Block of brain sodium channels by peptide mimetics of the isoleucine, phenylalanine, and methionine (IFM) motif from the inactivation gate.

Inactivation of sodium channels is thought to be mediated by an inactivation gate formed by the intracellular loop connecting domains III and IV. A hydrophobic motif containing the amino acid sequence isoleucine, phenylalanine, and methionine (IFM) is required for the inactivation process. Peptides containing the IFM motif, when applied to the cytoplasmic side of these channels, produce two types of block: fast block, which resembles the inactivation process, and slow, use-dependent block stimulated by strong depolarizing pulses. Fast block by the peptide ac-KIFMK-NH2, measured on sodium chann

A slow calcium-dependent component of charge movement in Rana temporaria cut twitch fibres.

1. Charge movement was studied in highly stretched frog cut twitch fibres in a double Vaseline-gap voltage-clamp chamber, with the internal solution containing either 0.1 mM EGTA or 20 mM EGTA plus 1. 8 mM total Ca2+. 2. Fibres were stimulated with TEST pulses lasting 100-400 ms. Replacement of the external Cl- with an 'impermeant' anion, such as SO42-, CH3SO3-, gluconate or glutamate, greatly reduced the calcium-dependent Cl- current in the ON segment and generated a slowly decaying inward OFF current in charge movement traces. 3. Application of 20 mM EGTA to the internal solution abolished t

Ion-transport properties of cultured bovine pancreatic duct epithelial cells.

Primary cultures of bovine pancreatic duct epithelial cells grown on permeable supports exhibit electrogenic transepithelial ion transport. The short-circuit current (I(sc)) generated by unstimulated duct cell monolayers and the increase in I(sc) elicited by increased levels of cyclic adenosine monophosphate (cAMP) were greater in monolayers bathed by bicarbonate-containing solution compared with monolayers bathed by nominally bicarbonate-free solution. An inhibitor of epithelial sodium channels (amiloride, 10 microM) had no effect on I(sc), whereas a Cl- channel blocker, N-phenylanthranilic a

Benzolamide inhibits low-threshold calcium currents in hippocampal pyramidal neurons.

1. Benzolamide is a poorly permeant sulfonamide inhibitor of the enzyme carbonic anhydrase. We studied the effect of benzolamide on low-threshold (LT) Ca currents in neonatal hippocampal CAl neurons. 2. In hippocampal slices, benzolamide (2-10 microM) inhibited the LT current 30-75% in voltage-clamped CAl pyramidal cells (n = 6). In slices bathed in N-2-hydroxypiperazine-N'-2-ethane-sulfonic acid (HEPES)-buffered Ringer, benzolamide also reduced the LT current, indicating that the action of the drug was not bicarbonate dependent. 3. Benzolamide inhibited LT Ca currents 20-75% in acutely dissoc

Modulation by stimulation rate of basal and cAMP-elevated Ca2+ channel current in guinea pig ventricular cardiomyocytes.

The modulation of L-type Ca2+ current (ICa) by changes in stimulation frequency was investigated in single ventricular cardiomyocytes isolated from guinea pig hearts. Electrical recordings were carried out at 21-25 degrees C and at 33-37 degrees C with the whole-cell patch clamp method, under K(+)-free conditions. A comparison is made between the response to frequency changes for ICa in the basal state and after the application of drugs which elevate the level of adenosine-3',5'-cyclic monophosphate (cAMP) within the cells. Peak basal ICa was reduced with an increase in stimulation rate from 0

Transcriptomic and RNAi analyses reveal chloride channel 3-associated osmoregulation in Litopenaeus vannamei under low-salinity stress.

Chloride channels and transporters are important for cellular volume regulation and salinity adaptation in euryhaline crustaceans, yet the intestinal transcriptional relationship between plasma-membrane and intracellular chloride pathways remains unclear in Litopenaeus vannamei. In this study, RNA interference of anoctamin 1 (ANO1) was combined with intestinal transcriptome sequencing under the production-relevant low-salinity condition of salinity 3. ANO1 silencing produced a focused transcriptional response, with 16 differentially expressed genes (DEGs) identified (11 upregulated and 5 downr

Quantum Dot-Based Dual-Fluorescence Aptasensing Platform Using Interface-Engineered MXene for Multiplex Protein Detection.

Antigen detection provides rapid and convenient diagnosis of respiratory infections. This study develops an innovative dual-fluorescence aptasensing method based on polydopamine-functionalized MXene (PDA-MXene) for the simultaneous detection of spike protein and hemagglutinin protein. The method employs green- and red-emitting quantum dot (QD) probes as fluorescence reporters, and the PDA-MXene as an effective adsorption and separation substrate. Coupled with a centrifugation-assisted separation strategy, this design method reduces background interference and enhances detection reliability. Th

Ferroelectric-Driven Nonvolatile Phase Transition in MoTe2 Transistors With an Expanded Memory Window.

Polymorphic MoTe2 offers a unique opportunity for phase-transition memristors due to its reversible structural transitions between semiconducting (2H) and metallic (1T') phases. Although gate-tunable phase transitions have been demonstrated, the presence of mobile ions in electrolyte gating leads to volatile behavior. Here, we demonstrate a phase-transition ferroelectric field-effect transistor (PT-FeFET) employing a polymorphic MoTe2 channel and a Hf0.5Zr0.5O2 (HZO) ferroelectric gate insulator. The ferroelectric polarization of the HZO layer induces an electrically driven, long-term nonvolat

Ionic Liquids at the Interface With the Transition Metal Dichalcogenide MoS2: Effect of the Chemical Structure on Induced Charge Carrier Density.

Transition metal dichalcogenides (TMDs) are emerging layered materials suited for applications in electronics, optoelectronics, and sensing thanks to their semiconductor nature. More recently, electric-field tuning of surface carrier density through carrier accumulation or depletion in TMDs-based field-effect-transistor devices has been attracting a great deal of interest as a new method to control superconductivity in this class of materials. Such results were achieved by means of ionic liquids (ILs) acting as a gating medium for MoS2 crystals. Despite the intensive use of ILs for gating TMDs

Channelopathies (Olmsted Syndrome)-causing point mutations of TRPV3 disrupt cellular thermal homeostasis in human keratinocytes.

TRPV3 is a thermosensitive and non-selective cation channel activated at physiological temperatures. It is abundantly expressed in skin keratinocytes, and point mutations in TRPV3 cause a rare genetic disorder, a channelopathy known as Olmsted Syndrome (OS). TRPV3-mutations that cause Olmsted Syndrome show major defects in lysosomal functions and many other cellular abnormalities, such as defects in cell adhesion, cell size and mitochondrial metabolism. In this work, we used a library of thermosensitive organelle probes to explore the sub-cellular thermal spectrum of the OS-causing TRPV3 mutan

Novel Allosteric Modulation of the Human 5-HT3A Receptor by the Clinically Used Drugs Piperazine and Hydroxyzine.

The 5-HT3 receptor is a serotonin-gated cation channel of the pentameric ligand-gated ion channel family and plays a key role in fast synaptic transmission within the central, peripheral, and enteric nervous systems. Dysregulation of this receptor has been linked to neurological, psychiatric, and gastrointestinal disorders. While competitive 5-HT3 antagonists, primarily setrons, are clinically used to alleviate gastrointestinal symptoms, there is a growing interest in developing new negative allosteric modulators (NAMs) for this receptor. Here, we applied a drug-repositioning strategy targetin

The mechanotransduction channel Piezo2 refines axonal projections to the accessory optic system and regulates the optokinetic reflex.

The optokinetic reflex (OKR) is an evolutionarily conserved reflexive behavior that ensures image stabilization on the retina during global motion. It consists of smooth eye tracking movements in the direction of the moving stimulus interspersed with rapid resetting saccades. This reflex is driven by retinal ON direction-selective ganglion cells (oDSGCs), which comprise distinct subtypes tuned either to vertical or horizontal motion. oDSGCs convey directional signals to the brain via precise axonal projections to specific accessory optic system (AOS) nuclei. However, the mechanisms that establ

Manual Massage Therapy for Neuropathic Pain: Clinical Evidence and Mechanistic Insights.

Neuropathic pain (NP) is a complex chronic pain condition arising from a lesion or disease of the somatosensory nervous system, presenting significant clinical challenges with limited therapeutic efficacy from conventional pharmacological interventions. Massage, as a specific form of manual therapy, is a pivotal therapeutic modality in traditional Chinese medicine (TCM) and has been increasingly adopted for the management of NP due to its noninvasive nature and favorable safety profile. Recent advances in clinical and basic research indicate that massage not only effectively mitigates pain sym

Muscarinic Receptor Antagonism and TRPM3 Activation as Stimulators of Mitochondrial Function and Axonal Repair in Diabetic Sensorimotor Polyneuropathy.

Diabetic sensorimotor polyneuropathy (DSPN) is the most prevalent complication of diabetes, affecting nearly half of all persons with diabetes. It is characterized by nerve degeneration, progressive sensory loss and pain, with increased risk of ulceration and amputation. Despite its high prevalence, disease-modifying treatments for DSPN do not exist. Mitochondrial dysfunction and Ca2+ dyshomeostasis are key contributors to the pathophysiology of DSPN, disrupting neuronal energy homeostasis and initiating axonal degeneration. Recent findings have demonstrated that antagonism of the muscarinic a

Inflammatory response in CF airway epithelial cells: a comparative study of modulators and wild-type CFTR rescue.

The combination of pharmacological modulators such as lumacaftor, tezacaftor, and elexacaftor restore CFTR activity at the plasma membrane and improve lung function in patients carrying CFTR mutations such as F508del, their effects on inflammation are less clear. This study aimed to investigate whether the inflammatory response in CF airway epithelial cells depends solely on Cl- transport or also on the structural integrity of the CFTR protein. We examined the effects of several CFTR modulators and wild-type CFTR overexpression on CFTR expression, trafficking, Cl- channel activity, and inflamm

A New Toolbox to Label Zinc-MTF1 Responsive Neuronal Populations Unravels Cellular Congruence between MTF1 Responses and T-type Calcium Channelopathies in an Experimental Model of Epilepsy.

Temporal lobe epilepsy (TLE) is one of the most common forms of epilepsy and usually manifests after serious brain insults, including traumatic brain injury, stroke, and status epilepticus (SE). Previously, we identified a new epilepsy-associated transcriptional control mechanism that relies on the zinc-dependent transcriptional activator metal-regulatory transcription factor-1 (MTF1). MTF1 is one of the key transcription factors involved in intracellular metal ion balance. Upon excessive increase in the concentration of zinc in the cytoplasm, zinc can bind to MTF1, inducing its translocation

Epithelial sodium channels: contributions to the regulation of vascular function and blood pressure.

The epithelial sodium channel (ENaC) is a sodium channel expressed in renal tubules that, by selectively allowing the transmembrane passage of sodium ions, plays a crucial role in maintaining extracellular fluid volume and regulating blood pressure. Recent data have indicated that ENaC also exists in the cells of the vascular wall and contributes to the direct regulation of vascular function and blood pressure. The expression and activation of ENaC are both known to be regulated through mineralocorticoid receptor activation and serum glucocorticoid-regulated kinase 1. In addition, insulin meta

Potassium-sensitive loss of muscle force in the setting of reduced inward rectifier K+ current: Implications for Andersen-Tawil syndrome.

Andersen-Tawil syndrome (ATS) is an ion channelopathy with variable penetrance for the triad of periodic paralysis, arrhythmia, and dysmorphia. Dominant-negative mutations of KCNJ2 encoding the Kir2.1 potassium channel subunit are found in 60% of ATS families. As with most channelopathies, episodic attacks in ATS are frequently triggered by environmental stresses: exercise for periodic paralysis or stress with adrenergic stimulation for arrhythmia. Fluctuations in K+, either low or high, are potent triggers for attacks of weakness in other variants of periodic paralysis (hypokalemic periodic p

Cystic Fibrosis: A Journey through Time and Hope.

Just over thirty years is the span of a generation. It is also the time that has passed since the discovery of the gene responsible for cystic fibrosis. Today, it is safe to say that this discovery has revolutionized our understanding, research perspectives, and management of this disease, which was, thirty years ago, a pediatric condition with a grim prognosis. The aim of this review is to present the advances that science and medicine have brought to our understanding of the pathophysiology of the disease and its management, which in many ways, epitomizes modern molecular genetic research.
