Plant-derived exosomes (PDEs) are gaining attention owing to their key implications in cross-kingdom communication, facilitating bioactive entities among plants and animals. PDEs are tiny nanoscale vesicles generally comprised of RNAs, proteins, and secondary metabolites and are involved in the regulation of physiological processes (immune modulation, cell regeneration, and stress response). An important feature of PDEs is to enable cross-kingdom regulation in skin wound repair. This is because PDEs can modulate several signaling pathways (PI3K-Akt, TGF-β, and mitogen-activated protein kinase)

Heparan sulfate (HS) proteoglycans are essential regulators of cell signaling, development, and extracellular matrix organization. Central to the HS biosynthesis pathway is the exostosin family of enzymes that control commitment to HS assembly and catalyze formation of the polymer backbone in the Golgi apparatus. Recent structural studies have substantially revised our understanding by demonstrating that the HS co-polymerase is a tightly organized EXT1-EXT2 heterodimer, wherein the GT-B domain of EXT1 and the GT-A domain of EXT2 provide the β1,4-GlcA and α1,4-GlcNAc transferase activities, res

Autosomal recessive spinocerebellar ataxia type 10 (SCAR10) is a rare, slowly progressive neurodegenerative disorder characterised by ataxia, cerebellar atrophy, and oculomotor abnormalities, caused by variants in the ANO10 (anoctamin 10) gene. While ANO10-mediated calcium dysregulation in Purkinje cells has been proposed to cause SCAR10, the precise pathogenic mechanism remains unclear. To investigate cellular responses to ANO10 deficiency, four ANO10-mutant neuronal cell lines - a knockout and three lines harbouring recurrent SCAR10-associated pathogenic variants - were engineered using CRIS

Solid tumors are highly dynamic environments, in which endothelial cells (ECs) interface with blood, cancer cells, and the extracellular matrix (ECM), thereby regulating tumor behavior and metastatic dissemination. We recently showed that aberrant Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) activation promotes tumor progression through ECM remodeling. Here we investigated whether VEGFR2-driven ECM remodeling affects EC behavior and vessel stability. Using decellularized ECM from Sk-Mel-31 melanoma cells expressing either VEGFR2WT or the aberrant VEGFR2R1032Q mutant, we found that VE

Osteoarthritis (OA) is a degenerative joint disease characterized by cartilage degradation and chondrocyte senescence. Emerging evidence implicates NADPH oxidase 4 (Nox4) in OA pathogenesis. This study investigated the role of Nox4 in regulating extracellular matrix (ECM) homeostasis and chondrocyte senescence through the p38 mitogen-activated protein kinase (p38MAPK) signaling pathway. We found that Nox4 expression was significantly upregulated in OA chondrocytes and cartilage. In a post-traumatic OA mouse model (Anterior cruciate ligament transection, ACLT), Nox4 knockout attenuated cartilag

The pharmacological activities of traditional herbal medicine-derived carbon dots (THM-CDs) render them highly promising for applications in biomedicine. However, the interactions between THM-CDs and intracellular proteins have yet to be fully elucidated. Herein, inspired by the traditional vinegar-processing of Artemisia argyi (A. argyi), acetic acid-treated Artemisia argyi-derived carbon dots (AA-CDs) demonstrate excellent resistance to iron death via efficient ROS scavenging and Fe2+ chelating. Notably, multi-omics analysis uncovers a novel whereby AA-CDs directly target and stabilize heat

Lumican, an extracellular matrix (ECM) component highly expressed in placenta, is implicated in preeclampsia (PE) pathogenesis, though its precise mechanisms in trophoblast function and PE development remain unclear. Transcriptomic analysis of a PE rat model induced by L-NAME revealed altered placental gene expression. Lumican expression progressively increased throughout rat and human gestation but was significantly downregulated in PE patient and model placentas. Functional assays showed shRNA-mediated lumican knockdown inhibited trophoblast proliferation, migration, and invasion while promo

Liver fibrosis is defined as the excessive accumulation of extracellular matrix proteins in the liver due to chronic liver injury. Targeted ferroptosis of activated hepatic stellate cells (HSCs) is considered a promising therapeutic strategy for liver fibrosis. Rubimaillin (Rub), a naphthoquinone compound extracted from traditional Chinese medicine Rubia cordifolia L., exhibits various activities in multiple diseases. This study aimed to investigate the anti-hepatic fibrosis effect, the direct protein target, and molecular mechanism of Rub. Here, our results demonstrated that Rub effectively a…

Asthma is characterized by persistent airway epithelial dysfunction and remodelling of the reticular basement membrane (RBM). In healthy airways, the RBM is primarily composed of the extracellular matrix (ECM) proteins laminin and collagen-IV, but in remodelled asthmatic airways, the RBM has increased deposition of collagen-I, -III and fibronectin. Here, we systematically compared the effects of collagen-I, -III, -IV, fibronectin, laminin, and bovine serum albumin (BSA) control on bronchial epithelial cells (BECs) from six healthy controls and seven individuals with asthma. Epithelial attachme

Although mouse models of acute allergic inflammation (AAI) have long been used to study asthma mechanisms, their ability to reproduce human disease remains controversial and long-term studies of chronic asthma remain scarce. We compared lung function and tissue changes in three experimental asthma models after four months of recovery. Using x-ray-based lung function (XLF) analysis, we assessed lung parameters and diaphragm motion, alongside lung expression of α-smooth muscle actin and key extracellular matrix proteins, collagen and elastin. A severe ovalbumin (OVA) model (SAA) produced stronge…

Chronic inflammatory signaling contributes to cartilage degeneration across multiple joint diseases, yet the molecular consequences of sustained cytokine exposure remain incompletely understood. We investigated how prolonged interleukin-1β (IL-1β) stimulation remodels the chondrocyte proteome and whether these changes are associated with senescence-associated traits. Primary human articular chondrocytes were exposed to IL-1β (10 ng/mL) for up to four days. Time-resolved data-independent acquisition (DIA) proteomics was integrated with immunofluorescence, quantitative PCR, multiplex metalloprot

Hyaluronan (HA) is a major glycosaminoglycan of the extracellular matrix that regulates cell migration, signaling, and tissue homeostasis. Its turnover is controlled by coordinated synthesis by HA synthases and degradation by hyaluronidases. Among these hyaluronidases, TMEM2, the only known transmembrane hyaluronidase, plays a unique role in HA degradation at the cell surface; however, the cellular conditions that support its activity remain incompletely understood. To address this, we developed a cell-based HA turnover assay to examine TMEM2-mediated degradation of endogenously synthesized HA

Neonatal hypoxia-ischemia (HI) remains a major global cause of neonatal morbidity and mortality. To develop effective therapeutic strategies, a deeper understanding of the acute inflammatory processes following HI is required. As the role of peripheral immune cell infiltration is poorly understood we used single nuclei RNA sequencing to investigate acute sequalae post-HI. This revealed a distinct neutrophil population characterized by increased expression of markers associated with neutrophil extracellular trap (NET) formation. The study evaluated the therapeutic potential of inhibiting NETosi

The homeobox (HOX) gene family is a highly conserved group of transcription factors, important for embryonic patterning, positional identity, and lineage commitment. Although many HOX genes are largely silenced in adult endocrine tissues, emerging evidence suggests that their aberrant reactivation contributes to tumorigenic processes in certain endocrine neoplasms, such as thyroid cancer and adrenocortical carcinoma. This reactivation affects cell differentiation, proliferation, and lineage plasticity. Mechanistically, HOX proteins govern transcriptional networks involved in cell fate determin

Multiple sclerosis (MS) is a chronic inflammatory disease characterized by demyelinating lesions in the central nervous system. While animal models have provided insights into lesion development, human-based studies remain limited. This study investigated the molecular landscape of gray matter lesions at different stages using spatial transcriptomics. Post-mortem cortical tissue from MS patients was analyzed to examine gene expression in lesions classified as early active, chronic active, or chronic inactive based on myelin integrity and macrophage/microglia activity. Distinct transcriptomic p

Fibrosis is a significant barrier to drug delivery in pancreatic ductal adenocarcinoma (PDAC) and contributes to its dismal prognosis. Pancreatic stellate cells (PSCs) drive fibrosis by excessively secreting extracellular matrix proteins such as collagen I. Collagen I is thought to physically obstruct the delivery of macromolecules, such as albumin, antibodies, and nanomedicines. Apart from its structural role, collagen signals through dedicated cell surface receptors, such as the discoidin domain receptors (DDR) 1/2. However, whether and how collagen signaling contributes to fibrotic barrier …
