Redox Control of Seed Germination is Mediated by the Crosstalk of Nitric Oxide and Reactive Oxygen Species.

Significance: Seed germination and seedling establishment are characterized by changes in the intracellular redox state modulated by accelerated production of nitric oxide (NO) and reactive oxygen species (ROS). Redox regulation and enhanced accumulation of NO and ROS, approaching excessively high levels during seed imbibition, are critically important for breaking endodormancy and inducing germination. Recent Advances: Upon depletion of oxygen under the seed coat, NO is produced anaerobically in the reductive pathway associated mainly with mitochondria, and it participates in the energy metab

Comparative analysis of structural modifications induced by monocular retinal inactivation and monocular deprivation in the developing cat lateral geniculate nucleus.

During a critical period of postnatal life, monocular deprivation (MD) by eyelid closure reduces the size of neurons in layers of the dorsal lateral geniculate nucleus (dLGN) connected to the deprived eye and shifts cortical ocular dominance in favor of the non-deprived eye. Temporary inactivation of the non-deprived eye can promote superior recovery from the effects of long-term MD compared to conventional occlusion therapy. In the current study, we assessed the modification of neuron size in the dLGN as a means of measuring the impact of a brief period of monocular inactivation (MI) imposed

Ferritin and Neurotoxicity: A Contributor to Deleterious Outcomes for Subarachnoid Hemorrhage.

BACKGROUND: Ferritin is a protein that is critical for storing iron. Ferritin has recently been shown to play a role in iron homeostasis, immunomodulation, inflammation, and antioxidation. Previously, it was believed that ferritin was exclusively an intracellular peptide. However, there is significant evidence that ferritin is also in the serum, cerebral spinal fluid, and synovial fluid. SUMMARY: Within the brain, ferritin can bind to oligodendrocytes adjacent to the blood-brain barrier to allow a docking point for ferritin to be engulfed by microglia in the brain parenchyma. When iron suppli

Carbonic anhydrase-8 regulates inflammatory pain by inhibiting the ITPR1-cytosolic free calcium pathway.

Calcium dysregulation is causally linked with various forms of neuropathology including seizure disorders, multiple sclerosis, Huntington's disease, Alzheimer's, spinal cerebellar ataxia (SCA) and chronic pain. Carbonic anhydrase-8 (Car8) is an allosteric inhibitor of inositol trisphosphate receptor-1 (ITPR1), which regulates intracellular calcium release fundamental to critical cellular functions including neuronal excitability, neurite outgrowth, neurotransmitter release, mitochondrial energy production and cell fate. In this report we test the hypothesis that Car8 regulation of ITPR1 and cy

Unlocking Myofibroblast Plasticity to Reverse Salivary Gland Fibrosis.

Salivary gland (SG) fibrosis represents a critical pathological feature of exocrine hypofunction driven by the persistent activation of myofibroblasts (MFs). However, the translational gap between the convoluted in vivo regulatory networks governing these cells and the deployment of precise, disease-modifying interventions remains a formidable challenge. This review systematically examines current mechanistic evidence undergirding MF activation, survival, and its phenotypic reversal, focusing on restoring glandular function. We synthesize recent literature mapping in vivo canonical profibrotic

The XPO7-NPAT axis represents key vulnerabilities in TP53-mutated acute myeloid leukemia.

Acute myeloid leukemia (AML) with TP53 mutations is almost universally refractory to chemotherapy, molecular-targeted therapies, and hematopoietic stem cell transplantation, leading to dismal clinical outcomes. The lack of effective treatments underscores the urgent need for novel therapeutic strategies. Using genome-wide CRISPR/Cas9 dropout screens in isogenic Trp53-wild-type (WT) and Trp53-knockout mouse AML models, combined with transcriptomic and proteomic analyses of AML samples from mice and humans, we identify the XPO7-NPAT (exportin 7-nuclear protein, coactivator of histone transcripti

HBx-mediated GPT2 suppression promotes liver cancer development by downregulating ADH1A.

OBJECTIVE: Hepatitis B virus (HBV) is the primary driver of liver cancer (LC), a malignancy characterized by extensive metabolic reprogramming. However, the specific mechanisms linking viral oncoproteins to metabolic dysregulation in LC remain incompletely understood. This study was aimed at investigating the function of the metabolic enzyme GPT2 in hepatocarcinogenesis, focusing on its regulation by HBV X protein (HBx) and its effects on downstream signaling pathways. METHODS: We combined analysis of multiple patient cohorts with in vitro and in vivo functional assays to determine the clinic

Vascular calcification: pathogenic mechanisms, signaling crosstalk and translational therapeutics.

Vascular calcification (VC) is a multifactorial pathological process characterized by ectopic deposition of hydroxyapatite in the arterial wall, which is closely associated with increased cardiovascular morbidity and mortality. This review provides a comprehensive synthesis of current insights into the pathogenesis of VC, emphasizing its nature as a dynamically regulated process driven by intricate crosstalk among diverse molecular mechanisms, signaling cascades, and pathological pathways. Under specific pathological stimuli, this intricate regulatory network disrupts the homeostatic balance b

Barcoded mutant library enables high-throughput functional genomics in a filamentous fungus.

Advances in sequencing technology enabling rapid and inexpensive whole-genome sequencing highlight how few genes are functionally characterized. This problem is particularly acute in filamentous fungi, where even in the best studied organisms upward of half of genes are poorly characterized or unannotated. High-throughput tools to identify gene function exist for single-celled organisms, like yeast and bacteria. However, filamentous fungi present challenges to high-throughput gene characterization, including low transformation efficiency and multinucleate cells. Filamentous fungi are critical

Penicillium-mediated toxicity and microbial dysbiosis are associated with fruiting body abortion in artificially cultivated Chinese cordyceps.

Chinese cordyceps consists of fruiting body and sclerotia (larvae part) formed through the parasitism of the insect Hepialidae by the fungus Ophiocordyceps sinensis, but artificial cultivation is hindered by fruiting body abortion, severely impacting yield and quality. The interactions between the fungus, its host insects, and soil are critical for its development. Multi-omics analyses were conducted to compare normally and abnormally developing samples, examining microbial communities and metabolites in fungus-colonized host larvae and the mycosphere soil. Abnormal fruiting body development w

Deletion of RhoGDI Protects Against Hepatic Steatosis via Improved Mitochondrial Metabolism in Mice.

The global incidence of metabolic dysfunction-associated steatotic liver disease (MASLD) is rising alongside epidemics of diabetes and obesity. Rho GDP-dissociation inhibitor (RhoGDI) is now recognized to play dual regulatory roles in disease. A deeper understanding of its mechanistic contributions in MASLD could offer critical insights for developing novel therapies against this growing health burden. Immunohistochemical staining was used to examine RhoGDI expression in liver tissues from patients with MASLD. Hepatocyte-specific deletion of Arhgdia (the gene encodes RhoGDI) was generated in m

Plant-Based Proteins and Renal Protection in Acute Kidney Injury: Nutritional and Metabolic Perspectives.

Acute kidney injury (AKI) is a frequent complication in critically ill patients and is associated with high morbidity, mortality, and an increased risk of progression to chronic kidney disease (CKD). In this context, nutritional management represents a key component of supportive therapy, as AKI is commonly characterized by hypercatabolism, negative nitrogen balance, and protein-energy wasting. Current nutritional strategies primarily focus on the quantity of protein intake required to compensate for catabolic losses, particularly in patients undergoing renal replacement therapy (RRT). However

Hepatitis B virus X protein induces E6-associated protein-mediated proteasomal degradation of p53 phosphorylated at Ser-15.

The hepatitis B virus (HBV) X protein (HBx) activates the ataxia telangiectasia mutated (ATM)-checkpoint kinase 2 (Chk2) pathway, leading to phosphorylation of p53 at multiple sites including Ser-15. This phosphorylation largely contributes to p53 accumulation by preventing its degradation via MDM2. This study further shows that HBx facilitates E6-associated protein (E6AP)-mediated proteasomal degradation of p53 during HBV replication. In the presence of HBx, E6AP expression induced p53 ubiquitination, reduced its stability and decreased p53 levels, whereas E6AP knockdown elevated p53 levels.

Ginkgetin alleviates cisplatin-induced muscle atrophy via inhibition of the macrophage cGAS-STING pathway.

Chemotherapy-induced muscle atrophy is a severe side effect, impairing patients' quality of life and overall survival. However, the persistence of muscle atrophy in cancer survivors long after treatment completion suggests that it is driven not only by the agent's direct toxicity, but also by a persistent, chemotherapy-induced pathological immune microenvironment. Elucidating the interplay between chemotherapy drugs, the immune microenvironment, and muscle cells is essential for identifying mechanisms and potential therapeutic targets. In this study, we investigated the critical role of macrop

A Splice Acceptor Variant in DLL3 Is Associated with Spondylocostal Dysostosis in a Litter of Mixed-Breed Dogs.

BACKGROUND/OBJECTIVES: Spondylocostal dysostosis (SCDO) is a rare disorder characterized by congenital malformations of the spine and ribs. SCDO affects 1 in 40,000 human births, with rare cases also reported in dogs. Mutations in DLL3, encoding a critical Notch signaling pathway ligand, account for a majority of human SCDO cases. The remaining cases have variants in HES7, LFNG, MESP2, RIPPLY2, TBX6, and DLL1, which code for proteins in the Notch pathway. A mixed-breed litter of three dogs presented with varying degrees of spinal malformations and underwent comprehensive phenotyping including

Hippo Pathway-YAP/TAZ Signaling: Molecular Mechanisms, Biological Function, Diseases, and Therapeutic Targets.

The Hippo pathway and its downstream effectors Yes-associated protein/transcriptional coactivator with PDZ-binding motif-TEA domain transcription factor (YAP/TAZ-TEAD) play critical roles in organ-size control, tissue homeostasis, regeneration, and stem-cell biology. Their aberrant activation drives malignancies, fibrosis, cardiovascular disease, and immune dysregulation. Once regarded undruggable, the YAP/TAZ-TEAD complex has become one of the most actively pursued target classes in oncology and beyond. In this review, we delineate the biological functions of the Hippo-YAP/TAZ-TEAD axis and i

Progress on Mechanisms of Therapeutic Potential of Oxygen Delivery in Brain Disorders.

Oxygen is both essential and potentially toxic to the brain, which consumes 20-25% of the body's resting metabolic rate despite comprising only 2% of total body weight. The exquisite sensitivity of neural tissue to oxygen imbalance positions oxygen delivery as a critical determinant of both pathophysiology and therapeutic opportunity in brain disorders. This review synthesizes recent advances in our understanding of how oxygen sensing and delivery can be harnessed for therapeutic benefit across the spectrum of neurological conditions. We examine the molecular machinery of oxygen sensing, inclu
