Lipid homeostasis plays a critical role in inherited and acquired retinal diseases.

The photoreceptor layer of the retina converts light stimuli into electrical signals and transmits them to the visual cortex for image processing. Photoreceptor cells rely on a constant supply of nutrients, including lipids, to replenish their high demand of membrane remodelling. Dysregulated retinal lipid homeostasis results in ectopic lipid deposition that triggers oxidative stress, inflammatory response, mitochondrial dysfunction, and impaired signal transduction, thereby contributing to the pathogenesis of retinal degeneration in several eye diseases. In this review, we discuss the importa…

Spatial dynamics of mTOR pathway activity during bovine embryo development.

The mTOR pathway regulates cell proliferation, growth, survival, and metabolism by integrating nutritional and growth factor signaling. In embryos, its activity is influenced by the availability of nutrients in the culture medium, and it can affect the first cellular differentiation event, driving trophectoderm (TE) formation in mice. We hypothesized that mTOR activity is increased in cells poised to become TE and in differentiated TE cells of early bovine embryos. To test this, we assessed mTOR pathway activity through immunofluorescence detection of phospho-S6 (pS6) using confocal microscopy…

Double-edged dispersal: animal-mediated transport in the Anthropocene.

Animals play important roles in dispersing seeds, nutrients, and microbes across Earth's surface. These transport functions connect ecosystems and shape fundamental dynamics, including community composition, productivity, and carbon storage. Yet humans have substantially altered resource availability, wildlife assemblages, and movement patterns, leading to some undesirable outcomes, including the spread of nonnative species, pollutants, and zoonotic pathogens. Although animals disperse multiple natural and anthropogenic objects simultaneously, people tend to champion or criticize animal disper…

Bewitching the host: Striga disrupts host stomatal ABA signaling by impairing the tryptophan lock.

Parasitic plants of the Orobanchaceae family, particularly Striga hermonthica, rank among the world's most devastating agricultural weeds (Jamil et al., 2021). They inflict massive yield losses on staple cereal crops across Africa, including sorghum, millet, and rice, as well as globally on other essential crops such as maize, rapeseed, tomato, and legumes (Parker et al., 2009; Bouwmeester, 2018). These root parasites have evolved a specialized organ called the 'haustorium' that connects to the vascular system of the host to siphon water, minerals, and nutrients (Yang et al., 2015). A critical…

Vitamin D and Fracture Healing.

Maintaining bone health involves complex interactions among nutrients, hormones, and physiological mechanisms, with vitamin D playing a pivotal role. Vitamin D is essential for the absorption of calcium and phosphate, which are critical minerals for bone development, maintenance, and repair. Disruptions in mineral ion homeostasis can lead to conditions such as hypocalcemia, osteoporosis, osteomalacia, and rickets, which are prevalent worldwide. Inadequate vitamin D impairs bone health, increasing the risk of fractures and catabolic bone diseases. This chapter elaborates on the synthesis, activ…

A waxy path: Lipid dynamics during tuberculosis.

Infectious pathogens must adapt to the metabolic landscapes of their hosts, often exploiting host-derived nutrients while mitigating the stresses these resources impose. Among these challenges, host lipids are both a rich energy source and a potential liability, requiring tight regulation during infection. Mycobacterium tuberculosis, the causative agent of tuberculosis and the leading cause of death from a single infectious agent, exemplifies an extreme adaptation to lipid-rich environments. Recent studies have uncovered coordinated host and bacterial pathways that mediate fatty acid transfer,…

Culture of the Human Embryo.

Fundamental to the success of the IVF laboratory is the use of an optimized embryo culture system that facilitates the production of high-quality blastocysts. This is achieved through the establishment of a culture system that encompasses the provision of essential nutrients by use of appropriate culture media, the maintenance of a stable environment in vitro and establishment of effective quality control protocols within the laboratory. While there is considerable variability in the culture systems used in IVF laboratories worldwide, as no standardized system exists, all modern embryo culture…

Somatic cells compartmentalise their metabolism to sustain germ cell survival.

To ensure success in reproduction, organisms dedicate substantial resources to supporting the germline. In testes, somatic gonadal cells form a barrier that isolates germ cells from circulating nutrients, raising the question of how germ cell metabolism is sustained and how somatic cells ensure sufficient resources are directed to the germline. We use lineage-specific manipulations and metabolite reporters to show in vivo that somatic gonadal cells break down circulating sugars to produce and shuttle lactate to the germline, sustaining germ cell survival. We show that somatic cells ensure that…

Nutraceuticals in Psychiatric Disorders: A Systematic Review.

Correct nutrition and diet are directly correlated with mental health, functions of the immune system, and gut microbiota composition. Diets with a high content of some nutrients, such as fibers, phytochemicals, and short-chain fatty acids (omega-3 fatty acids), seem to have an anti-inflammatory and protective action on the nervous system. Among nutraceuticals, supplementation of probiotics and omega-3 fatty acids plays a role in improving symptoms of several mental disorders. In this review, we collect data on the efficacy of nutraceuticals in patients with schizophrenia, autism spectrum diso…

Deep-Dive Directions for Dietitians (DDDDs): a roadmap for the nutritional management of poorly responsive coeliac disease patients.

A substantial number of coeliac disease patients fail to respond to treatment with a gluten-free diet. Non-responsiveness might be multifactorial and the spectrum ranges from intentional or inadvertent gluten contamination as the main aetiology, to sensitivity to other nutrients (in addition to additives and preservatives). If the diagnosis of coeliac disease is correctly made and cross contamination and other factors have been excluded, then the aetiology behind the symptoms of a small group of coeliac patients might be refractory coeliac disease. The journey to ensure gluten contamination is…

Lysosomal lipid peroxidation mediates immunogenic cell death.

Cancer cells rely on lysosome-dependent degradation to recycle nutrients that serve their energetic and biosynthetic needs. Despite great interest in repurposing the antimalarial hydroxychloroquine as a lysosomal inhibitor in clinical oncology trials, the mechanisms by which hydroxychloroquine and other lysosomal inhibitors induce tumor-cell cytotoxicity remain unclear. In this issue of the JCI, Bhardwaj et al. demonstrate that DC661, a dimeric form of chloroquine that inhibits palmitoyl-protein thioesterase 1 (PPT1), promoted lysosomal lipid peroxidation, resulting in lysosomal membrane perme…

Epigenetic basis of diabetic vasculopathy.

Type 2 diabetes mellitus (T2DM) causes peripheral vascular disease because of which several blood-borne factors, including vital nutrients fail to reach the affected tissue. Tissue epigenome is sensitive to chronic hyperglycemia and is known to cause pathogenesis of micro- and macrovascular complications. These vascular complications of T2DM may perpetuate the onset of organ dysfunction. The burden of diabetes is primarily because of a wide range of complications of which nonhealing diabetic ulcers represent a major component. Thus, it is imperative that current research help recognize more ef…

The Multifaceted Role of Autophagy in Endometrium Homeostasis and Disease.

Autophagy is a conserved fundamental cellular process with a primary function of catabolizing harmful or surplus cellular contents such as protein aggregates, dysfunctional/long-lived organelles, intracellular pathogens, and storage nutrients. An increasing body of evidence reveals that basal autophagy is essential for maintaining endometrial homeostasis and mediating endometrial-specific functions, including menstrual cycle, embryo implantation, and decidualization. However, perturbed levels of autophagy can lead to severe endometrial pathologies, including endometriosis, endometrial hyperpla…

[Progress of researches on the interaction between parasitic infections and host cell autophagy].

Autophagy, a conserved intracellular degradation system, is a specific life phenomenon in eukaryocytes. Autophagy is widely accepted as a pathway that double-membrane autophagosomes envelop and sequester intracellular cytoplasmic components and then fuse with lysosomes to form autolysosomes, which degrade their contents to regenerate nutrients. Autophagy may be triggered by starvation and a diverse range of pathogens, including parasites. Following infection with intracellular parasites, host cells may eliminate parasites by autophagy. However, parasites may develop self-defense mechanisms, an…

Removal of biogenic compounds from the post-fermentation effluent in a culture of Chlorella vulgaris.

Wastewater rich in organic carbon, nitrogen and phosphorus may serve as a convenient source of carbon and nutrients for a year-long microalgae production. Scientific reports indicate that some single-cell microalgae such as Chlorella and Scenedesmus, are highly tolerant to wastewater environments and efficiently remove biogenic compounds. The aim of this study was to determine the possibility of using the effluent produced in the process of anaerobic degradation of whey as a culture medium for the multiplication of Chlorella vulgaris algae biomass and to characterise their growth efficiency an…
