PCK1 attenuates intrahepatic cholangiocarcinoma progression by suppressing lactate accumulation and PI3K-AKT signaling.

Intrahepatic cholangiocarcinoma (iCCA) is a highly malignant liver cancer with limited treatment options. Recent evidence implicates lactate metabolism as playing a crucial role in tumor progression, but its precise contribution in iCCA remains unclear. In this study, lactate metabolism-related genes (LMRGs) in iCCA were identified through analyses of bulk and single-cell RNA sequencing data, diagnostic models were developed using machine learning algorithms, and the functional significance of candidate genes was validated through a combination of in vitro and in vivo experiments. 38 different

Ultra-Early Blood Pressure Control in Acute Intracerebral Hemorrhage.

Acute intracerebral hemorrhage (ICH) remains one of the most devastating neurological emergencies, with blood pressure (BP) management in the early hours playing a pivotal role in determining patient outcomes. Emerging evidence suggests that ultra-early BP control-intervening within minutes to 2 hours of symptom onset-has the potential to reduce secondary brain injury and improve survival. Yet, despite this evidence, current clinical practice is often inconsistent, and there is no universally agreed-upon approach for managing BP in the acute phase of ICH. We advocate for ultra-early BP interve

Antioxidant Molecules in the Human Vitreous Body During Prenatal Eye Development.

The structures of the developing eye may be damaged as a result of the impact of reactive oxygen species (ROS) interacting with different cellular components. The antioxidant molecules found in the eye, especially in the vitreous body-the largest component of the eye, playing a crucial role in the formation of structures and functions of the developing eye-provide protection to the eye tissues from ROS. This review considers various antioxidant molecules (ascorbic acid, lutein, bilirubin, uric acid, catecholamines, erythropoietin, albumin, and alpha-fetoprotein) that have been found in the hum

PRPF31 reduction causes ciliary defects of photoreceptors via inhibiting expression of FSCN2.

PRPF31 and FSCN2 are both recognized as causative genes of retinitis pigmentosa (RP). PRPF31 encodes an ubiquitously expressed splicing factor essential for pre-mRNA processing across all tissues, whereas FSCN2 encodes a protein responsible for actin cross-linking, playing a crucial role in the development of photoreceptor and retinal architecture. Despite their known roles, the retina-specific pathogenic mechanisms linking these genes remain incompletely understood. This study investigates how PRPF31 regulates FSCN2 expression and contributes to ciliary dysfunction in photoreceptors. PRPF31 h

Increasing resilience to stress by home-based transcranial stimulation.

Mental health disorders, especially anxiety and depression, affect nearly one billion people worldwide, with chronic stress playing a major role in their onset and severity. Despite growing demand, access to care remains limited, underscoring the need for scalable, preventive interventions. This study investigates the feasibility, safety, and preliminary effects of a Home-Based transcranial Direct Current Stimulation (HB-tDCS) protocol targeting the left dorsolateral prefrontal cortex (L-DLPFC) to enhance stress resilience in healthy middle-aged adults. Thirty-one participants completed ten se

Netrin-1 as a molecular mediator linking APOE ε4 to alzheimer's disease pathogenesis.

The ɛ4 allele of the apolipoprotein E (APOE) gene stands as the most significant genetic risk factor for Alzheimer's disease (AD), although the underlying mechanism linking APOE to the disease remains elusive. Netrin-1, has been found to inhibit Aβ production, reduce neuroinflammation, and regulate synaptic plasticity, thereby playing a role in AD. Recent reports suggest that Netrin-1 could be a key protein connecting APOE ɛ4 with Aβ and tau tangles. This study aims to investigate whether APOE regulates the expression or function of Netrin-1, thus influencing the risk of developing AD. The ser

Use of Clinical Public Databases in Hidradenitis Suppurativa Research.

In this viewpoint, we argue that recent studies using clinical public databases have revolutionized our understanding of hidradenitis suppurativa (HS), a chronic inflammatory skin condition with significant impacts on patients' quality of life. Our key messages are as follows: (1) these databases enable large-scale studies integrating genetic, epidemiological, and clinical data, providing crucial insights into HS's genetic predispositions, comorbidities, and treatment outcomes; (2) findings highlight a strong genetic component, with mutations in the γ-secretase complex playing a key role in HS

Harnessing Targeted Photodynamic Therapy to Synergistically Activate T Cell and NK Cell Responses in Multiple Myeloma.

Multiple myeloma (MM) remains an incurable malignancy, with immune suppression in the bone marrow microenvironment playing a critical role in disease progression and relapse. Restoring both adaptive and innate immunity represents a promising therapeutic strategy. In this study, a novel photodynamic therapy (PDT) platform is developed that simultaneously activates both adaptive immunity through T cells and innate immunity through natural killer (NK) cells, thereby fostering a synergistic anti-tumor immune response in MM. A series of aggregation-induced emission luminogen (AIEgen)-based photosen

Autocrine interferon poisoning mediates ADAR1-dependent synthetic lethality in BRCA1/2-mutant cancers.

ADAR1 is an RNA editing enzyme which prevents autoimmunity by blocking interferon responses triggered by cytosolic RNA sensors, and is a potential target in immuno-oncology. However, predictive biomarkers for ADAR1 inhibition are lacking. Using multiple in vitro and in vivo systems, we show that BRCA1/2 and ADAR1 are synthetically lethal, and that ADAR1 activity is upregulated in BRCA1/2-mutant cancers. ADAR1 depletion in BRCA1-mutant cells causes an increase in R-loops and consequently, an upregulation of cytosolic nucleic acid sensing pattern recognition receptors (PRR), events which are ass

Eosinophils and Bioactive Lipid Mediators Regulate Skin Inflammation and Cancer Growth.

Cutaneous squamous cell carcinoma (cSCC) is the second most common form of skin cancer, with local immunity playing a key role in regulating outcome. Nonresolving inflammation has been suggested to drive cancer growth; however, a fundamental understanding of the tumor immune microenvironment in cSCC is still lacking. In this study, we demonstrate tissue-associated tumor eosinophilia in both human and murine cSCC and show that eosinophils promote epithelial hyperplasia and tumor outgrowth. The skin eosinophils display tissue adaptation and dynamically alter their transcriptome according to the

Dual effect of targeting LSD1 on the invasiveness and the mechanical response of acute lymphoblastic leukemia cells.

Epigenetic alterations are hallmarks of cancer, with histone modifiers playing critical roles in gene transcription, DNA homeostasis, and other nuclear functions. Lysine-specific demethylase 1 (LSD1), a key regulator of H3K4 methylation, has emerged as a promising pharmacological target in cancer treatment, including leukemia. Acute lymphoblastic leukemia (ALL), the most common pediatric cancer, remains a significant therapeutic challenge due to limited understanding of how epigenetic therapy impacts leukemia dissemination. In this study, we demonstrate that targeting LSD1 enhances the invasiv

Low-Dose Caffeine Exposure Protects the Human Genome from Ionizing Radiation-Induced Damage and Prolongs Mouse Longevity.

Coffee is one of the most widely consumed beverages in the world and is a rich source of caffeine, a methylxanthine. Here we show that exposure to caffeine significantly reduces ionizing radiation (IR) induced DNA breaks and resulted in no or minimal G2/M arrest within the human cell, in contrast to IR alone. At the molecular level, we demonstrate that when naked plasmid DNA or oligomeric DNA was irradiated, the number of breaks was significantly less in the presence of caffeine. The observed radioprotection was irrespective of its sequence and was due to quenching of ROS by caffeine. Besides,

Plasma 4-hydroxynonenal estimates severity and prognosis in patients with acute exacerbation of chronic obstructive pulmonary disease.

4-Hydroxynonenal (4-HNE), a product of lipid peroxidation, is recognized as a biomarker of oxidative stress. However, its relationship with the severity and prognosis of acute exacerbation in chronic obstructive pulmonary disease (AECOPD) remains unclear. This prospective cohort study aimed to investigate the associations between plasma 4-HNE levels and disease severity and prognosis in AECOPD patients. A total of 150 AECOPD patients, 80 stable COPD (SCOPD) patients, and healthy volunteers were enrolled. Plasma 4-HNE and inflammatory cytokines were measured using enzyme-linked immunosorbent as

Selection criteria in the talent identification of triathlon.

Triathlon is a very complex sport, as the athlete has to master the characteristics of three sports (swimming, cycling and running), and the tasks of coaches are increased by the need to get the most out of all three. This sport improves endurance, has a positive effect on muscle development, movement coordination, breathing, and circulation. For talent identification factors determining the performance in triathlon are essential (physiological, anthropometric, psychosocial and tactical factors).Scientific literature concerning performance in triathlon sport is rather scarce. Although there are

Improving Sensory Differentiation: Refining the 'Fruitiness' Descriptor in Extra Virgin Olive Oil.

Sensory analysis is a fundamental tool in evaluating extra virgin olive oil (EVOO) quality, playing an essential role in both consumer markets and international competitions that recognize and promote high-quality olive oils. Among the key attributes assessed, the fruitiness descriptor-subcategorized as green or ripe-is particularly significant, especially considering that higher green fruitiness is often associated with greater prestige. However, a clear methodological approach to distinguish between green fruitiness and ripe fruitiness perceptions, particularly in their overlapping zone, is

Temporal Dynamics and Surveillance of Highly Pathogenic H5 Avian Influenza in Wild Birds in Northern Serbia (2016-2025).

Wild birds, particularly migratory waterfowl (family Anatidae), are the primary reservoir of avian influenza viruses, playing a key role in their transboundary spread. In the winter of 2016/2017, the H5N8 strain was first recorded in Serbia, with 20 outbreaks confirmed in wild birds by 2021, mostly involving mute swans in the Vojvodina region. From 2021 onward, the dominant strain was H5N1, with 38 confirmed outbreaks up to 2023, primarily affecting mute swans, black-headed gulls, and common cranes. Sporadic H5N2 outbreaks were also detected, with two cases in 2021 and 2023. Due to its rich or

Potassium-Doped MnO2 Nanoparticles Reprogram Neutrophil Calcium Signaling to Accelerate Healing of Methicillin-Resistant Staphylococcus aureus-Infected Diabetic Wounds.

Neutrophils, as first-line immune cells, typically lose their edge within the diabetic wounds accompanied by methicillin-resistant Staphylococcus aureus (MRSA) infections (the D/M setting), playing the role of "more foe than friend" during the healing process. Specifically, reduced influx of calcium ions (Ca2+) and impaired calcium homeostasis yield the dysfunction of neutrophil sequential behaviors in pathogen killing and wound healing, manifesting as suppressed chemotaxis, decreased intracellular reactive oxygen species (ROS) generation, prolonged apoptosis, and retention of neutrophil extra

Microglial TREM2 deficiency causes E/I imbalance and triggers ASD-like behaviors by altering potassium channel Kv1.3 activation.

Autism Spectrum Disorder (ASD) is a type of neurodevelopmental disorder characterized by deficits in social interaction and stereotyped behavior, with synaptic dysfunction playing a causal role in its pathogenesis. Microglia, as innate immune cells in CNS, are involved in regulation of synaptic transmission and plasticity through directly pruning spines and indirectly releasing bioactive substances. Interestingly, loss-of-function of triggering receptor expressed on myeloid cells 2 (TREM2), a key receptor in regulation of microglial immune functions, has been implicated in neurodevelopmental a
