Molecular evolution of wound healing-related genes during cetacean secondary aquatic adaptation.

The marine environment presents challenges for wound healing in cetaceans, despite their remarkable recovery abilities with minimal infections or complications. However, the molecular mechanism underlying this efficient wound healing remains underexplored. To better understand the molecular mechanisms behind wound healing in cetaceans, we investigated the evolutionary patterns of 37 wound healing-related genes in representative mammals. We found wound healing-related genes experience adaptive evolution in cetaceans: (1) Three extrinsic coagulation pathway-related genes-tissue factor (F3), coag

Predictors of a relapsing course in myelin oligodendrocyte glycoprotein antibody-associated disease.

BACKGROUND: Myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD) is a recently described demyelinating disorder, and children represent about 50% of all cases. Almost half of the patients experience relapses, but very few studies have evaluated predictors of relapse risk, challenging clinical management. The study aimed to identify predictors at MOGAD onset that are associated with a relapsing course. METHODS: Prospectively collected data from paediatric patients with MOGAD seen by the US Network of Paediatric MS Centres were leveraged. Univariable and adjusted multivariabl

Immediate Interferon Gamma Induction Determines Murine Host Compatibility Differences between Toxoplasma gondii and Neospora caninum.

Rodents are critical for the transmission of Toxoplasma gondii to the definitive feline host via predation, and this relationship has been extensively studied as a model for immune responses to parasites. Neospora caninum is a closely related coccidian parasite of ruminants and canines but is not naturally transmitted by rodents. We compared mouse innate immune responses to N. caninum and T. gondii and found marked differences in cytokine levels and parasite growth kinetics during the first 24 h postinfection (hpi). N. caninum-infected mice produced significantly higher levels of interleukin-1

Indications for use and safety of rituximab in childhood renal diseases.

Rituximab was initially developed for the treatment of patients with B cell lymphoma but has during the last decade proven to be quite effective in treating a range of kidney diseases including lupus nephritis, nephrotic syndrome, and also in different situations before and after a renal transplant. We will here review the scientific basis for the use of rituximab in children with renal diseases and give recommendations both regarding its clinical use and need for further research.

Localized cutaneous Cryptococcus albidus infection in a 14-year-old boy on etanercept therapy.

Immune-modulating medications, such as the tumor necrosis factor-alpha inhibitors, are establishing a niche in the dermatologic therapeutic armamentarium as novel, effective, and relatively safe alternatives in the treatment of numerous inflammatory dermatoses. The benefits of anti-tumor necrosis factor-alpha inhibitor therapy are well known. However, these agents are also associated with an increased risk of infectious complications--most commonly granulomatous infections with mycobacteria and fungi. We present a teenaged boy with localized, cutaneous Cryptococcus albidus infection on the sca

Tumour necrosis factor-alpha as a new therapeutic target for rheumatoid arthritis: an update.

In rheumatoid arthritis (RA), the conventional therapies (first-line, second-line, third-line drugs) provide more or less effective symptomatic relief for a decade or so from the onset of the disease. However, the chronic inflammatory destructive processes involving connective tissue, cartilage and bone with their attendant disability progress relentlessly in majority of patients. Secondly, use of 'second-line' and 'third-line' drugs in RA are limited due to their side effects. Studies in animals and RA patients have confirmed that tumour necrosis factor-alpha (TNFalpha), an inflammatory cytok

Long-term risks associated with biologic response modifiers used in rheumatic diseases.

PURPOSE OF REVIEW: The introduction of tumor necrosis factor-alpha antagonists in 1998 has had a significant impact on the treatment of rheumatoid arthritis. However, as use of these agents has increased worldwide, infrequent adverse events that were not apparent in pivotal controlled clinical trials required for registration have emerged. RECENT FINDINGS: These adverse events include serious infections, particularly tuberculosis, which may be atypical in presentation. Concern regarding increased risk of lymphoma has also emerged, although it remains unclear whether the risk exceeds that obse

Inhibiting OX40 Restores Regulatory T-Cell Function and Suppresses Inflammation in Pulmonary Sarcoidosis.

BACKGROUND: Pulmonary sarcoidosis (PS) is a noncaseating granulomatous disease of unknown origin. Despite conflicting reports, it is considered that the regulatory T (Treg) cells are functionally impaired in PS, but the underlying mechanisms remain unclear. OX40, a pivotal costimulatory molecule, is essential for T-cell functions and memory development, but its impact on Treg cells is ambiguous. RESEARCH QUESTION: Does the OX40 pathway influence the suppressive functions of Treg cells in PS? STUDY DESIGN AND METHODS: Fifty treatment-naïve patients with PS and 30 healthy control participants

A novel model for IFN-γ-mediated autoinflammatory syndromes.

Autoinflammatory disease and hyperinflammatory syndromes represent a growing number of diseases associated with inappropriately controlled inflammation in multiple organs. Systemic inflammation commonly results from dysregulated activation of innate immune cells, and therapeutic targeting of the IL-1β pathway has been used to ameliorate some of these diseases. Some hyperinflammatory syndromes, however, such as hemophagocytic lymphohistiocytosis and the newly classified proteasome disability syndromes, are refractory to such treatments, suggesting that other factors or environmental stressors m

N-acetyl-L-cysteine exhibits antitumoral activity by increasing tumor necrosis factor alpha-dependent T-cell cytotoxicity.

Because of its anticarcinogenic and antimutagenic properties, N-acetyl-L-cysteine (NAC) has been proposed for cancer treatment. Here we present a mechanism of action for NAC in cancer. Our data show that NAC (1) induces an early and sustained increase of membrane tumor necrosis factor alpha (TNF alpha) expression on human stimulated-peripheral blood (PB) T cells and (2) increases membrane TNF-RI and TNF-RII on tumoral cell lines and on T cells after stimulation. These effects result from an early inhibition of both TNF alpha and TNF-R shedding, as well as a later increase of the respective mRN

Colorectal cancer landscape from gut microbiota: Insights into mutations, epigenetic dysregulation, and immune microenvironment alterations.

This review explores how gut microbiota reshapes colorectal cancer (CRC) molecular landscape, driving initiation/progression via key mechanisms. Microbes/metabolites cause driver mutations (DNA damage, signaling interference) and alter epigenetics (DNA methylation, histone modifications, ncRNAs) to boost tumor cell proliferation/survival. Dysbiosis disrupts tumor immune microenvironment (TIME) by impairing immune cells and increasing immunosuppressive factors, fostering immune evasion.Integrating molecular biology, microbiology, and immunology, we show microbial changes are causal in oncogenes

Mechanisms driving immunopathogenesis of viral exacerbations in chronic respiratory disease.

BACKGROUND: Exacerbations are major causes of morbidity in individuals with chronic respiratory diseases such as chronic obstructive pulmonary disease, asthma and bronchiectasis. Increasing evidence implicates respiratory viruses as predominant triggers, though the underlying immunopathogenic mechanisms remain poorly understood. NARRATIVE: This review synthesises current knowledge on the interplay between viral pathogens at the airway epithelial barrier, including structural and immunological mechanisms that may dysregulate antiviral immunity in chronic respiratory diseases. Furthermore, we d

Immune response to rotavirus vaccines in low- and middle-income countries: Challenges and perspectives.

Rotavirus is a highly contagious pathogen and a leading cause of severe diarrhea cases in children under 5 y of age worldwide. Although effective oral rotavirus vaccines have been developed and implemented globally, their performance varies substantially by setting. In high-income countries (HICs), these oral vaccines exhibit 85-90% efficacy in preventing rotavirus-associated hospitalizations, with protection lasting until at least 2 y of age. In contrast, vaccine efficacy in low- and middle-income countries (LMICs) is considerably lower, ranging from approximately 45% to 65%. To address this

The emerging roles of NFIL3 in immunity.

NFIL3 (E4BP4) is a basic leucine zipper transcription factor best known for its roles in circadian regulation. It functions primarily as a transcriptional repressor through competitive DNA binding and the recruitment of histone-modifying complexes. More recently, NFIL3 has emerged as a critical regulator of immunity. It is essential for the development of multiple immune cell subsets, including NK cells and CD8+ dendritic cells, and modulates the activity of many others. For instance, NFIL3 influences B cell class switching and cytokine production in T helper subsets and macrophages. Its expre

Molecular mechanisms of streptococcal disruption of the blood-brain barrier and their pathogenic role in bacterial meningitis.

Bacterial meningitis represents a devastating inflammatory disease of the central nervous system (CNS), characterized by the invasion of pathogens across the blood-brain barrier (BBB) and subsequent dysregulated immune responses. Key inflammatory mechanisms include pathogen recognition by microglial TLRs and NLRP3, neutrophil infiltration, and cytokine storms such as IL-1β and TNF-α, leading to BBB disruption, cerebral edema, and neuronal injury. Despite antimicrobial therapy, excessive inflammation often results in neurological sequelae. Emerging strategies target immunomodulation through inf

A systematic review on the role of gut microbiome in inflammatory bowel disease: Spotlight on virome and plant metabolites.

Inflammatory bowel diseases (IBD), including ulcerative colitis and Crohn's disease, arise from various factors such as dietary, genetic, immunological, and microbiological influences. The gut microbiota plays a crucial role in the development and treatment of IBD, though the exact mechanisms remain uncertain. Current research has yet to definitively establish the beneficial effects of the microbiome on IBD. Bacteria and viruses (both prokaryotic and eukaryotic) are key components of the microbiome uniquely related to IBD. Numerous studies suggest that dysbiosis of the microbiota, including ba

Parvovirus B19-associated pure red cell aplasia in the setting of HIV and competing health risks.

An HIV positive, early adolescent girl from rural Southern Africa required multiple hospital admissions for severe normocytic anaemia. As she was virologically suppressed on antiretrovirals, the initial anaemia workup focused on nutritional deficiencies and excluding chronic haemolysis. A lack of readily available expertise and resources contributed to a delay in investigations, during which she was treated symptomatically with blood transfusions. She was later transferred to a tertiary hospital where she was diagnosed with parvovirus B19 (B19V) associated pure red cell aplasia. Intravenous im

An approach to Repeat Breeder Syndrome. Cervical mucus immunoglobulin content does not alter sperm motility.

Repeat breeder (RB) syndrome plays a detrimental role on fertility and economic performance of dairy farms. Regarding its multifactorial origin, it has been stated that an immunological response in the female genital tract may impair sperm viability. Therefore, the aim of this study was to evaluate the concentration of immunoglobulins in cervical mucus of RB cows and its influence on sperm motility. Fifteen fertile cows and 32 RB cows were included in the study. Cervical mucus samples were collected at the time of artificial insemination (AI). The concentration of IgG and IgA was determined by
