Image-guided multiscale modeling of trabecular bone: lacuna-resolved stress amplification and interaction effects.

Osteocyte lacunae are ubiquitous microstructural cavities in bone that perturb local stress and strain fields, shaping the mechanical microenvironment relevant to mechanotransduction and microdamage initiation. Yet lacunar-scale mechanical fields are not directly measurable in situ, and existing numerical studies are often limited by idealized geometries, incomplete verification, generic material properties, and a lack of specimen-matched experimental validation. Here, we present an image-guided multiscale framework integrating synchrotron radiation µCT in situ compression, convolutional neura

High-resolution genomics uncovers region-specific evolution and virulence of ocular Streptococcus pneumoniae.

UNLABELLED: Streptococcus pneumoniae is the leading cause of sight-threatening ocular infection. However, comprehensive molecular epidemiological analyses that focus on ocular pneumococci are limited. We performed a high-resolution whole-genome analysis (WGS) of ocular pneumococcal isolates from the Zhejiang province of China and a global data set (n = 58). Using latex agglutination and the Quellung reaction, nine serotypes were identified, with PCV13 covering 54.5% of the Chinese isolates, and no non-typeable (NT) strains were found in conjunctivitis. Illumina short-read data were assembled d

A quantum machine learning framework for predicting drug sensitivity in multiple myeloma using proteomic data.

In this paper, we introduce QProteoML, a new quantum machine learning (QML) framework for predicting drug sensitivity in Multiple Myeloma (MM) using high-dimensional proteomic data. MM, an extremely heterogeneous condition, displays often mixed responses to treatment, with a large number of patients showing drug resistance to proteasome inhibitors and immune modulatory agents. However, the methods previously used for genomic and proteomic data analysis techniques are plagued by issues of high dimensionality, imbalanced class distribution and feature redundancy, which work against the accurate

Plasma haem oxygenase-1 and interleukin-6 as adjunct host biomarkers associated with malaria.

INTRODUCTION: Malaria pathogenesis involves both parasite burden and host inflammatory and oxidative stress responses that contribute to haematological alterations. Full blood count (FBC)-derived indices have been explored as accessible surrogate biomarkers, reflecting systemic inflammatory changes; however, they provide limited mechanistic insight into underlying haem-driven oxidative stress pathways. Haem oxygenase-1 (HO-1) reflects cellular responses to free haem, while interleukin-6 (IL-6) mediates systemic inflammation. However, their combined role in relation to parasitaemia and haematol

Application of nanozyme-based drug delivery systems in the treatment of ROS-related ophthalmic diseases.

ROS-related ophthalmic diseases span the ocular surface as well as the anterior and posterior segments of the eye. Dry eye disease (DED), glaucoma, diabetic retinopathy (DR) and age-related macular degeneration (AMD) have distinct anatomical targets but share common pathophysiological features, including oxidative stress, inflammation, vascular dysfunction, epithelial injury or neurodegeneration. Conventional ocular drug delivery and therapeutic modalities are hindered by severe limitations, including low bioavailability in the posterior segment of the eye and the requirement for frequent inva

Genome-Wide Identification and Expression Profiling of the GATA Gene Family Reveals Candidate Genes Associated with GA3-Induced Shoot Elongation in Citrus grandis 'Wanbai'.

Plant GATA transcription factors are zinc-finger proteins involved in signal transduction, growth and development. However, their potential involvement in plant height and gibberellin-associated shoot growth in pummelo insufficiently characterised. In this study, members of the CgGATA gene family were identified genome-wide and characterised through analyses of physicochemical properties, chromosomal distribution, phylogenetic relationships, gene structures, conserved motifs, collinearity and predicted cis-acting elements in putative upstream regulatory regions. A muscle-enriched mitochondrial protein regulates postnatal heart growth through mitonuclear ubiquinone-PPAR signaling.

Increased mitochondrial biogenesis and aerobic respiration in postnatal mammalian organs are essential for adaptation to extrauterine life, yet the functions of many tissue-enriched, developmentally regulated mitochondrial proteins remain poorly defined. By integrating mitochondrial proteome annotations with mouse and human transcriptomic datasets, we identified Coenzyme Q10a (Coq10a), a striated muscle-enriched mitochondrial protein, as a regulator of postnatal cardiac growth. Coq10a, the mammalian homolog of the yeast ubiquinone-binding protein Coq10p, is highly induced in the heart after bi

Toward regionally equitable precision medicine: microbiota‑associated mechanisms and emerging personalized strategies in multiple sclerosis.

Multiple sclerosis (MS) is a chronic immune-mediated disorder of the central nervous system in which dysregulated immune signalling and barrier dysfunction play central pathogenic roles. Increasing evidence implicates the gut microbiota as a key modulator of host immune homeostasis; however, current understanding is largely derived from Western cohorts and remains fragmented at the mechanistic level. This review synthesizes global evidence alongside emerging data from underrepresented regions, including Egypt, Iran, and India, to delineate molecular pathways linking MS-associated dysbiosis to

Metformin Enhances the Pro-Regenerative Function of Human Adipose-Derived Stromal Cells for Diabetic Wound Healing.

Chronic wounds associated with diabetes remain a major challenge due to impaired angiogenesis, reduced fibroblast activity and dysfunction of regenerative cells. Adipose-derived mesenchymal stromal cells (ASCs) represent a promising therapeutic option; however, their regenerative potential is limited under hyperglycaemic conditions. This study investigated whether metformin could enhance ASC function and improve their therapeutic potential for tissue regeneration. Transcriptomic analysis of wound tissue identified pathways associated with angiogenesis, inflammation, extracellular matrix remode

The emerging landscape of lncRNAs in gastric cancer immunity: From immune escape to ferroptosis and metabolic reprogramming.

Gastric cancer (GC) remains a major global health challenge, particularly in East Asia, and the clinical impact of immune checkpoint inhibitors is limited by prevalent immune evasion and heterogeneous tumor immune microenvironments (TIME). Long noncoding RNAs (lncRNAs) have rapidly emerged as critical regulators of these processes, functioning far beyond their originally presumed nonfunctional roles. Through intricate interactions with chromatin regulators, RNA-binding proteins, microRNAs, metabolic enzymes, and RNA modification machinery, lncRNAs orchestrate diverse pathways that collectively

Mechanisms and therapeutic prospects of DNA methylation-mucosal innate immunity crosstalk in inflammatory bowel disease.

Inflammatory bowel disease (IBD) comprises a group of chronic and relapsing intestinal inflammatory disorders whose pathogenesis and progression are closely associated with disruption of the intestinal mucosal barrier, dysregulated immune responses, and altered epigenetic regulation. The innate immune system is a central component of mucosal host defense and plays a pivotal role in pathogen recognition, inflammatory signal transduction, immune-cell functional regulation, and maintenance of barrier homeostasis. In recent years, DNA methylation has been increasingly recognized as an important me

Multimodal approach to identify neuropsychophysiological subgroups in myalgic encephalomyelitis/chronic fatigue syndrome and their relevance for rehabilitation: protocol for a mechanistic cross-sectional and longitudinal study.

INTRODUCTION: Myalgic Encephalomyelitis (ME)/Chronic Fatigue Syndrome (CFS) is a debilitating condition characterized by severe fatigue and post-exertional malaise (PEM). Reported neuropsychophysiological abnormalities suggest ME/CFS is multifactorial, but current knowledge remains fragmented. This study protocol outlines a multimodal investigation designed to (1) compare neuropsychophysiological mechanisms between ME/CFS patients and healthy participants, (2) test an integrative model of ME/CFS, (3) identify neuropsychophysiological subgroups within the patient population, and (4) identify pr

The Intersection of Immunosenescence, Cellular Senescence, and Type 1 Diabetes: Paving the Way for Future Interventions.

UNLABELLED: Immunosenescence, characterized by functional decline and altered phenotypes of immune cells with age, can disturb immune tolerance and promote autoreactive responses. Type 1 diabetes (T1D) has traditionally been viewed as a disease of immune dysregulation that leads to the autoimmune destruction of pancreatic β-cells. Emerging evidence suggests that senescence contributes to both the onset and progression of T1D. Autoreactive T cells are associated not only with β-cell death but also with the induction of β-cell senescence. In parallel, senescent β-cells acquire a senescence-assoc

The lactate-lactylation axis in tumor radioresistance: metabolic, epigenetic, and immune mechanisms with emerging links to RNA regulation.

Radiotherapy remains a cornerstone treatment for solid tumors, but its efficacy is frequently limited by intrinsic and acquired radioresistance. Increasing evidence indicates that lactate metabolism and protein lactylation are not merely by-products of glycolysis, but context-dependent regulators of tumor adaptation to irradiation. In irradiated tumor models, lactate-associated metabolic rewiring has been linked to DNA damage repair, redox buffering, and clonogenic survival. Other mechanisms, including chromatin remodeling, immune suppression, and RNA-level regulation, should be interpreted ac

The red blood cell proteome and interactome identify a Band 3-BLVRB axis regulating hypoxic metabolic adaptation.

Red blood cells (RBCs) are transcriptionally silent yet dynamically remodel metabolism in response to oxygen tension. Using ultrapure human RBCs, we generated the deepest contamination-free proteome to date (3775 proteins) and mapped the oxygen-dependent interactome. These data sets revealed an oxygen-responsive metabolon centered on the Band 3 (SLC4A1) N terminus. We identified biliverdin reductase B (BLVRB) as a previously unrecognized Band 3 interactor that dissociates under hypoxia, coincident with increased Band 3-deoxyhemoglobin contacts. This reversible assembly functions as an oxygen-s

Amiodarone-mediated thyroid hormone disruption induces mitochondrial apoptosis and G1 arrest during chick embryogenesis.

Thyroid hormones (THs) coordinate proliferation, apoptosis, and tissue remodeling during vertebrate development, yet how TH signaling constrains embryonic cell death in avian embryos remains unclear. Here, we used amiodarone, a pharmacological disruptor of TH signaling with reported effects on TH receptors and deiodinase-mediated hormone metabolism, to perturb TH signaling at the onset of incubation in chick embryos and integrate systems-level and functional readouts. In silico docking predicted favorable binding of amiodarone to all three deiodinase isoforms (DIO1-3), while whole-embryo assay

Monothiol glutaredoxins in microbial iron metabolism: From iron-sulfur cluster trafficking to iron-dependent regulation.

Iron-sulfur (Fe-S) clusters are essential cofactors that support a wide range of metabolic and regulatory processes across all domains of life. The assembly and distribution of these chemically labile cofactors require tightly coordinated biosynthetic and trafficking systems that respond dynamically to cellular iron availability and redox conditions. Monothiol CGFS-type glutaredoxins (Grxs) have emerged as central components of these networks. Once considered primarily thiol-disulfide oxidoreductases, these proteins are now recognized as versatile Fe-S cluster carriers that coordinate and exch

Hypoxia induces microtubule rearrangement in intestinal epithelial cells.

Microtubules are integral components of the highly regulated and dynamic cytoskeleton, which is vital for cell function and the maintenance of homeostasis. Microtubule disruption is associated with multiple disease states including colorectal cancer and neurodegenerative disorders. Although much is known about the mechanisms by which microtubules are regulated under physiologic conditions, the effect of pathological stimuli and how this contributes to disease progression is less clear. Hypoxia is a prominent microenvironmental feature of a range of pathological states including inflammation, i
