CD14+CD16- monocytes exhibit NF-κB hyperactivation in biliary atresia: Clinical association and murine therapeutic validation.

BACKGROUND: Classical CD14+CD16- monocytes are elevated in biliary atresia (BA); however, their specific role in bile duct injury and the underlying regulatory mechanisms remain unclear. This study aimed to define their contribution to BA pathogenesis, focusing on the NF-κB signaling pathway. METHODS: Liver tissues and blood samples from patients with BA and controls were analyzed by single-cell RNA sequencing, flow cytometry, and immunofluorescence. A rhesus rotavirus-induced BA mouse model was used for anti-Ly6C monocyte depletion and NF-κB inhibition (dehydroxymethylepoxyquinomicin). Trans

Pathological depositions in human disease: converging mechanisms in atherosclerosis, Alzheimer's disease, and related disorders.

Pathological deposition of endogenous material within tissues represents a central, unifying mechanism across a broad spectrum of prevalent human diseases. Atherosclerosis and Alzheimer's disease (AD) exemplify this paradigm: atherosclerosis is driven by subendothelial accumulation of apolipoprotein B-containing lipoproteins, cholesterol crystals, and hydroxyapatite, while AD is characterized by cerebral deposition of misfolded amyloid-β (Aβ) and/or hyperphosphorylated tau. Emerging evidence implicates substantial lipid dyshomeostasis in AD pathogenesis, including lipid-droplet-accumulating mi

The processing of pro-inflammatory cytokines in post-traumatic stress disorder: Inflammation as a central mechanism in PTSD pathophysiology.

Post-traumatic stress disorder (PTSD) is a severe psychiatric condition associated with persistent emotional dysregulation, cognitive impairment, and structural alterations in limbic and prefrontal brain regions. Growing evidence indicates that chronic inflammation and abnormal processing of pro-inflammatory cytokines are central components of PTSD pathophysiology. Activation of the NOD-like receptor family pyrin domain containing 3 inflammasome (NLRP3) and caspase-1 promotes the proteolytic maturation of pro-interleukin-1β (pro-IL-1β) and pro-interleukin-18 (pro-IL-18) into their active forms

Nanoparticle-driven macrophage polarization as a therapeutic strategy for chronic diabetic wounds.

Chronic diabetic wounds, including diabetic foot ulcers, present significant clinical challenges due to persistent inflammation, oxidative stress, impaired angiogenesis, infection, and defective tissue regeneration. Central to these pathological features is macrophage dysfunction, wherein immune cells become locked in a pro-inflammatory state that impedes the transition to reparative phenotypes necessary for resolution of inflammation and tissue repair. Nanomedicine-based strategies have emerged as promising approaches to restore macrophage plasticity and promote regenerative healing. Multifun

Polystyrene microplastics and hepatic fibrosis-related indices in type 2 diabetes: a cross-sectional analysis with experimental validation.

BACKGROUND: Microplastics (MPs) are emerging environmental contaminants with potential hepatotoxicity, yet direct epidemiological evidence linking internal MP exposure to liver injury is scarce, particularly in individuals with type 2 diabetes (T2D). In this cross-sectional study nested within the METAL2 cohort, we aimed to characterize blood microplastic profiles in patients with T2D and to determine whether specific MP polymers are associated with hepatic steatosis and fibrosis risk, with experimental validation of the identified polymer. RESULTS: In patients with T2D, multiple MP polymers

Roles of inflammation and metabolic dysfunction in cardiovascular diseases: molecular mechanisms and therapeutic targets.

Cardiovascular disease progression is shaped by a close interaction between metabolic adaptation and inflammatory signaling. In injured cardiac and vascular tissues, ischemia, pressure overload, lipid stress and metabolic excess reshape how cardiomyocytes, endothelial cells, immune cells and fibroblasts sense damage and communicate with their microenvironment. Cardiovascular inflammation follows a staged tissue response, from danger-signal sensing and nuclear factor-κB-related priming to inflammasome activation, cytokine amplification, leukocyte recruitment, efferocytosis and chronic remodelin

Dopamine receptors in ocular surface biology: Current evidence and future directions.

Dopamine, a classical catecholamine neurotransmitter, exerts diverse regulatory functions in the central nervous system, retina, and intraocular signaling. Recent research has highlighted its emerging role in ocular surface biology, which includes the cornea, conjunctiva, tear film, and lacrimal glands. The ocular surface serves as a neuroepithelialimmune interface where neuromodulators such as dopamine influence epithelial homeostasis, wound repair, inflammation, and sensory signaling. Molecular and functional studies have confirmed the expression of dopamine receptor subtypes (DRD1-DRD5) and

Intraparenchymal B cell therapy protects neurological function and modulates local neuroinflammation in experimental hemorrhagic stroke.

Intracerebral hemorrhage (ICH) is a devastating subtype of stroke, with a 5-year mortality of up to 70 %. Disease-modifying treatments to improve neurological outcomes in ICH survivors represent a critical unmet need. Neuroinflammation contributes significantly to ongoing secondary brain injury and long-term morbidity in ICH. We and others have shown that B cells are potent modulators of the inflammatory response, capable of acquiring a regulatory phenotype within injured microenvironments. Here, we investigated the effects of a single intraparenchymal application of mature naïve splenic B lym

Transcriptomic and proteomic signatures following AS03-adjuvanted Influenza A/H7N9 vaccine.

INTRODUCTION: Vaccines targeting avian influenza virus A/H7N9 are poorly immunogenic. While the immune responses can be improved with oil-in-water emulsion adjuvants such as Adjuvant System 03 (AS03), the cellular mechanisms underpinning the adjuvant effect are incompletely characterized and poorly understood. METHODS: We enrolled 30 healthy adult participants and used RNA sequencing and quantitative proteomics to characterize the response to two doses of the influenza A/H7N9 vaccine, with and without AS03, in six immune cell types. These responses were compared to those seen after administra

Sympathetic regulation of bone homeostasis and remodeling: molecular mechanisms, pathophysiological roles, and therapeutic implications.

Bone metabolic disorders, such as osteoporosis, osteoarthritis, and rheumatoid arthritis, are characterized by disrupted skeletal homeostasis and represent major challenges in musculoskeletal health. Bone remodeling has long been interpreted as a coordinated process between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. However, emerging evidence indicates that bone is not merely a structural tissue, but a dynamic regulatory organ influenced by neural, endocrine, immune, metabolic, and vascular signals. Among these regulatory systems, the sympathetic nervous system

Mesenteric denervation ameliorates post‑infarction heart failure alongside alterations in the gut-nerve-microbiota axis.

Sympathetic overactivity and gut microbiota dysbiosis drive chronic heart failure (CHF) progression. This study explored the associations of mesenteric denervation (MDN) with cardioprotection and potential involvement of the nerve-microbiota-heart axis in MI-induced CHF mice. C57BL/6 mice were randomized into control, CHF, and CHF_MDN groups. CHF was induced by LAD ligation, and MDN was performed via phenol ablation of the superior mesenteric artery. MDN was associated with suppressed mesenteric sympathetic activity, improved left ventricular ejection fraction, and reduced myocardial fibrosis

Colchicine attenuates myocardial inflammation, endothelial dysfunction, and apoptosis in a rat model of alcohol-related cardiac damage.

BACKGROUND: Chronic alcohol consumption is a recognized contributor to non-ischemic dilated cardiomyopathy, particularly in individuals with an underlying genetic predisposition, through myocardial inflammation, oxidative stress, and apoptosis. Colchicine, an NLRP3 inflammasome inhibitor, has cardioprotective potential, but its role in alcohol-induced injury is unknown. This study aimed to evaluate the dose-dependent effects of colchicine (0.1 mg/kg and 0.3 mg/kg) on alcohol-induced cardiac damage in rats. METHODS: Overall, 40 Sprague-Dawley rats were randomized into five groups: control, alc

The Oxidative-Mitochondrial-Inflammatory Axis in Retinitis Pigmentosa: Extracellular mtDNA as Biomarker and Therapeutic Read-Out.

Retinitis pigmentosa (RP) is the most common inherited retinal dystrophy (prevalence ~1:4000) and a leading Mendelian cause of working-age blindness. Despite marked genetic heterogeneity, its progression converges on a common secondary cascade of outer-retinal hyperoxia, increased reactive oxygen species (ROS), and mitochondrial dysfunction that drives cone degeneration and central vision loss. Because this oxidative cascade is largely genotype-independent and pharmacologically tractable, oxidative stress is a cross-cutting therapeutic target. Within it, mitochondrial DNA (mtDNA) is a key elem

Human Umbilical Cord Mesenchymal Stem Cells and Their Extracellular Vesicles in Neurological Disorders: Mechanisms, Delivery Strategies, and a Translational Roadmap.

BACKGROUND: Neurological disorders-including ischemic and hemorrhagic stroke, traumatic brain injury, spinal cord injury, neurodegenerative disease, chronic neuropathic and inflammatory pain, peripheral nerve injury, and selected psychiatric and neurodevelopmental disorders-remain major causes of death and disability. Human umbilical cord mesenchymal stromal cells (hUC-MSCs) and their extracellular vesicles (EVs) are being investigated as regenerative and immunomodulatory products, but translation is constrained by product heterogeneity, limited direct clinical evidence, uncertain CNS exposure

Convergent Astrocytic Failure in Parkinson's Disease: A System-Level Model of Pathogenesis and Treatment.

A paradigm shift from a purely neurodegenerative disorder to a multicellular failure of the neuroglial unit has fundamentally redefined Parkinson's disease (PD). This review synthesizes compelling evidence that positions astrocyte dysfunction, or astrocytopathy, as a central and active driver of pathogenesis, extending far beyond a passive bystander role. We delineate how pathological α-synuclein triggers a vicious cycle of astrocytic failure, encompassing impaired proteostasis via ubiquitin-proteasome and autophagy-lysosomal pathways, glymphatic system collapse due to aquaporin-4 (AQP4) depol

SIRT3 mediates mitochondrial protection and attenuates mtROS-TXNIP-NLRP3 signaling activation in dry eye disease.

BACKGROUND/AIM: Dry eye disease (DED) is a chronic inflammatory disorder of the ocular surface in which mitochondrial dysfunction and oxidative stress have been increasingly implicated, yet the molecular mechanisms linking mitochondrial redox imbalance to inflammatory signaling remain unclear. This study aimed to investigate the role of Sirtuin 3 (SIRT3)-mediated mitochondrial protection in DED and to elucidate the involvement of the mtROS-TXNIP-NLRP3 signaling pathway. METHODS: We established animal model of benzalkonium chloride (BAC) induced mice, and cellular model of hypertonic medium-ch

Targeting Macrophage-Mediated Mechanisms in Sepsis-Induced Cardiomyopathy: From Pathophysiology to Therapeutic Strategies.

Sepsis-induced cardiomyopathy (SICM) is a life-threatening complication of sepsis, characterized by dysregulated inflammation, metabolic disturbances, and complex cellular crosstalk. As central innate immune cells, macrophages exert a dual role in this pathology. This review focuses on the key mechanisms by which macrophages contribute to SICM. We discuss macrophage-driven pathogenesis across three interconnected themes. First, inflammatory activation. Macrophages amplify the inflammatory storm through dynamic M1/M2 polarization and activation of Toll-like receptor 4 (TLR4) / nuclear factor κB

Thrombo-inflammation as a missing variable in thoracic aortic intervention timing: toward a biology-informed threshold.

Aortic dissection and aneurysm have long been framed as mechanical failures of a degenerating wall. A growing body of evidence suggests that they may also reflect a shared thrombo-inflammatory process, in which coagulation and innate immunity are increasingly proposed to contribute actively to wall destruction rather than merely to indicate it. This review synthesizes that shift across the dissection and aneurysm spectrum. We first delineate the core machinery, including tissue-factor-driven coagulation, platelet and leukocyte interactions, neutrophil extracellular traps, inflammasome signalin

Genetic diversity of collaborative cross mice enables the establishment of a novel Chlamydia muridarum female genital tract infection model.

Chlamydial infection in women displays wide variation in bacterial burden, persistence, and risk of upper genital tract pathology, yet the host genetic factors underlying this heterogeneity remain poorly defined. We evaluated genital tract infection with Chlamydia muridarum across 20 Collaborative Cross (CC) strains, a recombinant inbred mouse panel that captures broad genetic diversity with high within-strain reproducibility. CC strains exhibited striking differences in early bacterial burden, time to clearance, and oviduct pathology, including prolonged low-inflammatory infections and burden

HMGB1 signalling in Alzheimer's disease: pathogenic roles and therapeutic prospects.

Alzheimer's disease (AD) is a progressive neurodegenerative condition marked by the gradual loss of neurons, especially in the hippocampus and cerebral cortex. This neuronal loss results in cognitive decline, memory problems, and changes in behaviour. It accounts for roughly 90% of all cases, making it the most common reason for dementia worldwide, with a marked rise in its occurrence as one ages. AD is pathologically marked by the presence of intracellular neurofibrillary tangles composed of hyperphosphorylated tau protein and the formation of extracellular amyloid-β plaques. Along with these
