Immunological parameters, viral antibodies, and biochemical and histological findings in relatives of patients with chronic active hepatitis and primary biliary cirrhosis.

The familial occurrence of immunological, virological, biochemical and histopathological abnormalities has been studied in 39 first degree relatives of patients with primary biliary cirrhosis (PBC) and in 58 first degree relatives of patients with chronic active hepatitis (CAH). Tissue antibodies were more frequent in relatives than in age- and sex-matched controls. Abnormal immunoglobulin levels were demonstrated in about half of the relatives. Very high antibody titres to measles and rubella occurred only in the relatives of the patients with CAH. Moreover the raised titres tended to be clus

Monkeypox and whitepox viruses in West and Central Africa.

Prospects for the eradication of smallpox are now highly encouraging. With the cessation of man-to-man transmission, the question of possible animal reservoirs of smallpox becomes increasingly important. During the period 1970-1975, 20 cases of a smallpox-like disease were detected in smallpox-free areas of tropical rain forest in West and Central Africa. Epidemiological and virological investigations revealed that the disease was caused by an animal poxvirus termed monkeypox virus, a member of the orthopox virus group. The disease spread with difficulty even among susceptible close contacts a

[Role of viral-bacterial associations in meningitis in children].

A total of 50 section cases of meningitis in children were investigated. All patients revealed acute viral respiratory infections with generalization, including lesions of the central nervous system (CNS). In 14 children moreover meningococcal infection was diagnosed. In 30 children lesions of the CNS were due to mixed bacterial microflora. In 6 children along with acute viral respiratory infections (AVRI) mycoplasmosis was also revealed. Etiology of the process was determined on the basis of characteristic structural changes in the CNS and other organs, findings of virological and bacteriolog

Some properties of an established fish cell line from the marine fish, Caranx mate (Omaka).

A monolayer culture of fibroblast-like cells was initiated and established from a trypsin digest of Omaka fish larvae (Caranx mate). The cell culture has been subcultured 105 times over a period of 30 mo and appears to be contact-inhibited. The Omaka cells grew optimally at 27 degrees in Eagle's basal medium plus 10% fetal calf serum but failed to grow at 16 and 37 degrees. At the lower temperature the cells remained viable and began to divide when the temperature was raised to 27 degrees. Unlike certain marine fish cell lines the Omaka cell did not require a higher osmolarity medium for optim

Long-Acting HIV Therapy in African Adults with Adherence Challenges.

BACKGROUND: In eastern and southern Africa, approximately 1 million people living with human immunodeficiency virus (HIV) and receiving oral therapy do not have viral suppression because of adherence challenges. METHODS: We conducted a phase 3b, open-label, randomized trial to compare long-acting injectable cabotegravir plus rilpivirine with oral dolutegravir-based therapy among adults with virally suppressed HIV type 1 (viral load, <200 copies per milliliter) and suboptimal adherence or care engagement. The participants were assigned in a 1:1 ratio to receive intramuscular injections of cabo

Cost-Effectiveness of Point-of-Care Hepatitis C Virus RNA Testing in the US.

IMPORTANCE: The standard of care (SOC) laboratory-based hepatitis C virus (HCV) diagnostic algorithm is associated with high rates of undiagnosed cases, patients lost to follow-up, and low rates of treatment initiation, particularly among high-risk populations. Improving diagnostic efficiency is critical for HCV elimination. OBJECTIVE: To evaluate the cost-effectiveness of a point-of-care (POC) HCV RNA-first diagnostic strategy compared with SOC across care settings in the US serving people at high-risk of HCV infection. DESIGN, SETTING, AND PARTICIPANTS: This economic evaluation used a hybr

An analysis of WHO FluNet and FluID influenza surveillance data for South East Asia Region, 2015-2023.

BACKGROUND: Influenza is known to cause seasonal epidemics and recurrent pandemics and demands robust surveillance systems for strain detection and trend monitoring. There are global surveillance systems (FluNet and FluID) to monitor the trends in influenza virus strains and epidemiology. SEAR has been vulnerable to influenza outbreaks and hence, efforts to enhance surveillance have been ongoing. However, there is limited analysis of data trends from influenza surveillance systems from SEAR member states (MS). OBJECTIVES: To describe the virological and epidemiological characteristics of infl

Safety and Efficacy of Therapeutic Agents in Viral Pneumonia in Immunosuppressed Patients.

INTRODUCTION/OBJECTIVE: Immunosuppressed Patients (ISP) - defined by congenital or acquired deficiencies in systemic defense mechanisms - are disproportionately susceptible to viral respiratory infections. Agents such as SARS-CoV-2, Respiratory Syncytial Virus (RSV), influenza A and B, and Cytomegalovirus (CMV) cause significant morbidity in both immunocompetent and immunosuppressed individuals. Clinical manifestations range from mild influenza-like illness to severe pneumonia, respiratory failure, multiorgan dysfunction, and concurrent bacterial infections. METHODS: A narrative synthesis of

Understanding spatiotemporal clustering of seasonal influenza in the United States.

BACKGROUND: Seasonal influenza exhibits distinct spatiotemporal patterns across the United States, yet the geographic clustering of influenza activity remains incompletely understood. This study aims to identify jurisdictions with similar patterns of seasonal influenza epidemics by exploring spatiotemporal dynamics across the United States after the 2009 H1N1 pandemic. METHODS: We analyzed data from U.S. influenza surveillance systems, including outpatient illness surveillance and virologic surveillance. The outpatient illness data included weekly proportions of outpatient visits for influenza

Single-cell analyses of liver and blood reveal distinct immune cell signatures of HBV-related HCC patients.

Chronic hepatitis B virus (HBV) infection is a major etiological driver of hepatocellular carcinoma (HCC) and is accompanied by profound immune dysregulation that shapes disease progression and therapeutic responsiveness. However, the extent to which systemic immunity reflects the tumor immune microenvironment (TIME) in HBV-related HCC (HBV-HCC) remains incompletely defined. Here, we used mass cytometry (CyTOF) to resolve immune heterogeneity across peripheral blood mononuclear cells (PBMCs), tumor tissues, and matched adjacent non-tumor (paracancer) tissues in treatment-naïve HBV-HCC. PBMCs,

HIV dominates T-cell immune responses in HIV-HBV co-infection and is associated with increased HIV reservoir transcription.

Due to the effectiveness of antiretroviral therapy (ART), Human Immunodeficiency Virus (HIV) infection is now managed as a chronic condition. However, people with HIV (PWH) remain susceptible to co-infections, including Hepatitis B virus (HBV), with 15% of PWH in Africa being co-infected. While co-infections may accelerate disease progression and increase AIDS-related mortality, their effects on T-cell responses and HIV reservoir size are unclear. This study compared T-cell responses in people with HIV-HBV co-infection, HIV or HBV alone, and assessed the effects of HBV on the HIV reservoir siz

Role of viral and host factors in determining the outcome of HBV-associated acute liver failure.

BACKGROUND: Acute hepatitis B virus (HBV) infection encompasses a broad clinical spectrum, from self-limited hepatitis to acute liver injury (ALI), acute liver failure (ALF), subacute liver failure, and ALF on chronic HBV reactivation. This study investigates how host and viral factors interact to shape the clinical spectrum of HBV-associated ALF. METHODS: We analyzed 231 serial serum samples from 49 well-characterized patients including 36 with HBV-associated ALF and 13 with acute hepatitis B. Among ALF patients, 7 had ALI, 7 acute and 11 subacute liver failure, and 11 ALF on chronic HBV. Vi

Viral load dynamics among adults receiving HIV care in rural North-Eastern South Africa, 2015-2020: insights from a population-based record linkage study.

INTRODUCTION: The Joint United Nations Programme on HIV/AIDS set ambitious-but-reachable targets to have 95% of HIV-positive people diagnosed, 95% on ART, and 95% virally suppressed by 2030. To address the latter, post-2016, South Africa's HIV treatment guidelines aimed to deliver maximal and durable viral load (VL) suppression through extensive antiretroviral therapy (ART) scale-up. Yet, standard suppression one-off measurement conceals viral response trajectories with high onward transmission potential for HIV patients on lifelong treatment. We investigated the dynamics of periodic VL patter

Characteristics and risk factors of acquired drug resistance among people living with HIV: A retrospective study in Guangzhou, China (2018-2023).

BACKGROUND: Acquired drug resistance (ADR) in people living with HIV (PLWH) reduces treatment efficacy, restricts drug options, and increases the risk of human immunodeficiency virus (HIV) transmission. The resistance characteristics and associated risk factors of PLWH suffering from virological failure (VF) in Guangdong remain unclear, thus highlighting the need for further research in this area. This retrospective study examines the prevalence, resistance characteristics, and contributing factors of ADR among PLWH who experienced VF in Guangzhou, China, between 2018 and 2023. METHODS: Blood

Evaluation of respiratory virus transmissibility and resilience from fomites: the case of 11 SARS-CoV-2 clinical isolates.

The emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) caused a global pandemic, significantly impacting social and economic aspects of society. While belonging to the Coronaviridae family, SARS-CoV-2 exhibited unique characteristics that facilitated its widespread transmission and pathogenicity. This study aimed to investigate the molecular characterization and infection kinetics of emerging SARS-CoV-2 variants, focusing on four key virological aspects: viral entry mechanisms, replication dynamics, immune evasion strategies, and surface persistence. We isolated 11 SARS-
