Xanthan gum intake modifies the colon microbiota profile and causes mild colon inflammation in rats.

Xanthan gum is commonly used in the food industry to adjust food consistency and to improve the safety of swallowing liquids and food in peo … d processed for cytokine and immunohistochemical analysis. Fecal matter from the colon was used for microbiota analysis. In general, the addition of xanthan gum at all doses promoted an inflammatory state as demonstrated by the high presence of lymphocytes. Also, it modified the content of the pro-inflammatory cytokines IL-1β and TNF-α compared to the contro …

Regulation Progresses of Selenium Improving Intestinal and Extra-Intestinal Tissues Health Through Regulating Gut Microbiota.

Selenium (Se) is an essential trace element that exerts pleiotropic effects on host physiology, yet the mechanisms by which it coordinates systemic health remain incompletely understood. Emerging evidence regards the gut microbiota as a key mediator of Se biological functions, giving rise to the Se-gut-tissue axis. This review synthesizes the current research progresses on how dietary Se may shape gut microbial composition and metabolism, and how these microbial shifts are associated with protective effects in both intestinal and extra-intestinal tissues. Se sources (particularly organic or ne…

Selenium-enriched tea polysaccharide treatment ameliorates walnut protein allergy by regulating gut microbiota and metabolism.

Selenium-enriched polysaccharides conventionally possess multiple benefits for human health. To investigate the anti-allergic ability of selenium-enriched tea polysaccharide (Se-TPS) and its effect on the gut microbiota and metabolism, a walnut protein (WP)-induced allergic BALB/c mouse model was established. In vivo, Se-TPS (250 mg kg-1) alleviated the clinical allergic symptoms of WP sensitization and repaired the intestinal barrier. Furthermore, Se-TPS can inhibit the over-secretion of IgE, HIS, and IL-4 and promote the normal secretion of TGF-β and IFN-γ to ameliorate the WP-induced immune…

Natural products for depression: Preclinical insights into microbiota-gut-brain axis modulation and pharmacological potential.

The microbiota-gut-brain axis (MGBA) is progressively associated with the pathophysiology of depression. This analysis consolidates existing preclinical findings on MGBA mechanisms associated with depression and assesses natural-product therapies through three key metabolite-related pathways: bile acid-responsive signaling via the farnesoid X receptor, G protein-coupled bile acid receptor 1, vitamin D receptor, and pregnane X receptor; metabolites from the kynurenine pathway and their connection to N-methyl-D-aspartate receptor signaling; and microbial indole-aryl hydrocarbon receptor signalin…

High-dose folic acid therapy ameliorates gut microbiota dysbiosis in reproductive-aged women with hyperhomocysteinemia.

BACKGROUND: Hyperhomocysteinemia (HHCY) in reproductive-aged women is associated with adverse pregnancy outcomes. Although folic acid effectively lowers serum homocysteine (HCY), its relationship with the gut microbiota remains unclear. This study investigated gut microbial alterations associated with HHCY and explored microbiota changes accompanying high-dose folic acid intervention. METHODS: Fecal samples from 38 reproductive-aged women, including 19 patients with HHCY and 19 normal controls, were analyzed using 16S rRNA gene sequencing. Three patients with HHCY receiving high-dose folic ac…

Gut microbiota dysbiosis and metabolic abnormalities promote oxidative stress and fibrosis in idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive lung disease marked by declining pulmonary function and excessive fibrosis. Recent studies suggest that gut microbiota and their metabolites influence systemic inflammation and fibrotic processes via the gut-lung axis. We conducted a comprehensive analysis involving pulmonary function tests, gut microbiota profiling, fecal metabolomics, and serum oxidative stress marker measurements in IPF patients and matched healthy controls. Additionally, a bleomycin-induced rat model of IPF were used to assess the effects of tryptophan-glycine (Trp-Gly) …

Sucralose Exposure During Pregnancy Elevates Gestational Diabetes Risk via Gut Microbiota-Metabolic Axis in Mice.

Sucralose, a widely used nonnutritive artificial sweetener, has gained increasing popularity during pregnancy due to its low-calorie properties. The influence of sucralose on gestational diabetes mellitus (GDM) risk via alterations in gut microbiota composition is not yet well understood. This study sought to explore how gestational sucralose exposure influences GDM development in mice through gut microbiota dysbiosis, impaired intestinal barrier, and metabolic disorders. As an exploratory extension, we also preliminarily assessed early growth in offspring. In our experimental design, pregnant…

Gut microbiota regulation of the IL-6/STAT3 pathway: Linking hematopoiesis to colorectal cancer progression.

BACKGROUND AND AIM: Chronic gut microbiota-driven inflammation fuels colorectal cancer (CRC) progression, but how microbial signals influence systemic immunity and tumor growth remains poorly defined. This review synthesizes evidence on gut microbiota regulation of the interleukin-6 (IL-6)/signal transducer and activator of transcription 3 (STAT3) pathway as the central mechanistic link connecting intestinal dysbiosis, altered hematopoiesis, and CRC progression. Other inflammatory pathways and therapies are discussed only as they intersect with this core axis. METHODS: A structured search of …

Adjunctive fecal microbiota transplantation for major depressive disorder: A randomized, double-blind, placebo-controlled trial.

Gut microbiota may influence antidepressant treatment outcomes, yet whether targeted modulation can enhance efficacy remains unclear. We conducted a randomized, double-blind, placebo-controlled trial in patients with major depressive disorder, administering a 2-week course of fecal microbiota transplantation (FMT) capsules or placebo as an adjunct to escitalopram (ChiCTR2300071421). Remission rates at week 8 did not differ significantly between groups, but FMT produced greater reductions in Hamilton Depression Rating Scale, 17-item version (HAMD-17) scores at weeks 2 and 8. FMT was well-tolera…

Reconstitution of gut microbiota by medicinal plant isoflavones ameliorates heart failure with preserved ejection fraction.

Heart failure with preserved ejection fraction (HFpEF) is a heterogeneous syndrome with limited therapeutic options. Gut microbiota and microbiota-derived metabolites impact the progression and severity of HFpEF-related cardiometabolic dysfunctions. Here, we demonstrated that the isoflavone extract of Pueraria lobata (Iso-Pl), puerarin, or its microbial metabolite S-equol, improved left ventricular diastolic dysfunctions, reduced hypertension, and ameliorated metabolic disorders in an HFD and L-NAME-induced HFpEF mouse model. Antibiotic depletion and fecal microbiota transplantation experiment…

Gut microbiota-derived 5-HTP penetrates the host blood-brain barrier and ameliorates autism symptoms.

Autism spectrum disorder (ASD), a highly prevalent neurodevelopmental condition, is increasingly recognized for its strong association with the intestinal microbiome. However, the development of gut microbiota-targeted therapies has been impeded by a limited understanding of the molecular mechanisms underlying interactions between commensal bacteria and the host nervous system. In this study, shotgun metagenomic sequencing and UPLC-MS/MS targeted metabolic analyses identified altered tryptophan metabolites in the gut microbiota of both human ASD patients and ASD mouse models. Notably, we demon…
