Transcriptome sequencing identifies genes associated with invasion of ovarian cancer.

OBJECTIVE: To identify key genes in ovarian cancer using transcriptome sequencing in two cell lines: MCV152 (benign ovarian epithelial tumour) and SKOV-3 (ovarian serous carcinoma). METHODS: Differentially expressed genes (DEGs) between SKOV-3 and MCV152 were identified. Candidate genes were assessed for enrichment in gene ontology function and Kyoto Encyclopaedia of Genes and Genomes pathway. Candidate gene expression in SKOV-3 and MCV152 cells was validated using Western blots. RESULTS: A total of 2020 upregulated and 1673 downregulated DEGs between SKOV3 and MCV152 cells were identified t

Advances in the prevention and treatment of radiation-induced brain necrosis: a narrative review.

Radiation-induced brain necrosis (RBN) is a serious and often debilitating complication of radiotherapy for intracranial and head and neck malignancies, with an incidence of 5-25%. It can lead to significant cognitive impairment, neurological deficits, and increased mortality. As radiotherapy techniques advance and patient survival improves, effective prevention and management of RBN have become critical clinical priorities. This review systematically summarizes the current understanding of RBN pathogenesis, highlighting the central roles of vascular injury (endothelial damage, HIF-1α/VEGF dys

Immune profiling in a living human recipient of a gene-edited pig kidney.

Xenotransplantation of gene-edited pig kidneys offers a promising solution to the shortage of kidneys for organ transplantation. We recently performed a gene-edited pig kidney transplantation into a living human recipient with end-stage kidney disease. Here, using transcriptomics, proteomics, metabolomics and multiplexed imaging, we conducted high-dimensional immune profiling in this individual. Despite profound depletion of circulating T cells, early T cell-mediated rejection occurred within 1 week after transplantation, likely driven by subtherapeutic immunosuppression and the presence of re

Distinct Immunomodulatory Strategies Guide Mesenchymal Stromal/Stem Cell-Mediated Bone Regeneration.

The bone regenerative potential of bone marrow-derived mesenchymal stromal/stem cells (MSC) is critically shaped by their interaction with the host immune system. Here, we compare two MSC priming strategies: coculture with regulatory T cell (Treg) or exposure to the proresolving mediator Resolvin E1 (RvE1) to enhance ectopic osteogenesis in mice. Both approaches enhance MSC-mediated bone formation through distinct local and systemic immunomodulatory mechanisms. Treg priming promotes a regulated anti-inflammatory environment characterized by reduced IL-6 protein levels (3.6-fold), sustained IL-

CSF3R compound mutations produce a hyperproliferative leukemia driven by overexpression of PIM kinases.

Colony Stimulating Factor 3 Receptor (CSF3R) mutations are the molecular hallmark of chronic neutrophilic leukemia (CNL). A subset of these patients (~25%) have two mutations in CSF3R on the same allele. These "compound" CSF3R mutations include both an activating point mutation and a cytoplasmic truncating mutation. While compound mutations are prevalent, we have a limited understanding of their molecular characteristics. In this study, we interrogated the mechanisms driving the evolution of compound mutations. We found that CSF3R compound mutations induce a hyperproliferative leukemia compare

Pancytopenia Misattributed to Parvovirus B19 Infection Revealing Acute Promyelocytic Leukemia.

Pancytopenia is a diagnostic challenge due to its broad differential diagnosis, including infections and hematological malignancies. Parvovirus B19 infection is a recognized but uncommon cause of transient pancytopenia. However, positive viral serology or viremia may lead to diagnostic anchoring and delay the identification of underlying malignant disorders. We report the case of a 17-year-old previously healthy female presenting with persistent fever and tonsillitis. Laboratory investigations revealed pancytopenia with agranulocytosis. Blood serology and polymerase chain reaction (PCR) were p

Immune Endotype-Guided Precision Immunotherapy in Sepsis: From Transcriptomics to Bedside Application.

Sepsis is a life-threatening syndrome characterized by profound biological heterogeneity and dynamic immune dysregulation. Traditional uniform treatment strategies have failed to account for distinct immune endotypes ranging from hyperinflammation to immunoparalysis. Advances in transcriptomic, proteomic, and cellular profiling have enabled the identification of reproducible immune phenotypes with differing prognoses and therapeutic responsiveness. Hyperinflammatory states may benefit from targeted cytokine inhibition, whereas immunosuppressed phenotypes marked by reduced monocyte HLA-DR (huma

Lighting up Macrophage Reprogramming Assists Immunosuppressive Niche Modulation in Primary Tumors and Tumor-Draining Lymph Nodes of Breast Cancer.

The recurrence and metastasis of breast cancer are driven by immunosuppressive myeloid cells in tumors and lymph nodes. The inherent heterogeneity of myeloid cells poses a significant challenge for real-time imaging and precise treatment. In this study, we developed microenvironment-responsive second near-infrared (NIR-II) biomimetic nanoparticles (PA NPs) to spatiotemporally modulate macrophage-mediated immunotherapies. These NIR-II activatable "off-on" PA NPs accumulate in tumors and lymph nodes, enabling controlled immune activation. By incorporating colony-stimulating factor 1 receptor inh

Thrombotic thrombocytopenic purpura and macrophage activation syndrome secondary to Sjögren's syndrome: a case report.

Sjögren's syndrome (SS) is a systemic autoimmune disorder frequently associated with diverse hematological manifestations. Immune-mediated thrombotic thrombocytopenic purpura (iTTP) and macrophage activation syndrome (MAS) represent rare, life-threatening complications, yet the consecutive occurrence of both conditions in the same SS patient is exceedingly rare. We report a 20-year-old female diagnosed with SS following drug-induced liver injury. In July 2023, she developed acute iTTP (0% ADAMTS13 activity, with positive inhibitory antibodies) shortly after the initiation of immunomodulatory t

Voriconazole-Induced Agranulocytosis in a Cirrhotic Patient with Influenza-Associated Pulmonary Aspergillosis: A Case Report.

The diagnosis and treatment of influenza-associated pulmonary aspergillosis (IAPA) present significant challenges, and voriconazole, as the first-line treatment for IAPA, rarely causes the serious adverse event of agranulocytosis. We first report a case of voriconazole-associated agranulocytosis in a patient with IAPA complicated by cirrhosis and systematically describe the complete process of diagnosis, treatment, and adverse event management. A 68-year-old male with a history of liver cirrhosis presented with cough, dyspnea, and fever. Testing confirmed influenza A, and chest computed tomogr

Updates on intratumoral therapies in melanoma.

Intratumoral therapies provide an opportunity for novel strategies in the management of advanced melanoma. These approaches deliver concentrated doses of therapeutic immune agents directly into individual tumor(s), achieving local tumor control and induction of systemic antitumor immune responses while minimizing the risk of immune-related adverse events. Talimogene laherparepvec, a genetically modified herpes simplex virus type 1-encoding granulocyte-macrophage colony-stimulating factor, was the first oncolytic viral therapy approved in a solid tumor indication. In pivotal trials of patients

Targeted E3 Region Engineering Boosts Antitumor Efficacy of Conditionally Replicating Adenoviruses in an Immunocompetent Tumor Model.

The adenovirus E3 region's immune-modulating genes (gp19K, Adenovirus Death Protein [ADP], E3B) are frequently modified in oncolytic adenoviruses (OAds) through deletion and transgene insertion like granulocyte-macrophage colony-stimulating factor (GM-CSF). However, the synergistic effects of dual-gene deletions on antitumor efficacy and transgene capacity remain unexplored. To address this, we constructed three E3-modified OAds including OAd5-delgp19K (delgp19K), M20 (delgp19K and ADP), M22-0 (delgp19K and E3B), and their GM-CSF-armed derivatives, systematically evaluating the impact of ADP a

Induced pluripotent stem cell treatment for microglia replacement in progressive MS.

BACKGROUND: Induced pluripotent stem cells (iPSCs) offer a novel approach to target microglia, key drivers of multiple sclerosis (MS) pathology. A promising therapeutic strategy involves depleting resident microglia using colony-stimulating factor 1 receptor (CSF-1R) inhibitors, followed by differentiating iPSCs into microglia in vitro and transplanting them into the central nervous system (CNS). However, no studies have yet specifically examined iPSC-derived microglial replacement in MS. OBJECTIVES: This review evaluates the feasibility of replacing pathogenic microglia in MS with iPSC-deriv

Potential Pharmacologic Treatments in Spinal Cord Injury: A Narrative Review.

Spinal cord injury (SCI) remains one of the most devastating neurological conditions with poor prognosis and limited therapeutic options. Early surgical decompression has been established as the cornerstone of acute management, yet multiple pharmacologic strategies have been extensively investigated to improve neurological outcomes. High-dose methylprednisolone sodium succinate, once widely adopted, ultimately failed to demonstrate consistent efficacy, underscoring the challenges of translating preclinical promise into clinical benefit. Advances in understanding the sequential pathophysiology

Cyclooxygenase 2 overexpression suppresses Smad3 and augments ERK1/2 signaling activated by TGFβ1 in endometrial stromal cells: A novel insight into endometriosis pathogenesis.

RESEARCH QUESTION: To investigate the underlying mechanisms driving the opposing effects of transforming growth factor-beta 1 (TGFβ1) on the proliferation of control (CESCs) and ectopic (EESCs) endometrial stromal cells. DESIGN: Cell proliferation assays (CCK-8 and colony formation) were employed to assess the effects of TGFβ1 on CESC and EESC proliferation. An immortalized human endometrial stromal cell line (HESC) was used to elucidate the mechanisms behind cytostatic effect of TGFβ1 and the potential role of cyclooxygenase (COX)-2 in mediating the modulation of TGFβ1 signaling. RESULTS: T

Additivity of Transfer Free Energies Enables the Description of Complex Protein Formulations in Implicit Solvent Molecular Dynamics Simulations.

A complex 3D structure and the surrounding environment determine the function and stability of a protein. Various osmolytes can be added to a protein drug formulation to stabilize the native protein structure by preventing unfolding and aggregation. In this context, the concept of transfer free energy, which represents the change in chemical potential of a protein being transferred from water to an osmolyte solution, has emerged as a powerful tool to elucidate the energetics involved in the protein-osmolyte interaction. In the present work, we experimentally determine the transfer free energie

Case study of resecabtagene autoleucel in a subject with diffuse cutaneous systemic sclerosis treated in the RESET-SSc trial.

Under compassionate use, CAR T cells have elicited durable remissions in patients with refractory systemic sclerosis (SSc). Here, we report on the safety, efficacy, and correlative data of the first SSc subject treated with a fully human CD19-CART cell therapy (resecabtagene autoleucel) in the RESET-SSc trial (NCT06328777). Nine days post-infusion, the subject experienced a brief grade 2 episode of cytokine release syndrome that resolved promptly with intravenous hydration and indomethacin. Immune-effector-cell-associated neurotoxicity syndrome was not observed. Seven days post-infusion, the s
