Artesunate Targets MBL2 to Modulate the TLR4/NRF2/HO‑1 Axis and Ameliorate Podophyllotoxin‑Induced Liver Injury.

BACKGROUND: Drug-induced liver injury (DILI) presents a significant clinical challenge with limited preventive and therapeutic options. Artesunate (ART) holds potential for DILI treatment; however, its precise hepatoprotective mechanisms and direct molecular target(s) remain unclear. METHODS: We evaluated ART in a podophyllotoxin (PPT)-induced rat model of DILI. Its direct target was identified through integrated chemical proteomics (pull-down/LC-MS/MS) followed by biophysical validation (SPR, MST) and molecular dynamics simulations. Mechanistic insights from hepatic transcriptomics (RNA-seq)

An Integrated Multi-omics Single Cell Atlas of the Human RPE and Choroid.

The retinal pigment epithelium and choroid are critical for supporting the function and maintaining the homeostasis of the outer retina, and their dysfunction underlies a range of inherited and complex ocular diseases. To comprehensively characterize the cellular, transcriptomic, and epigenomic heterogeneity and dynamics within these tissues, we assembled an integrated multi-omics reference atlas comprising 719,813 single-cell/single-nucleus transcriptomes and 234,007 snATAC-seq profiles from 102 ancestrally diverse donors spanning 0 to 99 years of age, including cells from both the macula and

scRNA sequencing revealed HIV-associated inflammation-mediated lung epithelial dysregulation and fibroblast remodeling.

BACKGROUND: HIV infection is risk factor for a wide spectrum of pulmonary diseases, with incidence rates significantly higher in people living with HIV (PLWH). Despite antiretroviral therapy, persistent immune activation and recurrent injury continue to compromise lung integrity in this population. However, the mechanism by which HIV disrupts alveolar homeostasis and affects epithelial, immune, and stromal compartments in the lungs remain undefined. OBJECTIVE: To characterize the cellular and molecular landscape of HIV infection within the lungs, the focus of the study was to assess epithelia

Nanosystem-Mediated Phototherapy (PDT/PTT) - Chemodynamic Therapy for Synergistic Antitumor Therapy: Strategies and Advances.

Chemodynamic therapy (CDT) eradicates tumor cells by generating hydroxyl radicals (•OH) through Fenton/Fenton-like reactions, yet its therapeutic efficacy is severely constrained by intrinsic deficiencies of the tumor microenvironment (TME), including insufficient endogenous hydrogen peroxide(H2O2), suboptimal pH, robust glutathione (GSH)-mediated reactive oxygen species (ROS) scavenging, and tumor hypoxia. As potent synergistic modalities, photodynamic therapy (PDT) elevates oxidative stress via ROS overproduction, while photothermal therapy (PTT) accelerates Fenton reaction kinetics and impr

RNA modifications as determinants of cancer cell death: from epitranscriptomic mechanisms to therapeutic targeting.

Epitranscriptomics has rapidly evolved into a central layer of post-transcriptional gene regulation in cancer, yet its mechanistic contribution to regulated cell death remains incompletely resolved. This review critically examines how RNA modifications principally N⁶-methyladenosine (m⁶A), alongside 5-methylcytosine (m5C), N⁷-methylguanosine (m⁷G), pseudouridylation (Ψ), and adenosine-to-inosine (A-to-I) editing reprogram apoptotic and ferroptotic thresholds in malignant cells. It highlights that m⁶A does not exert uniform effects; rather, outcomes are dictated by site specificity, reader stoi

Assembly of a biomimetic copper-based nanocomplex for alleviating hypoxia to enhance cuproptosis against osteosarcoma and lung metastasis.

Osteosarcoma tissues demonstrated elevated expression of proteins (FDX1 and DLAT) integral to cuproptosis in our preliminary study, indicating the potential effectiveness of anti-tumor strategies predicated on this process. Nevertheless, the overexpression of copper export proteins and the challenge of copper ion penetration may contribute to insufficient local copper ion concentration for inducing cuproptosis. Herein, we engineered a biomimetic copper-elesclomol-polyphenol network for the efficient delivery of copper ions and the copper ionophore elesclomol. Simultaneously, we integrated cata

Gut Microbial Perturbation and Host Response Induce Redox Pathway Upregulation along the Gut-Liver Axis during Giardiasis in C57BL/6J Mouse Model.

Apicomplexan infections, such as giardiasis and cryptosporidiosis, negatively impact a considerable proportion of human and commercial livestock populations. Despite this, the molecular mechanisms of disease, particularly the effect on the body beyond the gastrointestinal tract, are still poorly understood. To highlight host-parasite-microbiome biochemical interactions, we utilised integrated metabolomics-16S rRNA genomics and metabolomics-proteomics approaches in a C57BL/6J mouse model of giardiasis and compared these to Cryptosporidium and uropathogenic Escherichia coli (UPEC) infections. Addition of Reduced Glutathione (GSH) to Freezing Medium Reduces Intracellular ROS Levels in Donkey Sperm.

In donkeys, the use of frozen-thawed sperm for artificial insemination (AI) leads to low fertility rates. Furthermore, donkey sperm produce a large amount of reactive oxygen species (ROS), and post-AI inflammation induces the formation of neutrophil extracellular traps (NETosis), which further generates many more ROS. These high ROS levels may induce lipid peroxidation in the sperm plasma membrane, thus affecting its integrity. Enzymatic and non-enzymatic antioxidants, mainly found in the seminal plasma (SP), are responsible for maintaining the redox balance. However, this fluid is removed pri

Novel concepts on pregnancy clocks and alarms: redundancy and synergy in human parturition.

The signals and mechanisms that synchronize the timing of human parturition remain a mystery and a better understanding of these processes is essential to avert adverse pregnancy outcomes. Although our insights into human labor initiation have been informed by studies in animal models, the timing of parturition relative to fetal maturation varies among viviparous species, indicative of phylogenetically different clocks and alarms; but what is clear is that important common pathways must converge to control the birth process. For example, in all species, parturition involves the transition of t

Yingmu San ameliorates pathological myopia by protecting retinal pigment epithelium and enhancing mitophagy-associated responses.

BACKGROUND: Pathological myopia is associated with progressive axial elongation and retinal pigment epithelium (RPE) injury, in which oxidative stress, inflammation, and mitochondrial dysfunction are important pathological contributors. Yingmu San (YMS) is a defined multi-component formulation composed of peony seed oil (PSO), lutein, and zeaxanthin at fixed proportions. However, whether YMS protects the RPE during myopia progression and whether mitophagy-associated responses are involved in this effect remain unclear. PURPOSE: This study aimed to determine whether YMS protects RPE cells and

Pathophysiological mechanisms of post-exertional malaise: an integrative analysis based on the metabolism-immune-neuro interaction model.

Post-exertional malaise (PEM) is a common core symptom in various chronic debilitating conditions, such as Post COVID-19 Condition (PCC, also known as Long COVID) and Chronic Fatigue Syndrome (CFS). It is characterized by the delayed and persistent exacerbation of symptoms following even mild physical or cognitive activities. This review presents a systematic review of the pathophysiological mechanisms involved in PEM, proposing a dynamic framework of multi-system interactions that may lead to homeostatic imbalance. The etiology of PEM is multifactorial, potentially involving factors such as t

Pilot longitudinal integrated transcriptomic-metabolomic study reveals immune and metabolic signatures in non-hospitalized healthcare workers with long COVID.

INTRODUCTION: Long COVID affects hundreds of millions of individuals worldwide, yet its underlying biological mechanisms remain incompletely understood, and the absence of validated biomarkers continues to limit diagnosis and clinical management. Most biomarker studies have focused on hospitalized patients with severe disease, leaving non-hospitalized populations, particularly healthcare workers, who are at high occupational risk, underrepresented. This gap may constrain the identification of biomarkers relevant to milder but persistent post-acute phenotypes. METHODS: We performed integrated

Research trends and cellular targets of NF-κB pathway in acute lung injury [2005-2024]: a combined bibliometric analysis and systematic review.

BACKGROUND: The nuclear factor-κB (NF-κB) pathway is a key regulator of inflammation and immune responses in acute lung injury (ALI). Despite significant progress in elucidating the molecular and cellular mechanisms of the NF-κB pathway in ALI, current research is scattered across different disciplines, experimental models, and cell types. Therefore, this study aims to integrate bibliometric trends with an in-depth review of molecular mechanisms to characterize publication trends, collaboration networks, and evolving research themes over the past two decades. METHODS: Literature related to th

Beyond Anti-VEGF: Toward Integrated Precision Care in Diabetic Retinal Disease.

Diabetic retinal disease (DRD) remains one of the leading causes of preventable vision loss worldwide and continues to represent a major global public health challenge despite remarkable therapeutic advances over the past two decades. Intravitreal anti-vascular endothelial growth factor (VEGF) therapy has revolutionized the management of diabetic macular edema and proliferative diabetic retinopathy, significantly improving visual outcomes and reducing vision-threatening complications. Nevertheless, a substantial proportion of patients exhibit incomplete anatomical or functional responses, requ

Family History of Substance Use and Stressful Life Events Impact Adolescent Maturation of Cerebral White Matter.

Family history (FH) of substance use disorders (SUDs) and stressful life events (SLEs) are known risk factors for SUDs in adolescents and young adults. Cross-sectional studies suggest that FH and SLEs affect adolescent white matter (WM) development and form abnormal WM patterns. Here, we examined the effects of FH, SLEs and their interaction on WM integrity in youths in the Adolescent Cognitive Brain Development (ABCD) study at baseline and 2- and 4-year follow-ups. ABCD youths (N = 8939, age ± SD = 9.9 ± 0.6 years, 4302 female) completed baseline diffusion tensor imaging, of which 5661 repeat

Costunolide mitigates inflammation and promotes extracellualr matrix integrity of thoracic aortic dissection by inhibiting NF-κB signaling.

BACKGROUND: Thoracic aortic dissection (TAD) is one of the most fatal cardiovascular diseases. One of its important pathological characteristics is the local inflammatory response. Many studies have found that Macrophage polarization plays an extremely critical role in the inflammatory progression and tissue remodeling of TAD. Costunolide (CTD) has an improving effect on oxidative stress and inflammation in the body. However, whether it can promote the integrity of extracellular matrix in Aortic dissection and its mechanism are still unclear. METHODS: The male C57BL/6J mice were used to const

Calcium-channel-blockers exhibit divergent regulation of cancer extravasation through the mechanical properties of cancer cells and underlying vascular endothelial cells.

Cardiovascular and cancer illnesses often co-exist, share pathological pathways, and complicate therapy. In the context of the potential oncological role of cardiovascular-antihypertensive drugs (AHD), here we examine the role of calcium-channel blocking drugs on mechanics of extravasating cancer cells, choosing two clinically-approved calcium-channel blockers (CCB): Verapamil-hydrochloride and Nifedipine, as model AHD to simultaneously target cancer cells (MCF7 and or MDA231) and an underlying monolayer of endothelial cells (HUVEC). First, live-cell microscopy shows that exposure to Nifedipin

EGR1 mediates neuronal damage via suppressing HIF1A-induced mitophagy following traumatic brain injury.

Traumatic brain injury (TBI) remains a leading cause of neurological morbidity and mortality, characterized by complex pathophysiological cascades. Here, we investigate the role of the transcription factor EGR1 (early growth response 1) in modulating mitochondrial homeostasis via the HIF1A (hypoxia inducible factor 1, alpha subunit)-BNIP3 (BCL2/adenovirus E1B interacting protein 3) axis following TBI. Using integrated transcriptomic and epigenomic analyses, we identified EGR1 as a critical regulator of TBI pathology, with its expression acutely upregulated in neurons post-injury. Genetic ablat

[Integrative transcriptomics-metabolomics approach to identify metabolic pathways regulated by glutamine synthetase activity].

Glutamine synthetase (GS), the only enzyme responsible for de novo glutamine synthesis, plays a significant role in cancer progression. As an example of the consequences of GS mutations, the R324C variant causes congenital glutamine deficiency, which results in brain abnormalities and neonatal death. However, the influence of GS-deficient mutations on cancer cells remains relatively unexplored. In this study, we investigated the effects of GS and GS-deficient mutations, including R324C and previously unreported K241R, which serve as models for GS inactivation. This study provided intriguing in
