Metabolomics reveals pre-eclamptic-protective mechanisms within individuals.

OBJECTIVES: Pre-eclampsia is a common and dangerous hypertensive pregnancy complication, with recurrence in as many as 80% of subsequent pregnancies. However, it is often difficult to predict recurrence. Further complicating risk assessment and biomarker development, the genetic and environmental drivers of pre-eclampsia are varied and numerous. In this study, a longitudinal, repeated-measures design was used to control for many of these drivers, and to isolate factors that may be associated with pre-eclampsia in subsequent pregnancies. STUDY DESIGN: In an exploratory cohort (n = 14) of indiv

Morphine and codeine in racing horse feed: is there reason for concern?

Opiates such as morphine and codeine are substances often misused to improve the performance of racing horses during competitions and are therefore on the International Federation for Equestrian Sports' list of prohibited substances. However, a positive antidoping test may be due to the consumption of feed (mainly lucerne or oats) contaminated by opium poppy containing the alkaloids morphine and codeine. In order to determine whether a positive antidoping test is the result of an intentional abuse of opiates or consumption of feed contaminated by poppies, we optimised conditions for the extrac

Inhibition of ferroptosis rescues BMP osteogenic differentiation impaired by iron overload in the osteoporotic microenvironment.

Bone morphogenetic proteins (BMPs) are effective for treating various orthopedic conditions and are widely used clinically. However, their therapeutic efficacy is limited in osteoporosis patients. Iron overload represents a key risk factor for osteoporosis, inducing ferroptosis and suppressing the osteogenic differentiation of bone marrow stromal cells (BMSCs). This mechanism likely contributes to the suboptimal response to BMP therapy in these patients. Using ovariectomized (OVX) mouse models and sequencing analysis of human BMSCs, we confirmed ferroptosis occurs in BMSCs from both OVX mice a

Glycocalyx-targeted therapy prevents age-related muscle loss and declines in maximal exercise capacity.

Age-related declines in cardiovascular function contribute to reduced physical capacity, both of which are independent predictors of mortality. We have previously demonstrated that glycocalyx-targeted therapy with Endocalyx™ that contains high-molecular-weight hyaluronan (HMW-HA) improves cardiovascular health in old age, raising the possibility that HMW-HA also plays a role in age-related physical dysfunction. Here, we first demonstrate that tamoxifen-inducible deletion of Has2, which produces HMW-HA, leads to glycocalyx depletion, decreases exercise capacity, and impairs skeletal muscle resp

[Glutamine metabolic reprogramming in regulating the occurrence and development of osteosarcoma].

Excessive cellular proliferation and metabolic reprogramming are important characteristics of cancer cells. Cancer cells promote excessive proliferation and growth by altering coordinated metabolic pathways. In terms of glucose metabolism, most cancer cells exhibit increased glucose uptake and lactate production even under aerobic conditions, known as the Warburg effect. Increased glucose uptake not only provides energy but also supplies essential carbon sources for the biosynthesis of nucleotides, lipids, and proteins. During this process, decreased pyruvate dehydrogenase activity leads to di

The mediating role of key amino acid and vitamin metabolite ratios in the effects of 5 dietary habits on psoriatic arthritis: A Mendelian randomization study.

The causal relationship between dietary habits and psoriatic arthritis (PsA) remains unclear, and the mediating role of human plasma metabolites in this relationship is unexamined. This study aims to elucidate the causal relationship between 80 dietary patterns and PsA using a 2-sample Mendelian randomization (MR) analysis. A 2-step MR approach was employed to investigate whether 1400 human plasma metabolites could serve as potential mediators between dietary habits and PsA. To ensure the reliability of the results, heterogeneity and pleiotropy tests were conducted. Our MR analysis identified

Urinary metabolic alterations associated with occupational exposure to metals and polycyclic aromatic hydrocarbons based on non-target metabolomics.

Long-term occupational exposure to metals and organics have been reported to be under great health risks. However, limited data are available on the molecular mechanism between combined exposure to metals and polycyclic aromatic hydrocarbons (PAHs) and harmful health effects. In present work, non-target metabolomics study was conducted based on urine samples from nonferrous metal smelting workers (n = 207), surrounding residents (n = 180), and the control residents (n = 187) by using ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-QT

In vitro and in vivo immune-enhancing effects of polysaccharides with different molecular weights and structural characteristics from Gastrodia elata Blume.

Research on high-molecular-weight polysaccharides tends to be more difficult and lag in terms of their fine structures and bioavailability. We focused on Gastrodia elata Blume polysaccharides (GEPs) with different molecular weights and structural characteristics to reveal their bioactivities, especially those abundant high-molecular-weight GEPs. A novel high-yield polysaccharide (GEP1-2) with the high molecular weight of 3.21 × 106 Da was first purified. Through conventional and enzymolysis-assisted analyses, GEP1-2 was an α-D-(1,4)(1,6)-glucan with unique linkages of →2)-β-D-Frucf-(1→, →4)-β-

Disturbances in mitochondrial quality control and mitochondria-lysosome contact underlie the cerebral cortex and heart damage of mucopolysaccharidosis type II mice.

Mucopolysaccharidosis type II (or Hunter syndrome) is a lysosomal disease caused by mutations in the IDS gene, which encodes the enzyme iduronate 2-sulfatase. MPS II patients present with systemic clinical manifestations and, in the most severe cases, with severe central nervous system abnormalities. Cardiac alterations are also commonly observed. In this study, we evaluated the communication between mitochondria and lysosomes, as well as mitochondrial dynamics and bioenergetics, mitophagy/autophagy, and redox homeostasis in the cerebral cortex and heart of 6-month-old MPS II mice. Our finding

Fenitrothion exposure induces hyperglycemia in juvenile rats: Impeded glycolysis and promoted gluconeogenesis mediated by acetyl-CoA and oxaloacetate metabolism dysregulation.

Fenitrothion (FNT), an organophosphorus insecticide widely used in agriculture and household pest control, is closely associated with several glucose metabolism diseases. However, scientific knowledge about the mechanisms of FNT disturbance of glucose metabolism in puberty models remains poorly understood. Here, we show that 4-week-old SD rats exposed to 3.7 or 37 mg/kg bw/day FNT for 28 days exhibited hyperglycemia and hyperphagia, as well as impaired glucose tolerance and insulin resistance, which resulted from FNT's interference with insulin secretion, inhibition of glycolysis, and promotio

Exploring Etiologies of Hypokalemic Paralysis: A Case Series.

BACKGROUND: Hypokalemic paralysis is a rare but potentially life-threatening neuromuscular emergency characterized by the sudden onset of flaccid weakness due to reduced serum potassium. While inherited channelopathies represent primary causes, secondary etiologies-particularly endocrine, renal, and metabolic disorders-are more frequently encountered and require timely recognition to avoid complications. CASE SUMMARY: We report four patients presenting with acute flaccid paralysis associated with documented hypokalemia (serum potassium <3.5 mmol/L). The first case involved a pregnant woman wi

An Unusual Presentation of Multifactorial Hypocalcemia as Myopathy: A Case Report.

Hypocalcemia is a common metabolic disturbance with a broad spectrum of clinical presentations, ranging from asymptomatic laboratory findings to severe, life-threatening complications. While neuromuscular irritability is a well-known feature, myopathy as a primary presentation is rare and often under-recognized. We report the case of a 45-year-old female with type 2 diabetes and hypertension who presented with progressive lower limb weakness and generalized body aches. She had a recent history of an abdominal skin abscess and had received blood transfusions without calcium correction. On exami

Emerging Risk of Flea-Borne Bartonella in Tropical Cities: Evidence from Stray Cats in the Klang Valley, Malaysia.

Urban stray cats are often associated with ectoparasites and zoonotic pathogens due to their unsanitary living conditions and lack of veterinary care. Fleas, especially Ctenocephalides spp., are competent vectors of Bartonella spp., a genus of emerging bacterial pathogens with both public health and veterinary relevance. This study investigated the presence of Bartonella DNA in ectoparasitic fleas infesting stray cats in various urban habitats within the Klang Valley, Malaysia. A total of 204 fleas were collected from 89 stray cats. Fleas were identified morphologically using established taxon

Identification of Candidate Male-Reproduction-Related Genes from the Testis and Androgenic Gland of Macrobrachium nipponense, Regulated by PDHE1, through Transcriptome Profiling Analysis.

The previous publication identified that pyruvate dehydrogenase E1 (PDHE1) positively regulated the process of male reproduction in M. nipponense through affecting the expressions of insulin-like androgenic gland hormone. The present study aimed to identify the potential male-reproduction-related genes that were regulated by PDHE1 through performing the transcriptome profiling analysis in the testis and androgenic gland after the knockdown of the expressions of PDHE1 by the injection of dsPDHE1. Both RNA-Seq and qPCR analysis identified the significant decreases in PDHE1 expressions in the tes

Hepatotoxicity evaluations of different surface charged carbon quantum dots in vivo and in vitro.

Recently, carbon quantum dots (CQDs) have become popular because of their simple synthesis and potential applications. Although CQDs have high biocompatibility, their biotoxicity must be verified to reduce the possible risks associated with large-scale application. In this study, the hepatotoxicity of three CQD types, namely diammonium citrate (AC)-based (CQDs-AC), spermidine trihydrochloride (Spd)-based (CQDs-Spd), and AC- and Spd-based CQDs (CQDs-AC/Spd), were evaluated in vivo and in vitro. It was observed in vivo that CQDs-Spd and CQDs-AC/Spd, but not CQDs-AC, caused histopathological dama
