores were assessed for first perception, pain pe

Invasive guests: the role of non-oral bacteria in oral dysbiosis and systemic health.

BACKGROUND: The human oral cavity harbors a diverse microbiota essential for maintaining oral and systemic health. While most resident microorganisms are commensals adapted to specific oral niches, increasing evidence highlights the presence of non-oral bacteria, including Helicobacter pylori, Pseudomonas aeruginosa, Klebsiella pneumoniae, Enterobacteriaceae, Staphylococcus aureus, Acinetobacter baumannii, Enterococcus faecalis and Clostridioides difficile. Traditionally associated with other body sites, these bacteria may enter the oral cavity and, under favorable ecological conditions, persi

Thymoquinone exerts anti-inflammatory effects in multiple disease models by regulating immune-inflammatory homeostasis.

Thymoquinone (TQ), an abundant phytochemical identified from Nigella sativa seeds of the Ranunculaceae family, is recognized for its multifaceted biological effects involving inflammatory regulation, oxidative stress control, and immune modulation. This review summarizes the anti-inflammatory effects of TQ across inflammation-related diseases and integrates bibliometric analysis, network pharmacology, and molecular docking approaches to explore its potential cross-disease immunoregulatory mechanisms. Accumulating evidence suggests that TQ may exert immunoregulatory effects through modulation o

Ferroptosis-related hub genes and immune infiltration characteristics in Duchenne muscular dystrophy: a molecular framework with implications for aging-related muscle degeneration.

Sarcopenia, the age-related loss of skeletal muscle mass and function, is a major driver of frailty in older adults, yet its molecular basis remains incompletely defined. Ferroptosis, an iron-dependent regulated cell death, has emerged as a link between iron dysregulation and muscle wasting in aging. However, ferroptosis-related signatures and their interplay with immune remodeling in degenerating muscle remain uncharacterized. DMD shares selected molecular and pathological features with aging-related muscle degeneration, including iron overload, oxidative stress, and impaired regenerative cap

Biomarkers in heart failure.

Heart failure (HF) is a leading cause of morbidity and mortality worldwide, characterized by a complex pathophysiology and heterogeneous clinical trajectories. Circulating biomarkers have progressively moved from ancillary diagnostic tools to indispensable instruments that inform diagnosis, prognostication, and therapeutic guidance. B-type natriuretic peptide (BNP) and N-terminal pro-B-type natriuretic peptide (NT-proBNP) remain the cornerstones of biomarker-guided management, yet their interpretation is influenced by age, renal function, obesity, and atrial fibrillation. Cardiac troponins, or

From molecules to field: Integrated insights into cuticle-mediated drought tolerance in plants.

Drought stress exacerbates non-stomatal water loss, which hinders agricultural growth and global food security. The cuticle, a layer of cutin, waxes, and other polymers, protects plants from the negative impact of drought. One crucial adaptation that has enabled terrestrial plants to survive and flourish in drought-prone regions is the development of cuticle structures. In this review, we (1) provide an overview of the molecular and enzymatic pathways involved in the biosynthesis of cutin and wax; (2) analyze important regulatory networks that control the formation of cuticles, such as abscisi

Dipsacoside B alleviates experimental colitis by reshaping gut microbiota and metabolically regulating the balance of macrophage polarization.

Gut dysbiosis-driven macrophage polarization plays a critical role in the pathogenesis of ulcerative colitis (UC). Dipsacoside B (DB), a natural saponin, possesses potential anti-inflammatory properties; however, its influence on mucosal immunity and the gut microbiota remains to be elucidated. To evaluate the therapeutic effects of DB, this study employed a DSS-induced colitis model in C57BL/6 mice, testing three different doses. The role of the gut microbiota was investigated through antibiotic-induced depletion and fecal microbiota transplantation (FMT). Both in vivo and in vitro experiment

Genome-Wide Identification and Analysis of BAHD Acyltransferases Involved in Anthocyanin Biosynthesis in Perilla frutescens.

Perilla frutescens (L.) Britt., a medicinal and edible herb, is valued for its diverse leaf coloration, attributed to varying levels of anthocyanin accumulation. The primary anthocyanins in P. frutescens are acylated cyanidin glycosides; however, the enzymes facilitating the acylation process have yet to be characterized. BAHD acyltransferases, known to catalyze such modifications, remain uncharacterized in P. frutescens. To systematically identify potential genes that may be associated with this function, we performed a comprehensive genome-wide analysis of the BAHD acyltransferase family in

Development and Characterization of a Fully Human Recombinant MPO-ANCA Monoclonal Antibody as Candidate Quality Control Material.

AIM: MPO-ANCA detection suffers prominent inter-assay variability due to a shortage of standardized, sustainable quality control (QC) materials. Existing patient-pooled sera have limited supply, while chimeric monoclonal antibodies fail to fully mimic human autoantibodies. This study aimed to construct a fully human recombinant MPO-ANCA monoclonal antibody and systematically characterize its analytical performance as a candidate QC material for multi-platform MPO-ANCA immunoassay evaluation. METHODS: A human anti-MPO scFv phage display library was constructed using peripheral blood mononuclea

Molecular immunopharmacology of traditional Chinese medicine-derived compounds in membranous nephropathy: mechanistic insights into immune aging and kidney essence deficiency.

Membranous nephropathy (MN) is a prototypical immune-mediated glomerular disease characterized by the formation of autoantibodies targeting podocyte antigens, deposition of subepithelial immune complexes, and activation of complement pathways leading to podocyte injury and proteinuria. The discovery of target antigens such as the M-type phospholipase A2 receptor (PLA2R) and thrombospondin type-1 domain-containing 7A (THSD7A) has substantially advanced the understanding of MN immunopathogenesis. Despite these advances, current therapeutic approaches remain limited by incomplete response rates,
