Combination of EPC-EXs and NPC-EXs with miR-126 and miR-210 overexpression produces better therapeutic effects on ischemic stroke by protecting neurons through the Nox2/ROS and BDNF/TrkB pathways.

BACKGROUNDS/AIMS: Neural progenitor cells (NPCs) and endothelial progenitor cell (EPCs) exhibit synergistical effects on protecting endothelial cell functions. MiR-126 and miR-210 can protect cell activities by regulating brain-derived neurotrophic factor (BDNF) and reactive oxygen species (ROS) production. Exosomes (EXs) mediate the beneficial effects of stem cells via delivering microRNAs (miRs). Here, we investigated the combination effects of EXs from EPCs (EPC-EXs) and NPCs (NPC-EXs), and determined whether these EXs with miR-126 (EPC-EXsmiR-126) and miR-210 overexpression (NPC-EXsmiR-210

Midbrain control of breathing and blood pressure: The role of periaqueductal gray matter and mesencephalic collicular neuronal microcircuit oscillators.

Источник пометил публикацию как отозванную

Neural circuitry residing within the medullary ventral respiratory column nuclei and dorsal respiratory group interact with the Kölliker-Fuse and medial parabrachial nuclei to generate the core breathing rhythm and pattern during resting conditions. Triphasic eupnea consists of inspiratory [I], post-inspiratory [post-I], and late-expiratory [E2] phases. Mesencephalic zones exert modulatory influences upon respiratory rhythm-generating circuitry, sympathetic oscillators, and parasympathetic nuclei. The earliest evidence supporting the existence of midbrain control of breathing derives from stud

Nitric oxide and synaptic dynamics in the adult brain: physiopathological aspects.

The adult brain retains the capacity to rewire mature neural circuits in response to environmental changes, brain damage or sensory and motor experiences. Two plastic processes, synaptic remodeling and neurogenesis, have been the subject of numerous studies due to their involvement in the maturation of the nervous system, their prevalence and re-activation in adulthood, and therapeutic relevance. However, most of the research looking for the mechanistic and molecular events underlying synaptogenic phenomena has been focused on the extensive synaptic reorganization occurring in the developing b

Spinal cord transection repair occurs when Nestin+ cells differentiate into neurons within a taxol-collagen-enhanced microenvironment in mice.

BACKGROUND: Previous studies have shown that taxol promotes axon regeneration in nerve repair, but fails to bridge the two ends of a completely transected spinal cord. Our prior in vitro research revealed that taxol, a microtubule-stabilizing agent, promotes neural stem cells (NSCs) differentiation into neurons while inhibiting astrocyte differentiation. In vivo studies further demonstrated that taxol-scaffold enhances functional recovery in animals with complete spinal cord injury (SCI). This study aims to directly validate the role of taxol-collagen in guiding NSCs to differentiate into neur

Synergistic effects of human umbilical cord mesenchymal stem cells/neural stem cells and epidural electrical stimulation on spinal cord injury rehabilitation.

Spinal cord injury (SCI) is a severe neurological condition marked by a complex pathology leading to irreversible functional loss, which current treatments fail to improve. Epidural electrical stimulation (EES) shows promise in alleviating pathological pain, regulating hemodynamic disturbances, and enhancing motor function by modulating residual interneurons in the lower spinal cord. Cell transplantation (CT), especially using human umbilical cord mesenchymal stem cells (hUCMSCs) and neural stem cells (NSCs), has significantly improved sensory and motor recovery in SCI. However, the limitation

Olfactory mucosa transplantation for spinal cord injury.

Carlos Lima et al. who are pioneers in this field reported their clinical pilot study of olfactory mucosa transplantation for chronic spinal cord injury. They showed the safety and feasibility of it. Olfactory mucosa contains the olfactory ensheathing cells and neural stem cells. Recent studies have demonstrated the potential therapeutic role of both cells in spinal cord injury. We have already reported the effectiveness of olfactory mucosa transplantation for rat spinal cord injury. Furthermore we indicated the reconstruction of cortico-spinal tract by BDA (biotinylated dextran amine) tracer

Si-Ni-San alleviates depressive-like behaviors in adolescent male mice by restoring DRN-NAc serotonergic circuit function and 5-HT4R-dependent synaptic plasticity.

BACKGROUND: Early-life stress (ELS) is a key risk factor for adolescent depression. Si-Ni-San (SNS), a classic traditional Chinese medicine formula, has shown antidepressant potential, yet its effects on the dorsal raphe nucleus (DRN)-nucleus accumbens (NAc) serotonergic circuit remain unclear. PURPOSE: This study aimed to investigate whether SNS alleviates adolescent depression by restoring DRN-NAc serotonergic circuit function and to identify the serotonin receptor mediating its synaptic effects in the NAc. STUDY DESIGN: Firstly, the antidepressant efficacy of SNS was evaluated in a mouse

Prenatal stress, excitatory-inhibitory imbalance, and ADHD risk: a hypothesis-driven perspective on psilocybin-induced neuroplasticity.

Attention-deficit/hyperactivity disorder (ADHD) is a prevalent neurodevelopmental condition in which prenatal stress and long-lasting disruptions of excitatory-inhibitory (E/I) balance have been implicated as key vulnerability factors. Although established pharmacological and behavioral treatments are effective for many individuals, they are not universally successful and do not directly target upstream neurodevelopmental mechanisms. In this hypothesis-driven perspective, we examine whether psilocybin-induced neuroplasticity could theoretically modulate stress-related neurodevelopmental risk p

Beyond Structural Pathology: Central Sensitization and Chronic Pain with Reference to Lumbar Disc Herniation-A Narrative Review.

Chronic pain is increasingly understood as a multidimensional condition in which, in a substantial subgroup of patients, a protective symptom can evolve into a persistent maladaptive disorder of the nervous system, while in others it may remain closely tied to ongoing mechanical or structural factors. Central sensitization (CS) represents a key mechanism underlying this transition, characterized by enhanced neural responsiveness and impaired endogenous pain inhibition, leading to a dissociation between pain and tissue pathology. The aim of this narrative review is to critically discuss current

Neuronal SPI1 suppression enhances axonal regeneration after spinal cord injury through Rassf10 downregulation.

BACKGROUND: Axonal regeneration following spinal cord injury (SCI) is hindered by the intrinsic inhibitory properties of spinal neurons. While the role of the transcription factor SPI1 (PU.1) has been well established in myeloid cells, its function in neurons, particularly in regulating axonogenesis, remains poorly defined. The data from recent single-cell sequencing studies have indicated that SPI1 is clearly expressed in spinal neurons. Thus, we aimed to explore the effect of SPI1 on axonal growth and its underlying molecular mechanisms. METHODS: Primary cultured rat spinal neurons, CRISPR/

Glutamatergic pathway from deep layers of the insular cortex to the rostral central medial thalamic nucleus participates in neuropathic pain and anxiety-like behaviors in mice.

BACKGROUND: The insular cortex (IC), a pivotal hub for pain signal integration, projects extensively to the thalamus. However, the functional role of its specific projection to the central medial thalamic nucleus (CM) in pain-affective comorbidities remains elusive. Elucidating the IC-CM pathway is thus critical for revealing endogenous pain regulation and pain-emotion comorbidity mechanisms. METHODS: A multidisciplinary approach combining neuroanatomical tracing, calcium imaging, electrophysiological recordings, optogenetics, chemogenetics and behavioral observations was used to investigate t

A neural substrate for targeted analgesia associated with attenuated serotonergic transmission in a mouse model of medication overuse headache.

BACKGROUND: Medication overuse headache (MOH) is a chronic disorder due to excessive acute headache treatment use. Neuroimaging studies suggest the involvement of the orbitofrontal cortex (OFC) in the pathophysiology of MOH, particularly its connections with the periaqueductal gray (PAG). The OFC-vlPAG circuit has been well-established as a critical neural substrate in pain regulation; while its role in triptan-related MOH pathogenesis remains unclear. This study aims to investigate the role of the OFC-vlPAG circuit and its underlying mechanisms in regulating mechanical hypersensitivity using

ENT1 inhibitor J4 restores cognitive function and white-matter integrity in a mouse model of tuberous sclerosis complex.

BACKGROUND: Tuberous sclerosis complex (TSC) is a hereditary disease caused by pathogenic mutations in the TSC1 or TSC2 genes, leading to overactivation of mTOR signaling and dysregulation of downstream pathways. The majority of individuals with TSC develop psychiatric and neurodevelopmental comorbidities-including intellectual disability, autism spectrum disorders, anxiety and other behavioral manifestations-collectively termed TSC-associated neuropsychiatric disorders. Over the past decade, increasing evidence from clinical and preclinical studies has highlighted a significant myelination de

Immediate-Term Dapoxetine Administration Suppressed Functional Connectivity of Subcallosal Cingulate Cortex in Premature Ejaculation: A Resting-State fMRI Study.

INTRODUCTION: Dapoxetine, a short-acting selective serotonin reuptake inhibitor (SSRI), is commonly prescribed to treat premature ejaculation (PE). Functional connectivity (FC) between dorsolateral prefrontal cortex (DLPFC) and subcallosal cingulate cortex (SCC) has been found to serve as neuroimaging biomarker for predicting SSRI treatment response. Despite the fact that altered FC has been observed in PE patients, the neural mechanisms underlying the rapid-acting effects on brain of immediate-term dapoxetine administration remain unknown. This study aimed to explore the mechanisms underlying

A mean field theory for pulse-coupled neural oscillators based on the spike time response curve.

A mean field method for pulse-coupled oscillators with delays used a self-connected oscillator to represent a synchronous cluster of N - 1 oscillators and a single oscillator assumed to be perturbed from the cluster. A periodic train of biexponential conductance input was divided into a tonic and a phasic component representing the mean field input. A single cycle of the phasic conductance from the cluster was applied to the single oscillator embedded in the tonic component at different phases to measure the change in the cycle length in which the perturbation was initiated, that is, the first
