CD24 in Melanoma: Biomarker, Innate Immune Checkpoint and Emerging Therapeutic Target.

Immune checkpoint inhibitors have transformed the treatment of advanced melanoma, yet many patients develop primary or acquired resistance. Although most work has focused on adaptive checkpoints (PD-1 and CTLA-4), accumulating evidence implicates innate immune suppression and stem-like, drug-resistant melanoma cell states. CD24, a small, heavily glycosylated glycosylphosphatidylinositol (GPI)-anchored surface protein, sits at the intersection of these processes and is emerging as a context-dependent biomarker and potential mediator of aggressive, therapy-resistant melanoma states. In this revi

CD155 links tumor immunotype to epithelial-directed precision therapy beyond checkpoint inhibition in cervical cancer.

BACKGROUND: Despite its viral etiology and immunogenic features, cervical cancer shows limited and often short-lived benefit from programmed cell death protein 1 blockade, currently the only approved immunotherapy for this disease. This limitation highlights the need for a deeper understanding of its immune microenvironment to uncover alternative or complementary immunotherapeutic targets that may improve outcomes. METHODS: We integrated spatial proteomic and bulk transcriptomic profiling of the cervical cancer immune landscape. Immune subset markers (CD8, CD4, CD68, FoxP3, NKp46) and clinica

Metabolite-driven dysfunction of intratumoral tertiary lymphoid structures: single-cell and spatial omics perspectives on cancer immune escape.

Intratumoral tertiary lymphoid structures (TLSs) are increasingly recognized as spatially organized immune niches associated with favorable … e-based interpretation of TLS competence. Here, we propose that metabolite-driven remodeling of the tumor microenvironment is a major cause of intratumoral TLS failure. However, the current literature lacks a cell-type- and spatially resolved framework linking individual metabolites to disruption of defined TLS modules and distinguishing early developmental deviation fr …

Automated Pathomic Analysis of Angiogenesis and Immune Profiles Unveils an Interpretable Prognostic Biomarker in Colon and Gastric Cancers.

Computational pathology enables the automatic tissue analysis of Whole Slide Images (WSIs), offering unmatched possibilities to capture quantitative tumor microenvironment (TME) characteristics that are essential for patients' prognosis and therapy response. The existing clinical and digital biomarkers do not encompass the morphometric features and spatial interactions between vascular networks and immunological compartment in the TME. To address this challenge, this work presents a high-throughput quantitative framework for automatic segmentation and assessment of aberrant phenotypes of blood…

Employing epigenetic protein degradation techniques to block CCL5-mediated photodynamic therapy via a programmed delivery platform.

Despite the significant potential of photodynamic therapy (PDT) in cancer treatment, further refinement is needed to address challenges such as poor tumor-specific accumulation of photosensitizers and the development of therapeutic resistance, which may be regulated by epigenetics. Here, a novel tumor microenvironment-responsive delivery platform was developed to co-deliver epigenetic protein degraders and photosensitizers, aiming to block the relevant regulatory mechanisms and enhance the effectiveness of combination therapy. Benefiting from the targeting ability, pH-triggered charge reversal…

FASN at the crossroads of tumor metabolism, immune evasion, and therapy resistance: Mechanistic insights and therapeutic opportunities.

Fatty acid synthase (FASN), the key enzyme driving de novo lipogenesis, has emerged as a central metabolic hub in cancer, linking aberrant lipid synthesis to tumor progression, immune escape, and therapy resistance. This review provides a comprehensive overview of the regulatory landscape and oncogenic functions of FASN, highlighting its modulation at transcriptional, post-transcriptional, and post-translational levels. We discuss how FASN-driven lipid remodeling supports tumor proliferation, disrupts antigen presentation, alters immune cell metabolism, and suppresses ferroptosis, thereby enab

The M1 Paradox: Pro-Tumorigenic Effect of Macrophage Cytotoxicity in Prostate Cancer.

Macrophages are highly plastic cells of the tumor microenvironment, and although classically activated M1 macrophages are generally regarded as anti-tumor effectors, their prolonged cytotoxic activity may also promote tumor adaptation. In this study, we investigated whether sustained exposure of prostate cancer cells to cytotoxic M1-like macrophages results in the selection of tumor cell populations with enhanced malignant properties. Macrophage-resistant derivatives of the human prostate cancer cell lines PC3 and DU145 were generated by repeated co-culture with cytotoxic THP-1-derived macroph…

Decoding Immune Mechanisms in BCG-unresponsive Non-muscle Invasive Bladder Cancer.

Non-muscle-invasive bladder cancer (NMIBC) accounts for roughly 75% of all bladder cancer cases. For patients with intermediate- and high-risk disease, intravesical Bacillus Calmette-Guérin (BCG) remains the standard treatment, yet it fails in up to 40% of cases. While radical cystectomy is the most effective salvage option, it carries significant morbidity and long-term quality-of-life consequences, highlighting the urgent need for bladder-sparing alternatives. Advancing such therapies requires a deep understanding of the immunologic mechanisms within the tumor microenvironment (TME). This re…

Concurrent P-Selectin Targeting Nanoparticle Orchestrates Tumor-Immune Dynamics for Advanced Immunochemotherapy.

Cancer therapy is often constrained by targeting single pathogenic mechanisms without addressing the complex tumor microenvironment (TME). Here, we introduce FINAL (Fucoidan-docetaxel Immunomodulatory Nanoparticles as an Antitumoral Lancer), a surface-engineered nanoplatform that simultaneously targets P-selectin-expressing cancer cells and tumor-associated macrophages (TAMs). Beyond targeting specificity, fucoidan surface modification provides intrinsic bioactivities that individually modulate both cell types while coordinately reshaping the TME. FINAL achieves dual-cell orchestration through…

Mimicking the Osteosarcoma Bone Microenvironment Using MEW-Printed PCL Scaffolds.

Osteosarcoma is the most common primary malignant bone tumor, typically affecting children and adolescents during rapid growth periods. With current treatments, the five-year survival rate for metastatic or recurrent cases remains only 20-30%, highlighting the need for new therapeutic approaches and better preclinical models. Our goal is to simulate the osteosarcoma bone microenvironment using melt electrowriting (MEW)-printed polycaprolactone (PCL) scaffolds with optimized calcium phosphate formation. To achieve this, we fabricated PCL scaffolds with distinct MEW micro-architectures, includin

Environmentally induced cell stiffening shapes metastatic site selection by tuning the immune vulnerability of cancer cells.

Metastatic microenvironments vary widely not only in their biochemical composition but also in their mechanical properties. Here, we examined how the mechanical rigidity of the metastatic niche affects metastases seeding and the local efficacy of antitumor immunosurveillance. Cancer cells stiffened in response to increasing environmental rigidity, a biophysical change that mechanically sensitized them to killing by cytotoxic lymphocytes. In immunodeficient mice, rigidity sensing by cancer cells yielded robust bone colonization, accompanied by marked stiffening of the cancer cells themselves. C

miR-155-5p drives coordinated tumor and macrophage reprogramming across multiple cancer entities.

BACKGROUND: T cell-based immunotherapies have improved cancer treatment, however their efficacy is frequently limited by tumor-intrinsic resistance mechanisms and by the immune suppressive tumor microenvironment, counteracting tumor specific immune responses through impaired antigen presentation and the accumulation of M2-like tumor-associated macrophages. MicroRNAs have emerged as regulators of tumor cell - immune cell interactions, yet the miRNAs' capacity to coordinate anti-tumor effects across different cell types is poorly understood. This study aimed to investigate whether miR-155-5p and…

Tertiary lymphoid structure topotypes in intrahepatic cholangiocarcinoma: spatial immunity, checkpoint-response associations, and therapeutic reprogramming.

Tertiary lymphoid structures (TLS) have evolved from descriptive histological findings into candidate biomarkers and targetable immune niches in solid tumors. In intrahepatic cholangiocarcinoma (ICC), however, similar lymphoid architectures may have different biological implications depending on their relationship to tumor nests, immune circuitry, and suppressive stroma. We define a "TLS topotype" as the multidimensional state of an individual TLS-centered region, including the TLS core, its perilesional neighborhood, and the nearest tumor-stroma interface. This state integrates anatomic topol

The nociceptor-CGRP-lactate axis in cancer immune escape: Linking neural signaling, immunometabolism, and immunotherapy resistance.

Cancer neuroscience has expanded the conceptual landscape of tumor biology by revealing that nerves are not passive bystanders within malignant tissues, but active regulators of tumor progression, immune surveillance, and therapeutic response. Among neural components of the tumor microenvironment, nociceptive sensory neurons have emerged as critical interfaces between tissue injury, inflammation, cancer-associated pain, and immune regulation. This review proposes that nociceptive neuron-driven immune escape represents a tissue-level pathological process positioned at the intersection of cancer…

Defects in the necroptosis machinery are a cancer resistance mechanism to checkpoint inhibitor immunotherapy.

BACKGROUND: Immune checkpoint inhibitors (ICIs) of programmed cell death protein-1 (PD-1) or cytotoxic T-lymphocytes-associated protein 4 (CTLA-4) reinvigorate strong polyclonal T-cell immune responses against tumor cells. For many patients, these therapies fail because the development of spontaneous immune responses is often compromised, as the tumor microenvironment (TME) lacks proinflammatory signals resulting in suboptimal activation of antigen-presenting cells (APCs). Necroptosis is a special form of programmed cell death associated with leakage of inflammatory factors that can lead to AP…

PPY-Induced iCAFs Cultivate an Immunosuppressive Microenvironment in Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) is characterized by cancer cells surrounded by affluent stromal components, which may underlie their limited response to various therapeutic interventions, including immunotherapy. Inflammatory cancer-associated fibroblasts (iCAFs), a crucial subset of CAFs within the PDAC microenvironment, play a pivotal role in shaping an immunosuppressive microenvironment. In this study, single-cell RNA sequencing analysis is performed to screen for cancer cells-secreted proteins associated with iCAF induction, and PPY (pancreatic polypeptide) is validated as a potent
