Incremental costs of introducing jet injection technology for delivery of routine childhood vaccinations: comparative analysis from Brazil, India, and South Africa.

BACKGROUND: Disposable-syringe jet injectors (DSJIs) have the potential to deliver vaccines safely and affordably to millions of children around the world. We estimated the incremental costs of transitioning from needles and syringes to delivering childhood vaccines with DSJIs in Brazil, India, and South Africa. METHODS: Two scenarios were assessed: (1) DSJI delivery of all vaccines at current dose and depth; (2) a change to intradermal (ID) delivery with DSJIs for hepatitis B and yellow fever vaccines, while the other vaccines are delivered by DSJIs at current dose and depth. The main advant…

Pattern differentiation in Traditional Chinese Medicine can help define specific indications for biomedical therapy in the treatment of rheumatoid arthritis.

OBJECTIVES: The objective of this study is to explore the role of Traditional Chinese Medicine (TCM) pattern differentiation in identifying a subset of patients with rheumatoid arthritis (RA) who are more likely to respond to biomedical combination therapy. METHODS: This study uses data from a previous multicenter randomized-controlled clinical trial. One hundred and ninety-four (194) patients were treated with biomedical combination therapy (diclofenac, methotrexate, and sulfasalazine). ACR20 response at 12 weeks and 24 weeks was used for evaluation of efficacy. Eight (8) symptoms, which are

Analysis of flavivirus NS5 methyltransferase cap binding.

The flavivirus 2'-O-nucleoside N-terminal RNA methyltransferase (MTase) enzyme is responsible for methylating the viral RNA cap structure. To increase our understanding of the mechanism of viral RNA cap binding we performed a detailed structural and biochemical characterization of the guanosine cap-binding pocket of the dengue (DEN) and yellow fever (YF) virus MTase enzymes. We solved an improved 2.1 A resolution crystal structure of DEN2 Mtase, new 1.5 A resolution crystal structures of the YF virus MTase domain in apo form, and a new 1.45 A structure in complex with guanosine triphosphate an…

Late-acting dominant lethal genetic systems and mosquito control.

BACKGROUND: Reduction or elimination of vector populations will tend to reduce or eliminate transmission of vector-borne diseases. One poten … ribed, so as a proof-of-principle we constructed a strain of the principal vector of the dengue and yellow fever viruses, Aedes (Stegomyia) aegypti, with the necessary properties of dominant, repressible, highly penetrant, late-acting lethality. CONCLUSION: Conventional SIT induces early-acting (embryonic) lethality, but genetic methods potentially allow the …

Multiple vaccine and pyridostigmine bromide interactions in the common marmoset Callithrix jacchus: immunological and endocrinological effects.

Following active service during the 1990/1991 Gulf Conflict, a number of UK and US veterans presented with a diverse range of symptoms, collectively known as Gulf Veterans Illnesses (GVI). The administration of vaccines and/or the pretreatment against possible nerve agent poisoning, pyridostigmine bromide (PB), given to Armed Forces personnel during the Gulf Conflict has been implicated as a possible factor in the aetiology of these illnesses. The possibility that adverse health effects may result from the administration of these vaccines (anthrax, pertussis, plague, yellow fever, polio, typho…

RAG1 core and V(D)J recombination signal sequences were derived from Transib transposons.

The V(D)J recombination reaction in jawed vertebrates is catalyzed by the RAG1 and RAG2 proteins, which are believed to have emerged approxi … frican malaria mosquito genomes. Transib transposons also are present in the genomes of sea urchin, yellow fever mosquito, silkworm, dog hookworm, hydra, and soybean rust. We demonstrate that recombination signal sequences (RSSs) were derived from terminal inverted repeats of an ancient Transib transposon. Furthermore, the critical DDE catalytic triad of RAG1 …

Hepatic amebiasis.

Amebiasis can be considered the most aggressive disease of the human intestine, responsible in its invasive form for clinical syndromes, ranging from the classic dysentery of acute colitis to extra-intestinal disease, with emphasis on hepatic amebiasis, unsuitably named amebic liver abscess. Found worldwide, with a high incidence in India, tropical regions of Africa, Mexico and other areas of Central America, it has been frequently reported in Amazonia. The trophozoite reaches the liver through the portal system, provoking enzymatic focal necrosis of hepatocytes and multiple micro-abscesses th

Mutagenesis of the NS2B-NS3-mediated cleavage site in the flavivirus capsid protein demonstrates a requirement for coordinated processing.

Analysis of flavivirus polyprotein processing has revealed the presence of a substrate for the virus-encoded NS2B-NS3 protease at the carboxy-terminal end of the C (capsid or core) protein. Cleavage at this site has been implicated in the efficient generation of the amino terminus of prM via signal peptidase cleavage. Yellow fever virus has four basic residues (Arg-Lys-Arg-Arg) in the P1 through P4 positions of this cleavage site. Multiple alanine substitutions were made for these residues in order to investigate the substrate specificity and biological significance of this cleavage. Mutants w…

Analysis of disulfides present in the membrane proteins of the West Nile flavivirus.

Recently the primary structure of the structural proteins of the flaviviruses West Nile (WN) virus (Castle et al., 1985; Wengler et al., 1985) and yellow fever (YF) virus (Rice et al., 1985) have been determined. As a first step in a further characterization of the organization of the structural proteins we have now studied the disulfide bridges present in the WN virus membrane proteins. All three membrane proteins, pre M, M, and E, were analyzed. The results obtained can be summarized as follows: The pre M proteins of both WN and YF virus each contain 6 cysteine residues and the position of a…

Dengue virus infections in Nigeria: a survey for antibodies in monkeys and humans.

A retrospective serological survey for dengue immunity was conducted in Nigeria to determine the prevalence of infection in man and non-human primates. Preliminary haemagglutination-inhibition (HI) tests revealed that 63% of persons tested had HI antibodies against one or more of the following flaviviruses: dengue type 1, yellow fever, West Nile and Wesselsbron. Parallel HI and neutralization (N) tests on 179 human sera showed that six of 20 sera (30%) negative for flavivirus HI antibody contained dengue N antibody. This finding emphasized the advantage of the N test over HI in screening for d…

Pathogen transmission in relation to feeding and digestion by haematophagous Arthropods.

The blood feeding habit, especially among opportunist feeders such as tabanids and Stomoxys is known to result in transmission of diseases f … ds and transovarial transmission to succeeding generations is frequent in mites and ticks. Although Yellow Fever virus is not transovarially transmitted by its vector Aedes aegypti, the mosquito only becomes infective some time after ingesting an infected blood meal (Chandler, 1955). Thus, metabolic or biochemical changes or adaptations in the virus or in the …

Thermal stress responses on developmental time, survival, and wing morphometrics in desert and coastal strains of Aedes aegypti (Diptera: Culicidae) from India.

BACKGROUND: Aedes aegypti is a primary vector of dengue, chikungunya, Zika, and yellow fever diseases. Despite its global invasion, little is known about how populations from climatically contrasting regions adapt to varied thermal environments. Understanding survival strategies is critical for predicting population dynamics and disease transmission under climate change. To address this gap, we assessed ecological adaptations in terms of development, survival, and wing length, along with molecular variations, in the Thar Desert (Jodhpur) and the east coastal (Kolkata) strains of Ae. aegypti. …

Aedes aegypti Hsp70 has enhanced ATPase and protein protection under oxidative stress.

Aedes aegypti mosquitoes are responsible for transmitting several viral diseases in humans, including Dengue, Zika, Yellow Fever, and Chikungunya. Despite significant efforts to control the mosquito vector and the viruses, the issue has intensified. This is mainly due to the mosquito's successful adaptation to urban environments and its expanding geographic range, driven by rising global temperatures. To effectively combat vector-borne diseases, we need a comprehensive understanding of mosquito physiology and cellular regulatory mechanisms, as these could reveal new molecular targets for inter…

Evaluation of Escherichia coli-derived Silver Nanoparticles for Aedes Vector Control in Southern India.

BACKGROUND: Aedes aegypti and Aedes albopictus are the principal mosquito vectors in Tamil Nadu transmitting the major arboviral diseases such as Dengue, Chikungunya, Zika and Yellow Fever. Aedes aegypti serves as the primary vector, followed by Aedes albopictus. Due to the limitations and environmental concerns associated with predictable chemical larvicides, there is an emergency need for alternative and sustainable vector control strategies. METHODS: In this study, environmentally friendly silver nanoparticles were synthesized using Escherichia coli through a biogenic approach that develop…
