Germinal center B cell development has distinctly regulated stages completed by disengagement from T cell help.

To reconcile conflicting reports on the role of CD40 signaling in germinal center (GC) formation, we examined the earliest stages of murine GC B cell differentiation. Peri-follicular GC precursors first expressed intermediate levels of BCL6 while co-expressing the transcription factors RelB and IRF4, the latter known to repress Bcl6 transcription. Transition of GC precursors to the BCL6hi follicular state was associated with cell division, although the number of required cell divisions was immunogen dose dependent. Potentiating T cell help or CD40 signaling in these GC precursors actively repr

A review of quantitative modeling of B cell responses to antigenic challenge.

A key role of B cells in the mammalian immune response is the generation of antibodies that serve to protect the organism against antigenic challenges. The same process may also be detrimental in the context of autoimmunity. Several modeling approaches have been applied to this aspect of the immune response, from predicting potential epitopes to describing B cells progress through developmental models and simulating antibody production. Here we review some of the modeling techniques, and summarize models that describe different activation mechanisms for B cells, including T cell dependent and

Common variable immunodeficiency and autoimmunity--an inconvenient truth.

Coexisting morbidities in CVID include bronchiectasis, autoimmunity and malignancies. The incidence of autoimmune disease in CVID patients may approach 20% of cases. The most common autoimmune disease found in CVID patients is autoimmune cytopenia, but rheumatoid arthritis, lupus, and now primary biliary cirrhosis have also been reported. The coexistence of immunodeficiency and autoimmunity appears paradoxical, since one represents a hypoimmune state and the other a hyperimmune state. However, this paradox may not actually be all that implausible due to the complex nature of immune cells, sign

Peculiar tonsil-like structure near vagina of the laboratory shrew, Suncus murinus.

After finding tonsil-like structures near the entrance of vagina of a laboratory shrew (Suncus murinus), which we subsequently designated as vaginal tonsils, we performed detailed immunohistochemical and developmental studies. The location of T and B cells in the vaginal tonsils differed from that in the palatine tonsils or that in the lymphoid nodes of other animals. The boundary between the germinal center region and the region encompassing follicular interfollicular tissue was not clear. B cells were widely distributed and very dense in the parenchyma, but they were scattered in the epithel

[Pathological mechanisms in rheumatoid arthritis].

Rheumatoid arthritis(RA) is a representative autoimmune disease characterized by systemic, chronic, destructive inflammatory synovitis and multiple organ manifestations, resulting in severe disability and mortality rates. Immune cells such as T cells, B cells, dendritic cells and macrophages and cellular interaction among them through cytokines and cell surface molecules plays a pivotal role in the pathological processes of RA. The accumulation of inflammatory cells, the self-perpetuation of inflammation and induction and/or activation of osteoclasts lead to cartilage and bone destruction and

[Role of the immune system in the pathophysiology of postmenopausal osteoporosis].

The immune system plays a critical role in the pathophysiology of postmenopausal osteoporosis. Estrogen deficiency induces mild increase in production of proinflammatory cytokines, such as IL-1, IL-6, and TNF-alpha, systemically and locally, which promotes osteoclastogenesis in the bone marrow. The most important cytokine in the context of estrogen deficiency-induced bone loss has been shown to be TNF-alpha produced by bone marrow T-lymphocytes that are stimulated through a complex mechanism involving antigen-presenting cells and various cytokines including IL-7, IFN-gamma, and TGF-beta. Immunohistochemical aspects of the evaluation of the inflammatory answer of the dental pulp.

The inflammatory answer of the dental pulp in front of an antigen presents a series of particularities caused by its special anatomical cond … ory aspects of the dental pulp. We did not find positive answers for the CD20 protein, specific for B-lymphocytes, for the fragments of normal pulp tissue taken into study. Even if they are in small number among the pulp cells, the T-lymphocytes that express the protein CD45Ro may be also found in the normal dental pulp. Of the two subsets of T-lymphocytes, we …

Polyclonal B cell activation in infections: infectious agents' devilry or defense mechanism of the host?

Polyclonal B cell activation is not a peculiar characteristic to a particular infection, as many viruses, bacteria, and parasites induce a strong polyclonal B cell response resulting in hyper-gamma-globulinemia. Here, we discuss the different roles proposed for polyclonal B cell activation, which can be crucial for early host defense against rapidly dividing microorganisms by contributing antibodies specific for a spectrum of conserved structures present in the pathogens. In addition, polyclonal B cell activation can be responsible for maintenance of memory B cell responses because of the cont

Highlights of Mediterranean clinical immunology.

The immune system continues to fascinate by the complexity of its intricacies. At the First Mediterranean Workshop on Clinical Immunology held in Evora (Portugal), recently identified mechanisms of immune defense and immunoregulation were put under a magnifying glass by an international cast of immunologists. Studies of Bacillus anthracis revealed that this anaerobic bacterium can inhibit type-II A phospholipase synthesis and secretion by alveolar macrophages, thereby subverting the pulmonary host immune response. Investigation of the mode of action of regulatory T cells indicated that FOXP3 b

Stem cell transplantation for autoimmune disorders. Immune reconstitution.

Lymphocyte recovery is delayed following autologous haematopoietic stem cell transplantation (HSCT). B-cells recover before T-cells and CD8+ before CD4+ T-cells. The initial phase of T-cell recovery is dependent upon the expansion of mature host T-cells that have survived conditioning or are transferred back with the graft. This phase is therefore quicker when the graft is not CD34+ selected. Subsequently, naïve T-cells appear. Naïve CD4+ T-cell recovery is thymus dependent and starts at around 6-9 months. Naïve CD8+ recovery occurs earlier and seems less thymus dependent. Immune function reco

[Pathology of Hodgkin's lymphoma].

Hodgkin's lymphoma can be divided with the help of morphologic and immunohistochemical techniques into classical and nodular lymphocyte predominant Hodgkin's lymphoma. By single cell analyses it could be established that the tumor cells of Hodgkin's lymphoma are clonal B-cells with germinal center origin. In rare cases (less than 5 %), the Hodgkin and Reed Sternberg cells represent clonal tumor cells that derive from T-cells. By gene expression analyses it can be shown that Hodgkin cell lines represent an entity independently of their B- or T-cell-origin. Hodgkin cell lines show similarities t

Selective IgM deficiency in a patient with Hashimoto's disease.

A 70-year-old man with Hashimoto's disease had selective IgM deficiency, while other immunoglobulin levels were normal. In vitro mixing experiments were carried out in which B cells and T cells from the patient and from a healthy control donor were co-cultured in the presence of pokeweed mitogen, in order to investigate the etiology of IgM hypoproduction. The results indicated that the patient had B-cell dysfunction, involving the impairment of B-cell differentiation. In addition, both IgG of the healthy control donor and thyroid hormone may play important roles in the pathogenesis of this cas

[Scanning electron microscopic examination of 50 cases of non-Hodgkin's lymphoma (NHL)].

Fifty cases of NHL were studied by scanning electron microscopy to study the possible differences in the surface morphology of tumor cells between T cell and B cell NHL. Acid naphthyl acetate esterase (ANAE) activity test, the stain of antiserum samples of gamma, mu, alpha, kappa, lambda chains with the peroxidase antiperoxidase immune complex method (PAP method) and classification under light microscope were done for all the cases. It was found that the surface morphology of T cell lymphoma cells differed from that of B cell lymphoma. There were a lot of granular protrusions with a few pseudo

Cellular immunity in systemic lupus erythematosus.

Production of pathogenic autoantibodies in patients with systemic lupus erythematosus (SLE) is the cornerstone in the pathogenesis of the disease. From the etiologic point of view, there is evidence that genetic, infectious, hormonal and environmental factors are essential to the development of SLE. The relative contribution of each group of factors to the expression of disease is individualized to each patient or group of patients with identical clinical disease. The cellular events which precede the B cell overactivity are crucial. T cells may produce excessive amounts of factors promoting B

Classification of non-Hodgkin malignant lymphomas of the skin.

The authors subdivide the primary non-Hodgkin cutaneous malignant lymphomas into "proper" and "non-proper" types. "Proper" lymphomas are those which have in the skin their proper site of localization, and include mycosis fungoides, Pagetoid reticulosis, Baccaredda-Sézary syndrome and possibly lymphomatoid papulosis. They are T-cell lymphomas arising in the papillary dermis, characterized by epidermotropism, having a specific clinical feature in that they are unlikely to be simulated by other cutaneous malignant lymphomas. "Non-proper" lymphomas are those which do not usually arise in the skin,

[Immunological changes in patients with colorectal cancer].

Sera, duodenal liquid and biopsy materials of small intestine and of mesenteric lymph nodes were collected from 22 patients with colorectal cancer (intestinal lymphoma and adenocarcinoma) and 10 control patients with gastric ulcer were found in all of patients with cancer. The increasing of B type lymphocytes and decreasing of T cells were detected in biopsy materials and high level IgA was found in the sera and duodenal liquid of these patients. The immunological alterations were not seen in control patients with gastric ulcer. These findings suggest that there is a role of cellular and humor

Designing a conjugate vaccine targeting Klebsiella pneumoniae ST258 and ST11.

Klebsiella pneumoniae (K. pneumoniae) is a common bacterium that can cause iatrogenic infection. Recently, the rise of antibiotic resistance among K. pneumoniae strains is one key factor associated with antibiotic treatment failure. Hencefore, there is an urgent need for effective K. pneumoniae vaccines. This study aimed to design a multi-epitope vaccine (MEV) candidate against K. pneumonia by utilizing an immunoinformatics method. In this study, we obtained 15 cytotoxic T lymphocyte epitopes, 10 helper T lymphocyte epitopes, 6 linear B-cell epitopes, and 2 conformational B-cell epitopes for f

Novel BTK Mutation in Patient with Late Diagnosis of X-Linked Agammaglobulinemia.

X-linked agammaglobulinemia (XLA) is a genetic disorder with mutation in Bruton's tyrosine kinase (BTK). Defects in B cell development and immunoglobulin production lead to recurrent infections following loss of maternal IgG at 6 months of age. A 55-year-old male with a longstanding common variable immunodeficiency diagnosis on infusion therapy presented to the clinic with cutaneous T-cell lymphoma, which inspired overall repeat evaluation. Immunoglobulin levels and lymphocyte markers, family history, and genetic testing prompted a true diagnosis of XLA and novel mutation in the BTK gene. Dise
