Genomic profiling, implications for genotype-based treatment of 131 patients with phenylketonuria and characterization of novel p.Pro416Leu PAH variant.

Phenylketonuria (PKU) is the most common inborn disorder of amino acid metabolism caused by biallelic pathogenic variants in phenylalanine hydroxylase (PAH) gene. This study comprised genomic profiling and phenotypic characterization of 131 Serbian PKU patients along with implications for BH4 therapy. By combining Sanger sequencing, MLPA and WES re-analysis of previously unsolved cases, we identified 38 different disease causing variants in the PAH gene and classified them using ACMG guidelines. The most frequent variant was p.Leu48Ser (30.92%), followed by p.Arg408Trp (12.21%) and p.Ile306Val

Chemical Synthesis, Biological Evaluation, and Cheminformatics Analysis of a Group of Chlorinated Diaryl Sulfonamides: Promising Inhibitors of Cholesteryl Ester Transfer Protein.

BACKGROUND: Hyperlipidemia is characterized by an abnormally elevated serum cholesterol, triglycerides, or both. The relationship between an elevated level of LDL and cardiovascular diseases is well-established. Cholesteryl ester transfer protein (CETP) is an enzyme that moves cholesterol esters and triglycerides between LDL, VLDL, and HDL. CETP inhibition leads to a reduction in cardiovascular disease by raising HDL and minimizing LDL. OBJECTIVE: This study synthesized ten meta-chlorinated benzene sulfonamides 6a-6j and explored their structure-activity relationship. METHODS: The synthesize

A Dual-Template Molecularly Imprinted Polymer to Inhibit Quorum Sensing Molecules: Theoretical Design, Optimized Synthesis, Physicochemical Characterization and Preliminary Microbiological Analysis.

Molecularly imprinted polymers (MIPs) have emerged as promising materials for selectively targeting biomolecules, including quorum sensing autoinducers that regulate bacterial communication and biofilm formation. In this study, both single-template and dual-template strategies were employed to design and synthesize MIPs capable of capturing autoinducer-2 analogs using (3R,4S)-tetrahydro-3,4-furandiol (T1) or (R/S) 2,2-dimethyl-1,3-dioxolane-4-methanol (T2) as the templates. This approach offers translational potential of a complementary or non-antibiotic strategy to conventional antimicrobial

The "Missing Link", Allostery and SynergismHosting of Metal Cations by Regular and Partial Cone Calix[4]arene Isomers.

The influence of the tetra-O-2-oxopropyl-substituted calix-[4]-arene conformation on its binding affinity toward first- and second-group metal cations, as well as on the solvent molecule (acetonitrile or methanol) inclusion in the calixarene hydrophobic cavity, was investigated experimentally and computationally. Misorientation of one monomeric subunit in the partial cone ligand (L p ) led to incomplete cation desolvation and significantly reduced its cation-binding ability compared to the regular cone isomer (L c ). Aromatic ring inversion also precluded solvent inclusion in the calixarene ba

A multifaceted examination of the action of PDE4 inhibitor rolipram on MMP2/9 reveals therapeutic implications.

A PDE4 (phosphodiesterase 4) inhibitor, Rolipram, was previously found to down-regulate (in a manner dependent on cAMP (cyclic adenosine monophosphate)-PKA (protein kinase A)) MMP2 (matrix metalloproteinase 2) and MMP9 protein expression levels, important markers of epithelial-to-mesenchymal transition in human breast cancer cell lines. However, zymographic studies revealed that rolipram could also alter the enzymatic activities of these MMPs, even in the presence of the PKA inhibitor H89. This calls for more detailed investigations of the inhibitory mechanism of rolipram on MMP2 and MMP9. The

Centimeter-Wave Rotational Spectroscopy of Ethynylbenzonitriles: Structural Analysis and Astronomical Search.

The identification of individual molecular species in the interstellar medium (ISM) via radio astronomical observations relies on precise laboratory measurement of the species' rotational spectrum. Following the detections of benzonitrile and ethynylbenzene in the ISM, we perform a search for the disubstituted benzene derivatives that feature both a nitrile and an ethynyl substituent: the ortho (o), meta (m), and para (p) isomers of ethynylbenzonitrile (EBN). Having previously been measured in the millimeter-wave regime, we extend the rotational spectroscopy of the isomeric family down to the

Synthesis and characterization of nanocapsules containing anti-inflammatory drugs: in vitro and in silico biological activity.

Cancer constitutes a major cause of death globally. Many current treatments are not very selective and often harm healthy cells. Inflammation is known to be associated with tumor growth, yet anti-inflammatory drugs alone are rarely used in a targeted manner. The aim of this study was to examine the synergic activity of two frequently used anti-inflammatory drugs, dexamethasone acetate (DA), and nimesulide (NIME) in nanoencapsulated form to diminish toxicity but enhance therapeutic effectiveness. The stability of the nanocapsules was established by applying light scattering, zeta potential, ele

Targeted design, synthesis, molecular simulation, ADME-T and in-vitro anticancer assessment of phenyl-substituted-thioxo-tetrahydro-pyrimidin-benzenesulfonamide derivatives as potential BRAFV600E/WT inhibitors.

BRAF is one of the most commonly mutated oncogenes in human cancers, with more than 90 % of mutations occurring at codon 600. Among these, BRAFV600E is the most prevalent, accounting for nearly 90 % of all BRAF codon 600 mutations, while the less frequent variants are collectively referred to as BRAFV600WT. BRAF mutations are reported across several cancers, with the highest frequency in malignant melanoma (70-90 %). To target these mutations, a series of phenyl-substituted thioxo-tetrahydro-pyrimidine-benzenesulfonamide hybrids in the [αC-OUT/DFG-IN] conformation were designed. Fourteen deriv

Electrostatically tuning radical addition and atom abstraction reactions with distonic radical ions.

Although electrostatic catalysis can enhance the kinetics and selectivity of reactions to produce greener synthetic processes, the highly directional nature of electrostatic interactions has limited widespread application. In this study, the influence of oriented electric fields (OEF) on radical addition and atom abstraction reactions are systematically explored with ion-trap mass spectrometry using structurally diverse distonic radical ions that maintain spatially separated charge and radical moieties. When installed on rigid molecular scaffolds, charged functional groups lock the magnitude a
