The WSD-type wax ester synthase is widely conserved in streptophytes and crucial for floral organ formation under high humidity in land plants.

Cuticular waxes are a complex mixture of long-chain aliphatic compounds, including alkanes, aldehydes, alcohols, ketones, and esters, that cover the outer surface of most terrestrial plants. While cuticular waxes play a pivotal role in plant adaptation to various environmental stresses, the specific roles of individual molecular species, particularly wax esters (WEs), remain poorly understood. In this study, we investigate the evolution and function of wax ester biosynthesis in land plants, focusing on the WSD (wax synthase/DGAT) enzyme family. We demonstrate that the ancestral origin of WSD e

Revisiting the critical roles of reactive microglia in traumatic brain injury.

Traumatic brain injury (TBI) triggers a complex neuroinflammatory cascade, with microglia serving as key regulators of both pathological damage and tissue structural restoration. Despite extensive research, the precise temporal evolution of microglial activation and its implications for long-term neurological outcomes remain incompletely understood. Here, we provide a comprehensive review of the molecular and cellular mechanisms underlying microglial responses in TBI, highlighting their role in neuroinflammation, neurogenesis, and tissue remodeling. We systematically compare clinical and precl

Development and Application of MLB Human Astrovirus Reverse Genetics Clones and Replicons.

Human astroviruses pose a significant public health threat, especially to children, the elderly, and immunocompromised individuals. Nevertheless, these viruses remain largely understudied, with no approved antivirals or vaccines. This protocol focuses on leveraging reverse genetics (RG) and replicon systems to unravel the biology of MLB genotypes, a key group of neurotropic astroviruses. Using reverse genetics and replicon systems, we identified critical genetic deletions linked to viral attenuation and neurotropism, pushing forward vaccine development. We also uncovered novel replication mech

The Remediation of Arsenic-Contaminated Soil by Pteris vittata L. Facilitates the Recovery of Soil Bacterial Diversity and Network Complexity.

The remediation of contaminated soils is essential for restoring land productivity and soil health. Pteris vittata L., an arsenic hyperaccumulator, has been widely used for phytoremediation, yet its ecological effects on soil systems remain insufficiently understood. In this field study, we evaluated the influence of Pteris vittata L. remediation on soil physico-chemical properties, microbial diversity, and molecular ecological networks. The results showed that long-term arsenic contamination significantly reduced soil total carbon, total nitrogen, and available phosphorus, simplified bacteria

Design principles of ciliary signaling.

Primary cilia are microtubule-based sensory organelles that have been conserved throughout eukaryotic evolution. As discussed in this Review, a cilium is an elongated and highly specialized structure, and, together with its ability to selectively traffic and concentrate proteins, lipids and second messengers, it creates a signaling environment distinct from the cell body. Ciliary signaling pathways adopt a bow-tie network architecture, in which diverse inputs converge on shared effectors and second messengers before diverging to multiple outputs. Unlike other cellular bow-tie systems, cells ex

Patterned wireless transcranial optogenetics generates artificial perception.

Synthesizing perceivable artificial neural inputs independent of typical sensory channels remains a fundamental challenge in developing next-generation brain-machine interfaces. Establishing a minimally invasive, wirelessly effective and miniaturized platform with long-term stability is crucial for creating research methods and clinically meaningful biointerfaces capable of mediating artificial perceptual feedback. Here we demonstrate a miniaturized, fully implantable transcranial optogenetic neural stimulator designed to generate artificial perceptions by patterning large cortical ensembles w

Toxic Effects and Adaptive Response Mechanisms of Lignocellulose-Derived Phenolic Aldehydes in Saccharomyces cerevisiae during Cellulosic Ethanol Fermentation.

Lignocellulose-derived phenolic aldehydes, including p-hydroxybenzaldehyde (P), vanillin (V), and syringaldehyde (S), exert synergistic inhibition on the fermentation performance of Saccharomyces cerevisiae. In this study, a robust S. cerevisiae S91 was obtained through adaptive laboratory evolution under increasing concentrations of a mixed phenolic aldehyde cocktail (P + V + S). Phenotypic analyses of S. cerevisiae S91 revealed enhanced phenolic aldehyde detoxification, improved cell morphology, and increased membrane integrity under inhibitory conditions. The evolved strain exhibited over 6

Human parvovirus B19 vertical infection and hydrops fetalis. A case report.

Non-immune hydrops fetalis represents a diagnostic challenge in high-risk pregnant women. Vertical infection with human parvovirus B19 (B19V) is a possible cause. National guidelines propose maternal serologic screening (IgG/IgM), which may be insufficient in some situations. We report a case of vertical B19V infection with difficulties in prenatal diagnosis. Preterm newborn, normal weight (2950 g), born to a 30-year-old mother with anemia and hydrops fetalis (week 17). Cardiac, chromosomal, isoimmunization-Rh, and usual infectious causes (TORCH) were ruled out. Maternal serology for B19V show

Cerebral Venous Thrombosis in Pediatric Oncology: A Case of Burkitt Lymphoma.

Cerebral venous thrombosis (CVT) is an uncommon but serious complication in pediatric patients with cancer. We report the case of a nine-year-old boy diagnosed with Burkitt lymphoma involving the nasopharyngeal region, who developed superior sagittal sinus thrombosis during chemotherapy. Neurological examination prior to treatment revealed no signs of intracranial hypertension. However, two weeks after completing the first course of chemotherapy, the patient presented with severe headaches and intermittent vomiting. Neuroimaging confirmed cerebral venous thrombosis, while the tumor mass was re

How Neuromodulation Changed the Landscape of Epilepsy Surgery.

BACKGROUND: Epilepsy is one of the most prevalent chronic neurological disorders, with approximately 30% of patients not responding to medical treatment. In selected cases, drug-resistant epilepsy can be safely managed with neuromodulation, leading to a significant reduction in disease burden. SUMMARY: Experimental evidence has demonstrated that the primary neuromodulation modalities, vagus nerve stimulation (VNS), deep brain stimulation (DBS), and responsive neurostimulation (RNS), can modulate various brain circuits and reduce epileptic activity by decreasing neuronal hypersynchronization t

Is Eosinophilic Colitis a Forerunner of Inflammatory Bowel Disease?

PURPOSE OF REVIEW: Eosinophilic colitis (EoC) has an increasing association with inflammatory bowel disease (IBD), and there is a paucity of data on the natural history of EoC. Here, we provide a comprehensive overview of EoC and critically evaluate the evidence supporting its potential association with, or evolution into IBD. RECENT FINDINGS: Experts currently classify EoC as one of the eosinophilic gastrointestinal disorders (EGID) beyond eosinophilic esophagitis (EoE). Transcriptomic analyses have demonstrated a distinct molecular signature for EoC. However, recent studies suggest that a s

Mpox virus: virology, molecular epidemiology, and global public health challenges.

Monkeypox (Mpox), a zoonotic disease caused by the Mpox virus (MPXV), has re-emerged as a significant global health concern, particularly since the 2022 outbreaks in non-endemic countries. MPXV shares close virological and genetic similarities with other orthopoxviruses, notably variola virus. The current circulating strains, primarily of clade IIb, exhibit enhanced human-to-human transmissibility. This review synthesizes recent advances in MPXV virology, pathogenesis, molecular evolution, clinical features, and diagnostic techniques. In addition, we highlight the mounting public health challe

Identification of a Novel HIV-1 Circulating Recombinant Form (CRF184_BC) Among Heterosexual Populations Along the China-Myanmar Border Region.

The China-Myanmar border region is recognized as a hotspot for the emergence of HIV-1 recombinant forms. This study identified a novel HIV-1 circulating recombinant form (CRF) in this area, designated CRF184_BC. Near-full-length genome sequences were obtained from four individuals infected through heterosexual contact: one Chinese individual and three Burmese individuals. These sequences formed a distinct monophyletic clade with strong bootstrap support, separate from all known subtypes/CRFs. Recombination analysis revealed a conserved mosaic structure comprising four subtype C segments and th

Case Report: Evolution and targeted therapy of an EGFR-mutant large-cell neuroendocrine carcinoma.

We report the case of a 47-year-old female non-smoker diagnosed with stage IV large-cell neuroendocrine carcinoma (LCNEC) of the lung harboring an EGFR exon 21 L858R mutation. The patient exhibited a sustained response to first-line osimertinib, with a progression-free survival of 20 months, followed by transformation to small-cell lung cancer (SCLC) confirmed via histopathological reassessment. Second-line treatment with etoposide and cisplatin combined with radiotherapy resulted in an additional 7 months of disease control. Subsequent progression was accompanied by features suggestive of ade

Converging and evolving immuno-genomic routes toward immune escape in breast cancer.

The interactions between tumor and immune cells along the course of breast cancer progression remain largely unknown. Here, we extensively characterize multiple sequential and parallel multiregion tumor and blood specimens of an index patient and a cohort of metastatic triple-negative breast cancers. We demonstrate that a continuous increase in tumor genomic heterogeneity and distinct molecular clocks correlated with resistance to treatment, eventually allowing tumors to escape from immune control. TCR repertoire loses diversity over time, leading to convergent evolution as breast cancer progr

Characterization of self-templating catalytic amyloids.

Amyloid aggregates with unique periodic structures have garnered significant attention due to their association with numerous diseases, including systemic amyloidoses and the neurodegenerative diseases Parkinson's, Alzheimer's, and Creutzfeld-Jakob. However, more recent investigations have expanded our understanding of amyloids, revealing their diverse functional biological roles. Amyloids have also been proposed to have played a significant role in prebiotic molecular evolution because of their exceptional stability, spontaneous formation in a prebiotic environment, catalytic and templating a

A Prebiotic Genetic Nucleotide as an Early Darwinian Ancestor for Pre-RNA Evolution.

Prebiotic genetic nucleotides (PGNs) often outcompete canonical alphabets in the formation of nucleotides and subsequent RNA oligomerization under early Earth conditions. This indicates that the early genetic code might have been dominated by pre-RNA that contained PGNs for information transfer and catalysis. Despite this, deciphering pre-RNAs' capacity to acquire function and delineating their evolutionary transition to a canonical RNA World has remained under-researched in the origins of life (OoL) field. We report the synthesis of a prebiotically relevant nucleotide (BaTP) containing the no
