The antinociceptive and anti-inflammatory activities of caulerpin, a bisindole alkaloid isolated from seaweeds of the genus Caulerpa.

The antinociceptive and anti-inflammatory activity of caulerpin was investigated. This bisindole alkaloid was isolated from the lipoid extract of Caulerpa racemosa and its structure was identified by spectroscopic methods, including IR and NMR techniques. The pharmacological assays used were the writhing and the hot plate tests, the formalin-induced pain, the capsaicin-induced ear edema and the carrageenan-induced peritonitis. Caulerpin was given orally at a concentration of 100 micromol/kg. In the abdominal constriction test caulerpin showed reduction in the acetic acid-induced nociception at

Local gene delivery of tumor necrosis factor alpha can impact primary tumor growth and metastases through a host-mediated response.

TNFerade is a replication incompetent adenovector designed to express human TNFalpha under control of the Egr-1 radiation and chemotherapy enhanced promoter, and is currently in Phase II/III clinical testing. Data from Phase I clinical testing of TNFerade in a limited set of melanoma patients suggested the potential to impact distal metastases following intratumoral injections of TNFerade. These clinical observations and the multiple potential mechanisms of TNFerade led us to hypothesize local treatment with TNFerade + radiation may impact metastatic disease. We explored this hypothesis in pre

Layer-specific touch-dependent facilitation and depression in the somatosensory cortex during active whisking.

Brains adapt to new situations by retuning their neurons. The most common form of neuronal adaptation, typically observed with repetitive stimulations of passive sensory organs, is depression (responses gradually decrease until stabilized). We studied cortical adaptation when stimuli are acquired by active movements of the sensory organ. In anesthetized rats, artificial whisking was induced at 5 Hz, and activity of individual neurons in layers 2-5 was recorded during whisking in air (Whisking condition) and whisking against an object (Touch condition). Response strengths were assessed by spike

Ovine model for clear-cut study on the role of cholecystokinin in antral, small intestinal and gallbladder motility.

Cholecystokinin (CCK) is one of the major gastrointestinal hormones involved in the control of digestive tract and gallbladder motility. Its action involves several mechanisms. The ovine model was developed in order to further explore the role of CCK in gastric, small intestinal and gallbladder motility under various experimental conditions. Five Merino sheep were used with bipolar electrodes implanted to their antrum, entire small intestine and gallbladder as well as strain gauge force transducers were attached to the duodenum and gallbladder fundus, near the electrodes. In the course of chro

Apoptotic activity of ursolic acid may correlate with the inhibition of initiation of DNA replication.

Ursolic acid (UA), a pentacyclic triterpene acid, has been reported to exhibit anti-tumor activity. In this study, we investigated the pro-a … ted predominantly in the G(0) and G(1) phases with a concomitant decrease in the cell population of S phase. Moreover, expression of p21(WAF1), a cell-cycle regulator, was increased by UA, indicating that p21(WAF1) might mediate UA-induced cell-cycle arrest. However, UA markedly inhibited SV40 DNA replication in the initiation stage in vitro and significa …

Comparison of the rate of excision of major UV photoproducts in the strands of the human HPRT gene of normal and xeroderma pigmentosum variant cells.

Xeroderma pigmentosum (XP) variant patients are genetically predisposed to sunlight-induced skin cancer. Fibroblasts from such patients are … al fibroblasts, irradiated them in early G1-phase, 12 or more hours prior to the scheduled onset of S phase, harvested them immediately or after allowing various times for repair, and analyzed the DNA for photoproducts in the HPRT gene, using quantitative Southern blotting. DNA damage in human B cells can induce apoptosis, proceeding from G1/S when p53 is transactivation competent and G2/M when it is transactivation defective.

Cisplatin treatment of Epstein-Barr virus-immortalized human B lymphoblastoid cell lines (LCLs) results in p53-mediated apoptosis which occurs largely in a population of cells at the G1/S boundary of the cell cycle. Cell cycle progression appears to be required for this apoptosis because arresting cells earlier in G1 inhibited apoptosis despite the accumulation of p53. Overexpression of wild-type p53 also induces apoptosis in an LCL. Therefore six mutant genes derived from Burkitt's lymphoma (BL) cells were assayed for their ability to induce apoptosis when similarly overexpressed. The same ge

The potential role of bacterial superantigens in the pathogenesis of Kawasaki syndrome.

Kawasaki syndrome is an acute multisystem vasculitis of infancy and early childhood associated with high fever, mucocutaneous inflammation, and the development of coronary artery abnormalities. Despite the widely held belief that Kawasaki syndrome is an infectious disease, investigations have failed to identify a causal organism. Previous studies have demonstrated that this illness is associated with marked activation of monocyte/macrophages and the selective expansion of V beta 2-, less so, of V beta 8.1/8.2-expressing T cells in the peripheral blood from Kawasaki syndrome patients during the

From HIV infection to AIDS: are the manifestations of effective immune resistance misinterpreted?

Viewing the immune system as part of an adaptive communication network, rather than merely a coalition of effectors, we argue that the alter … ssified and evoke selective activities. The relative stability of the virus-host relationship in this phase involves a degree of mutual adaptation. However, an excessively perturbed microenvironment is the core of unstable cellular organization in which the resistance to infection gradually deteriorates. We suggest that this is due to "overadaptation" of …

Prolactin as an inhibitor of granulosa cell luteinization: implications for hyperprolactinemia-associated luteal phase dysfunction.

Chronic and transient hyperprolactinemia have been associated with luteal phase dysfunction in both spontaneously cycling women and those subjected to ovarian hyperstimulation by exogenous gonadotropins. It is the objective of the in vitro and in vivo studies reported herein to examine the possibility that prolactin (PRL), a known constituent of ovarian follicular fluid, may account, at least in part, for the luteinization inhibitory (LI) activity exerted by follicular fluid (FF) under both physiologic and pathophysiologic circumstances. In vitro treatment with human PRL (100 ng/ml) reduced by

Expression of beta-adrenergic receptors in synchronous and asynchronous S49 lymphoma cells. I. Receptor metabolism after irreversible blockade of receptors and in cells traversing the cell division cycle.

We have examined the metabolism of beta-adrenergic receptors in intact S49 lymphoma cells. Centrifugal elutriation was used to prepare synchronized cells enriched in particular phases of the cell division cycle. In these synchronized cells, the rate of appearance of beta-adrenergic receptors (i.e., [125I]iodocyanopindolol binding sites) was continuous, approximately 75 sites/cell/hr, and receptor number per cell increased in proportion to the increase in cell size. Thus, receptor number, normalized for cell size, remains constant throughout the cell cycle. Receptors on cells in G1, S, and G2/M

Antibody kinetics following confirmed Shigella infection: A longitudinal study in Dhaka, Bangladesh.

BACKGROUND: Shigella causes an estimated 200 million infections annually, disproportionately affecting children in low- and middle-income countries. Accurate incidence measurement is hindered by brief pathogen shedding and limited diagnostics. Serological approaches can detect infections missed by clinical surveillance but require well-characterized post-infection antibody kinetics. METHODOLOGY/PRINCIPAL FINDINGS: We analyzed archived sera from the Study of Shigella Antibody Responses (SOSAR), a longitudinal cohort of 48 individuals with culture- or molecular-confirmed Shigella infection in D

Use of hysteroscopy in diagnosis and follow-up of acquired uterine enhanced myometrial vascularity.

OBJECTIVE: To describe the role of hysteroscopy in diagnosis and subsequent follow-up of uterine enhanced myometrial vascularity (EMV). Uterine EMV, previously known as arteriovenous malformation (AVM), is a rare but cannot-miss finding often associated with prior pregnancy or uterine surgery and is typically suspected when a vascular mass is found on ultrasound. Color Doppler imaging will demonstrate high-velocity, low-impedance flow, with more significant shunts demonstrating higher peak systolic velocity (PSV). If not already diagnosed by ultrasound, accurate recognition during hysteroscopy

Dynamic brain network states in human generalized spike-wave discharges.

Generalized spike-wave discharges in idiopathic generalized epilepsy are conventionally assumed to have abrupt onset and offset. However, in rodent models, discharges emerge during a dynamic evolution of brain network states, extending several seconds before and after the discharge. In human idiopathic generalized epilepsy, simultaneous EEG and functional MRI shows cortical regions may be active before discharges, and network connectivity around discharges may not be normal. Here, in human idiopathic generalized epilepsy, we investigated whether generalized spike-wave discharges emerge during

STZ‑induced sporadic Alzheimer's in rats: behavioral, histopathological, and MiRNA profiles.

Alzheimer's disease (AD) is a multifactorial disease, resulting from a combination of environmental and genetic predisposing factors, leading to the build-up of Amyloid (Aβ) plaques and tangles of phosphorylated Tau (p-Tau). MicroRNAs such as Let-7 s have been shown to exert neurotoxic effects. MiR-107 endogenously suppresses Let-7 s, and it was found to be downregulated in the brains of AD patients. However, miR-107 expression in the hippocampus in the STZ-induced sAD model has yet to be investigated. Thus, the current study aimed to investigate the levels of selected Let-7 s and miR-107 in t

Commentary on "Trace cisplatin adsorption by thiol-functionalized sponge (TFS) and Sn/SnO₂-coated TFS: Adsorption study and mechanism investigation".

Cisplatin is an environmentally relevant cytostatic contaminant, and the thiol-functionalized sponge materials reported by Han et al. represent a valuable contribution to the development of selective adsorbents for trace platinum-based pharmaceuticals. The present Commentary does not question the synthetic merit of the materials, their adsorption performance, or the surface-sensitive evidence supporting local Pt-S coordination. The scientific concern is narrower and methodological. It concerns whether the reported thermodynamic parameters and mechanistic terminology are justified by the equati

HONO chemistry and its impact on the atmospheric oxidizing capacity over the Indo-Gangetic Plain.

Chemical processes involving nitrous acid (HONO) play a pivotal role as it is a notable source of hydroxyl (∙OH) radicals, influencing the oxidation capacity of the atmosphere. We conduct a comprehensive investigation into the temporal dynamics of HONO, other gases (nitrogen oxides (NOx), ozone (O3), ammonia (NH3), sulphur dioxide (SO2), and nitric acid (HNO3)), particulate matter (PM2.5), and meteorological parameters using measurements that took place during the Winter Fog Experiment (WiFEx) campaign in Delhi, India, during the winter of 2017-2018. Energy Expenditure of Dynamic Submaximal Human Plantarflexion Movements: Model Prediction and Validation by in-vivo Magnetic Resonance Spectroscopy.

To understand the organization and efficiency of biological movement, it is important to evaluate the energy requirements on the level of individual muscles. To this end, predicting energy expenditure with musculoskeletal models in forward-dynamic computer simulations is currently the most promising approach. However, it is challenging to validate muscle models in-vivo in humans, because access to the energy expenditure of single muscles is difficult. Previous approaches focused on whole body energy expenditure, e.g., oxygen consumption (VO2), or on thermal measurements of individual muscles b
