Role of the Kupffer cell in mediating hepatic toxicity and carcinogenesis.

Kupffer cells are resident macrophages of the liver and play an important role in its normal physiology and homeostasis as well as participating in the acute and chronic responses of the liver to toxic compounds. Activation of Kupffer cells directly or indirectly by toxic agents results in the release of an array of inflammatory mediators, growth factors, and reactive oxygen species. This activation appears to modulate acute hepatocyte injury as well as chronic liver responses including hepatic cancer. Understanding the role Kupffer cells play in these diverse responses is key to understanding

Mogroside V Promotes Brown Adipose Thermogenesis via FTO-m6A-LETMD1-UCP1 Pathway to Combat Obesity.

Obesity is a chronic, recurrent, and progressive disease caused by energy imbalance. N6-methyladenosine (m6A) modification regulates fat thermogenesis, potentially counteracting obesity. Mogroside V (MV) is a triterpene glycoside extracted from Siraitia grosvenorii and has shown beneficial effects in models of obesity. However, its underlying mechanism remains unknown. The objective of this study is to investigate the effects of MV on adipose thermogenesis and explore the mechanism of m6A modifications in mediating these effects. Mitochondrial respiration analysis, histological and molecular b

Transcriptomic insights into renal osmoregulation and environmental adaptation in two hammerhead shark species (Sphyrna zygaena and Sphyrna lewini).

Elasmobranchs maintain osmotic balance through specialized renal mechanisms regulating ion transport, nitrogen conservation, and epithelial integrity. Although euryhaline species exhibit adaptive physiological responses to salinity variation, the molecular bases of these strategies remain poorly understood at the species level. Here, we performed comparative transcriptomic analysis of kidney tissues from two sympatric hammerhead sharks, Sphyrna lewini and Sphyrna zygaena, collected under comparable marine salinity conditions (~ 26-28‰). RNA-seq identified 16,543 differentially expressed genes

A One-Year Retrospective Observational Study Reveals the Transient Nature of Pemafibrate-Induced LDL-C Elevation: Implications Beyond the PROMINENT Trial.

PURPOSE: Pemafibrate, a selective peroxisome proliferator-activated receptor-α modulator (SPPARMα), is widely used for hypertriglyceridemia; however, concerns persist regarding its potential to elevate low-density lipoprotein cholesterol (LDL-C). Previous studies monitored LDL-C for only 3-6 months, leaving the long-term trajectory unclear. This one-year retrospective observational study aimed to determine whether pemafibrate-induced LDL-C elevation is sustained or transient and to identify factors associated with this response. PATIENTS AND METHODS: A total of 110 patients receiving pemafibr

[Research progress on traditional Chinese medicine for metabolic-associated fatty liver disease via regulating MAPK signaling pathway].

Metabolic-associated fatty liver disease(MAFLD) is one of the most prevalent chronic liver diseases worldwide. Its complex pathogenesis involves multiple pathological processes, including insulin resistance, lipid metabolism disorder, inflammation, oxidative stress, and apoptosis. The mitogen-activated protein kinase(MAPK) signaling pathway serves as a critical hub linking these sections. It plays a central role in MAFLD occurrence and progress, identified as a key therapeutic target. Recent studies have shown that active compounds, single herbs, and traditional Chinese medicine(TCM) formulas

Lipid emulsion modulates myogenic and collagen-related gene expression in skeletal muscle of preterm fetal sheep.

Although intravenous lipid emulsions are routinely administered to preterm infants, their specific effects on skeletal muscle development remain unclear. In this study, a soybean oil-based lipid emulsion (Intralipid 20®) was administered via intravenous infusion to fetal sheep (gestational day 88-90) at a dose rising from 1 g/kg/day (day 0) to 3 g/kg/day (days 2-8). Intralipid infusion did not alter overall fetal body weight, tibialis anterior (TA) muscle mass or serum testosterone levels. Histological analyses revealed no significant differences in muscle fibre diameter or collagen content in

PPARβ/δ contributes to the antidiabetic effect and the increase in GDF15 caused by metformin.

Metformin, the most prescribed drug for the treatment of type 2 diabetes mellitus, increases the circulating levels of the metabolic regulator growth differentiation factor 15 (GDF15) via transcriptional regulation, with the kidneys being responsible for this increase. Since peroxisome proliferator-activated receptor (PPAR)β/δ agonists mimic many of the effects of metformin, including the rise in circulating GDF15 levels, we herein investigated whether the metformin-mediated antidiabetic effects and GDF15 upregulation were dependent on this nuclear receptor. Maternal Metabolic Dysfunction and Uteroplacental Adaptation in Polyendocrine Metabolic Ovarian Syndrome Pregnancy.

Polyendocrine metabolic ovarian syndrome (PMOS), formerly known as polycystic ovary syndrome (PCOS), is a multisystem endocrine-metabolic disorder that significantly increases the risk of adverse pregnancy outcomes, partly independently of obesity. PMOS patients frequently exhibit a characteristic metabolic-endocrine milieu of insulin resistance, hyperandrogenemia, dyslipidemia, and chronic low-grade inflammation. The physiological insulin resistance of pregnancy, driven by placental hormones, exacerbates these pre-existing disturbances and disrupts endometrial receptivity and placental functi

Probiotic Combination of Lactiplantibacillus plantarum M1 and Limosilactobacillus reuteri K4 Alleviates Early Weaning-Induced Intestinal Injury in Lambs via Modulation of Oxidative and Inflammatory Pathways.

Early weaning in intensive lamb production improves reproductive efficiency but predisposes lambs to diarrhea, oxidative stress, and intestinal barrier dysfunction, highlighting the need for non-antibiotic strategies to protect gut health. This study evaluated whether a sheep-derived mixed probiotic could alleviate early weaning-induced intestinal injury and clarified its potential molecular mechanisms. Early weaning reduced body weight, average daily gain and feed efficiency, increased diarrhea, decreased plasma and colonic catalase (CAT), glutathione peroxidase (GSH-PX), and superoxide dismu

Human dermal fibroblast-derived exosomes attenuate UVB-induced photoaging phenotypes by reducing oxidative DNA damage and modulating PGC-1α/Nrf2 signaling.

BACKGROUND: Aging, particularly photoaging induced by ultraviolet B (UVB) exposure, is characterized by oxidative DNA damage and extracellular matrix degradation. Exosomes are small vesicles secreted by cells and may mitigate oxidative stress and protect against photoaging. This study investigated the role of human dermal fibroblast-derived exosomes (HDFs-EXO) in alleviating UVB-induced photoaging and the underlying mechanisms. METHODS: HDFs-EXO were isolated and characterized by size, morphology, and exosomal markers. HDFs received a single ultraviolet B (UVB) exposure followed immediately b

Electroacupuncture modulates P300-mediated STAT6/PPARγ binding to promote M2 polarization of microglia following cerebral ischemic injury.

Microglia-mediated neuroinflammation is a key determinant of ischemic brain injury. Electroacupuncture (EA) is known to promote M2 polarization of microglia by upregulating Signal Transducer and Activator of Transcription 6 (STAT6) / Peroxisome Proliferator-Activated Receptor γ (PPARγ), however, the epigenetic mechanism governing the transcriptional stability and recruitment efficiency of this pathway remain poorly understood. We hypothesized that the transcriptional coactivator E1A-binding protein P300 (P300) may serve as a functional partner involved in facilitating this process. To test thi

Integrated transcriptomic and metabolomic profiling reveals coordinated regulatory networks associated with mosaic disease resistance in sugarcane.

INTRODUCTION: Sugarcane mosaic disease (SCMD) poses a severe threat to global sugarcane yield. Since conventional field management is insufficient to restrict viral transmission, unraveling the underlying defense mechanisms is imperative for targeted breeding. The primary objective of this study was to delineate the molecular and metabolic networks governing SCMD resistance by comparing highly resistant (XIDAZHE10-19, YT94-128) and susceptible (HP, XTT22) cultivars. METHODS: We employed metabolomic profiling and integrated transcriptomic data to investigate the genetic basis driving host resp

Melatonin Enhances Thermal Resilience and Extends Worker Lifespan in Apis cerana via Redox-Metabolic Reprogramming.

Apis cerana is widely managed in apiculture in Southern China but experiences substantial colony losses during prolonged summer heat. Developing effective strategies to support colony over-summering is therefore critical. This study demonstrates that dietary supplementation with melatonin significantly enhances thermal tolerance and extends worker lifespan in A. cerana under heat stress. Laboratory bioassays revealed that melatonin supplementation (20 µg/mL) markedly improved worker survival at both 35 °C and 37 °C, with the most pronounced effect at 37 °C, where mortality was significantly re
