The effects and physiological consequences of raised levels of cortisol on rainbow trout (Oncorhynchus mykiss) erythrocyte beta-adrenoreceptors.

We have investigated the influence of cortisol on the beta-adrenoreceptor population of rainbow trout [Oncorhynchus mykiss (Walbaum)] erythrocytes and determined what impact it has on the adrenergic responsiveness of erythrocytes in vitro to exogenous catecholamines. To do so, the erythrocyte beta-adrenoreceptors were characterized in fish with chronically elevated plasma cortisol levels (118 +/- 5.9 ng ml-1, greater than 10 days) and compared with shams, using radioreceptor assay techniques. The number of 'internalized', low-affinity receptors was increased when cortisol levels were raised, b

Glucocorticoid receptors in normal leukocytes: effects of age, gender, season, and plasma cortisol concentrations.

We measured glucocorticoid receptors (GR) in mononuclear leukocytes (MNL) isolated from peripheral blood of 145 apparently healthy volunteers (86 men and 59 women). An age-related decrease in the number of GR was suggested between subjects younger than 20 years and elderly subjects; there was no apparent seasonal variation in GR. Gender difference in the number of GR was not significant, although women showed slightly fewer GR. Eight patients with dermatomyositis/polymyositis were examined to determine whether the number of GR in MNL could be down-regulated by their cognate ligands. The number

Salsalate cross-sensitivity in aspirin-sensitive patients with asthma.

Ten aspirin (ASA)-sensitive patients with asthma underwent double-blind, placebo-controlled oral challenges with salsalate followed by ASA-sensitive confirmatory challenges. All 10 patients sustained asthmatic reactions to ASA, but only two developed respiratory reactions to 2 gm of salsalate. In these two patients, repeat confirmatory challenges with 2 gm of salsalate reproduced the same asthmatic reactions. Both patients were desensitized to ASA, and cross-desensitization with 2 gm of salsalate was then achieved. We conclude that salsalate, a weak inhibitor of cyclooxygenase in vitro, is les

[The role of glucocorticoids in the post-vibration changes in inhibition mediation in the brain structures].

Low-frequency vertical vibration for 2 hours together with three-fold increase of glucocorticoids is accompanied by the disruption of typical relationships of neuromediator processes (norepinephrine- and serotoninergic mediation) as well as the reduction of the intensity of choline trapping with glucocorticoid perception. Glucocorticoid-related conformation changes in plasma membranes and a drastic increase of the cAMP level provide plasticity of both nonspecific (hypothalamus, raphe bluish sites and nuclei) and specific (Deiters' nuclei) structures. The relationships of neuromediator processe

Pregnancy in panhypopituitarism.

The case of a 30-year-old pregnant patient suffering from panhypopituitarism is presented. Pregnancy induced by HMG/HCG had been uneventful. Induction of labor by PGE2 vaginal tablets and by intravenous oxytocin at 42 gestational weeks failed and the patient was operated. Following the operation severe exacerbation of her diabetes insipidus occurred. Fluid balance mechanism failed and electrolyte status deteriorated rapidly. The importance of pitressin in maintenance of fluid and electrolyte balance in panhypopituitaric patients in labor and surgical trauma is stressed.

[Is it possible to predict the development of Nelson's syndrome in Cushing's disease after bilateral adrenalectomy?].

A detailed analysis of clinical and laboratory findings in a group of 66 patients with Cushing's disease treated with bilateral adrenalectomy, out of whom 15 developed Nelson's syndrome, gave evidence that Nelson's syndrome mostly affects children and young patients compared to older persons. Bilateral adrenalectomy performed in patients aged over 40 years was never accompanied by Nelson's syndrome. Basal plasma ACTH values prior to adrenalectomy (30.9 +/- 4.53 pmol/l in the absence of Nelson's syndrome and 31.3 +/- 5.41 pmol/l in patients with Nelson's syndrome) as well as the degree of their

Haemolytic uraemic syndrome with shigella.

A 49-year-old male developed bloody diarrhoea whilst on a visit to India. Sigmoidoscopy and rectal biopsy showed acute colitis. Shigella dysentery type I was isolated from stool culture. Cytotoxin production by the organism was demonstrated. The patient developed acute renal failure, thrombocytopaenia and microangiopathic haemolytic anaemia. He required mechanical ventilation, haemodialysis, blood transfusion and antibiotic therapy and achieved a complete recovery. This is an unusual case of haemolytic uraemic syndrome complicating shigellosis in an adult.

Plasma adrenocorticotropic hormone and cortisol in pigs: effects of time of day on basal and stressor-altered concentrations.

An initial study was conducted to establish the presence in plasma of diurnal rhythms of immunoreactive porcine adrenocorticotropic hormone (pACTH) and cortisol in castrated male pigs (barrows). Fourteen barrows with jugular catheters were bled at 6-hr intervals for 24 hr. Significant changes in plasma pACTH were evident with peak levels (61 +/- 6 pg/ml) at 0100-0700 hr and a trough (38 +/- 4 pg/ml) at 1900 hr. Changes (P less than 0.05) in plasma cortisol were also present in barrows with a peak (44 +/- 6 ng/ml) at 0700 hr and a trough (21 +/- 5 ng/ml) at 1900 hr. Plasma norepinephrine and ep

Effects of the opiate agonist loperamide on pituitary-adrenal function in patients with suspected hypercortisolism.

In the present work the possible use of loperamide, an opiate agonist, in the dynamic evaluation of patients with suspected hypercortisolism was investigated. The effects of loperamide on plasma ACTH and cortisol levels were evaluated in normal subjects and in 58 patients with suspected Cushing's syndrome. The results were compared to those obtained after the overnight dexamethasone suppression test. In normal subjects plasma ACTH and cortisol levels were significantly (p less than 0.005) suppressed by both loperamide (16 mg po) and dexamethasone (1 mg po). In 17 patients, in whom the diagnosi

Adrenal cortical reserve in patients undergoing muscle biopsy for malignant hyperthermia screening.

Stress has been suggested as a factor involved in the development of a malignant hyperthermia (MH) reaction. This study has compared the hormonal response of a group of patients susceptible to MH (MHS), with that of a control group, during diagnostic muscle biopsy under general anaesthesia. Preoperative adrenal cortical reserve was assessed using the Short Synacthen Test (SST). There was no significant difference between the groups in their hormonal response to the procedure, both groups showing a peroperative reduction in plasma cortisol concentration from an initially high value, and the SST

Behavioral and endocrine consequences of heterosexual pair formation in squirrel monkeys.

The South American squirrel monkey (Saimiri sciureus) typically lives in large social groups containing several individuals of all age/sex categories. When living in established heterosexual pairs, reproduction in this seasonally breeding primate is poor. We attempted to induce breeding activity in pair-housed monkeys by forming new heterosexual pairs just prior to the breeding season. Breeding readiness, as reflected in behavior and gonadal hormones, was induced in males, but not in females. Males also showed persistent increases in cortisol levels following formation of new heterosexual pair

In vitro studies in primary aldosteronism: baseline steroid production and aldosterone responses to ACTH and angiotensin II.

The spontaneous glucocorticoid production in control adrenal cells (N = 10) and in the adenoma cells (N = 15) exhibited comparable geometric mean values: 1.896 nmol/ml/4-5 x 10(5) cells per 2 h (confidence limits: 0.428-8.391) and 1.852 nmol/ml (0.326-12.241), respectively. The same results were obtained for the three samples of nodular hyperplasia cells. When cortisol and corticosterone were measured separately, there was no significant difference between the outputs for control cells and those for pathological cells. Baseline aldosterone production in control cells showed a geometric mean of

Plasma cortisol in sickle cell disease.

The plasma cortisol levels of 108 children aged 1-16 years with various Hb genotypes (AA, AS and SS) were measured. The mean plasma cortisol levels of both the AS and SS groups (during steady state) fell within the normal range but the mean of the SS group is significantly lower than the AA group. In fact, 9 of the sickle cell children had plasma cortisol levels below the normal range while no child in the HbAS and HbAA groups had plasma cortisol below the normal range. These results indicate that in sickle cell disease, there is low cortisol production while the secretion is increased during

Effects of electroacupuncture and gonadotropin-releasing hormone treatments on hormonal changes in anoestrous sows.

The therapeutic effects of acupuncture in treating reproductive disturbances of man and animals have been proven in the past and is used clinically already. However, the mechanism of such therapy is not known yet. In this study, anoestrous sows were used to investigate the mechanism. Anoestrous sows with luteal ovaries were allocated to three groups. Four sows received electroacupuncture treatment at Pai-Hui and Wei-Ken (acupuncture treated group; group 1). Three sows received electroacupuncture treatment at Chiang-Feng and Chou-Shu (acupuncture control group; group 2). Four received 50 microg

Hormonal and metabolic mechanisms in the development of cerebral pseudoatrophy in eating disorders.

Cranial computed tomography (CT) examinations performed on patients with schizophrenia, affective disorders or on patients with anorexia and bulimia nervosa revealed morphological brain alterations. In patients with eating disorders these structural changes were characterized by enlarged ventricles and sulci. Malnourishment-induced hormonal and metabolic disturbances may be responsible for this morphological brain alteration which, due to its reversibility after clinical remission, is frequently called 'pseudoatrophy'. As patients with alcohol dependency also display a cerebral pseudoatrophy,

[Study of the profiles of prolactin and other hormones in both maternal plasma and amniotic fluid during labor].

The effects of a dopamine antagonist and stress of labor on prolactin (PRL) concentrations were studied in maternal plasma and amniotic fluid during term delivery. In procedure 1, maternal blood and amniotic fluid samples were obtained from 5 normal full-term deliveries at 20-minute intervals for 180 minutes after the intravenous bolus of 10 mg metoclopramide (MCP) during labor just before delivery. Amniotic fluid samples were drawn through a transcervical intrauterine pressure catheter to avoid contamination with maternal blood. PRL, 17 beta-estradiol (E2), progesterone (P) and cortisol were

Midwinter insomnia in the subarctic region: evening levels of serum melatonin and cortisol before and after treatment with bright artificial light.

"Midwinter insomnia" (MI), mainly characterized by difficulties in falling asleep at night, is a common complaint during the period of obscuration or "dark period" north of the arctic circle. We hypothesize that MI is a result of a phase delay of the sleep-wake cycle due to insufficient exposure to daylight. In the present study based on this hypothesis, we wanted to find out whether otherwise healthy subjects with MI show abnormalities in the endocrine markers melatonin and cortisol late in the evening, and whether exposure to intensive light for one half hour in the morning for 5 days has an
