Expression of beta 1 integrins in human dental pulp in vivo: a comparative immunohistochemical study on healthy and chronic marginal periodontitis samples.

AIM: The objective of this study was to determine the tissue distribution of beta 1 integrin chains in sound human dental pulps and to compare the findings with connective tissue compartments of other organs and to pulp tissue in teeth extracted due to periodontal disease. METHODOLOGY: Freshly frozen pulp tissue samples from teeth extracted for orthodontic reasons were examined and compared to samples from teeth extracted due to chronic (marginal) periodontitis. beta 1 integrin chains were determined using an indirect-immunoperoxidase technique. Seven monoclonal antibodies recognizing alpha 1

Adaptive responses to peroxynitrite: increased glutathione levels and cystine uptake in vascular cells.

We and others recently demonstrated increased glutathione levels, stimulated cystine uptake, and induced gamma-glutamylcysteinyl synthase (gamma-GCS) in vascular cells exposed to nitric oxide donors. Here we report the effects of peroxynitrite on glutathione levels and cystine uptake. Treatment of bovine aortic endothelial and smooth muscle cells with 3-morpholinosydnonimine (SIN-1), a peroxynitrite donor, resulted in transient depletion of glutathione followed by a prolonged increase beginning at 8-9 h. Concentration-dependent increases in glutathione of up to sixfold occurred 16-18 h after 0

Connective tissue growth factor: what's in a name?

Connective tissue growth factor (CTGF) is a member of the recently described CCN gene family which contains CTGF itself, cyr61, nov, elm1, Cop1, and WISP-3. CTGF is transcriptionally activated by several factors although its stimulation by transforming growth factor beta (TGF-beta) has attracted considerable attention. CTGF acts to promote fibroblast proliferation, migration, adhesion, and extracellular matrix formation, and its overproduction is proposed to play a major role in pathways that lead to fibrosis, especially those that are TGF-beta-dependent. This includes fibrosis of major organs

Exercise- and cold-induced asthma.

Exercise- and cold-induced asthma are commonly recognized respiratory disorders. The asthmatic response includes several factors contributing to airway narrowing, and thus increased airway resistance. These include airway smooth muscle contraction, mucus accumulation, and bronchial vascular congestion as well as epithelial damage and vascular leakage. The etiology for these disorders is nonantigenic. The primary stimulus is probably a combination of airway cooling and drying (leading to hypertonicity of airway lining fluid). Symptoms generally do not occur during the stimulus period (e.g., exe

Low-density lipoproteins inhibit histamine and NaNO2 relaxations of the coronary vasculature and reduce contractile function in isolated rat hearts.

In the present study we examined the action of native and oxidized low-density lipoproteins (LDL) on coronary vascular and cardiac function and ultrastructure in rat hearts perfused isovolumically in the Langendorff mode. Responses of the coronary resistance vessels to the endothelium-dependent vasodilator, histamine, and the endothelium-independent vasodilator, NaNO2, were measured together with contractile function (rate-pressure product) before and after perfusion for 20 min with native - or oxidized-LDL at a concentration of 100 mu g protein/ml. Ultrastructural damage was assessed via elec

[Plasma levels of atriopeptin and hemodynamics during major vascular surgery: comparison between isoflurane and propofol+fentanyl].

Atrial natriuretic factor (ANF) is a peptide hormone released from atrial cardiocytes in response to atrial stretch. It has potent and selective effects on vascular smooth muscle, fluid and electrolyte balance, and may interact with other vasoactive substances. The influence of anaesthesia and major vascular surgery on the release and circulation of ANF is unknown. Therefore the relationships between haemodynamic variables, volume expansion and plasma ANF were studied in patients undergoing resection of abdominal aortic aneurysm, randomly assigned to receive isoflurane or propofol+fentanyl ana

Endothelium-mediated coronary blood flow modulation in humans. Effects of age, atherosclerosis, hypercholesterolemia, and hypertension.

The effects of age, atherosclerosis, hypertension, and hypercholesterolemia on vascular function of the coronary circulation were studied by subselective intracoronary infusions of acetylcholine, which releases endothelium-derived relaxing factor, and papaverine, which directly relaxes vascular smooth muscle, in normal patients (n = 18; no risk factors for coronary artery disease), in patients with evidence of early atherosclerosis but normal cholesterol levels and normal blood pressure (n = 12), in patients with hypertension without left ventricular hypertrophy (n = 12), and in patients with

Dietary prevention of stroke and its mechanisms in stroke-prone spontaneously hypertensive rats--preventive effect of dietary fibre and palmitoleic acid.

Previously it was reported that dietary protein, some amino acids and potassium are effective in preventing stroke in stroke-prone spontaneously hypertensive rats (SHRSP). The present study revealed that other dietary factors could also prevent cerebral lesions, and the mechanism of this effect was studied. In SHRSP given 1% NaCl in their drinking water, a diet containing 10% active fibre (powdered brown seaweed converted to K+ form) significantly lowered blood pressure (BP) and markedly reduced the incidence of stroke (0 versus 100% in controls on the 30th day of experiment). Since the faecal

Elucidation of mechanisms underlying the therapeutic effects of cordycepin on pulmonary hypertension, with a focus on cell senescence and gut microbiota.

INTRODUCTION: Pulmonary hypertension (PH) is a progressive cardiopulmonary disorder characterized by excessive pulmonary vascular remodeling and aberrant proliferation of pulmonary artery smooth muscle cells (PASMCs). Emerging evidence suggests that gut microbiota dysbiosis contributes to PH development. Cordycepin, a natural adenosine analogue derived from Cordyceps militaris, has demonstrated antiproliferative and microbiota-modulating properties; however, its mechanism of action in PH remains unclear. OBJECTIVE: Elucidate the mechanisms underlying the therapeutic effects of cordycepin on P

USP53 regulates glycolytic reprogramming and accelerates Abdominal aortic aneurysm progression and rupture.

Abdominal aortic aneurysm (AAA) rupture remains a fatal vascular event with limited mechanistic understanding beyond diameter-based risk assessment. Emerging evidence suggests that metabolic reprogramming contributes to aneurysm progression, yet regulators linking metabolism to rupture remain poorly defined. An AngII/BAPN-induced mouse model generated intact (iAAA) and ruptured AAA (rAAA) tissues. Bulk RNA sequencing and untargeted metabolomics profiled transcriptomic and metabolic alterations across sham, iAAA, and rAAA groups. Candidate ubiquitin-specific proteases (USPs) were screened by in

Apigenin Protects Hypertensive Intracerebral Hemorrhage by Regulating Trx1 Through Binding to PRDX2.

Intracerebral hemorrhage (ICH) remains the most severe stroke subtype with high mortality and disability rates. Although apigenin has shown cerebroprotective potential in preclinical studies, its mechanism of action, particularly in hypertensive ICH, remains poorly understood. This study investigates the molecular pathway through which apigenin confers protection against hypertensive ICH. We employed a hypertensive ICH mouse model treated with varying apigenin doses. Through transcriptomic profiling, molecular docking, surface plasmon resonance (SPR), and pull-down assay, we identified peroxir

Neurovascular Signaling in the Perivascular Space: Biophysical Principles and Disruption in Hypertension.

The brain's vasculature-from pial arteries through penetrating arterioles and the capillary bed to venous drainage-is enveloped by a narrow, compositionally elaborate perivascular space that provides the primary signaling interface between neural tissue and blood vessels. Here, we advance the principle that the geometry and composition of this space are not constant but change systematically along the vascular tree, and that perivascular space composition at these locations imposes biophysical constraints that dictate the fidelity and gain of neurovascular signaling. We argue that its narrow m

Two Pulses Per Beat: Parallel Energetics of Myocardium and Coronary Wall.

Each heartbeat is initiated by a sinoatrial node action potential that propagates through the myocardial syncytium, triggering sequential contraction of the four chambers and propelling blood through the coronary, pulmonary, and systemic circulations. This mechanical event is fueled by two coupled energetic pulses: one in the working myocardium, where ATP is consumed by Ca2+ cycling, cross-bridge cycling and ion pumping, and a second in the coronary vascular wall, where smooth muscle cells (SMCs), endothelial cells, and pericytes expend ATP to maintain tone, manage Ca2+, and coordinate perfusi

Integrated single-cell and spatial transcriptomics reveal stromal-immune-vascular crosstalk in patients with interstitial cystitis/bladder pain syndrome.

Interstitial cystitis/bladder pain syndrome (IC/BPS) is characterized by complex cellular heterogeneity and inflammatory dysfunction in the bladder. However, the molecular pathogenesis of IC/BPS remains poorly understood, and few studies have provided comprehensive analyses. To elucidate the mechanisms underlying disease pathology, we performed integrated single-cell and spatial transcriptomic analyses of human bladder tissue. Comprehensive single-cell multiomics analyses have identified disease-specific alterations across multiple cell types. T helper 17 cells were significantly expanded in H

The dual role of extracellular vesicles in vascular calcification: from molecular mechanisms to clinical translation.

Vascular calcification (VC) is an active and regulated pathological process, which plays a central role in cardiovascular disease. Extracellular vesicles (EVs) are now recognized as crucial players in this pathology. EVs are nanoscale membrane vesicles secreted by cells. According to their biogenesis, they are mainly divided into exosomes, microvesicles and apoptotic bodies. They are rich in proteins, nucleic acids, lipids and other biologically active molecules. EVs play a dual role in VC. Regarding the pro-calcific role, EVs released by vascular smooth muscle cells (VSMCs), endothelial cells

RNF213 in Panvascular Disease: A Molecular Hub Linking Genetic Susceptibility to Systemic Vasculopathy.

Panvascular diseases, characterized by systemic vascular dysfunction across multiple organ systems, represent a complex interplay of genetic susceptibility and environmental triggers. Ring Finger Protein 213 (RNF213), initially identified as the principal susceptibility gene for moyamoya disease (MMD), has emerged as a central regulator of panvascular pathophysiology. This review synthesizes current evidence elucidating RNF213's multifaceted roles in vascular homeostasis, spanning its functions as an E3 ubiquitin ligase, mechanosensor, and immune modulator. The "second-hit" hypothesis posits t

Multi-trait and Gene-Based Analyses Identify Genetic Variants Associated with Spontaneous Coronary Artery Dissection.

BACKGROUND AND AIMS: Spontaneous coronary artery dissection (SCAD) is a non-atherosclerotic cause of acute myocardial infarction (MI) that predominantly affects young women. As an under-recognized cause of MI, large genome-wide association studies (GWAS) remain challenging. We aimed to leverage SCAD shared genetic basis with related vascular diseases to uncover genetically determined biological mechanisms. METHODS: Summary statistics for SCAD GWAS (1,917 cases, 9,293 controls) was harmonised with seven related vascular traits: fibromuscular dysplasia, intracranial aneurysm, cervical artery di

A rare case of intravenous leiomyomatosis with metastasis to the circulatory tract: case report and literature review.

INTRODUCTION: Intravenous leiomyomatosis (IVL) is a rare condition characterized by the proliferation of uterine smooth muscle cells within the lumen of vascular vessels. Despite its histologically benign nature, IVL can invade cardiac vessels, leading to severe complications such as circulatory failure and heart blockade. This underscores the critical need for accurate diagnosis and comprehensive preoperative management to prevent fatal outcomes. RESULTS: A previously healthy 41-year-old woman was admitted to Gynecology Department following abnormal findings on a routine abdominal ultrasound

Endothelial dysfunction promotes age-related reorganization of collagen fibers and alters aortic biomechanics in mice.

Endothelial dysfunction, defined as a reduction in the bioavailability of nitric oxide (NO), is a risk factor for the occurrence and progression of various vascular diseases. This study investigates the effect of endothelial dysfunction on age-related changes in aortic extracellular matrix (ECM) microstructure and the relationship between microstructural adaptation and the mechanical response. Here, we used groups of NOS3 knockout (KO), NOS3 heterozygotes (Het), and wild-type (WT) B6 mice (controls) to study changes in hemodynamic parameters, collagen fiber organization, and both active and pa

Polypoidal Choroidal Vasculopathy: From Clinicopathological and Molecular Perspectives.

<p>Background: Polypoidal choroidal vasculopathy (PCV) causes severe visual impairment in patients with neovascular age-related macular degeneration (nAMD). Currently, PCV is classified as a subtype or variant of type 1 macular neovascularization. Effective treatments for PCV remain limited, despite the variable efficacy of anti-vascular endothelial growth factor (VEGF) drugs in regressing polypoidal lesion(s) (PLs). A key contributing factor is the incomplete understanding of the disease mechanisms, which highlights the necessity for reliable animal models and a comprehensive understanding fr
